From eDNA to citizen science
Biological invasions are a form of global change threatening biodiversity, ecosystem stability, and human health, and cost government agencies billions of dollars in remediation and eradication programs. Attempts to eradicate introduced species are most successful when detection of newly established populations occurs early in the invasion process. We review existing and emerging tools – specifically environmental DNA (eDNA), chemical approaches, remote sensing, citizen science, and agency-based monitoring – for surveillance and monitoring of invasive species. For each tool, we consider the benefits provided, examine challenges and limitations, discuss data sharing and integration, and suggest best practice implementations for the early detection of invasive species. Programs that promote public participation in large-scale biodiversity identification and monitoring (such as iNaturalist and eBird) may be the best resources for early detection. However, data from these platforms must be monitored and used by agencies that can mount appropriate response efforts. Control efforts are more likely to succeed when they are focused on early detection and prevention, thereby saving considerable time and resources.
Who Benefits When Prescription Drug Manufacturers Offer Copay Coupons?
The rising cost of prescription drugs is a concern in the United States. To manage drug costs, insurance companies induce patients to choose less-expensive medications by making them pay higher copayments for more-expensive drugs, especially when multiple drug options are available to treat a condition. However, drug manufacturers have responded by offering copay coupons—coupons intended to be used by those already with prescription drug coverage. Recent empirical work has shown that such coupons significantly increase insurer costs without much benefit to patients, who incur lower out-of-pocket expenses with coupons but may eventually see higher costs passed to them. As a result, there is pressure from the insurance industry and consumer advocacy groups to ban copay coupons. In this paper we analyze how copay coupons affect patients, insurance companies, and drug manufacturers, while addressing the question of whether insurance companies would in fact always benefit from a copay coupon ban. We find that copay coupons tend to benefit drug manufacturers with large profit margins relative to other manufacturers, while generally, but not always, benefiting patients; insurer costs tend to increase with coupons from high-price drug manufacturers and decrease with coupons from low-price manufacturers. Although often helping drug manufacturers and increasing insurer costs, we also identify situations in which copay coupons benefit both patients and insurers. Thus, a blanket ban on copay coupons would not necessarily benefit insurance companies. In addition to the policy implications of our work, we make concrete managerial recommendations to insurers. We discuss how they should set formulary selection policies taking into account the fact that drug manufacturers may offer coupons; and we suggest how they can benefit from subsidizing coupons from drug manufacturers with low-price drugs, or from having drug manufacturers compete on price, to receive a favorable formulary position (i.e., copay). Tree growth response along an elevational gradient: climate or genetics?
Environment and genetics combine to influence tree growth and should therefore be jointly considered when evaluating forest responses in a warming climate. Here, we combine dendroclimatology and population genetic approaches with the aim of attributing climatic influences on growth of European larch (Larix decidua) and Norway spruce (Picea abies). Increment cores and genomic DNA samples were collected from populations along a ~900-m elevational transect where the air temperature gradient encompasses a ~4 °C temperature difference. We found that low genetic differentiation among populations indicates gene flow is high, suggesting that migration rate is high enough to counteract the selective pressures of local environmental variation. We observed lower growth rates towards higher elevations and a transition from negative to positive correlations with growing season temperature upward along the elevational transect. With increasing elevation there was also a clear increase in the explained variance of growth due to summer temperatures. Comparisons between climate sensitivity patterns observed along this elevational transect with those from Larix and Picea sites distributed across the Alps reveal good agreement, and suggest that tree-ring width (TRW) variations are more climate-driven than genetics-driven at regional and larger scales. We conclude that elevational transects are an extremely valuable platform for understanding climatic-driven changes over time and can be especially powerful when working within an assessed genetic framework.
Correlating Extremes in Wind Divergence with Extremes in Rain over the Tropical Atlantic
Air–sea fluxes are greatly enhanced by the winds and vertical exchanges generated by mesoscale convective systems (MCSs). In contrast to global numerical weather prediction models, space-borne scatterometers are able to resolve the small-scale wind variability in and near MCSs at the ocean surface. Downbursts of heavy rain in MCSs produce strong gusts and large divergence and vorticity in surface winds. In this paper, 12.5 km wind fields from the ASCAT-A and ASCAT-B tandem mission, collocated with short time series of Meteosat Second Generation 3 km rain fields, are used to quantify correlations between wind divergence and rain in the Inter-Tropical Convergence Zone (ITCZ) of the Atlantic Ocean. We show that when there is extreme rain, there is extreme convergence/divergence in the vicinity. Probability distributions for wind divergence and rain rates were found to be heavy-tailed: exponential tails for wind divergence (P∼e−αδ with slopes that flatten with increasing rain rate), and power-law tails for rain rates (P∼(R*)−β with a slower and approximately equal decay for the extremes of convergence and divergence). Co-occurring points are tabulated in two-by-two contingency tables from which cross-correlations are calculated in terms of the odds and odds ratio for each time lag in the collocation. The odds ratio for extreme convergence and extreme divergence both have a well-defined peak. The divergence time lag is close to zero, while it is 30 min for the convergence peak, implying that extreme rain generally appears after (lags) extreme convergence. The temporal scale of moist convection is thus determined by the slower updraft process, as expected. A structural analysis was carried out that demonstrates consistency with the known structure of MCSs. This work demonstrates that (tandem) ASCAT winds are well suited for air–sea exchange studies in moist convection.
Impacts of Zika emergence in Latin America on endemic dengue transmission
In 2015 and 2016, Zika virus (ZIKV) swept through dengue virus (DENV) endemic areas of Latin America. These viruses are of the same family, share a vector and may interact competitively or synergistically through human immune responses. We examine dengue incidence from Brazil and Colombia before, during, and after the Zika epidemic. We find evidence that dengue incidence was atypically low in 2017 in both countries. We investigate whether subnational Zika incidence is associated with changes in dengue incidence and find mixed results. Using simulations with multiple assumptions of interactions between DENV and ZIKV, we find cross-protection suppresses incidence of dengue following Zika outbreaks and low periods of dengue incidence are followed by resurgence. Our simulations suggest correlations in DENV and ZIKV reproduction numbers could complicate associations between ZIKV incidence and post-ZIKV DENV incidence and that periods of low dengue incidence are followed by large increases in dengue incidence. Dengue and Zika virus are related flaviviruses, and introduction of Zika in the Americas may have impacted dengue epidemiology. Here, Borchering et al. show that dengue incidence was unusually low in 2017 in Brazil and Colombia, and simulations incorporating immune-mediated interactions predict reductions in dengue following Zika outbreaks with subsequent rebounds.
Implementation of a preoperative exercise programme in lung cancer resection: protocol for a mixed-methods study
IntroductionPreoperative exercise training is recommended, when feasible, for people undergoing resection for lung cancer and has been shown to reduce the risk of postoperative pulmonary complications and improve preoperative exercise capacity. However, preoperative exercise training programmes are not commonly available in the Australian clinical practice setting due to a range of factors including resource and time restrictions. We aim to describe the protocol to evaluate the implementation of an existing preoperative exercise training programme in people undergoing lung cancer resection in an Australian setting.Methods and analysisThis is an evaluation of a secondary objective of a study examining the effect of lung cancer resection on exercise capacity, lung function and symptoms of dyspnoea and quality of life. Participants will be prospectively recruited at the time of lung cancer diagnosis and planned surgical treatment through the lung cancer multidisciplinary team of a metropolitan hospital in Sydney, Australia. All participants will be offered the choice of participating in the preoperative exercise training programme which encompasses a hybrid gym and telerehabilitation programme of up to five sessions/week from baseline until surgical date. The programme will be evaluated using the Reach, Effectiveness, Adoption, Implementation, Maintenance Framework including both quantitative and qualitative measures which will be analysed using descriptive statistics and qualitative analysis coded inductively.Ethics and disseminationThe study has received ethical approval through the Northern Sydney Local Health District reference 2023/ETH01643 and has been registered prospectively. Findings will be disseminated through peer-reviewed publication and scientific conference presentation.Trial registration numberACTRN12624000359538.
Establishment of Early Endpoints in Mouse Total-Body Irradiation Model
Acute radiation sickness (ARS) following exposure to ionizing irradiation is characterized by radiation-induced multiorgan dysfunction/failure that refers to progressive dysfunction of two or more organ systems, the etiological agent being radiation damage to cells and tissues over time. Radiation sensitivity data on humans and animals has made it possible to describe the signs associated with ARS. A mouse model of total-body irradiation (TBI) has previously been developed that represents the likely scenario of exposure in the human population. Herein, we present the Mouse Intervention Scoring System (MISS) developed at the Veterinary Sciences Department (VSD) of the Armed Forces Radiobiology Research Institute (AFRRI) to identify moribund mice and decrease the numbers of mice found dead, which is therefore a more humane refinement to death as the endpoint. Survival rates were compared to changes in body weights and temperatures in the mouse (CD2F1 male) TBI model (6-14 Gy, 60Co γ-rays at 0.6 Gy min-1), which informed improvements to the Scoring System. Individual tracking of animals via implanted microchips allowed for assessment of criteria based on individuals rather than by group averages. From a total of 132 mice (92 irradiated), 51 mice were euthanized versus only four mice that were found dead (7% of non-survivors). In this case, all four mice were found dead after overnight periods between observations. Weight loss alone was indicative of imminent succumbing to radiation injury, however mice did not always become moribund within 24 hours while having weight loss >30%. Only one survivor had a weight loss of greater than 30%. Temperature significantly dropped only 2-4 days before death/euthanasia in 10 and 14 Gy animals. The score system demonstrates a significant refinement as compared to using subjective assessment of morbidity or death as the endpoint for these survival studies.
Influenza vaccination in the elderly boosts antibodies against conserved viral proteins and egg-produced glycans
Seasonal influenza vaccination elicits a diminished adaptive immune response in the elderly, and the mechanisms of immunosenescence are not fully understood. Using Ig-Seq, we found a marked increase with age in the prevalence of cross-reactive (CR) serum antibodies that recognize both the H1N1 (vaccine-H1) and H3N2 (vaccine-H3) components of an egg-produced split influenza vaccine. CR antibodies accounted for 73% ± 18% of the serum vaccine responses in a cohort of elderly donors, 65% ± 15% in late middle-aged donors, and only 13% ± 5% in persons under 35 years of age. The antibody response to non-HA antigens was boosted by vaccination. Recombinant expression of 19 vaccine-H1+H3 CR serum monoclonal antibodies (s-mAbs) revealed that they predominantly bound to non-HA influenza proteins. A sizable fraction of vaccine-H1+H3 CR s-mAbs recognized with high affinity the sulfated glycans, in particular sulfated type 2 N-acetyllactosamine (Galβ1-4GalNAcβ), which is found on egg-produced proteins and thus unlikely to contribute to protection against influenza infection in humans. Antibodies against sulfated glycans in egg-produced vaccine had been identified in animals but were not previously characterized in humans. Collectively, our results provide a quantitative basis for how repeated exposure to split influenza vaccine correlates with unintended focusing of serum antibody responses to non-HA antigens that may result in suboptimal immunity against influenza.
