يتناول كتاب (مبادئ الحكم في الصين القديمة) والذي قامه بتأليفه (وي تشينغ) في حوالي (211) صفحة من القطع المتوسط موضوع (تاريخ الصين قديم) مستعرضا المحتويات التالية : قبل أكثر من ألف وربعمائة عام، أصدر الإمبراطور تاي تسونغ إمبراطور أسرة تانغ الملكية (618-907) مرسوما بأن يجمع أربعة من أشهر علماء الصين وسياسييها وأمهرهم وقتها المعارف التاريخية حول مبادئ الحكم الإمبراطوري من الكتب الكلاسيكية القديمة الستة، والمجموعات التاريخية الأربع، والمئات من مؤلفات المدارس الفكرية الصينية، وأن يرتبوها ويستخرجوا منها الدروس الاكثر اهمية حول تهذيب النفس وإصلاحها، وإدارة العائلة، والحكم الجيد للبلاد، وسبل جلب السلام للعالم. وكانت النتيجة مجموعة عنوانها \"تشيونشو تشيياو\" التي أقتبست بعناية من أربعة عشر ألف كتاب، وتسع وثمانين ألف مخطوطة من الكتابات القديمة، بإجمالي خمسمائة ألف فقرة مكتوبة، تغطي خمسة وشتين صنفا من الكتب. وقد ساعد هذا الكتاب الإمبراطور تاي سونغ والكثير من الأباطرة من بعده في أمور الحكم، ويعود الفضل في تحقيق الرخاء والسلام في المراحل اللاحقة بالصين القديمة بنسبة كبيرة إلى هذا الكتاب.
A kinetically controlled platform for ligand-oligonucleotide transduction

Ligand-oligonucleotide transduction provides the critical pathway to integrate non-nucleic acid molecules into nucleic acid circuits and nanomachines for a variety of strand-displacement related applications. Herein, a general platform is constructed to convert the signals of ligands into desired oligonucleotides through a precise kinetic control. In this design, the ligand-aptamer binding sequence with an engineered duplex stem is introduced between the toehold and displacement domains of the invading strand to regulate the strand-displacement reaction. Employing this platform, we achieve efficient transduction of both small molecules and proteins orthogonally, and more importantly, establish logical and cascading operations between different ligands for versatile transduction. Besides, this platform is capable of being directly coupled with the signal amplification systems to further enhance the transduction performance. This kinetically controlled platform presents unique features with designing simplicity and flexibility, expandable complexity and system compatibility, which may pave a broad road towards nucleic acid-based developments of sophisticated transduction networks.
Ligand-oligonucleotide interactions can integrate both small molecules and proteins into nucleic acid-based circuits. Here the authors design ligand-aptamer complexes to control strand-displacement reactions for versatile ligand transduction.

The relationship between expression of PD-L1 and HIF-1α in glioma cells under hypoxia

Hypoxia inducible factor-1α (HIF-1α) up-regulates the expression of programmed death ligand-1 (PD-L1) in some extracranial malignancies. However, whether it could increase PD-L1 expression in intracranial tumor is still unknown. Here, we explored the relationship between HIF-1α and PD-L1 expression in glioma, and investigated their clinical significance. In glioma patients, HIF-1α and PD-L1 were overexpressed in high grade glioma tissues and were significantly associated with poor survival. In glioma cells, PD-L1 expression was induced under hypoxia condition, and the enhanced PD-L1 expression was abrogated by either HIF-1α knock-down or HIF-1α inhibitor treatment. Furthermore, ChIP-qPCR analysis showed the direct binding of HIF-1α to PD-L1 proximal promoter region, providing evidence that HIF-1α up-regulates PD-L1 in glioma. In glioma murine model, the combination treatment with HIF-1α inhibitor and anti-PD-L1 antibody caused a more pronounced suppressive effect on tumor growth compared to either monotherapy. Immunologically, the combination treatment improved both dendritic cell (DC) and CD8
+
T cell activation. Overall, our results demonstrated that positive correlation between PD-L1 and HIF-1α in glioma, and provide an alternative strategy, inhibiting HIF-1α, as combination therapies with immunotherapies to advance glioma treatment.

Different stimuli can polarize macrophages into two basic types, M1 and M2. Tumor-associated macrophages (TAMs) in the tumor microenvironment (TME) are composed of heterogeneous subpopulations, which include the M1 anti-tumor and M2 pro-tumor phenotypes. TAMs predominantly play a M2-like tumor-promoting role in the TME and regulate various malignant effects, such as angiogenesis, immune suppression, and tumor metastasis; hence, TAMs have emerged as a hot topic of research in cancer therapy. This review focuses on three main aspects of TAMs. First, we summarize macrophage polarization along with the effects on the TME. Second, recent advances and challenges in cancer treatment and the role of M2-like TAMs in immune checkpoint blockade and CAR-T cell therapy are emphasized. Finally, factors, such as signaling pathways, associated with TAM polarization and potential strategies for targeting TAM repolarization to the M1 pro-inflammatory phenotype for cancer therapy are discussed.

Toward Hydrogen‐Free and Dendrite‐Free Aqueous Zinc Batteries: Formation of Zincophilic Protective Layer on Zn Anodes

Rechargeable aqueous Zn‐ion batteries (ZIBs) are regarded as one of the most promising devices for the next‐generation energy storage system. However, the uncontrolled dendrite growth on Zn metal anodes and the side hydrogen evolution reaction, which has not yet been well considered, hinder the practical application of these batteries. Herein, a uniform and robust metallic Sb protective layer is designed based on the theoretic calculation and decorated on Zn plate via in situ replacement reaction. Compared with the bare Zn plate, the as‐prepared Zn@Sb electrode provides abundant zincophilic sites for Zn nucleation, and homogenizes the electric field around the Zn anode surface, both of which promote the uniform Zn deposition to achieve a dendrite‐free morphology. Moreover, the Gibbs free energy (∆GH) calculation and in situ characterization demonstrate that hydrogen evolution reaction can be effectively suppressed by the Sb layer. Consequently, Sb‐modified Zn anodes exhibit an ultralow voltage hysteresis of 34 mV and achieve excellent cycling stability over 1000 h with hydrogen‐ and dendrite‐free behaviors. This work provides a facile and effective strategy to suppress both hydrogen evolution reaction and dendrite growth. A uniform and robust metallic Sb protective layer is decorated on Zn plate via in situ replacement reaction. With rich zincophilic sites for Zn nucleation, improved electrolyte wettability and homogenized electric field, the Sb layer promotes the uniform Zn deposition with a dendrite‐free morphology. Moreover, the Gibbs free energy calculation and in situ characterization demonstrate that hydrogen evolution reaction can be effectively suppressed by the Sb layer.

An ORFeome of rice E3 ubiquitin ligases for global analysis of the ubiquitination interactome

Background
Ubiquitination is essential for many cellular processes in eukaryotes, including 26S proteasome-dependent protein degradation, cell cycle progression, transcriptional regulation, and signal transduction. Although numerous ubiquitinated proteins have been empirically identified, their cognate ubiquitin E3 ligases remain largely unknown.
Results
Here, we generate a complete ubiquitin E3 ligase-encoding open reading frames (UbE3-ORFeome) library containing 98.94% of the 1515 E3 ligase genes in the rice (
Oryza sativa
L
.
) genome. In the test screens with four known ubiquitinated proteins, we identify both known and new E3s. The interaction and degradation between several E3s and their substrates are confirmed in vitro and in vivo. In addition, we identify the F-box E3 ligase OsFBK16 as a hub-interacting protein of the phenylalanine ammonia lyase family OsPAL1–OsPAL7. We demonstrate that OsFBK16 promotes the degradation of OsPAL1, OsPAL5, and OsPAL6. Remarkably, we find that overexpression of
OsPAL1
or
OsPAL6
as well as loss-of-function of
OsFBK16
in rice displayed enhanced blast resistance, indicating that OsFBK16 degrades OsPALs to negatively regulate rice immunity.
Conclusions
The rice UbE3-ORFeome is the first complete E3 ligase library in plants and represents a powerful proteomic resource for rapid identification of the cognate E3 ligases of ubiquitinated proteins and establishment of functional E3–substrate interactome in plants.
