High-fat diet promotes renal injury by inducing oxidative stress and mitochondrial dysfunction
Obesity has been recognized as a major risk factor for chronic kidney disease, but the underlying mechanism remains elusive. Here, we investigated the mechanism whereby long-term high-fat diet (HFD) feeding induces renal injury in mice. The C57BL/6 mice fed HFD for 16 weeks developed obesity, diabetes, and kidney dysfunction manifested by albuminuria and blood accumulation of BUN and creatinine. The HFD-fed kidney showed marked glomerular and tubular injuries, including prominent defects in the glomerular filtration barrier and increased tubular cell apoptosis. Mechanistically, HFD feeding markedly increased triglyceride and cholesterol contents in the kidney and activated lipogenic pathways for cholesterol and triglyceride synthesis. HFD feeding also increased oxidative stress and induced mitochondrial fission in tubular cells, thereby activating the pro-apoptotic pathway. In HK-2 and mesangial cell cultures, high glucose, fatty acid, and TNF-α combination was able to activate the lipogenic pathways, increase oxidative stress, promote mitochondrial fission, and activate the pro-apoptotic pathway, all of which could be attenuated by an inhibitor that depleted reactive oxygen species. Taken together, these observations suggest that long-term HFD feeding causes kidney injury at least in part as a result of tissue lipid accumulation, increased oxidative stress, and mitochondrial dysfunction, which promote excess programmed cell death.

The role of thioredoxin system in cancer: strategy for cancer therapy

PurposeCancer, a major public health problem, exhibits significant redox alteration. Thioredoxin (Trx) system, including Trx and Trx reductase (TrxR), as well as Trx-interacting protein (TXNIP) play important roles in controlling the cellular redox balance in cancer cells. In most cancers, Trx and TrxR are usually overexpressed and TXNIP is underexpressed. In recent years, some agents targeting Trx, TrxR, and TXNIP were used to explore a therapy approach for cancer patients.MethodsA systematic search of PMC and the PubMed Database was conducted to summarize the potential of Trx system inhibitors for cancer treatment.ResultsIn this article, we first summarize the functions of Trx, TrxR, and TXNIP in cancers. We also review some small molecule inhibitors of Trx/TrxR and d-allose (TXNIP inducer) and discuss their antitumor mechanisms. We highlight the combined inhibition of Trx system and GSH system in cancer therapy. We expect that a highly specific and selective antitumor agent with no cytotoxicity on human normal cells could be developed in the future.ConclusionIn conclusion, Trx system may be very promising for clinical therapy of cancer in the future.

One-point energy correlator inside jets

In this work, we introduce a new jet shape observable, the one-point energy correlator (EC), designed to characterize the intra-jet energy flow distribution by measuring energy deposition at a specific angle relative to the jet axis. Building upon the transverse momentum dependent (TMD) physics, we aim for the EC to provide novel insights into jet substructure and offer a new approach to study TMD physics, particularly gluon transverse momentum dependent fragmentation functions (TMDFFs) which are notoriously difficult to extract. We obtain the factorization of the EC jet function within Soft-Collinear Effective Theory and leverage the framework of semi-inclusive TMD fragmenting jet functions. We resum large global logarithms and non-global logarithms (NGLs) and show that the normalized EC jet function exhibits significantly reduced dependence on the factorization scale and is primarily sensitive to the jet scale. Finally, after incorporating non-perturbative effects, we present numerical calculations up to NNLL accuracy for global logarithms and LL accuracy for NGLs, and we compare these predictions with PYTHIA 8 simulations.

As people age, iron deposits in different areas of the brain may impair normal cognitive function and behavior. Abnormal iron metabolism generates hydroxyl radicals through the Fenton reaction, triggers oxidative stress reactions, damages cell lipids, protein and DNA structure and function, and ultimately leads to cell death. There is an imbalance in iron homeostasis in Alzheimer's disease (AD). Excessive iron contributes to the deposition of β-amyloid and the formation of neurofibrillary tangles, which in turn, promotes the development of AD. Therefore, iron-targeted therapeutic strategies have become a new direction. Iron chelators, such as desferoxamine, deferiprone, deferasirox, and clioquinol, have received a great deal of attention and have obtained good results in scientific experiments and some clinical trials. Given the limitations and side effects of the long-term application of traditional iron chelators, alpha-lipoic acid and lactoferrin, as self-synthesized naturally small molecules, have shown very intriguing biological activities in blocking Aβ-aggregation, tauopathy and neuronal damage. Despite a lack of evidence for any clinical benefits, the conjecture that therapeutic chelation, with a special focus on iron ions, is a valuable approach for treating AD remains widespread.

New media art design based on fast visual segmentation and 3D image processing

Acquiring innovative styles and compositions from intricate and heterogeneous artistic imagery has emerged as a pivotal research quandry within contemporary new media art image conception. In a concerted effort to adeptly distill the quintessence of artistic styles and elements embedded within these visuals, an innovative methodology is posited herein, underpinned by an enhanced U-net segmentation framework and harmoniously fused with the surface extraction image reconstruction algorithm. This meticulous amalgamation endeavors to attain accurate segmentation and tridimensional reconstruction of the artistry encapsulated in these images. Primarily, the imagery is meticulously partitioned, culminating in an output that artfully encapsulates the inherent artistic attributes. Subsequently, this segmentation outcome is adeptly reconstituted, bestowing form to a three-dimensional artistry model. Empirical validation substantiates the efficacy of this approach, with the method’s Mean Intersection over the Union (MIoU) parameter yielding an impressive score of 0.939 in segmentation performance. Moreover, the peak signal-to-noise ratio and structural similarity attain commendable zeniths of 38.16 and 0.9808, respectively, underscoring the excellence of the reconstruction process. The proposed methodology demonstrates its prowess in exacting segmentation and comprehensive reconstruction of semantic intricacies and nuanced features pervading the realm of artistic imagery. Consequently, this novel methodology augments artists’ capacity to discern diverse artistic paradigms and fabricate superlative new media art compositions of heightened caliber.

The cellular hallmarks of Parkinson’s disease (PD) are the loss of nigral dopaminergic neurons and the formation of α-synuclein-enriched Lewy bodies and Lewy neurites in the remaining neurons. Based on the topographic distribution of Lewy bodies established after autopsy of brains from PD patients, Braak and coworkers hypothesized that Lewy pathology primes in the enteric nervous system and spreads to the brain, suggesting an active retrograde transport of α-synuclein (the key protein component in Lewy bodies), via the vagal nerve. This hypothesis, however, has not been tested experimentally thus far. Here, we use a human PD brain lysate containing different forms of α-synuclein (monomeric, oligomeric and fibrillar), and recombinant α-synuclein in an in vivo animal model to test this hypothesis. We demonstrate that α-synuclein present in the human PD brain lysate and distinct recombinant α-synuclein forms are transported via the vagal nerve and reach the dorsal motor nucleus of the vagus in the brainstem in a time-dependent manner after injection into the intestinal wall. Using live cell imaging in a differentiated neuroblastoma cell line, we determine that both slow and fast components of axonal transport are involved in the transport of aggregated α-synuclein. In conclusion, we here provide the first experimental evidence that different α-synuclein forms can propagate from the gut to the brain, and that microtubule-associated transport is involved in the translocation of aggregated α-synuclein in neurons.
