Exploring Bioactive Compounds in Brown Seaweeds Using Subcritical Water: A Comprehensive Analysis
In this study, we characterized the bioactive properties of three important brown seaweed species, Sargassum thunbergii, Undaria pinnatifida, and Saccharina japonica, by subcritical water extraction (SWE), as these species are well known for their beneficial health effects. Their physiochemical properties, including potential antioxidant, antihypertensive, and α-glucosidase inhibitory activity, and the antibacterial activity of the hydroysates were also analyzed. The highest total phlorotannin, total sugar content, and reducing sugar content in the S. thunbergii hydrolysates were 38.82 ± 0.17 mg PGE/g, 116.66 ± 0.19 mg glucose/g dry sample, and 53.27 ± 1.57 mg glucose/g dry sample, respectively. The highest ABTS+ and DPPH antioxidant activities were obtained in the S. japonica hydrolysates (124.77 ± 2.47 and 46.35 ± 0.01 mg Trolox equivalent/g, respectively) and the highest FRAP activity was obtained in the S. thunbergii hydrolysates (34.47 ± 0.49 mg Trolox equivalent/g seaweed). In addition, the seaweed extracts showed antihypertensive (≤59.77 ± 0.14%) and α-glucosidase inhibitory activity (≤68.05 ± 1.15%), as well as activity against foodborne pathogens. The present findings provide evidence of the biological activity of brown seaweed extracts for potential application in the food, pharmaceutical, and cosmetic sectors.
Melanoma and brown seaweed: an integrative hypothesis
Although relatively rare, melanoma accounts for 2 % of cancer diagnoses globally and accounts for about 1 % of all cancer deaths. Worldwide, the annual incidence of melanoma is 272,000 cases which vary hugely, ranging from Japan where it is incredibly infrequent, to Queensland, Australia, where it is nearly 100 times higher. Based on epidemiology and laboratory studies, there is compelling evidence suggesting that seaweed might be protective against different types of cancers such as breast cancer in seaweed consuming populations. By comparing countries where melanoma is more common with countries where it is infrequent, it is possible to construct a hypothesis for how consuming brown seaweeds which may hold clues to the differences in melanoma susceptibility between Japanese and Western nations. Unlike in these other countries, where melanoma incidence has increased dramatically over the last two decades, in Japan, rates have remained remarkably low and stable. There is limited evidence from clinical studies and animal models that have used whole seaweed or isolated fractions from seaweed and measured changes in biomarkers. They have demonstrated the effectiveness of seaweed at inhibiting melanoma development and progression. In this review, the various results will be described. Although there are several effective fractions, it is proposed that consuming whole seaweeds may hold additional benefits that could be lost by consuming only a single extract.
Antioxidant and Anti-Inflammatory Activities of Sargassum macrocarpum Extracts
Oxidative stress and the inflammatory response are known to be the most important pathological factors for aging skin cells. Therefore, substances that protect skin cells from oxidative stress and inflammatory reactions of the skin have potential as functional ingredients for skin care. In the present study, we investigated the potential of Sargassum macrocarpum as an anti-inflammatory candidate for inflammatory skin disease. Antioxidant and anti-inflammatory activities are desirable properties in such functional materials. The total polyphenol content as well as antioxidant and anti-inflammatory activities were evaluated in hot-water (HES) and ethanol (EES) extracts of S. macrocarpum. The polyphenol content was higher in the HES (HES: 115.9 ± 15.3 mg GA/g, EES: 3.9 ± 0.5 mg GA/g), and the HES also had ABTS (HES: IC50 1.0 ± 0.0 mg/mL, EES: IC50 16.09 ± 0.7 mg/mL) and DPPH (HES: IC50 6.50 ± 0.3 mg/mL, EES: IC50 35.3 ± 3.1 mg/mL) radical scavenging capacities as well as FRAP activity (HES: IC50 18.8 ± 0.4 mg/mL, EES: IC50 n.d.). Compared with the EES at the equivalent concentration range (1.25–20 μg/mL), the HES exerted a more potent inhibitory activity on LPS-stimulated nitric oxide (10.3–43.1%), IL-6 (15.7–45.0%), and TNF-α (14.1–20.8%) in RAW 264.7 macrophage cells in addition to TNF-α and IFN-γ-facilitated IL-6 (10.9–84.1%) and IL-8 (7.7–73.2%) in HaCaT keratinocytes. These results suggested that water-soluble materials might be deeply involved in the antioxidant and anti-inflammatory activity in S. macrocarpum. General composition analysis indicated that the HES contains more carbohydrates and polyphenols than the EES, and the monosaccharide composition analysis suggested that fucose-containing sulfated polysaccharide and β-glucan might be potent anti-inflammatory candidates in the HES. The present study presents important preliminary results and a valuable strategy for developing novel anti-skin dermatitis candidates using a hot-water extract of S. macrocarpum.
Concentrations of Heavy Metals in Three Brown Seaweed (Phaeophyta: Phaeophyceae) Collected from Tourism Area in Sanur Beach, Coast of Denpasar, Bali and Public Health Risk Assessment

This study aims to examine the concentration of heavy metals (Pb, Cd, Hg, and As) in brown algae (Sargassum aquifolium, Padina australis, and Turbinaria ornata.) and the possible exposure to health risks caused by consumption.  Heavy metal concentrations were determined using Atomic Absorption Spectroscopy (AAS) on brown seaweed samples obtained from three different sites. The average concentration of heavy metals in the dry weight of brown seaweed remains within the guidelines established by The Food and Drug Supervisory Agency (BPOM) Number 32 of 2019 concerning the Safety and Quality of Traditional Medicines, which is then used to calculate the estimated daily intake (EDI), target hazard quotient (THQ and TTHQ), and target cancer risk (TCR) for arsenic associated with food exposure to potentially toxic metallic elements. Each species of brown seaweed has a THQ and TTHQ level of <1, indicating that one or more toxic metal elements in the same meal provide no significant non-carcinogenic risk. The TCR for arsenic in these seaweeds are all less than 1 x 10-4, indicating no cancer risk. There are no chronic health hazards related with the ingestion of brown seaweed harvested from the coast of Sanur Beach at Denpasar, Bali.
A Brief Review on the Development of Alginate Extraction Process and Its Sustainability
Alginate is an attractive marine resource-based biopolymer, which has been widely used in pharmaceutical, food and textile industries. This paper reviewed the latest development of the conventional and alternative processes for alginate extraction from brown seaweed. To improve extraction yield and product quality, various studies have been carried out to optimize the operation condition. Based on literature survey, the most commonly used protocol is soaking milled seaweed in 2% (w/v) formaldehyde, overnight, solid loading ratio of 1:10–20 (dry weight biomass to solution), then collecting the solid for acid pre-treatment with HCl 0.2–2% (w/v), 40–60 °C, 1:10–30 ratio for 2–4 h. Next, the solid residue from the acid pre-treatment is extracted using Na2CO3 at 2–4% (w/v), 40–60 °C, 2–3 h, 1:10–30 ratio. Then the liquid portion is precipitated by ethanol (95%+) with a ratio of 1:1 (v/v). Finally, the solid output is dried in oven at 50–60 °C. Novel extraction methods using ultrasound, microwave, enzymes and extrusion improved the extraction yield and alginate properties, but the financial benefits have not been fully justified yet. To improve the sustainable production of alginate, it is required to promote seaweed cultivation, reduce water footprint, decrease organic solvent usage and co-produce alginate with other value-added products.
The 'golden kelp' Laminaria ochroleuca under global change: Integrating multiple eco-physiological responses with species distribution models
1. The loss of marine foundation species, in particular kelps at temperate latitudes, has been linked to climatic drivers and co-occurring human perturbations. Ocean temperature and nutrients typically covary over local and regional scales and play a crucial role on kelp dynamics. Examining their independent and interactive effects on kelp physiological performance is essential to understand and predict patterns of kelp distribution, particularly under scenarios of global change. 2. Crossed combinations of ocean temperatures and availability of nutrients were experimentally tested on juveniles of the 'golden kelp', Laminaria ochroleuca, from the northwestern Iberian Peninsula. Eco-physiological responses included: survival, growth and total N content. Results were embedded into a Species Distribution Model (SDM), which relates presence records and climatic and non-climatic data to forecast distribution patterns of L. ochroleuca under different climate change scenarios. 3. Temperatures above 24.6°C were lethal irrespective of nutrients. Optimal growth of juvenile sporophytes occurred between 12 and 18°C and no nutrient limitation. The SDM, where ocean temperature was the main predictor of kelp distribution in line with temperature thresholds given by eco-physiological responses, suggests a future expansion towards northern latitudes and a retreat from the southern limit/ boundary of the current distribution. 4. Synthesis. Range-shifting of the golden kelp can have severe ecologipal impacts at regional and local scales. The expansion or retraction of the species along the European coast seems to be modulated mainly by temperature, but nutrient availability would be a key to maintain optimal physiological performance. Our work highlights that the combination of empirical and modelling approaches is accessible to researchers and crucial to building more robust predictions of ecological and biogeographic responses of habitat-forming species to forecasted environmental change.
A Comprehensive and Comparative Analysis of the Fucoidan Compositional Data Across the Phaeophyceae
In the current review, compositional data on fucoidans extracted from more than hundred different species were surveyed through the available literature. The analysis of crude extracts, purified extracts or carefully isolated fractions is included in tabular form, discriminating the seaweed source by its taxonomical order (and sometimes the family). This survey was able to encounter some similarities between the different species, as well as some differences. Fractions which were obtained through anion-exchange chromatography or cationic detergent precipitation showed the best separation patterns: the fractions with low charge correspond mostly to highly heterogeneous fucoidans, containing (besides fucose) other monosaccharides like xylose, galactose, mannose, rhamnose, and glucuronic acid, and contain low-sulfate/high uronic acid proportions, whereas those with higher total charge usually contain mainly fucose, accompanied with variable proportions of galactose, are highly sulfated and show almost no uronic acids. The latter fractions are usually the most biologically active. Fractions containing intermediate proportions of both polysaccharides appear at middle ionic strengths. This pattern is common for all the orders of brown seaweeds, and most differences appear from the seaweed source (habitat, season), and from the diverse extraction, purification, and analytitcal methods. The Dictyotales appear to be the most atypical order, as usually large proportions of mannose and uronic acids appear, and thus they obscure the differences between the fractions with different charge. Within the family Alariaceae (order Laminariales), the presence of sulfated galactofucans with high galactose content (almost equal to that of fucose) is especially noteworthy.
Ultrasound-assisted extraction of phlorotannins and polysaccharides from Silvetia compressa (Phaeophyceae)
Silvetia compressa is a brown seaweed native to the coast of Baja California, Mexico. It is a rich source of phlorotannins and polysaccharides, two compound families with important nutraceutical applications. Optimal conditions for obtaining highly concentrated phlorotannin and polysaccharide extracts from S. compressa were determined using Box-Behnken experimental design combined with response surface methodology (RSM). The effect of extraction temperature (X1: 50–65 °C), ultrasound power density (X2: 1.2–3.8 W cL−1), solvent/seaweed ratio (X3: 10–30 mL g−1 seaweed meal), and ethanol concentration (X4: 25–100% ethanol in water) on phlorotannin and polysaccharide yield was explored. Experimental results were fitted to a second-degree polynomial model, while model fitness was assessed using analysis of variance (ANOVA). From this analysis, optimal phlorotannin and polysaccharide extraction conditions were determined for the evaluated parameters (X1 = 50 °C, X2 = 3.8 W cL−1, X3 = 30 mL g−1 seaweed meal, and X4 = 32.33%). Under these conditions, experimental phlorotannin and polysaccharide yields were 0.73% and 23% (w/w), respectively. Phlorotannin extraction was significantly enhanced by ultrasound power density, while polysaccharide extraction improved when using low ethanol concentration in the solvent, therefore both families of compounds were obtained combining those parameters. Experimental data agreed with predictions from the RSM model, indicating suitability of the obtained model and the success of RSM in optimizing the extraction conditions. In addition, the identification of the main compounds in the phlorotannin extract was carried out by HPLC-MS-TOF; 8 of the 12 identified phenolic compounds belonged to the phlorotannin family, 4 from the fuhalol group, 3 being eckol derivatives, and 1 being a phloroglucinol subunit.
Applications of Antioxidant Secondary Metabolites of Sargassum spp
Sargassum is one of the largest and most diverse genus of brown seaweeds, comprising of around 400 taxonomically accepted species. Many species of this genus have long been a part of human culture with applications as food, feed, and remedies in folk medicine. Apart from their high nutritional value, these seaweeds are also a well-known reservoir of natural antioxidant compounds of great interest, including polyphenols, carotenoids, meroterpenoids, phytosterols, and several others. Such compounds provide a valuable contribution to innovation that can translate, for instance, into the development of new ingredients for preventing product deterioration, particularly in food products, cosmetics or biostimulants to boost crops production and tolerance to abiotic stress. This manuscript revises the chemical composition of Sargassum seaweeds, highlighting their antioxidant secondary metabolites, their mechanism of action, and multiple applications in fields, including agriculture, food, and health.
Green Synthesis of Copper Oxide Nanoparticles Using Protein Fractions from an Aqueous Extract of Brown Algae Macrocystis pyrifera
Amongst different living organisms studied as potential candidates for the green synthesis of copper nanoparticles, algal biomass is presented as a novel and easy-to-handle method. However, the role of specific biomolecules and their contribution as reductant and capping agents has not yet been described. This contribution reports a green synthesis method to obtain copper oxide nanoparticles (CuO-NPs) using separated protein fractions from an aqueous extract of brown algae Macrocystis pyrifera through size exclusion chromatography (HPLC-SEC). Proteins were detected by a UV/VIS diode array, time-based fraction collection was carried out, and each collected fraction was used to evaluate the synthesis of CuO-NPs. The characterization of CuO-NPs was evaluated by Dynamic Light Scattering (DLS), Z-potential, Fourier Transform Infrared (FTIR), Transmission Electron Microscope (TEM) equipped with Energy Dispersive X-ray Spectroscopy (EDS) detector. Low Molecular Weight (LMW) and High Molecular Weight (HMW) protein fractions were able to synthesize spherical CuO-NPs. TEM images showed that the metallic core present in the observed samples ranged from 2 to 50 nm in diameter, with spherical nanostructures present in all containing protein samples. FTIR measurements showed functional groups from proteins having a pivotal role in the reduction and stabilization of the nanoparticles. The highly negative zeta potential average values from obtained nanoparticles suggest high stability, expanding the range of possible applications. This facile and novel protein-assisted method for the green synthesis of CuO-NPs may also provide a suitable tool to synthesize other nanoparticles that have different application areas.
