The Air We Breathe: Air Pollution as a Prevalent Proinflammatory Stimulus Contributing to Neurodegeneration
Air pollution is regarded as an important risk factor for many diseases that affect a large proportion of the human population. To date, accumulating reports have noted that particulate matter (PM) is closely associated with the course of cardiopulmonary disorders. As the incidence of Alzheimer’s disease (AD), Parkinson’s disease (PD), and autoimmune disorders have risen and as the world’s population is aging, there is an increasing interest in environmental health hazards, mainly air pollution, which has been slightly overlooked as one of many plausible detrimental stimuli contributing to neurodegenerative disease onset and progression. Epidemiological studies have indicated a noticeable association between exposure to PM and neurotoxicity, which has been gradually confirmed by in vivo and in vitro studies. After entering the body directly through the olfactory epithelium or indirectly by passing through the respiratory system into the circulatory system, air pollutants are subsequently able to reach the brain. Among the potential mechanisms underlying particle-induced detrimental effects in the periphery and the central nervous system (CNS), increased oxidative stress, inflammation, mitochondrial dysfunction, microglial activation, disturbance of protein homeostasis, and ultimately, neuronal death are often postulated and concomitantly coincide with the main pathomechanisms of neurodegenerative processes. Other complementary mechanisms by which PM could mediate neurotoxicity and contribute to neurodegeneration remain unconfirmed. Furthermore, the question of how strong and proven air pollutants are as substantial adverse factors for neurodegenerative disease etiologies remains unsolved. This review highlights research advances regarding the issue of PM with an emphasis on neurodegeneration markers, symptoms, and mechanisms by which air pollutants could mediate damage in the CNS. Poor air quality and insufficient knowledge regarding its toxicity justify conducting scientific investigations to understand the biological impact of PM in the context of various types of neurodegeneration.
The Importance of 6-Aminohexanoic Acid as a Hydrophobic, Flexible Structural Element
6-aminohexanoic acid is an ω-amino acid with a hydrophobic, flexible structure. Although the ω-amino acid in question is mainly used clinically as an antifibrinolytic drug, other applications are also interesting and important. This synthetic lysine derivative, without an α-amino group, plays a significant role in chemical synthesis of modified peptides and in the polyamide synthetic fibers (nylon) industry. It is also often used as a linker in various biologically active structures. This review concentrates on the role of 6-aminohexanoic acid in the structure of various molecules.
Semi-field studies on biochemical markers of honey bee workers (Apis mellifera) after exposure to pesticides and their mixtures
Due to the fact that many different pesticides are used in crop production and their residues can accumulate in the environment, bees are in contact with various pesticides at the same time. Most studies on their influence on honey bees focus on single substances in concentrations higher than those found in the environment. Our study assessed the chronic effects of commonly used pesticides and their mixtures on selected biochemical markers in worker bee hemolymph. Workers developed in the hive and were provisioned with to pesticides in concentrations corresponding to residues detected in pollen, honey, and/or nectar. Colonies were exposed daily to 0.5L for 7 days by feeding a sugar syrup containing a formulation of acetamiprid (250 ppb) (insecticide), glyphosate (7200 ppb) (herbicide), and tebuconazole (147 ppb) (fungicide) administered alone, in a binary or ternary mixture. Administered alone, acetamiprid significantly decreased the level of urea in the hemolymph of worker honey bees. Glyphosate did not affect significantly the level/activity of any of the biochemical markers. Tebuconazole caused changes in the levels of most of the studied biochemical markers. We found that tebuconazole, which as a fungicide is generally considered safe for bees, may be harmful and more research is required. The impact of fungicides is a crucial element of the assessment of threats to honey bees.
Clinical benefit of cancer drugs approved in Switzerland 2010–2019
It is unknown to what extent cancer drugs approved in Switzerland by the Swissmedic fulfil criteria of clinical benefit according to the European Society of Medical Oncology Magnitude of Clinical Benefit Scale version 1.1 (ESMO-MCBS), the American Society of Clinical Oncology Value Framework version 2 (ASCO-VF) and the Swiss OLUtool v2 (OLUtool). An electronic search identified studies that led to marketing authorisations in Switzerland 2010-2019. Studies were evaluated according to ESMO-MCBS, ASCO-VF and OLUtool. Substantial benefit for ESMO-MCBS, was defined as a grade A or B for (neo)adjuvant intent and 4 or 5 for palliative intent. For ASCO-VF and OLUtool clinical benefit was defined as score ≥45 and A or B, respectively. Concordance between the frameworks was calculated with Cohen's Kappa (κ). Factors associated with clinical benefit were evaluated by logistic regression. In the study period, 48 drugs were approved for 92 evaluable indications, based on 100 studies. Ratings for ESMO-MCBS, ASCO-VF and OLUtool could be performed for 100, 86, and 97 studies, respectively. Overall, 39 (39%), 44 (51%), 45 (46%) of the studies showed substantial clinical benefit according to ESMO-MCBS v1.1, ASCO-VF, OLUtool criteria, respectively. There was fair concordance between ESMO-MCBS and ASCO-VF in the palliative setting (κ = 0.31, P = 0.004) and moderate concordance between ESMO-MCBS and OLUtool (κ = 0.41, P<0.001). There was no significant concordance between ASCO-VF and OLUtool (κ = 0.18, P = 0.12). Factors associated with substantial clinical benefit in multivariable analysis were HRQoL benefit reported as secondary outcome for ESMO-MCBS and the ASCO-VF and blinded studies for OLUtool. At the time of approval, only around half of the trials supporting marketing authorisation of recently approved cancer drugs in Switzerland meet the criteria for substantial clinical benefit when evaluated with ESMO-MCBS, ASCO-VF or OLUtool. There was at best only moderate concordance between the grading systems.
Spectroscopic characterization and in vitro studies of biological activity of bradykinin derivatives
Eleven multiple analogs of bradykinin—a peptide that is a natural ligand of B1 and B2 receptors but does not bind or activate the B1 receptor unless Arg 9 is removed from the sequence by the action of carboxypeptidase N—were synthesized. Their biological activity was examined on T-REx cell lines expressing B1 or B2 receptors using the intracellular IP1 assay. The mRNA expression of B1R and B2R in the lysate of tumor cell lines, e.g., U87-MG (human astrocytoma), SHP-77 (human small cell lung cancer), and H4 (human brain glioma), was determined. For five B1R antagonists, adsorption at the liquid/solid interface (Au nanoparticles (AuNPs) served as the solid surface) was discussed in terms of the vibrations of molecular fragments (structural factors) responsible for the biological properties of these analogs.
Effect of the electric field at 50 Hz and variable intensities on biochemical markers in the honey bee’s hemolymph
The amount of artificial electromagnetic fields of various parameters in the honey bee’s environment increases globally. So far, it had been proven that exposure to an E-field at 50 Hz can cause changes in bee’s behavior, alter the activity of proteases, and enzymatic antioxidants. Due to the potentially harmful effect of this factor on honey bees, we decided to investigate the activity of aspartate aminotransferase (AST), alanine aminotransferase (ALT), and alkaline phosphatase (ALP), and the concentration of albumin and creatinine in bee’s hemolymph after exposure to 50 Hz E-field. Honey bee workers were placed in wooden cages (200 × 150 × 70 mm) and exposed to the 50 Hz E-field with the intensity of <1, 5.0, 11.5, 23.0, or 34.5 kV/m for 1, 3, 6, or 12h. A homogeneous 50 Hz E-field was generated in the form of a plate capacitor. Hemolymph samples for analysis were taken immediately after the end of exposure to the E-field from 100 bees from each group. According to our study, the activity of AST, ALT, and ALP in honey bees’ hemolymph decreased after exposure to 50 Hz E-field with various intensities. The decrease in AST, ALT, and ALP activity intensified with prolonged exposure time. 50 Hz E-field may cause the impairment of crucial metabolic cycles in the honey bees’ organism (such as the citric acid cycle, ATP synthesis, oxidative phosphorylation, β-oxidation). Moreover, exposure to E-Field altered the concentration of creatinine and albumin, which are important non-enzymatic antioxidants. Such changes may indicate a disturbance in protein metabolism and increased muscle activity.
Structural fire performance of wood-steel-wood bolted connections with and without perpendicular-to-wood grain reinforcement
PurposeIn fire condition, the time to failure of a timber connection is mainly reliant on the wood charring rate, the strength of the residual wood section, and the limiting temperature of the steel connectors involved in the connection. The purpose of this study is to experimentally investigate the effects of loaded bolt end distance, number of bolt rows, and the existence of perpendicular-to-wood grain reinforcement on the structural fire behavior of semi-rigid glued-laminated timber (glulam) beam-to-column connections that used steel bolts and concealed steel plate connectors.Design/methodology/approachIn total, 16 beam-to-column connections, which were fabricated in wood-steel-wood bolted connection configurations, in eight large-scale sub-frame test assemblies were exposed to elevated temperatures that followed CAN/ULC-S101 standard time-temperature curve, while being subjected to monotonic loading. The beam-to-column connections of four of the eight test assemblies were reinforced perpendicular to the wood grain using self-tapping screws (STS). Fire tests were terminated upon achieving the failure criterion, which predominantly was dependent on the connection’s maximum allowed rotation.FindingsExperimental results revealed that increasing the number of bolt rows from two to three, each of two bolts, increased the connection’s time to failure by a greater time increment than that achieved by increasing the bolt end distance from four- to five-times the bolt diameter. Also, the use of STS reinforcement increased the connection’s time to failure by greater time increments than those achieved by increasing the number of bolt rows or the bolt end distance.Originality/valueThe invaluable experimental data obtained from this study can be effectively used to provide insight and better understanding on how mass-timber glulam bolted connections can behave in fire condition. This can also help in further improving the existing design guidelines for mass-timber structures. Currently, beam-to-column wood connections are designed mainly as axially loaded connections with no guidelines available for determining the fire resistance of timber connections exerting any degree of moment-resisting capability.
The Role of Digital Platforms in Enhancing Customer Relationships and Building Strategic Advantage in Beekeeping
This study examines how beekeepers perceive the benefits of digital platforms for customer relationships and strategic outcomes, and whether these perceptions differ based on their digital experience. Using a mixed-method approach, data were collected in 2024 in Poland through a survey of 949 beekeepers and 30 expert interviews. Quantitative results show that digitally experienced beekeepers reported higher perceived benefits across all customer relationship dimensions and most strategic ones, particularly in transparency and honey quality, as confirmed by Mann-Whitney U tests and a random forest model. No significant difference was found for customer loyalty, indicating that loyalty depends more on long-term relationships than digital proficiency. Qualitative insights revealed that digital platforms foster trust, support quality improvement, and strengthen beekeeper–customer connections. The study enhances our understanding of how digital experiences shape perceived benefits, and it also provides practical guidance for prioritizing platform features that promote transparency, quality monitoring, and effective communication.
Magnetization enhanced the multiple magnetic domain-dominated resonance modes in stripe domain films
FeCoHf films with different Hf contents and thicknesses were deposited by composition-gradient sputtering. The results showed that the stripe domain (SD) structure emerges when the film thickness exceeds a critical thickness (220–330 nm), but the increase in introduced Hf leads to a decrease in magnetization, and in turn, the stripe phase weakens and eventually disappears. Dynamic measurements revealed magnetic domain-dependent resonant absorption spectra with up to seven resonance peaks, which have rarely been observed in magnetic film with an established SD structure. The number of resonant peaks can be controlled by the saturation magnetization of the film. Micromagnetic simulations indicate that multiple magnetic domain resonance modes can be attributed to the enhanced magnetization, which induces an increase in the magnetic domain wall volume. These results emphasize the pivotal role of magnetic domain dynamics in the framework of spintronic and microwave devices.
