Workplace diversity initiatives: more than ticking a box
There can be a disconnect between Diversity, Equity, and Inclusion (DEI) initiatives implemented at organizations and the perceptions of DEI in the workplace by their employees. Our study sought to determine whether employee experiences within healthcare organizations were dependent on demographic compositions of employee populations. While we did not find a relationship between perceptions of workplace diversity and demographic heterogeneity, we did find a strong association between perceptions of diversity and employee engagement. This relationship suggests that token hiring of a diverse workforce will not improve perceptions of DEI in the workplace. Instead, organizations must also implement cultural policies and procedures to help all employees feel included, valued, and heard. Ultimately, performing well on measures of diversity and equity requires organizations to put in the effort to improve the work experience of their staff.
Targeting of Formyl Peptide Receptor 2 for in vivo imaging of acute vascular inflammation
Inflammatory conditions are associated with a variety of diseases and can significantly contribute to their pathophysiology. Neutrophils are recognised as key players in driving vascular inflammation and promoting inflammation resolution. As a result, neutrophils, and specifically their surface formyl peptide receptors (FPRs), are attractive targets for non-invasive visualization of inflammatory disease states and studying mechanistic details of the process. : A small-molecule Formyl Peptide Receptor 2 (FPR2/ALX)-targeted compound was combined with two rhodamine-derived fluorescent tags to form firstly, a targeted probe (Rho-pip-C1) and secondly a targeted, pH-responsive probe (Rho-NH-C1) for applications. We tested internalization, toxicity and functional interactions with neutrophils for both compounds, as well as the fluorescence switching response of Rho-NH-C1 to neutrophil activation. Finally, imaging (fluorescent intravital microscopy [IVM]) and therapeutic efficacy studies were performed in an inflammatory mouse model. : studies showed that the compounds bound to human neutrophils via FPR2/ALX without causing internalization at relevant concentrations. Additionally, the compounds did not cause toxicity or affect neutrophil functional responses (e.g. chemotaxis or transmigration). studies using IVM showed Rho-pip-C1 bound to activated neutrophils in a model of vascular inflammation. The pH-sensitive (\"switchable\") version termed Rho-NH-C1 validated these findings, showing fluorescent activity only in inflammatory conditions. : These results indicate a viable design of fluorescent probes that have the ability to detect inflammatory events by targeting activated neutrophils.
Shaping the future of kidney genetics in Australia: proceedings from the KidGen policy implementation workshop 2023
The KidGen Collaborative's Policy Implementation Workshop 2023 celebrated the 10th anniversary of Australia's first kidney genetics clinic in Brisbane. This event marked the establishment of a national network now comprising 19 kidney genetics clinics across Australia, all dedicated to providing equitable access to genomic testing for families affected by genetic kidney diseases. The workshop reflected on past progress and outlined future objectives for kidney genetics in Australia, recognising the collaborative efforts of clinical teams, researchers, and patients. Key insights from the workshop are documented in the proceedings.
Bryostatin-1 Attenuates Ischemia-Elicited Neutrophil Transmigration and Ameliorates Graft Injury after Kidney Transplantation
Ischemia reperfusion injury (IRI) is a form of sterile inflammation whose severity determines short- and long-term graft fates in kidney transplantation. Neutrophils are now recognized as a key cell type mediating early graft injury, which activates further innate immune responses and intensifies acquired immunity and alloimmunity. Since the macrolide Bryostatin-1 has been shown to block neutrophil transmigration, we aimed to determine whether these findings could be translated to the field of kidney transplantation. To study the effects of Bryostatin-1 on ischemia-elicited neutrophil transmigration, an in vitro model of hypoxia and normoxia was equipped with human endothelial cells and neutrophils. To translate these findings, a porcine renal autotransplantation model with eight hours of reperfusion was used to study neutrophil infiltration in vivo. Graft-specific treatment using Bryostatin-1 (100 nM) was applied during static cold storage. Bryostatin-1 dose-dependently blocked neutrophil activation and transmigration over ischemically challenged endothelial cell monolayers. When applied to porcine renal autografts, Bryostatin-1 reduced neutrophil graft infiltration, attenuated histological and ultrastructural damage, and improved renal function. Our novel findings demonstrate that Bryostatin-1 is a promising pharmacological candidate for graft-specific treatment in kidney transplantation, as it provides protection by blocking neutrophil infiltration and attenuating functional graft injury.
On the frontiers of collaboration and conflict: how context influences the success of collaboration
The increasing scale and interconnection of many environmental challenges - from climate change to land use - has resulted in the need to collaborate across borders and boundaries of all types. Traditional centralized, top-down and sectoral approaches to governance of single-issue areas or species within social-ecological systems often have limited potential to alleviate issues that go beyond their jurisdiction. As a result, collaborative governance approaches have come to the forefront. A great deal of past research has examined the conditions under which collaborative efforts are likely to achieve desired outcomes. However, few studies have analyzed how the means to achieve successful collaborative outcomes differ based on context when examined across multiple studies. In this research, we begin to chart a means for doing this. Building onto a Context-Mechanism-Outcome (CMO) Framework, we provide a coding manual to analyse how contextual variables mediate the effects of mechanism variables on outcomes of the collaborative governance of social-ecological systems. Through the examination of four cases, we provide a proof-of-concept assessment and show the utility of the CMO framework and coding manual to draw comparisons across cases for understanding how collaborative outcomes are contingent on the social-ecological context in which they occur.
Unity in Service: Cultivating Shared Wisdom for Effective Chaplaincy in the United States Army
Effective Army chaplaincy is forged not only through doctrine and training, but also through shared wisdom refined by experience, hardship, and service. This project sought to capture the collective insight from senior and seasoned chaplains and religious affairs specialists across the United States Army Chaplain Corps to create a field-ready guide designed to develop and sustain effective chaplaincy for future Army operations. Using the Delphi method, two iterative rounds of structured and anonymous feedback were conducted with thirty chaplains and religious affairs specialists, representing 719 combined years of service. The process yielded a six-domain model of effective chaplaincy leadership, encompassing spiritual authenticity, relational credibility, institutional competence, operational resilience, proactive ministry and advocacy, and personal and professional development. Together, these domains describe how chaplains sustain moral readiness, spiritual presence, and trusted advisement across all echelons of the force. This project presents a theologically grounded and practically applied model of chaplaincy leadership rooted in unity and shared wisdom. It demonstrates that effective chaplains are shaped by the convergence of faith, experience, and purpose that strengthens both individual leaders and the collective soul of the United States Army Chaplain Corps.
