Review: Assessing fish welfare in research and aquaculture, with a focus on European directives
The number of farmed fish in the world has increased considerably. Aquaculture is a growing industry that will in the future provide a large portion of fishery products. Moreover, in recent years, the number of teleost fish used as animal models for scientific research in both biomedical and ecological fields has increased. Therefore, it is increasingly important to implement measures designed to enhance the welfare of these animals. Currently, a number of European rules exist as requirements for the establishment, care and accommodation of fish maintained for human purposes. As far as (teleost) fish are concerned, the fact that the number of extant species is much greater than that of all other vertebrates must be considered. Of further importance is that each species has its own specific physical and chemical requirements. These factors make it difficult to provide generalized recommendations or requirements for all fish species. An adequate knowledge is required of the physiology and ecology of each species bred. This paper integrates and discusses, in a single synthesis, the current issues related to fish welfare, considering that teleosts are target species for both aquaculture and experimental models in biological and biomedical research. We first focus on the practical aspects, which must be considered when assessing fish welfare in both research and aquaculture contexts. Next, we address husbandry and the care of fish housed in research laboratories and aquaculture facilities in relation to their physiological and behavioural requirements, as well as in reference to the suggestions provided by European regulations. Finally, to evaluate precisely which parameters described by Directive 2010/63/EU are reported in scientific papers, we analysed 82 articles published by European researchers in 2014 and 2015. This review found that there is a general lack of information related to the optimal environmental conditions that should be provided for the range of species covered by this directive.
Acute environmental temperature variation affects brain protein expression, anxiety and explorative behaviour in adult zebrafish
This study investigated the effect of 4-d acute thermal treatments at 18 °C, 26 °C (control) and 34 °C on the nervous system of adult zebrafish ( Danio rerio ) using a multidisciplinary approach based on behavioural tests and brain proteomic analysis. The behavioural variations induced by thermal treatment were investigated using five different tests, the novel tank diving, light and dark preference, social preference, mirror biting, and Y-Maze tests, which are standard paradigms specifically tailored for zebrafish to assess their anxiety-like behaviour, boldness, social preference, aggressiveness, and explorative behaviour, respectively. Proteomic data revealed that several proteins involved in energy metabolism, messenger RNA translation, protein synthesis, folding and degradation, cytoskeleton organisation and synaptic vesiculation are regulated differently at extreme temperatures. The results showed that anxiety-like behaviours increase in zebrafish at 18 °C compared to those at 26 °C or 34 °C, whereas anxiety-related protein signalling pathways are downregulated. Moreover, treatments at both 18 °C and 34 °C affect the exploratory behaviour that appears not to be modulated by past experiences, suggesting the impairment of fish cognitive abilities. This study is the continuation of our previous work on the effect of 21-d chronic treatment at the same constant temperature level and will enable the comparison of acute and chronic treatment effects on the nervous system function in adult zebrafish.
Increase in environmental temperature affects exploratory behaviour, anxiety and social preference in Danio rerio
The aim of this work is to investigate the effect of a temperature increase on the behaviour of adult zebrafish ( Danio rerio ) maintained for 21 days at 34 °C (treatment) and 26 °C (control). The temperatures chosen are within the vital range of zebrafish and correspond to temperatures that this species encounters in the natural environment. Previous results showed that the same treatment affects the brain proteome and the behaviour of adult zebrafish by producing alterations in the proteins involved in neurotransmitter release and synaptic function and impairing fish exploratory behaviour. In this study, we have investigated the performance of treated and control zebrafish during environmental exploration by using four behavioural tests (novel tank diving, light and dark preference, social preference and mirror biting) that are paradigms for assessing the state of anxiety, boldness, social preference and aggressive behaviour, respectively. The results showed that heat treatment reduces anxiety and increases the boldness of zebrafish, which spent more time in potentially dangerous areas of the tank such as the top and the uncovered bright area and at a distance from the social group, thus decreasing protection for the zebrafish. These data suggest that the increase in ambient temperature may compromise zebrafish survival rate in the natural environment.
Comparison of Surgical Outcomes Associated With Compression Secondary to Hemorrhage and Intervertebral Disk Extrusions in Dogs
Acute intervertebral disk extrusion (IVDE) is one of the most commonly reported neurologic disorders seen in veterinary practice. There is a recognized subset of IVDE cases that have a hemorrhagic inflammatory reaction within the epidural space that causes compression in addition to compression from herniated disk material. Previous reports have been conflicting in the outcomes of these cases. The goals of this retrospective case-control cross-sectional study are to (1) compare the success rate of routine surgical decompression in dogs with DEEH compression compared to Modified Frankel Score (MFS) matched dogs with non-hemorrhagic disk extrusions; (2) evaluate the extent of spinal cord compression on MRI compared to final patient outcomes in DEEH compression and (3) determine the surgical compression to decompression ratio and its relation to patient outcomes in cases of DEEH compression. A total of 143 dogs were included in this study and divided into two groups: DEEH compression dogs ( n = 78) and non-hemorrhagic IVDE dogs ( n = 65). Outcomes were assigned for each patient [0 = deceased, 1 = alive and non-ambulatory (MFS 0–3), 2 = alive and ambulatory (MFS 4 or 5)] in both groups. Outcomes of DEEH and non-hemorrhagic IVDE did not differ when taken to surgery with comparable success rates when stratified by MFS. Similarly, outcomes did not differ between DEEH and non-hemorrhagic IVDE dogs when assessed by compression to decompression ratio. Dogs with DEEH compression had more compressed sites than dogs with non-hemorrhagic IVDE ( P = 0.001) and had more sites decompressed surgically than dogs with non-hemorrhagic IVDE ( P < 0.001). Consequently, the compression to decompression ratio did not differ between the two groups ( P = 0.52). Our results support the finding that when a similar level of surgical decompression is achieved, dogs with DEEH compression have similar outcomes to dogs with non-hemorrhagic IVDE for similar degrees of neurological dysfunction.
Organochlorine and Heavy-Metal Contaminants in Wild Mammals and Birds of Urbino-Pesaro Province, Italy: An Analytic Overview for Potential Bioindicators
Tissue samples from 56 bird and 11 mammal species of different trophic levels, collected from 1994 to 1995 from the Urbino-Pesaro area in the Marche region of central Italy, were analyzed for the presence of organochlorine compounds (polychlorinated biphenyls and p,p'-DDE) and heavy metals (Pb, Cd, Cr, and Hg). Results revealed interspecies differences in pollutant residue concentrations. A clear relationship between contaminant and trophic levels emerged depending on several factors specific to the chemicals and the organisms, the importance of dietary accumulation, and metabolic capacity as it increased toward higher trophic levels. Polychlorinated biphenyls and p,p'-DDE were found in all of the bird and mammal species analyzed (bird- or fish-eating birds), and insectivore mammals showed the highest level of these contaminants. Pb and Hg residues were also widely detected and reflected trophic-level differences. The highest concentration of Pb was found in herbivorous or bird-eating aquatic invertebrates and in insectivorous mammals, particularly in the hedgehog (Erinaceus europaeus), whereas the highest Hg levels were found in fish-eating birds. All of the other heavy metals were detected at low concentrations and represented background levels for birds and mammals, with the exception of increased amounts of Cd and Cr, respectively, found in stone marten (Martes foina) and fox (Vulpes vulpes). Data from this study provided information on baseline levels of interest to monitor status and trends in chemical residue in biota in this specific area, and therefore they represent a tool to evaluate potential ecologic, wildlife, and human health exposure.
Social withdrawal, neophobia, and stereotyped behavior in developing rats exposed to neonatal asphyxia
Perinatal asphyxia is a concern for public health and may promote subtle neuropsychiatric disorders. Anoxic insults to neonatal rats cause long-lasting neurobehavioral deficits. In the present study, we focussed on changes in emotional behaviors as a consequence of neonatal asphyxia in Wistar rats. Newborn pups (24 h after birth) underwent a single 30-min exposure to a 100% N2 atmosphere (or air). The offspring was tested for a) locomotor and exploratory activity with or without a d-amphetamine challenge (0, 1, or 2 mg/kg) on postnatal day (pnd) 15; b) social interactions and novelty seeking during adolescence; c) levels of the brain-derived neurotrophic factor (BDNF). In the open-field test (pnd 15), N2-exposed pups injected with the high (2 mg/kg) amphetamine dose exhibited reduced levels of locomotor hyperactivity, and a more marked involvement in stereotyped behaviors. Individual differences emerged in the locomotor response to the novelty-seeking test: two subgroups of rats (separated on the basis of the median value) showed either arousal/attraction or avoidance/inhibition in response to free-choice novelty. The N2-exposed group showed a more marked novelty-induced avoidance and inhibition. Time devoted to allogrooming and play-soliciting behaviors was reduced, whereas object exploration was increased. Levels of BDNF were reduced in the striatum of N2-exposed rats, suggesting poorer synaptic performance of dopamine pathways. In conclusion, these findings suggest an increased risk of developing social withdrawal, neophobia and behavioral stereotypies (common symptoms found in schizophrenia and autism) as a consequence of neonatal asphyxia in preterm humans.
Emotional Stress Induced by Parachute Jumping Enhances Blood Nerve Growth Factor Levels and the Distribution of Nerve Growth Factor Receptors in Lymphocytes
We examined the plasma nerve growth factor (NGF) level and the distribution of NGF receptors in peripheral lymphocytes of young soldiers (mean age, 20-24 yr) experiencing the thrill of a novice about to make their first parachute jumps. Blood was collected from soldiers who knew they were selected to jump (n = 26), as well as from soldiers who knew they were not selected (n = 17, controls). The former group was sampled the evening before the jump and 20 min after landing. Compared with controls, NGF levels increased 84% in prejump and 107% in postjump sampling. Our studies also showed that the increase of NGF levels preceded the increase of plasma cortisol and adrenocorticotropic hormone. No changes in the baseline levels of circulating interleukin 1β or tumor necrosis factor were found, suggesting that the increased levels of NGF were not correlated with change in these cytokines. Moreover, immunofluorescence analysis demonstrated that parachuting stress enhances the distribution of low-affinity p75LNGFRand high-affinity p140trkA NGF receptors in circulating peripheral blood mononuclear cells. These observations suggest that the release of NGF might be involved in the activation of cells of the immune system and is most probably associated with homeostatic adaptive mechanisms, as previously shown for stressed rodents.
Use of the Milan Pet Quality of Life Instrument (MPQL) to Measure Pets’ Quality of Life during COVID-19
The COVID-19 pandemic caused lifestyle changes, with unknown effect on pets’ quality of life (QoL). Between May and July 2020, we distributed an online survey to investigate the role of several factors on feline and canine QoL, including lockdown-related factors. We used existing scales to measure human and pets’ personalities (Reinforcement Sensitivity Theory Personality Questionnaire, RST-PQ; RST-Dog; RST-Cat) and the human–animal relationship (Lexington Attachment to Pets Scale, LAPS) and the Milan Pet Quality of Life instrument (MPQL). Overall, 235 participants reported about 242 adult pets (Ncats = 78, Ndogs = 164). Factor analysis confirmed the structure and internal reliability of the existing scales (RST-PQ, RST-Dog, RST-Cat, LAPS) and suggested a four-factor structure for the MPQL (physical, psychological, social, environmental). The results indicate that the pets’ psysical QoL was largely explained by pet-related elements (pets’ demographics and life experience, and pets’ personality). Conversely, the pets’ psychological QoL was explained mostly by owner-related elements, such as the owners’ demographics, COVID-19-related changes, and the owners’ personality. Predictably, the pets’ environmental QoL is mostly explained by environmental factors, such as the outdoor access in the home environment and the country. Finally, the pets’ social QoL was explained by the larger combination of models: pets’ characteristics and personality, environment and COVID-19-related changes, and the pet–human relationship. These findings can be explained by two non-mutually exclusive mechanisms. The reported changes may be a by-product of the COVID-19 pandemic’s psychological and lifestyle effects on the owners, which in turn alter the way the owners interact with their pets and look after them. However, the owners’ characteristics and mood may bias their answers regarding their pets.
A Retrospective Performance Assessment of the Developmental Neurotoxicity Study in Support of OECD Test Guideline 426
Objective: We conducted a review of the history and performance of developmental neurotoxicity (DNT) testing in support of the finalization and implementation of Organisation of Economic Co-operation and Development (OECD) DNT test guideline 426 (TG 426). Information Sources And Analysis: In this review we summarize extensive scientific efforts that form the foundation for this testing paradigm, including basic neurotoxicology research, interlaboratory collaborative studies, expert workshops, and validation studies, and we address the relevance, applicability, and use of the DNT study in risk assessment. Conclusions: The OECD DNT guideline represents the best available science for assessing the potential for DNT in human health risk assessment, and data generated with this protocol are relevant and reliable for the assessment of these end points. The test methods used have been subjected to an extensive history of international validation, peer review, and evaluation, which is contained in the public record. The reproducibility, reliability, and sensitivity of these methods have been demonstrated, using a wide variety of test substances, in accordance with OECD guidance on the validation and international acceptance of new or updated test methods for hazard characterization. Multiple independent, expert scientific peer reviews affirm these conclusions.
Aerial hunting behaviour and predation success by peregrine falcons Falco peregrinus on starling flocks Sturnus vulgaris
Predators use diverse hunting strategies to maximize hunting success, while preys adopt anti-predator strategies to maximize escape chances, among which flocking, communal roosting, and the related collective responses are a common pattern in gregarious species. Prey-predator interactions involving a single predator and flocks, a common situation in birds, have received little attention. We studied predation behaviour and success of peregrine falcons Falco peregrinus on starlings Sturnus vulgaris, a highly gregarious species, in proximity of two winter roosts. A total of 328 hunting sequences, with an overall success of 23.1% were recorded. They usually consisted of several attacks, predation success being higher when hunting sequences lasted less than 1.5 min, included less than 3 attacks and no other falcons were hunting simultaneously. Predation success was higher when hunts were directed on singletons than on flocks. However, most hunting sequences were directed towards flocks. Nine hunting strategies on flocks were identified. The most frequent was the 'surprise attack', which was also the most successful. We suggest that this strategy minimizes the amount of anti-predator display elicited by flocks and economizes energy spent in hunting. The constant predation pressure did not seem to affect the use of roosts by starlings, consistent with the 'dilution' hypothesis, while falcons captured at least one prey item every evening. Communal roosting may benefit predator and prey, as both sides could have reached a mutual local equilibrium.
