Interferons (IFNs) are cytokines with powerful immunomodulatory and antiviral properties, but less is known about how they induce cell death. Here, we show that both type I (α/β) and type II (γ) IFNs induce precipitous receptor-interacting protein (RIP)1/RIP3 kinase-mediated necrosis when the adaptor protein Fas-associated death domain (FADD) is lost or disabled by phosphorylation, or when caspases (e.g., caspase 8) are inactivated. IFN-induced necrosis proceeds via progressive assembly of a RIP1–RIP3 “necrosome” complex that requires Jak1/STAT1-dependent transcription, but does not need the kinase activity of RIP1. Instead, IFNs transcriptionally activate the RNA-responsive protein kinase PKR, which then interacts with RIP1 to initiate necrosome formation and trigger necrosis. Although IFNs are powerful activators of necrosis when FADD is absent, these cytokines are likely not the dominant inducers of RIP kinase-driven embryonic lethality in FADD-deficient mice. We also identify phosphorylation on serine 191 as a mechanism that disables FADD and collaborates with caspase inactivation to allow IFN-activated necrosis. Collectively, these findings outline a mechanism of IFN-induced RIP kinase-dependent necrotic cell death and identify FADD and caspases as negative regulators of this process.

Financing acquisitions: to lease or to borrow

A financial lease is an important method of acquiring the use of a wide variety of capital assets. The noncancelable nature of the contract makes this form of asset acquisition very similar to a borrow-to-buy method of financing. The data needed to determine the cost of leasing an asset are: 1. the nature and timing of cash flows, 2. the borrowing rate, and 3. the tax rate. The determination of which alternative lease is best in a given situation will depend on the timing of the cash outflows for each method. A hypothetical example shows the operation of each of these alternatives and presents a coherent method for comparison of the alternative forms of financing. Leasing has increased in popularity in recent years, mainly because of its flexibility in dividing the rights and risks of ownership. Since various leasing arrangements exist, cash flows associated with each arrangement must be analyzed to ensure the choice of a proper financing alternative. Once this analysis is completed, the alternative with the lowest present value of cost should be accepted.

The Rule of 72: Why It Works

The Rule of 72 is probably the best-known rule of thumb in finance. The rule states that the number of years it takes to double an amount invested can be estimated by dividing 72 by the annual interest rate earned, expressed as a percentage. In this article, the two forms of the rule are explained. Also presented is a brief analysis to compare results of the rule so as to assess its accuracy. Finally, a mathematical explanation of why the rule works is provided.

Comparison of Cases Captured in the National Cancer Data Base with Those in Population-based Central Cancer Registries

Background
The National Cancer Data Base (NCDB) is a large, geographically diverse hospital-based cancer registry that has been used to study factors related to cancer diagnosis, treatment, and survival. The primary purpose of this study was to compare the case counts and characteristics of patients in NCDB with population-based registries reported in the United States Cancer Statistics (USCS).
Methods
Cancer case counts from NCDB were compared to case counts from USCS to measure NCDB’s case coverage, or the percentage of cases captured. Case coverage was examined by a variety of characteristics, including state of residence, race/ethnicity, age, and primary cancer site.
Results
The overall NCDB case coverage was 67.4 %, ranging from a high of 88.7 % for Delaware to a low of 27.1 % for Arizona. Case coverage for white, black, and Asian/Pacific Islander cases was high (64.7 % to 67.4 %), but it was much lower for American Indians/Alaskan Natives (32.8 %) and those of Hispanic ethnicity (51.1 %). Among the elderly (aged 65 + years), case coverage is much lower compared to persons younger than 65 (63.0 % and 73.0 %, respectively). Case coverage also varied widely by site, with the highest being cervix (77.9 %) and the lowest being melanoma (50.6 %).
Conclusions
This study highlights the geographic- and site-specific variation in NCDB case coverage, primarily as a result of NCDB facility presence and data collection and processing protocols. These findings illustrate the strengths and limitations of NCDB as a resource for nationwide data on cancer diagnosis, treatment, and survival.
