Characterization of Multidrug-Resistant Staphylococcus aureus Isolates and Comparison of Methods of Susceptibility to Vancomycin
S. aureus are among the main bacteria causing problems related to multidrug resistance in nosocomial infections. Therefore, it is necessary to carry out a reliable and rapid diagnosis for the identification of the bacteria and characterization of its susceptibility profile, especially vancomycin, which is an alternative treatment against multidrug-resistant (MDR) S. aureus. Thus, the goal of this study was to characterize isolates of S. aureus regarding the resistance and virulence and to check the susceptibility to vancomycin, through different methods, for comparative purposes. Seventeen antimicrobials were tested to assess the susceptibility profile. It was evaluated the presence of identification (nuc), resistance (mecA and blaZ), biofilm (icaA and icaD) and siderophore (sfaD and sbnD) genes. The susceptibility to vancomycin was evaluated by Minimum Inhibitory Concentration (MIC) by broth microdilution (BMD), E-test, commercial panel (Kit), and Phoenix equipment. Most S. aureus (93,33%) was classified as MDR. These isolates were 100% positive for nuc, mecA, icaA, icaD, and sfaD genes; 96.67% for sbnD and 33.33% for blaZ. In relation to BMD, all methods correctly classified the susceptibility of the isolates; however, regarding the exact MIC value for vancomycin, Phoenix showed agreement of 63.33%, E-test (33.33%) and Kit (26.66%). In conclusion, most of S. aureus was considered MDR. Also, they presented resistance, biofilm production, and siderophores genes, showing the pathogenic potential of these bacteria. Besides, the Phoenix test was considered the most effective, as it presents advantages, such as identification of the microorganism and a greater number of antimicrobials tested at a time.
Climate justice beliefs related to climate action and policy support around the world
Climate justice is increasingly prominent in climate change communication and advocacy but little is known about public understanding of the concept or how widely it resonates with different groups. In our global survey of 5,627 adults in 11 countries spanning the global north and south, most participants (66.2%) had never heard of climate justice. Nonetheless, endorsement of climate justice beliefs was widespread (for example, acknowledging the disproportionate impact of climate change on poor people and the underpinning roles of capitalism and colonialism in the climate crisis). Climate justice beliefs were also associated with various indices of climate action and policy support. These associations tended to be stronger in countries with high GHG emissions and where social inequality is also more politically salient. The results highlight the value of climate justice as a motive for climate action across diverse geographical contexts. Little is known about public understanding of climate justice, despite its increasing prominence in climate change communication. Here a global survey reveals that, although awareness of climate justice is low, beliefs in climate justice are widely supported across countries.
Response to mRNA vaccination for COVID-19 among patients with multiple myeloma
Multiple myeloma (MM) patients are at higher risk for severe COVID-19. Their mRNA vaccination response against SARS-CoV-2 is unknown. Thus, we analyzed responses to mRNA vaccination against COVID-19 among these patients. Using an ELISA-based assay that detects IgG antibodies to SARS-CoV-2 spike protein, we determined serum antibody levels prior to immunization and 12–21 and 14–21 days following the first and second vaccinations, respectively, with mRNA-1273 (Moderna) or BNT162b2 (Pfizer/BioNTech) among 103 MM patients (96 and 7 with active and smoldering disease, respectively). We stratified patients into clinically relevant responders (>250 IU/mL), partial responders (50–250 IU/mL, which was above pre-COVID-19 background), and nonresponders (<50 IU/mL). Smoldering MM patients responded better than those with active disease. Only 45% of active MM patients developed an adequate response, while 22% had a partial response. Lower spike antibody levels were associated with older age, impaired renal function, low lymphocyte counts, reduced uninvolved immunoglobulin levels, > second line of treatment, and among those not in complete remission. Patients who received mRNA-1273 vaccine had higher anti-spike antibody levels than those who were vaccinated with BNT162b2. Thus, most MM patients have impaired responses to mRNA vaccination against COVID-19, and specific clinical and myeloma-related characteristics predict vaccine responsiveness.
Transdiagnostic variations in impulsivity and compulsivity in obsessive-compulsive disorder and gambling disorder correlate with effective connectivity in cortical-striatal-thalamic-cortical circuits
Individual differences in impulsivity and compulsivity is thought to underlie vulnerability to a broad range of disorders and are closely tied to cortical-striatal-thalamic-cortical function. However, whether impulsivity and compulsivity in clinical disorders is continuous with the healthy population and explains cortical-striatal-thalamic-cortical dysfunction across different disorders remains unclear. Here, we characterized the relationship between cortical-striatal-thalamic-cortical effective connectivity, estimated using dynamic causal modelling of resting-state functional magnetic resonance imaging data, and dimensional phenotypes of impulsivity and compulsivity in two symptomatically distinct but phenotypically related disorders, obsessive-compulsive disorder and gambling disorder. 487 online participants provided data for modelling of dimensional phenotypes. These data were combined with 34 obsessive-compulsive disorder patients, 22 gambling disorder patients, and 39 healthy controls, who underwent functional magnetic resonance imaging. Three core dimensions were identified: disinhibition, impulsivity, and compulsivity. Patients’ scores on these dimensions were continuously distributed with the healthy participants, supporting a continuum model of psychopathology. Across all participants, higher disinhibition correlated with lower bottom-up connectivity in the dorsal circuit and greater bottom-up connectivity in the ventral circuit, and higher compulsivity correlated with lower bottom-up connectivity in the dorsal circuit. In patients, higher clinical severity was also linked to lower bottom-up connectivity in the dorsal circuit, but these findings were independent of phenotypic variation, demonstrating convergence towards behaviourally and clinically relevant changes in brain dynamics. Effective connectivity did not differ as a function of traditional diagnostic labels and only weak associations were observed for functional connectivity measures. Together, our results demonstrate that cortical-striatal-thalamic-cortical dysfunction across obsessive-compulsive disorder and gambling disorder may be better characterized by dimensional phenotypes than diagnostic comparisons, supporting investigation of quantitative liability phenotypes. •We model transdiagnostic phenotypes of impulsivity and compulsivity.•We examine effective connectivity in obsessive-compulsive and gambling disorders.•Phenotypes better explain variance in effective connectivity than group comparisons.•Effective connectivity more sensitive than functional connectivity.•Results highlight strength of applying dimensional phenotypes trans-diagnostically.
Age modifies the association between sex and the plasma inflammatory proteome in treated HIV
BACKGROUNDAmong antiretroviral therapy-suppressed (ART-suppressed) people with HIV (PWH), women have higher levels of some inflammatory markers than men, but the effect of sex on the inflammatory proteome, and whether age modifies these differences, remain unclear.METHODSPlasma inflammatory protein levels were assessed in ART-suppressed PWH from the Center for AIDS Research Network of Integrated Clinical Systems. The relationship between sex and plasma proteins - including 22 interferon-α response pathway proteins - was assessed, adjusting for confounders and assessing interactions by age.RESULTSOf 922 participants, 162 (18%) were female. The median age was 47, above which the majority of women had undetectable plasma anti-Müllerian hormone levels, a biomarker of ovarian reserve. Age modified the influence of sex on the inflammatory proteome. Older age (>47) was associated with greater increases among women than men in 194 proteins. Interferon-α response proteins were higher in men in those ≤ 47 but higher in women in those > 47 (interaction P < 0.001). Among the 131 proteins associated with mortality risk (q < 0.05), only 5 differed by sex among those ≤ 47, while 79 differed by sex in those > 47, with nearly all being higher in women. Women had decreased mortality than men ≤ 47 (P < 0.001) but had similar mortality > 47 (P = 0.84).CONCLUSIONThe menopausal transition appears to increase systemic type I interferon responses and inflammation in women with HIV, which may contribute to a loss of female advantage in mortality.FUNDINGNIH National Heart, Lung, and Blood Institute; National Institute of Neurological Disorders and Stroke; National Institute of Allergy and Infectious Diseases.
Personality predictors of career exploration: A meta‐analysis
In this meta‐analysis, correlations of personality traits and career decision‐making self‐efficacy with self and environmental career exploration were estimated across 71 samples and a pairwise sample of 19,846. Traits included the Big Five personality traits, shyness, locus of control, vocational decision styles, and stress. Many measures of career exploration were based on scores of self and environmental exploration on the Career Exploration Survey. Results varied by characteristics with the largest effects for locus of control, vocational decision style: thinking‐feeling and self‐efficacy. Environmental exploration and self‐exploration were moderately correlated and demonstrated different relations with traits that were consistent with theory. Self‐exploration was related to openness to experience and unrelated to extraversion, while environmental exploration was correlated with extraversion. Both were similarly correlated with conscientiousness such that conscientious individuals were more likely to engage in career exploration.
The global spectrum of tree crown architecture
Species’ traits and environmental conditions determine the abundance of tree species across the globe. The extent to which traits of dominant and rare tree species differ remains untested across a broad environmental range, limiting our understanding of how species traits and the environment shape forest functional composition. We use a global dataset of tree composition of >22,000 forest plots and 11 traits of 1663 tree species to ask how locally dominant and rare species differ in their trait values, and how these differences are driven by climatic gradients in temperature and water availability in forest biomes across the globe. We find three consistent trait differences between locally dominant and rare species across all biomes; dominant species are taller, have softer wood and higher loading on the multivariate stem strategy axis (related to narrow tracheids and thick bark). The difference between traits of dominant and rare species is more strongly driven by temperature compared to water availability, as temperature might affect a larger number of traits. Therefore, climate change driven global temperature rise may have a strong effect on trait differences between dominant and rare tree species and may lead to changes in species abundances and therefore strong community reassembly.
Potential areas of richness, endemism and conservation of the birds of Queretaro State, Mexico/Areas potenciales de riqueza, endemismo y conservacion de las aves del estado de Queretaro, Mexico/Areas potenciais de riqueza, endemismo e conservacao das aves do estado de Queretaro, Mexico
The distribution of bird species in the Mexican state of Queretaro was analyzed, based on the determination of potential distribution areas using ecological niche modeling. The potential distributions were also used to define areas of endemism for the birds in the state, applying a parsimony analysis of endemicity, and the protective effectiveness of core areas within the Sierra Gorda Biosphere Reserve (RBSG) was evaluated. Presence data points for 376 species of birds across the state, obtained from scientific collections, the literature and field work, were used in the analyzes. The potential distribution of species richness, endemism and threatened species showed the highest concentration in the bio-geographical province of Sierra Madre Oriental. Although there are areas in the south of the state that have a large number of species, they have not been sufficiently evaluated in terms of their conservation. The identification of areas of endemism revealed two major groups: 1) the northern region of the eastern slope of the Sierra Madre Oriental, characterized by humid climates; and 2) the areas that correspond to the western slope of the Sierra Madre Oriental, the Mexican Plateau and the Transmexican Volcanic Belt, characterized by dry and temperate climates and medium-low moisture. On the other hand, the results demonstrated that the core zones within the RBSG are not covered effectively in order to protect areas with a high hypothetical concentrations of species, including the endemic and threatened birds.
