Enhancing DLG2 Implications in Neuropsychiatric Disorders: Analysis of a Cohort of Eight Patients with 11q14.1 Imbalances
Neurodevelopmental disorders (NDDs) are considered synaptopathies, as they are due to anomalies in neuronal connectivity during development. DLG2 is a gene involved insynaptic function; the phenotypic effect of itsalterations in NDDs has been underestimated since few cases have been thoroughly described.We report on eight patients with 11q14.1 imbalances involving DLG2, underlining its potential effects on clinical presentation and its contribution to NDD comorbidity by accurate neuropsychiatric data collection. DLG2 is a very large gene in 11q14.1, extending over 2.172 Mb, with alternative splicing that gives rise to numerous isoforms differentially expressed in brain tissues. A thorough bioinformatic analysis of the altered transcripts was conducted for each patient. The different expression profiles of the isoforms of this gene and their influence on the excitatory–inhibitory balance in crucial brain structures could contribute to the phenotypic variability related to DLG2 alterations. Further studies on patients would be helpful to enrich clinical and neurodevelopmental findings and elucidate the molecular mechanisms subtended to NDDs.

On the relations between affordance and representation of the agent’s effector
The present study aimed to determine whether the representation of object affordances requires specification of the effector potentially interacting with the object: specifically, in this study, vision of the interacting hand. In Experiment 1 we used an apparatus by which a fruit to be reached and grasped was identified by word reading, whereas another (interfering) fruit was visually perceived at the same location as the target. The apparatus allowed visual presentation of the agent's interacting hand or prevented it. When visually presented, the hand was perceived as still at the start position even when it moved to grasp the fruit. An interference effect on the grasp congruent with the distractor size was observed only when the hand was visible. In Experiment 2, interference was observed also when a hand different from the agent's own was visually presented. In both Experiments 1 and 2 the visible fruit interfered with the arm's reach, but the effect was independent of its size and less dependent on the visually-presented hand. A control experiment (Experiment 3) enabled comparison of the interference of visual stimuli on targets identified by word reading (Experiments 1 and 2) with that of objects identified by word reading on visually-presented targets (Experiment 3). The interference induced by visual stimuli was stronger than the interference induced by objects identified by words (i.e. affordances evoked by visual stimuli were stronger than affordances evoked by semantics). Taken together, the results of the present study suggest that the specification of the agent's effector is necessary for the elicitation of affordances. However, the elicitation of these affordances was observed for interactions between object and hand (grasp), rather than for interactions between object and arm (reach). Finally, our data confirm the influence of semantics on the control of arm movements, though less strong than that due to visual input.
Early Behavioral Development in Autistic Children: The First 2 Years of Life through Home Movies
Objective: The main aim of the research is to study the early behavioral development in autistic children through home movies. Methods: fifteen home movies, regarding the first 2 years of life of autistic children are compared with the home movies of 15 normal children. The films of the two groups were mixed and rated by blind ratings with the Grid for the Assessment of Normal Behavior in Infants and Toddlers. The grid is composed of 17 items grouped into three developmental areas: social competence, intersubjectivity and symbolic activity. For every area, we have identified specific children’s behaviors. Results: Significant differences between the two groups are found both in the range of age 0–6 for intersubjectivity, and in the ranges of age 6–12 and 18–24 for symbolic activity. Conclusions: The authors pose some hypotheses about an early-appearing impairment of intersubjectivity, the ability to represent other’s state of mind, in subjects with autistic disorder.
Serum Levels of Perfluoroalkyl Substances (PFAS) in Adolescents and Young Adults Exposed to Contaminated Drinking Water in the Veneto Region, Italy: A Cross-Sectional Study Based on a Health Surveillance Program
In spring 2013, groundwater of a vast area of the Veneto Region (northeastern Italy) was found to be contaminated by perfluoroalkyl substances (PFAS) from a PFAS manufacturing plant active since the late 1960s. Residents were exposed to high concentrations of PFAS, particularly perfluorooctanoic acid (PFOA), through drinking water until autumn 2013. A publicly funded health surveillance program is under way to aid in the prevention, early diagnosis, and treatment of chronic disorders possibly associated with PFAS exposure. The objectives of this paper are: ) to describe the organization of the health surveillance program, ) to report serum PFAS concentrations in adolescents and young adults, and ) to identify predictors of serum PFAS concentrations in the studied population. The health surveillance program offered to residents of municipalities supplied by contaminated waterworks includes a structured interview, routine blood and urine tests, and measurement of 12 PFAS in serum by high-performance liquid chromatography-tandem mass spectrometry. We studied 18,345 participants born between 1978 and 2002, 14-39 years of age at recruitment. Multivariable linear regression was used to identify sociodemographic, lifestyle, dietary, and reproductive predictors of serum PFAS concentrations. The PFAS with the highest serum concentrations were PFOA [median , interquartile range (IQR) 19.3-84.9], perfluorohexanesulfonic acid (PFHxS) (median , IQR 1.9-7.4), and perfluorooctanesulfonic acid (PFOS) (median , IQR 2.6-5.8). The major predictors of serum levels were gender, municipality, duration of residence in the affected area, and number of deliveries. Overall, the regression models explained 37%, 23%, and 43% of the variance of PFOA, PFOS, and PFHxS, respectively. Serum PFOA concentrations were high relative to concentrations in populations with background residential exposures only. Flavour physics from an approximate U(2)3 symmetry
A bstract The quark sector of the Standard Model exhibits an approximate U(2) 3 flavour symmetry. This symmetry, broken in specific directions dictated by minimality, can explain the success of the Cabibbo-Kobayashi-Maskawa picture of flavour mixing and CP violation, confirmed by the data so far, while allowing for observable deviations from it, as expected in most models of ElectroWeak Symmetry Breaking. Building on previous work in the specific context of supersymmetry, we analyze the expected effects and we quantify the current bounds in a general Effective Field Theory framework. As a further relevant example we then show how the U(2) 3 symmetry and its breaking can be implemented in a generic composite Higgs model and we make a first analysis of its peculiar consequences. We also discuss how some partial extension of U(2) 3 to the lepton sector can arise, both in general and in composite Higgs models. An optimistic though conceivable interpretation of the considerations developed in this paper gives reasons to think that new physics searches in the flavour sector may be about to explore an interesting realm of phenomena.
Geodesy and metrology with a transportable optical clock
Optical atomic clocks, due to their unprecedented stability1–3 and uncertainty3–6, are already being used to test physical theories7,8 and herald a revision of the International System of Units9,10. However, to unlock their potential for cross-disciplinary applications such as relativistic geodesy11, a major challenge remains: their transformation from highly specialized instruments restricted to national metrology laboratories into flexible devices deployable in different locations12–14. Here, we report the first field measurement campaign with a transportable 87Sr optical lattice clock12. We use it to determine the gravity potential difference between the middle of a mountain and a location 90 km away, exploiting both local and remote clock comparisons to eliminate potential clock errors. A local comparison with a 171Yb lattice clock15 also serves as an important check on the international consistency of independently developed optical clocks. This campaign demonstrates the exciting prospects for transportable optical clocks.
Intercontinental comparison of optical atomic clocks through very long baseline interferometry
The comparison of distant atomic clocks is foundational to international timekeeping, global positioning and tests of fundamental physics. Optical-fibre links allow the most precise optical clocks to be compared, without degradation, over intracontinental distances up to thousands of kilometres, but intercontinental comparisons remain limited by the performance of satellite transfer techniques. Here we show that very long baseline interferometry (VLBI), although originally developed for radio astronomy and geodesy, can overcome this limit and compare remote clocks through the observation of extragalactic radio sources. We developed dedicated transportable VLBI stations that use broadband detection and demonstrate the comparison of two optical clocks located in Italy and Japan separated by 9,000 km. This system demonstrates performance beyond satellite techniques and can pave the way for future long-term stable international clock comparisons.Very long baseline interferometry is used to compare two optical clocks located in Japan and Italy through the observation of extragalactic radio sources. This approach overcomes limitations of the performance of satellite transfer techniques.
The Rise of Near-Infrared Emitters: Organic Dyes, Porphyrinoids, and Transition Metal Complexes
In recent years, the interest in near-infrared (NIR) emitting molecules and materials has increased significantly, thanks to the expansion of the potential technological applications of NIR luminescence in several areas such as bioimaging, sensors, telecommunications, and night-vision displays. This progress has been facilitated by the development of new synthetic routes for the targeted functionalization and expansion of established molecular frameworks and by the availability of simpler and cheaper NIR detectors. Herein, we present recent developments on three major classes of systems—i.e., organic dyes, porphyrinoids, and transition metal complexes—exhibiting the maximum of the emission band at λ > 700 nm. In particular, we focus on the design strategies that may increase the luminescence efficiency, while pushing the emission band more deeply in the NIR region. This overview suggests that further progress can be achieved in the near future, with enhanced availability of more robust, stronger, and cheaper NIR luminophores.
Non-Alcoholic Fatty Liver Disease Is Associated with an Increased Incidence of Atrial Fibrillation in Patients with Type 2 Diabetes
The relationship between non-alcoholic fatty liver disease (NAFLD) and atrial fibrillation (AF) in type 2 diabetes is currently unknown. We examined the relationship between NAFLD and risk of incident AF in people with type 2 diabetes. We prospectively followed for 10 years a random sample of 400 patients with type 2 diabetes, who were free from AF at baseline. A standard 12-lead electrocardiogram was undertaken annually and a diagnosis of incident AF was confirmed in affected participants by a single cardiologist. At baseline, NAFLD was defined by ultrasonographic detection of hepatic steatosis in the absence of other liver diseases. During the 10 years of follow-up, there were 42 (10.5%) incident AF cases. NAFLD was associated with an increased risk of incident AF (odds ratio [OR] 4.49, 95% CI 1.6-12.9, p<0.005). Adjustments for age, sex, hypertension and electrocardiographic features (left ventricular hypertrophy and PR interval) did not attenuate the association between NAFLD and incident AF (adjusted-OR 6.38, 95% CI 1.7-24.2, p = 0.005). Further adjustment for variables that were included in the 10-year Framingham Heart Study-derived AF risk score did not appreciably weaken this association. Other independent predictors of AF were older age, longer PR interval and left ventricular hypertrophy. Our results indicate that ultrasound-diagnosed NAFLD is strongly associated with an increased incidence of AF in patients with type 2 diabetes even after adjustment for important clinical risk factors for AF.
