Genetic and non-genetic clonal diversity in cancer evolution
The observation and analysis of intra-tumour heterogeneity (ITH), particularly in genomic studies, has advanced our understanding of the evolutionary forces that shape cancer growth and development. However, only a subset of the variation observed in a single tumour will have an impact on cancer evolution, highlighting the need to distinguish between functional and non-functional ITH. Emerging studies highlight a role for the cancer epigenome, transcriptome and immune microenvironment in functional ITH. Here, we consider the importance of both genetic and non-genetic ITH and their role in tumour evolution, and present the rationale for a broad research focus beyond the cancer genome. Systems-biology analytical approaches will be necessary to outline the scale and importance of functional ITH. By allowing a deeper understanding of tumour evolution this will, in time, encourage development of novel therapies and improve outcomes for patients.This Review discusses the role of functional (impacting tumour phenotype) and non-functional intra-tumour heterogeneity (ITH) in cancer evolution, highlighting the importance of considering genetic and non-genetic factors and their impact on patient outcomes.

Supporting the creative industries through the AI turn: A comparative analysis of Scottish policy and the needs of Scotland’s creative practitioners
In the creative industries (one of the largest UK sectors, which operates at the juncture of business, technology and the arts) creative practitioners are not only consumers of Artificial Intelligence (AI), but also its developers, originators and innovators. This has material consequences for how creative work is performed, valued and understood, as well as global consequences for business structures, the environment, equality and power discrepancies. We take Scotland as a case study for how governmental policy addresses the challenges of AI for the creative industries by comparing Scotland’s Artificial Intelligence Strategy with data gathered from grass roots creative industries practitioners in Scotland through workshops and a survey. We identify differences between the priorities of Scotland’s Artificial Intelligence Strategy and the creative industries, as well as a lack of advice specific to the needs of the creative industries, and offer recommendations on how to put creatives at the heart of AI developments.
Clinical implications of heterogeneity in PD-L1 immunohistochemical detection in hepatocellular carcinoma: the Blueprint-HCC study
Programmed cell death ligand-1 immunohistochemical detection (PD-L1 IHC) is a putative predictor of response to PD-1/PD-L1-targeted checkpoint inhibitors. However, there is no gold standard assay in hepatocellular carcinoma (HCC). We evaluated 5 PD-L1 IHC assay platforms (E1LN3, 28-8, 22c3, SP263 and SP142) in 100 HCCs reporting PD-L1 expression in malignant (M) and tumour-infiltrating immune cells (TICs) and non-tumorous cirrhotic tissues (NTICs). We found substantial inter-assay heterogeneity in detecting PD-L1 expression in M ( R 2  = 0.080–0.921), TICs (Cohen’s   κ  = 0.175–0.396) and NTICs ( κ  = 0.004–0.505). Such diversity may impact on the reliability and reproducibility of PD-L1 IHC assays as a predictor of response to immune checkpoint inhibitors.

Cancer gene identification from RNA variant allelic frequencies using RVdriver
Existing approaches to identifying cancer genes rely overwhelmingly on DNA sequencing data. Here, we introduce RVdriver , a computational tool that leverages paired bulk genomic and transcriptomic data to classify RNA variant allele frequencies (VAFs) of non-synonymous mutations relative to a synonymous mutation background. We analyze 7882 paired exomes and transcriptomes from 31 cancer types and identify novel, as well as known, cancer genes, complementing other DNA-based approaches. Furthermore, RNA VAFs of individual mutations are able to distinguish “driver” from “passenger” mutations within established cancer genes. This approach highlights the value of multi-omic approaches for cancer gene discovery.
