Roles of host and environment in shift of primary anthrax host species in Kruger National Park

Environmental and climatic factors, as well as host demographics and behaviour, significantly influence the exposure of herbivorous mammalian hosts to pathogens such as Bacillus anthracis, the causative agent of anthrax. Until the early 1990s in Kruger National Park (KNP), kudu (Tragelaphus strepsiceros) was the host species most affected by anthrax, with outbreaks occurring predominantly in the dry season, particularly during drought cycles. However, the most affected host species has shifted to impala (Aepyceros melampus), with more frequent anthrax outbreaks during the wet season. This study investigates the roles of environmental variation and other host species in this shift. Temporal trends in environmental variables such as precipitation, soil moisture, temperature, and normalised difference vegetation index (NDVI) were analyzed in relation to anthrax occurrence (presence/ absence and counts). Additionally, correlations between host species' densities and anthrax mortalities over time were examined. Anthrax cases in 1990 were concentrated in the central and northern regions of KNP (excluding Pafuri), primarily affected kudus; while subsequent mortalities affected mostly impala and were restricted to the far north, in Pafuri. Significant correlations were found between kudu anthrax mortality and a decrease in NDVI, average temperature, SPI-6 and SPI-12 (Standardised Precipitation Index in various time intervals. Conversely, anthrax occurrence in impalas was associated with a decline in SPI-3, and temperature rise, with increased mortality during the rainy season. Elephant density correlated negatively with kudu mortality, but a positive correlation with both impala mortality and impala density. The study concludes that environmental variables and species' densities may alter the diversity and frequency of hosts exposed to B. anthracis. Climate extremes and alterations therein may exacerbate anthrax severity by modifying species susceptibility and their probability of exposure over time.

Anthropogenic Influences on Conservation Values of White Rhinoceros

White rhinoceros (rhinos) is a keystone conservation species and also provides revenue for protection agencies. Restoring or mimicking the outcomes of impeded ecological processes allows reconciliation of biodiversity and financial objectives. We evaluate the consequences of white rhino management removal, and in recent times, poaching, on population persistence, regional conservation outcomes and opportunities for revenue generation. In Kruger National Park, white rhinos increased from 1998 to 2008. Since then the population may vary non-directionally. In 2010, we estimated 10,621 (95% CI: 8,767-12,682) white rhinos using three different population estimation methods. The desired management effect of a varying population was detectable after 2008. Age and sex structures in sink areas (focal rhino capture areas) were different from elsewhere. This comes from relatively more sub-adults being removed by managers than what the standing age distribution defined. Poachers in turn focused on more adults in 2011. Although the effect of poaching was not detectable at the population level given the confidence intervals of estimates, managers accommodated expected poaching annually and adapted management removals. The present poaching trend predicts that 432 white rhinos may be poached in Kruger during 2012. The white rhino management model mimicking outcomes of impeded ecological processes predicts 397 rhino management removals are required. At present poachers may be doing \"management removals,\" but conservationists have no opportunity left to contribute to regional rhino conservation strategies or generate revenue through white rhino sales. In addition, continued trends in poaching predict detectable white rhino declines in Kruger National Park by 2016. Our results suggest that conservationists need innovative approaches that reduce financial incentives to curb the threats that poaching poses to several conservation values of natural resources such as white rhinos.

Evidence that residues of tebuthiuron arboricide present in soil of Mokala National Park can be phytotoxic to woody and grass species

Mokala National Park (MoNP) has a history of arboricide use through South African National Parks (SANPs) having bought commercial game farmland for its establishment in 2007. Tebuthiuron arboricide is known to have been applied for controlling bush densification during the period 1996 to 2004. Persistent negative impacts on MoNP vegetation, which are ascribed to the historical arboricide use, have prompted this investigation from 2016 to 2017. Bioassay experiments employing as test plants the tree species Vachellia erioloba and Vachellia tortilis, the shrub species Senegalia mellifera and the grass Tragus berteronianus were conducted in a glasshouse. Growth responses of these species were assessed upon their exposure to a tebuthiuron concentration range that simulated expected levels in MoNP soil soon and long after application. Chemical analysis as well as bioassay with the test species Lycopersicon esculentum (tomato) were performed on soil samples collected from three depths (0−30, 30−60 and 60−90 cm) of the soil profile at two sites in MoNP where tebuthiuron was applied in the past. The three woody test species showed differential, negative growth response to tebuthiuron, and even growth of the grass species (T. berteronianus) was affected at the higher concentrations. Evidence provided by the tomato bioassay and analysis performed on soil samples collected in situ points to the putative presence of tebuthiuron, more than a decade after the last use of arboricides for controlling bush densification. CONSERVATION IMPLICATIONS : If the reported evidence of the presence of phytotoxic residue of tebuthiuron in soil of MoNP would be substantiated through further research, such findings could at least partly explain the failure of natural recruitment of vegetation in those areas where the woody component was degraded because of arboricide application more than a decade ago.

Differential herbivore occupancy of fire-manipulated savannas in the Satara region of the Kruger National Park, South Africa

The Kruger National Park's (KNP) long-running experimental burn plots (EBPs) have a history of research projects, which improve the understanding of fire in savanna ecosystems. Using data from KNP's aerial censuses (2005-2016) and in situ dung count data (2008-2017), this study assessed (1) herbivore densities on the Satara, N'Wanetsi and Marheya EBPs, on annual, triennial and no-burn treatments and across pre-, during and post-drought climate conditions; (2) herbivore densities of these EBPs relative to their non-manipulated surroundings and (3) the extent to which distance to water and rainfall influence ungulate densities. The results revealed that herbivore mean density differed significantly between the three EBPs of Satara and across their fire treatments. N'Wanetsi showed the highest density (0.30 animals/ha), whilst the lowest was found at Marheya (0.12 animals/ha). Overall, pre-drought density was higher on the annual plots (0.56 animals/ha), whilst higher post-drought density was evidenced on the triennial plots (0.80 animals/ha). On average, there were significantly higher herbivore densities on the EBPs (2.54 animals/ha) compared to the surrounding matrix at the larger scales of the Satara management section (0.15 animals/ha) and the central KNP (0.18 animals/ha). A positive correlation between herbivore mean density estimate and distance to water was shown. However, grazer mean density across fire treatments was strongly correlated to rainfall.Conservation implications: Given the variation in fire regimes and their application, and the non-uniform and elevated herbivore densities of the EBPs, inferences from the EBPs cannot be made to the larger KNP. The trials should rather be viewed as an isolated, fire herbivory experiment. It is also recommended to align the experiment with South African National Parks' mandate by including biodiversity parameters like small mammals and insects in the monitoring of the plots.

Low unspliced cell-associated HIV RNA in early treated adolescents living with HIV on long suppressive ART

Initiation of antiretroviral treatment (ART) in patients early after HIV-infection and long-term suppression leads to low or undetectable levels of HIV RNA and cell-associated (CA) HIV DNA and RNA. Both CA-DNA and CA-RNA, overestimate the size of the HIV reservoir but CA-RNA as well as p24/cell-free viral RNA can be indicators of residual viral replication. This study describes HIV RNA amounts and levels of cytokines/soluble markers in 40 well-suppressed adolescents who initiated ART early in life and investigated which viral markers may be informative as endpoints in cure clinical trials within this population.
Forty adolescents perinatally infected with HIV on suppressive ART for >5 years were enrolled in the CARMA study. HIV DNA and total or unspliced CA-RNA in PBMCs were analyzed by qPCR/RT-qPCR and dPCR/RT-dPCR. Cell-free HIV was determined using an ultrasensitive viral load (US-VL) assay. Plasma markers and p24 were analyzed by digital ELISA and correlations between total and unspliced HIV RNA and clinical markers, including age at ART, Western Blot score, levels of cytokines/inflammation markers or HIV CA-DNA, were tested.
CA-RNA was detected in two thirds of the participants and was comparable in RT-qPCR and RT-dPCR. Adolescents with undetectable CA-RNA showed significantly lower HIV DNA compared to individuals with detectable CA-RNA. Undetectable unspliced CA-RNA was positively associated with age at ART initiation and Western Blot score. We found that a higher concentration of TNF-α was predictive of higher CA-DNA and CA-RNA. Other clinical characteristics like US-VL, time to suppression, or percent CD4+ T-lymphocytes were not predictive of the CA-RNA in this cross-sectional study.
Low CA-DNA after long-term suppressive ART is associated with lower CA-RNA, in concordance with other reports. Patients with low CA-RNA levels in combination with low CA-DNA and low Western Blot scores should be further investigated to characterize candidates for treatment interruption trials. Unspliced CA-RNA warrants further investigation as a marker that can be prioritized in paediatric clinical trials where the sample volume can be a significant limitation.
