Arbovirus vector dynamics and spread are influenced by climatic, environmental and geographic factors. Major Chikungunya and Dengue fever outbreaks occurring the last 10 years have coincided with the expansion of the mosquito vector Aedes albopictus to nearly all the continents. We characterized the ecological (larval development sites, population dynamics, insemination and daily survival rates) and genetic (diversity, gene flow, population structure) features of two Aedes albopictus populations from distinct environments (rural and urban) on Réunion Island, in the South-West Indian Ocean. Microsatellite analysis suggests population sub-structuring Ae. albopictus populations. Two genetic clusters were identified that were significantly linked to natural versus urban habitats with a mixed population in both areas. Ae. albopictus individuals prefer urban areas for mating and immature development, where hosts and containers that serve as larval development sites are readily available and support high population densities, whereas natural environments appear to serve as reservoirs for the mosquito.

Repeated pesticide contaminations of lentic freshwater systems located within agricultural landscapes may affect population evolution in non-target organisms, especially in species with a fully aquatic life cycle and low dispersal ability. The issue of evolutionary impact of pollutants is therefore conceptually important for ecotoxicologists. The impact of historical exposure to pesticides on genetic divergence was investigated in the freshwater gastropod Lymnaea stagnalis, using a set of 14 populations from contrasted environments in terms of pesticide and other anthropogenic pressures. The hypothesis of population adaptive divergence was tested on 11 life-history traits, using Q(ST)-F(ST) comparisons. Despite strong neutral differentiation (mean F(ST) = 0.291), five adult traits or parameters were found to be under divergent selection. Conversely, two early expressed traits showed a pattern consistent with uniform selection or trait canalization, and four adult traits appeared to evolve neutrally. Divergent selection patterns were mostly consistent with a habitat effect, opposing pond to ditch and channel populations. Comparatively, pesticide and other human pressures had little correspondence with evolutionary patterns, despite hatching rate impairment associated with global anthropogenic pressure. Globally, analyses revealed high genetic variation both at neutral markers and fitness-related traits in a species used as model in ecotoxicology, providing empirical support for the need to account for genetic and evolutionary components of population response in ecological risk assessment.

Methods for the evaluation and comparison of the structure of numerous honeybee colonies are needed for the development of applied and fundamental field research, as well as to evaluate how the structure and activity of honeybee colonies evolve over time. ColEval complements existing methods, as it uses an online reference image bank for (human) learning and training purposes. ColEval is based on the evaluation of the surface area percentage occupied by different components of a honeybee colony: adult worker bees, open and capped brood, honey, nectar, and pollen. This method is an essential tool for the description of the evolution in the size of honeybee colonies. The procedure makes allowances for tendencies between different observers and uses them to calculate accurate measurements of honeybee colony evaluation. ColEval thus allows for a posteriori comparison of under- or over-evaluation made by different observers working on the same project; it is thus possible to eliminate observer bias in the measurements and to conduct large surveys.

Due to their ability to explore whole genome response to drugs and stressors, omics-based approaches are widely used in toxicology and ecotoxicology, and identified as powerful tools for future ecological risk assessment and environmental monitoring programs. Understanding the long-term effects of contaminants may indeed benefit from the coupling of genomics and eco-evolutionary hypotheses. Next-generation sequencing provides a new way to investigate pollutants impact, by targeting early responses, screening chemicals, and directly quantifying gene expression, even in organisms without reference genome. Lymnaea stagnalis is a freshwater mollusk in which access to genomic resources is critical for many scientific issues, especially in ecotoxicology. We used 454-pyrosequencing to obtain new transcriptomic resources in L. stagnalis and to preliminarily explore gene expression response to a redox-cycling pesticide, diquat. We obtained 151,967 and 128,945 high-quality reads from control and diquat-exposed individuals, respectively. Sequence assembly provided 141,999 contigs, of which 124,387 were singletons. BlastX search revealed significant match for 34.6% of the contigs (21.2% protein hits). KEGG annotation showed a predominance of hits with genes involved in energy metabolism and circulatory system, and revealed more than 400 putative genes involved in oxidative stress, cellular/molecular stress and signaling pathways, apoptosis, and metabolism of xenobiotics. Results also suggest that diquat may have a great diversity of molecular effects. Moreover, new genetic markers (putative SNPs) were discovered. We also created a Ensembl-like web-tool for data-mining ( http://genotoul-contigbrowser.toulouse.inra.fr:9095/Lymnaea_stagnalis/index.html ). This resource is expected to be relevant for any genomic approach aimed at understanding the molecular basis of physiological and evolutionary responses to environmental stress in L. stagnalis.

Here we describe a novel, inexpensive and simple method for preserving RNA that reduces handling stress in aquatic invertebrates following ecotoxicogenomic experimentation. The application of the method is based on transcriptomic experiments conducted on Daphnia magna, but may easily be applied on a range of other aquatic organisms of a particular size with e.g. amphipod Gammarus pulex representing an upper size limit. We explain in detail how to apply this new method, named the “Cylindrical Sieve (CS) system”, and highlight its advantages and disadvantages.

Evidence of Habitat Structuring Aedes albopictus Populations in Réunion Island

yThe objective of our research was to study the movements the mosquito Aedes albopictus. This mosquito transmits more than 20 viruses to humans throughout the world and is the vector of the recent major epidemics of Dengue and Chikungunya on Reunion Island and the Indian Ocean Region and is, therefore, of great interest for human health. We set out to determine whether reservoirs of populations could be found in natural environments and whether or not these populations are capable of re-colonising urban areas. Until now, only limited data has been available on the population dynamics of Aedes albopictus in this part of the world, information critical for guiding vector control strategies and predicting or preventing epidemics. We chose two areas where a serious CHIKV epidemic occurred. We then used genetic markers and ecological data to estimate patterns of gene flow and behaviour. We were able to demonstrate that populations were structured with limited gene flow despite observing migration. We found that Ae. albopictus preferred urban areas for mating and to lay their eggs because of the availability of hosts and permanent containers that favoured higher mosquito densities. We also show, however, that natural environments are reservoirs for re-colonisation of urban areas.
