Advances in European agroforestry: results from the AGFORWARD project
In global terms, European farms produce high yields of safe and high quality food but this depends on the use of many off-farm inputs and the associated greenhouse gas emissions, loss of soil nutrients and other negative environmental impacts incur substantial societal costs. Farmers in the European Union receive support through a Common Agricultural Policy (CAP) that comprises direct payments to farmers (Pillar I) and payments related to rural development measures (Pillar II). This paper examines the ways in which agroforestry can support European agriculture and rural development drawing on the conclusions of 23 papers presented in this Special Issue of Agroforestry Systems which have been produced during a 4-year research project called AGFORWARD. The project had the goal of promoting agroforestry in Europe and focused on four types of agroforestry: (1) existing systems of high nature and cultural value, and agroforestry for (2) high value tree, (3) arable, and (4) livestock systems. The project has advanced our understanding of the extent of agroforestry in Europe and of farmers’ perceptions of agroforestry, including the reasons for adoption or non-adoption. A participatory approach was used with over 40 stakeholder groups across Europe to test selected agroforestry innovations through field trials and experiments. Innovations included improved grazing management in agroforestry systems of high nature and cultural value and the introduction of nitrogen fixing plants in high value timber plantations and olive groves. Other innovations included shelter benefits for arable crops, and disease-control, nutrient-retention, and food diversification benefits from integrating trees in livestock enterprises. Biophysical and economic models have also been developed to predict the effect of different agroforestry designs on crop and tree production, and on carbon sequestration, nutrient loss and ecosystems services in general. These models help us to quantify the potential environmental benefits of agroforestry, relative to agriculture without trees. In view of the substantial area of European agroforestry and its wider societal and environmental benefits, the final policy papers in this Special Issue argue that agroforestry should play a more significant role in future versions of the CAP than it does at present.
Current Status and Issues Regarding Pre-processing of fNIRS Neuroimaging Data: An Investigation of Diverse Signal Filtering Methods Within a General Linear Model Framework
Functional near-infrared spectroscopy (fNIRS) research articles show a large heterogeneity in the analysis approaches and pre-processing procedures. Additionally, there is often a lack of a complete description of the methods applied, necessary for study replication or for results comparison. The aims of this paper were (i) to review and investigate which information is generally included in published fNIRS papers, and (ii) to define a signal pre-processing procedure to set a common ground for standardization guidelines. To this goal, we have reviewed 110 fNIRS articles published in 2016 in the field of cognitive neuroscience, and performed a simulation analysis with synthetic fNIRS data to optimize the signal filtering step before applying the GLM method for statistical inference. Our results highlight the fact that many papers lack important information, and there is a large variability in the filtering methods used. Our simulations demonstrated that the optimal approach to remove noise and recover the hemodynamic response from fNIRS data in a GLM framework is to use a 1000th order band-pass Finite Impulse Response filter. Based on these results, we give preliminary recommendations as to the first step toward improving the analysis of fNIRS data and dissemination of the results.
Fifty Years of Prefrontal Cortex Research: Impact on Assessment
Our knowledge of the functions of the prefrontal cortex, often called executive, supervisory, or control, has been transformed over the past 50 years. After operationally defining terms for clarification, we review the impact of advances in functional, structural, and theoretical levels of understanding upon neuropsychological assessment practice as a means of identifying 11 principles/challenges relating to assessment of executive function. Three of these were already known 50 years ago, and 8 have been confirmed or emerged since. Key themes over this period have been the emergence of the use of naturalistic tests to address issues of “ecological validity”; discovery of the complexity of the frontal lobe control system; invention of new tests for clinical use; development of key theoretical frameworks that address the issue of the role of prefrontal cortex systems in the organization of human cognition; the move toward considering brain systems rather than brain regions; the advent of functional neuroimaging, and its emerging integration into clinical practice. Despite these huge advances, however, practicing neuropsychologists are still desperately in need of new ways of measuring executive function. We discuss pathways by which this might happen, including decoupling the two levels of explanation (information processing; brain structure) and integrating very recent technological advances into the neuropsychologist’s toolbox. The Rule of Lore in the Rule of Law: Putting the Problem of the Rule of Law in Context
In this article, I identify a number of commonly accepted assumptions from the literature associated with the Rule of Law and suggest that—whilst the assumptions are accepted as part of the conceptual narrative of the concept—the cogency of the assumptions falters when they are considered collectively. This article represents, in many respects, both a critique of current practices and a rallying cry in relation to future practices. Through illustrating that the form of conceptual change across canonical conceptions of the Rule of Law can impact the relative level of consistency in the assumptions that are used and relied on in the opening of so many Rule-of-Law-focused works, I demonstrate that there must—if we are to provide the strongest possible arguments relating to the contemporary idea of the Rule of Law—be consideration of the actual way in which change has occurred across conceptions. I argue that consideration of collective cogency is necessary for conceptual clarity and illustrate the essentiality of doing so by considering the assumptions in relation to two hypothetical mechanisms of change. This approach illustrates not only the general inconsistency, but also that inconsistency varies between the mechanisms. This variance leads to a fundamental problem: without the identification of the change mechanism that has operated across Rule of Law related ideas, there is no way to assess whether the Rule of Law’s common assumptions are, or can be considered to be, consistent with one another. I also suggest one way to solve this problem.
Acute physical activity supports inhibitory control in primary school children: a randomised cross-over trial
Studies have found inconsistent results regarding the acute effects of physical activity on inhibitory control in children. More naturalistic studies that have real-world validity and use objective measures of physical activity are essential. This study investigated the acute effects of a pre-existing school-based group physical activity session on inhibitory control. Fifty-five primary school children (9 ± 1 years old, 40% female) participated in a pre-existing school-based group physical activity session and a sedentary poster-making control condition, both lasting approximately 30 min, in a randomised, cross-over manner. Cognitive tasks were completed before and after both conditions, and measures of performance on these tasks were used as dependent variables in all analyses. Time engaged in moderate-to-vigorous physical activity during each condition was measured by accelerometry and used as a manipulation check. After physical activity there were faster reaction times (β = -176, 95%CI: -296.43, -59.51) and more commission errors (β = 1.06, 95%CI: 0.18, 1.95) in the simple reaction time task, but fewer commission errors (β = -4.64, 95%CI: -9.00 -0.28) in the inhibition task compared to the control condition. Using the same statistical approach, a supplementary analysis was run on a reduced sample of 32 (9 ± 1 years old, 44% female) whose adherence to conditions was confirmed by accelerometry. Results showed faster reaction times (β = -205, 95%CI: -384.58, -33.37) in the simple reaction time task and fewer commission errors (β = -6.10, 95%CI: -11.75, -0.45) in the inhibition task following physical activity. This suggests that an acute bout of physical activity facilitates inhibitory control in primary school children. Optimal effects are seen when participants achieved higher intensity physical activity during the intervention.
Assessing the carbon capture potential of a reforestation project
The number of reforestation projects worldwide is increasing. In many cases funding is obtained through the claimed carbon capture of the trees, presented as immediate and durable, whereas reforested plots need time and maintenance to realise their carbon capture potential. Further, claims usually overlook the environmental costs of natural or anthropogenic disturbances during the forest’s lifetime, and greenhouse gas (GHG) emissions associated with the reforestation are not allowed for. This study uses life cycle assessment to quantify the carbon footprint of setting up a reforestation plot in the Peruvian Amazon. In parallel, we combine a soil carbon model with an above- and below-ground plant carbon model to predict the increase in carbon stocks after planting. We compare our results with the carbon capture claims made by a reforestation platform. Our results show major errors in carbon accounting in reforestation projects if they (1) ignore the time needed for trees to reach their carbon capture potential; (2) ignore the GHG emissions involved in setting up a plot; (3) report the carbon capture potential per tree planted, thereby ignoring limitations at the forest ecosystem level; or (4) under-estimate tree losses due to inevitable human and climatic disturbances. Further, we show that applications of biochar during reforestation can partially compensate for project emissions.
Dabrafenib plus trametinib in patients with BRAFV600E-mutated biliary tract cancer (ROAR): a phase 2, open-label, single-arm, multicentre basket trial
Effective treatments for patients with cholangiocarcinoma after progression on gemcitabine-based chemotherapy are urgently needed. Mutations in the BRAF gene have been found in 5% of biliary tract tumours. The combination of dabrafenib and trametinib has shown activity in several BRAFV600E-mutated cancers. We aimed to assess the activity and safety of dabrafenib and trametinib combination therapy in patients with BRAFV600E-mutated biliary tract cancer. This study is part of an ongoing, phase 2, open-label, single-arm, multicentre, Rare Oncology Agnostic Research (ROAR) basket trial in patients with BRAFV600E-mutated rare cancers. Patients were eligible for the biliary tract cancer cohort if they were aged 18 years or older, had BRAFV600E-mutated, unresectable, metastatic, locally advanced, or recurrent biliary tract cancer, an Eastern Cooperative Oncology Group performance status of 0–2, and had received previous systemic treatment. All patients were treated with oral dabrafenib 150 mg twice daily and oral trametinib 2 mg once daily until disease progression or intolerance of treatment. The primary endpoint was the overall response rate, which was determined by Response Evaluation Criteria in Solid Tumors version 1.1 in the intention-to-treat evaluable population, which comprised all enrolled patients regardless of receiving treatment who were evaluable (ie, had progression, began a new anticancer treatment, withdrew consent, died, had stable disease for 6 weeks or longer, or had two or more post-baseline assessments). The ROAR trial is registered with ClinicalTrials.gov, NCT02034110. These results are based on an interim analysis; the study is active but not recruiting. Between March 12, 2014, and July 18, 2018, 43 patients with BRAFV600E-mutated biliary tract cancer were enrolled to the study and were evaluable. Median follow-up was 10 months (IQR 6–15). An investigator-assessed overall response was achieved by 22 (51%, 95% CI 36–67) of 43 patients. An independent reviewer-assessed overall response was achieved by 20 (47%, 95% CI 31–62) of 43 patients. The most common grade 3 or worse adverse event was increased γ-glutamyltransferase in five (12%) patients. 17 (40%) patients had serious adverse events and nine (21%) had treatment-related serious adverse events, the most frequent of which was pyrexia (eight [19%]). No treatment-related deaths were reported. Dabrafenib plus trametinib combination treatment showed promising activity in patients with BRAFV600E-mutated biliary tract cancer, with a manageable safety profile. Routine testing for BRAFV600E mutations should be considered in patients with biliary tract cancer. GlaxoSmithKline and Novartis.
