Aptamer-Based Imaging of Polyisoprenoids in the Malaria Parasite
Dolichols are isoprenoid end-products of the mevalonate and 2C-methyl-D-erythritol-4-phosphate pathways. The synthesis of dolichols is initiated with the addition of several molecules of isopentenyl diphosphate to farnesyl diphosphate. This reaction is catalyzed by a cis-prenyltransferase and leads to the formation of polyprenyl diphosphate. Subsequent steps involve the dephosphorylation and reduction of the α-isoprene unit by a polyprenol reductase, resulting in the generation of dolichol. The size of the dolichol varies, depending on the number of isoprene units incorporated. In eukaryotes, dolichols are synthesized as a mixture of four or more different lengths. Their biosynthesis is predicted to occur in the endoplasmic reticulum, where dolichols play an essential role in protein glycosylation. In this study, we have developed a selection of aptamers targeting dolichols and enhanced their specificity by incorporating fatty acids for negative selection. One aptamer showed high enrichment and specificity for linear polyisoprenoids containing at least one oxygen atom, such as an alcohol or aldehyde, in the α-isoprene unit. The selected aptamer proved to be a valuable tool for the subcellular localization of polyisoprenoids in the malaria parasite. To the best of our knowledge, this is the first time that polyisoprenoids have been localized within a cell using aptamer-based imaging techniques.

Cripowellins Pause Plasmodium falciparum Intraerythrocytic Development at the Ring Stage
Cripowellins from Crinum erubescens are known pesticidal and have potent antiplasmodial activity. To gain mechanistic insights to this class of natural products, studies to determine the timing of action of cripowellins within the asexual intraerythrocytic cycle of Plasmodium falciparum were performed and led to the observation that this class of natural products induced reversible cytostasis in the ring stage within the first 24 h of treatment. The transcriptional program necessary for P. falciparum to progress through the asexual intraerythrocytic life cycle is well characterized. Whole transcriptome abundance analysis showed that cripowellin B “pauses” the transcriptional program necessary to progress through the intraerythrocytic life cycle coinciding with the lack of morphological progression of drug treated parasites. In addition, cripowellin B-treated parasites re-enter transcriptional progression after treatment was removed. This study highlights the use of cripowellins as chemical probes to reveal new aspects of cell cycle progression of the asexual ring stage of P. falciparum which could be leveraged for the generation of future antimalarial therapeutics.

Picomolar antimalarial agent from a Chinese medicinal plant
In our long-term efforts searching for antimalarial agents from Chinese medicinal plants, seven novel dimeric sesquiterpenoids ( 1 – 7 ) with significant antimalarial activity were isolated and characterized from the whole plants of Sarcandra glabra subsp. brachystachys by solid data acquired by diverse methods. Among them, compound 3 with an EC 50 value of 4.3 pM against the chloroquine-resistant Plasmodium falciparum is the most potent antimalarial agent reported hitherto, about 1,000-fold stronger than artemisinin. This article further consolidates and refines our previously delineated structure-activity relationship for this antimalarial compound class.

A Kuhnian critique of Hume on miracles
In Part I of “Of Miracles,” Hume argues that belief in miracle-testimony is never justified. While Hume’s argument has been widely criticized and defended along a number of different veins, including its import on scientific inquiry, this paper takes a novel approach by comparing Hume’s argument with Thomas Kuhn’s account of scientific anomalies. This paper makes two arguments: first that certain types of scientific anomalies—those that conflict with the corresponding paradigm theory—are analogous to miracles in the relevant ways. Note, importantly, that the argument applies only to the first definition of ‘miracle’that Hume offers (i.e. ‘miracle’ as a “violation of the laws of nature.”) Second, it argues that we are sometimes rationally justified in believing testimony for scientific anomalies (that conflict with the corresponding paradigm theory), because there have been several cases of scientists accepting such anomalies and—assuming certain criteria are met—we are rationally justified in believing these scientists. If both arguments are successful, then it is possible to be rationally justified in believing miracle-testimony, though the extent of justification depends on various criteria and comes in degrees. After examining a few objections, the paper concludes by contextualizing this argument in relation to Part II of Hume’s essay and in relation to broader apologetic concerns. In short, it is vital to recognize that this paper’s focus is Hume’s first account of ‘miracle,’ rather than his argument against miracle-testimony more broadly, but the argument could be coupled with other arguments against Hume’s broader attack on miracle-testimony.

إن كتابة تاريخ الفتح العربي لمصر لم يعالج-من قبل المؤلف-على أنه حادث منقطع العلاقة بسائر حوادث التاريخ بل على أنه حادث لا يظهر خطره ولا تتضح حقيقته إلا إذا قرن بالأحداث التاريخية الكبرى التي ساقت دولتي الروم والفرس القديمتين إلى الأصطدام بالدولة العربية الناشئة إن الروح العادلة التي حدت بالمؤلف إلى نصرة الحق في جانب أمة القبط حدت به كذلك إلى نصرة الحق في جانب أمة العرب فلم يحاول أن يخفي من فضائلها شيئا أو يعكر من صفو سيرتها في مدة فتح مصر بل كان عادلا في وصف الأفراد والمجموع إن هذا الكتاب له قيمة خاصة لسبب آخر فوق ما سبق لنا بيانه وذلك أن تواريخ العرب وفتوحهم لم يتناولها إلى الآن كاتب حصر همه في ميدان محدود وبحث فيه بحثا مستفيضا كما فعل مؤلف هذا الكتاب.
Quantitative Trait Loci Associated with the Tocochromanol (Vitamin E) Pathway in Barley
The Genome-Wide Association Studies approach was used to detect Quantitative Trait Loci associated with tocochromanol concentrations using a panel of 1,466 barley accessions. All major tocochromanol types- α-, β-, δ-, γ-tocopherol and tocotrienol- were assayed. We found 13 single nucleotide polymorphisms associated with the concentration of one or more of these tocochromanol forms in barley, seven of which were within 2 cM of sequences homologous to cloned genes associated with tocochromanol production in barley and/or other plants. These associations confirmed a prior report based on bi-parental QTL mapping. This knowledge will aid future efforts to better understand the role of tocochromanols in barley, with specific reference to abiotic stress resistance. It will also be useful in developing barley varieties with higher tocochromanol concentrations, although at current recommended daily consumption amounts, barley would not be an effective sole source of vitamin E. However, it could be an important contributor in the context of whole grains in a balanced diet.
