Electrospinning and additive manufacturing (AM) are key technologies for fabricating bone tissue engineering scaffolds, each with unique strengths and limitations. Electrospinning produces nanoscale fibers that promote cell attachment and affinity on 2D surfaces but offer limited mechanical strength. In contrast, AM creates 3D scaffolds with enhanced mechanical properties through precise control of topological structures, but the capability to stimulate and guide cell growth is limited compared to electrospun nanoscale fibers. Combining both methods holds potential for next‐generation scaffold development with desirable mechanical and biological properties. This study investigates the fabrication of multi‐scale and multi‐material scaffolds by integrating extrusion‐based AM and solution electrospinning. Polycaprolactone (PCL), a biocompatible and biodegradable polymer, served as the base material, while graphene nanosheets were incorporated as functional fillers to enhance mechanical, electrical, surface, and biological properties. Solution electrospinning was first optimized, and hybrid scaffolds were fabricated, with an image‐based optimization method, obtaining 87% of the fibres well‐aligned with the designed direction. Optimal scaffold composition (PCL nanofibers with 1 wt.% graphene + PCL microfibers with 3 wt.% graphene) was also identified based on 2D mesh characterization results (186% enhancement of the mechanical property and 23% enhancement of the cell proliferation result, compared with neat PCL). The findings demonstrate the potential of this hybrid fabrication approach for developing advanced polymer‐carbon nanomaterial scaffolds for bone tissue regeneration applications. A multi‐scale and multi‐material PCL/graphene scaffold was successfully fabricated by integrating extrusion‐based AM and solution electrospinning. The process was optimized using an image‐based method, achieving 87% fiber alignment with the intended orientation. The combination of 3D printed nanofibers (1 wt.% graphene) and electrospun microfibers (3 wt.% graphene) was identified as optimal, based on comprehensive 2D mesh characterization.

Hyaluronidase Modulates Inflammatory Response and Accelerates the Cutaneous Wound Healing

Hyaluronidases are enzymes that degrade hyaluronan an important constituent of the extracellular matrix. They have been used as a spreading agent, improving the absorption of drugs and facilitating the subcutaneous infusion of fluids. Here, we investigated the influence of bovine testes hyaluronidase (HYAL) during cutaneous wound healing in in vitro and in vivo assays. We demonstrated in the wound scratch assay that HYAL increased the migration and proliferation of fibroblasts in vitro at low concentration, e.g. 0.1 U HYAL enhanced the cell number by 20%. HYAL presented faster and higher reepithelialization in in vivo full-thickness excisional wounds generated on adult Wistar rats back skin already in the early phase at 2nd day post operatory compared to vehicle-control group. Wound closured area observed in the 16 U and 32 U HYAL treated rats reached 38% and 46% compared to 19% in the controls, respectively. Histological and biochemical analyses supported the clinical observations and showed that HYAL treated wounds exhibited increased granulation tissue, diminished edema formation and regulated the inflammatory response by modulating the release of pro and anti-inflammatory cytokines, growth factor and eicosanoids mediators. Moreover, HYAL increased gene expression of peroxisome proliferator-activated receptors (PPAR) γ and PPAR β/δ, the collagen content in the early stages of healing processes as well as angiogenesis. Altogether these data revealed that HYAL accelerates wound healing processes and might be beneficial for treating wound disorders.

Critical bone defects are considered one of the major clinical challenges in reconstructive bone surgery. The combination of 3D printed conductive scaffolds and exogenous electrical stimulation (ES) is a potential favorable approach for bone tissue repair. In this study, 3D conductive scaffolds made with biocompatible and biodegradable polycaprolactone (PCL) and multi-walled carbon nanotubes (MWCNTs) were produced using the extrusion-based additive manufacturing to treat large calvary bone defects in rats. Histology results show that the use of PCL/MWCNTs scaffolds and ES contributes to thicker and increased bone tissue formation within the bone defect. Angiogenesis and mineralization are also significantly promoted using high concentration of MWCNTs (3 wt%) and ES. Moreover, scaffolds favor the tartrate-resistant acid phosphatase (TRAP) positive cell formation, while the addition of MWCNTs seems to inhibit the osteoclastogenesis but present limited effects on the osteoclast functionalities (receptor activator of nuclear factor
κβ
ligand (RANKL) and osteoprotegerin (OPG) expressions). The use of ES promotes the osteoclastogenesis and RANKL expressions, showing a dominant effect in the bone remodeling process. These results indicate that the combination of 3D printed conductive PCL/MWCNTs scaffold and ES is a promising strategy to treat critical bone defects and provide a cue to establish an optimal protocol to use conductive scaffolds and ES for bone tissue engineering.

Zoonotic transmission of novel Influenza A variant viruses detected in Brazil during 2020 to 2023

Zoonotic infections (swine-human) caused by influenza A viruses (IAVs) have been reported and linked to close contact between these species. Here, we describe eight human IAV variant infections (6 mild and 2 severe cases, including 1 death) detected in Paraná, Brazil, during 2020–2023. Genomes recovered were closely related to Brazilian swIAVs of three major lineages (1 A.3.3.2/pdm09, 1B/human-like, and H3.1990.5), including three H1N1v, two H1N2v, two H3N2v and one H1v. Five H1v were closely related to pdm09 lineage, one H1v (H1N2v) grouped within 1B.2.3 clade, and the two H3v grouped within a clade composed exclusively of Brazilian H3 swIAV (clade H3.1990.5.1). Internal gene segments were closely related to H1N1pdm09 isolated from pigs. IAV variant rarely result in sustained transmission between people, however the potential to develop such ability is of concern and must not be underestimated. This study brings into focus the need for continuous influenza surveillance and timely risk assessment.

Clastic sedimentary record impacted by carbonate bioclasts in the Late Ediacaran

Clastic sedimentary systems and their characteristics are assumed not to have been modified by carbonate bioclastic grains until the Phanerozoic. Here, we show that the presence of carbonate bioclasts produced by disintegrated biomineralizing metazoans modified fine-grained siliciclastic facies in the Late Ediacaran Tamengo Formation, Brazil, ca. 555–542 Ma. The analysis of both polished sections and thin sections shows that sand-sized carbonate bioclasts (< 2 mm) derived from the Ediacaran metazoan Corumbella created diverse sedimentary features later found in the Phanerozoic record, such as bioclastic-rich horizontal and low-angle cross-laminations, erosive pods and lenses, bioclastic syneresis cracks, ripples preserved by bioclastic caps, microbial lamination eroded and filled with bioclasts, and entrapped bioclasts within microbial mats. These sedimentary features would have hardly been recorded in fine siliciclastic facies without the sand-sized bioclasts. Based on these features, together with other sedimentary evidence, Corumbella depositional settings in the Tamengo Fm. are reinterpreted as mid-ramp, subtidal settings. The multi-component organization of the skeleton of Corumbella favoured disarticulation to yield a sand-sized bioclast, so in turn creating a new complexity to shallow marine clastic settings typical of Phanerozoic marine depositional systems.

Estimation of genetic parameters for longevity considering the cow’s age at last calving

The aim of this study was to estimate heritability and predict breeding values for longevity among cows in herds of Nellore breed, considering the trait cow’s age at last calving (ALC), by means of survival analysis methodology. The records of 11,791 animals from 22 farms were used. The variable ALC has been used by a criterion that made it possible to include cows not only at their first calving but also at their ninth calving. The criterion used was the difference between the date of each cow’s last calving and the date of the last calving on each farm. If this difference was greater than 36 months, the cow was considered to have failed and uncensored. If not, this cow was censored, thus indicating that future calving remained possible for this cow. The survival model used for the analyses was the proportional hazards model, and the base risk was given by a Weibull distribution. The heritability estimate obtained was equal to 0.25. It was found that the ALC variable had the capacity to respond to selection for the purpose of increasing the longevity of the cows in the herds.
