Microbial lipases and their industrial applications: a comprehensive review
Lipases are very versatile enzymes, and produced the attention of the several industrial processes. Lipase can be achieved from several sources, animal, vegetable, and microbiological. The uses of microbial lipase market is estimated to be USD 425.0 Million in 2018 and it is projected to reach USD 590.2 Million by 2023, growing at a CAGR of 6.8% from 2018. Microbial lipases (EC 3.1.1.3) catalyze the hydrolysis of long chain triglycerides. The microbial origins of lipase enzymes are logically dynamic and proficient also have an extensive range of industrial uses with the manufacturing of altered molecules. The unique lipase (triacylglycerol acyl hydrolase) enzymes catalyzed the hydrolysis, esterification and alcoholysis reactions. Immobilization has made the use of microbial lipases accomplish its best performance and hence suitable for several reactions and need to enhance aroma to the immobilization processes. Immobilized enzymes depend on the immobilization technique and the carrier type. The choice of the carrier concerns usually the biocompatibility, chemical and thermal stability, and insolubility under reaction conditions, capability of easy rejuvenation and reusability, as well as cost proficiency. Bacillus spp., Achromobacter spp., Alcaligenes spp., Arthrobacter spp., Pseudomonos spp., of bacteria and Penicillium spp., Fusarium spp., Aspergillus spp., of fungi are screened large scale for lipase production. Lipases as multipurpose biological catalyst has given a favorable vision in meeting the needs for several industries such as biodiesel, foods and drinks, leather, textile, detergents, pharmaceuticals and medicals. This review represents a discussion on microbial sources of lipases, immobilization methods increased productivity at market profitability and reduce logistical liability on the environment and user.
Tool and techniques study to plant microbiome current understanding and future needs: an overview
Microorganisms are present in the universe and they play role in beneficial and harmful to human life, society, and environments. Plant microbiome is a broad term in which microbes are present in the rhizo, phyllo, or endophytic region and play several beneficial and harmful roles with the plant. To know of these microorganisms, it is essential to be able to isolate purification and identify them quickly under laboratory conditions. So, to improve the microbial study, several tools and techniques such as microscopy, rRNA, or rDNA sequencing, fingerprinting, probing, clone libraries, chips, and metagenomics have been developed. The major benefits of these techniques are the identification of microbial community through direct analysis as well as it can apply in situ. Without tools and techniques, we cannot understand the roles of microbiomes. This review explains the tools and their roles in the understanding of microbiomes and their ecological diversity in environments.
Epidemic preparedness and innovations in digital healthcare: enhancing post-pandemic speech-language pathology services for child and adolescent mental health in Qatar
Background This paper discusses the critical importance of epidemic preparedness and innovations in digital health care by examining the transformative impact on speech-language pathology (SLP) services in a specialist outpatient child and adolescent service (CAMHS). Method This retrospective review analyzes referral data from three periods: pre-pandemic (15 March 2019–14 March 2020), pandemic (15 March 2020–14 March 2021), and post-pandemic (15 March 2021–14 March 2022). Statistical analyses assess trends in referrals and diagnoses during these periods. Feedback was also obtained from Parents of children who received virtual consultations during the pandemic. Results The results reveal an increase in the demand for SLP services during and after the pandemic, with a surge in referrals (increased from 9.7 to 12.9% when compared pre-pandemic to post-pandemic periods; Chi-Square value 3.33, P  = 0.068) for children with social communication challenges and autism spectrum disorder. Phone and video consultations were effectively adopted. Feedback from families shows a positive response (69%–98% of participants responded as strongly agreed and agreed on various items listed in feedback form specifically designed in line with the service objectives) to telehealth interventions, with many parents finding virtual consultations effective and helpful. Conclusions The study emphasizes the importance of telehealth SLP services in meeting the increasing demand for mental health interventions among children and adolescents. It suggests integrating telehealth into clinical practice beyond the pandemic and highlights the need for long-term evaluation and addressing potential barriers to access.
Use of Hyperion for Mangrove Forest Carbon Stock Assessment in Bhitarkanika Forest Reserve: A Contribution Towards Blue Carbon Initiative
Mangrove forest coastal ecosystems contain significant amount of carbon stocks and contribute to approximately 15% of the total carbon sequestered in ocean sediments. The present study aims at exploring the ability of Earth Observation EO-1 Hyperion hyperspectral sensor in estimating aboveground carbon stocks in mangrove forests. Bhitarkanika mangrove forest has been used as case study, where field measurements of the biomass and carbon were acquired simultaneously with the satellite data. The spatial distribution of most dominant mangrove species was identified using the Spectral Angle Mapper (SAM) classifier, which was implemented using the spectral profiles extracted from the hyperspectral data. SAM performed well, identifying the total area that each of the major species covers (overall kappa = 0.81). From the hyperspectral images, the NDVI (Normalized Difference Vegetation Index) and EVI (Enhanced Vegetation Index) were applied to assess the carbon stocks of the various species using machine learning (Linear, Polynomial, Logarithmic, Radial Basis Function (RBF), and Sigmoidal Function) models. NDVI and EVI is generated using covariance matrix based band selection algorithm. All the five machine learning models were tested between the carbon measured in the field sampling and the carbon estimated by the vegetation indices NDVI and EVI was satisfactory (Pearson correlation coefficient, R, of 86.98% for EVI and of 84.1% for NDVI), with the RBF model showing the best results in comparison to other models. As such, the aboveground carbon stocks for species-wise mangrove for the study area was estimated. Our study findings confirm that hyperspectral images such as those from Hyperion can be used to perform species-wise mangrove analysis and assess the carbon stocks with satisfactory accuracy.
Spatio-Temporal Monitoring of Atmospheric Pollutants Using Earth Observation Sentinel 5P TROPOMI Data: Impact of Stubble Burning a Case Study
The problems of atmospheric pollutants are causing significant concern across the globe and in India. The aggravated level of atmospheric pollutants in the surrounding environment poses serious threats to normal living conditions by deteriorating air quality and causing adverse health impacts. Pollutant concentration increases during harvesting seasons of Kharif/Rabi due to stubble burning and is aggravated by other points or mobile sources. The present study is intended to monitor the spatio-temporal variation of the major atmospheric pollutants using Sentinel-5P TROPOMI data through cloud computing. Land Use/Land Cover (LULC-categorization or classification of human activities and natural coverage on the landscape) was utilised to extract the agricultural area in the study site. It involves the cloud computing of MOD64A1 (MODIS Burned monthly gridded data) and Sentinel-5P TROPOMI (S5P Tropomi) data for major atmospheric pollutants, such as CH4, NO2, SOX, CO, aerosol, and HCHO. The burned area output provided information regarding the stubble burning period, which has seen post-harvesting agricultural residue burning after Kharif crop harvesting (i.e., rice from April to June) and Rabi crop harvesting (i.e., wheat from September to November). The long duration of stubble burning is due to variation in farmers’ harvesting and burning stubble/biomass remains in the field for successive crops. This period was used as criteria for considering the cloud computing of the Sentinel-5P TROPOMI data for atmospheric pollutants concentration in the study site. The results showed a significant increase in CH4, SO2, SOX, CO, and aerosol concentration during the AMJ months (stubble burning of Rabi crops) and OND months (stubble burning of Kharif crops) of each year. The results are validated with the ground control station data for PM2.5/PM10. and patterns of precipitation and temperature-gridded datasets. The trajectory frequency for air mass movement using the HYSPLIT model showed that the highest frequency and concentration were observed during OND months, followed by the AMJ months of each year (2018, 2019, 2020, and 2021). This study supports the role and robustness of Earth observation Sentinel-5P TROPOMI to monitor and evaluate air quality and pollutants distribution.
Clinical utility of ANA-ELISA vs ANA-immunofluorescence in connective tissue diseases
We investigated the performance of ANA-ELISA for CTDs screening and diagnosis and comparing it to the conventional ANA-IIF. ANA-ELISA is a solid-phase immune assay includes 17 ANA-targeted recombinant antigens; dsDNA, Sm-D, Rib-P, PCNA, U1-RNP (70, A, C), SS-A/Ro (52 and 60), SS-B/La, Centromere B, Scl-70, Fibrillarin, RNA Polymerase III, Jo-1, Mi-2, and PM-Scl. During the period between March till December 2016 all requests for ANA from primary, secondary, and tertiary care centers were processed with both techniques; ANA-IIF and ANA-ELISA. The electronic medical record of these patients was reviewed looking for CTD diagnosis documented by the Senior rheumatologist. SPSS 22 is used for analysis. Between March and December 2016, a total of 12,439 ANA tests were requested. 1457 patients were assessed by the rheumatologist and included in the analysis. At a cut-off ratio ≥ 1.0 for ANA-ELISA and a dilutional titre ≥ 1:80 for ANA-IIF, the sensitivity of ANA-IIF and ANA-ELISA for all CTDs were 63.3% vs 74.8% respectively. For the SLE it was 64.3% vs 76.9%, Sjogren’s Syndrome was 50% vs 76.9% respectively. The overall specificity of ANA-ELISA was 89.05%, which was slightly better than ANA-IIF 86.72%. The clinical performance of ANA-ELISA for CTDs screening showed better sensitivity and specificity as compared to the conventional ANA-IIF in our cohort.
Perceived stress and post-traumatic stress disorder symptoms among intensive care unit staff caring for severely ill coronavirus disease 2019 patients during the pandemic: a national study
Background Intensive care unit (ICU) staff have faced unprecedented challenges during the coronavirus disease 2019 (COVID-19) pandemic, which could significantly affect their mental health and well-being. The present study aimed to investigate perceived stress and post-traumatic stress disorder (PTSD) symptoms reported by ICU staff working directly with COVID-19 patients. Methods The Perceived Stress Scale was used to assess perceived stress, the PTSD Diagnostic Scale for the Diagnostic and Statistical Manual of Mental Disorders (5th edition) was used to determine PTSD symptoms, and a sociodemographic questionnaire was used to record different sociodemographic variables. Results Altogether, 124 participants (57.2% of whom were men) were included in the analysis. The majority of participants perceived working in the ICU with COVID-19 patients as moderately to severely stressful. Moreover, 71.4% of doctors and 74.4% of nurses experienced moderate-to-severe perceived stress. The staff with previous ICU experience were less likely to have a probable diagnosis of PTSD than those without previous ICU experience. Conclusions Assessing perceived stress levels and PTSD among ICU staff may enhance our understanding of COVID-19-induced mental health challenges. Specific strategies to enhance ICU staff’s mental well-being during the COVID-19 pandemic should be employed and monitored regularly. Interventions aimed at alleviating sources of anxiety in a high-stress environment may reduce the likelihood of developing PTSD.
Efficacy and cost effectiveness of intravenous ferric carboxymaltose versus iron sucrose in adult patients with iron deficiency anaemia
Iron deficiency anaemia (IDA) is a major health issues and common type of nutritional deficiency worldwide. For IDA treatment, intravenous (IV) iron is a useful therapy. To determine the efficacy and cost-effectiveness (CE) of intravenous (IV) Ferric Carboxymaltose (FCM) versus IV Iron Sucrose (IS) in treating IDA. Electronic medical record i.e. Cerner.sup.® system. IV Ferric Carboxymaltose versus IV Iron Sucrose. With regard to responses to treatment i.e., efficacy of treatment with FCM & IS in IDA patients, hemoglobin (Hgb), ferritin, and transferrin saturation (TSAT) levels were the primary outcomes. Additionally, the researchers also collected levels of iron, platelet, white blood cell (WBC), red blood cell (RBC), mean corpuscular hemoglobin (MCH), and mean corpuscular volume (MCV). The costs i.e. resources consumed (obtained from NCCCR-HMC) and the CE of FCM versus IS were the secondary outcomes. There was a significant improvement in Hgb, RBC and MCH levels in the IS group than the FCM group. The overall cost of IS therapy was significantly higher than FCM. The medication cost for FCM was approximately 6.5 times higher than IS, nonetheless, it is cheaper in terms of bed cost and nursing cost. The cost effectiveness (CE) ratio illustrated that FCM and IS were significantly different in terms of Hgb, ferritin and MCH levels. Further, Incremental Cost Effectiveness Ratio (ICER) indicated that further justifications and decisions need to be made for FCM when using Hgb, iron, TSAT, MCH and MCV levels as surrogate outcomes. Not applicable. The higher cost of FCM versus IS can be offset by savings in healthcare personnel time and bed space. Associations between maternal bacteremia during the peripartum period and early-onset neonatal sepsis: a retrospective cohort study
Introduction Early-onset neonatal sepsis (EONS) significantly impacts neonatal morbidity and mortality, with maternal bacteremia during the peripartum period being a potential risk factor. This study aims to explore the association between peripartum maternal bacteremia and EONS. Methods A retrospective cohort study at the Women's Wellness and Research Center in Doha, Qatar (2015–2019) compared women with and without bacteremia, based on blood cultures taken from up to seven days before to 48 h after delivery, examining the association with EONS. Results Among the 536 maternal blood cultures analyzed, 102 (19.0%) were positive. The most prevalent organisms were Group B streptococcus (GBS) (39.2%), followed by Escherichia coli (14.7%) and anaerobes (10.8%). Neonates from bacteremic mothers had lower birth weights (2913 ± 86 g vs. 3140 ± 745 g; MD 227.63 g; 95% CI 61.72 − 393.55; p  = 0.007), required more resuscitation (27.5% vs. 13.2%; OR 2.48; 95% CI 1.48 − 4.17; p  < 0.001), and received antibiotics for ≥ 7 days more frequently (41.2% vs. 16.6%; OR 3.51; 95% CI 2.20 − 5.62; p  < 0.001) compared to those from non-bacteremic mothers. Maternal Gram-positive (GP) organisms were more commonly isolated in term gestation (67.9%) compared to Gram-negative (GN) (22.2%) and anaerobic bacteremias (9.9%). During intrapartum, GP bacteremia was predominant (67.1%) vs. GN (21.4%) and Anaerobes (11.4%), with GN bacteremia being more common in postpartum samples. Culture-proven EONS occurred in 0.75% of the cohort, affecting 3.9% of infants from bacteremic mothers vs. none in controls (OR 2.34; 95% CI 1.27 − 4.31; p  < 0.001). Culture-negative EONS appeared in 14.7% of infants from bacteremic mothers vs. 7.8% in controls (OR 2.02; 95% CI, 1.05 − 3.88; p  = 0.03). Among 40 cases of maternal GBS bacteremia, culture-proven GBS EONS occurred in 3 neonates (7.5%), all from mothers with negative GBS screening, compared to none in the control group. A strong association was found between EONS and maternal bacteremia due to any organism (aOR 2.34; 95% CI, 1.24 − 4.41; p  = 0.009), GP bacteremia (aOR 3.66; 95% CI, 1.82 − 7.34; p  < 0.001), or GBS (aOR 5.74; 95% CI, 2.57 − 12.81; p  < 0.001). Bacteremia due to GN and Anaerobic organisms were not associated with EONS. Chorioamnionitis and antepartum fever were independent predictors for EONS associated with significant bacterial isolates. Conclusion This study underscores the significant impact of maternal GP bacteremia, particularly from GBS, on EONS. The strong association highlights the need for vigilant monitoring and interventions in pregnancies complicated by bacteremia to reduce adverse neonatal outcomes.
