Ice Inhibition for Cryopreservation: Materials, Strategies, and Challenges

Cryopreservation technology has developed into a fundamental and important supporting method for biomedical applications such as cell‐based therapeutics, tissue engineering, assisted reproduction, and vaccine storage. The formation, growth, and recrystallization of ice crystals are the major limitations in cell/tissue/organ cryopreservation, and cause fatal cryoinjury to cryopreserved biological samples. Flourishing anti‐icing materials and strategies can effectively regulate and suppress ice crystals, thus reducing ice damage and promoting cryopreservation efficiency. This review first describes the basic ice cryodamage mechanisms in the cryopreservation process. The recent development of chemical ice‐inhibition molecules, including cryoprotectant, antifreeze protein, synthetic polymer, nanomaterial, and hydrogel, and their applications in cryopreservation are summarized. The advanced engineering strategies, including trehalose delivery, cell encapsulation, and bioinspired structure design for ice inhibition, are further discussed. Furthermore, external physical field technologies used for inhibiting ice crystals in both the cooling and thawing processes are systematically reviewed. Finally, the current challenges and future perspectives in the field of ice inhibition for high‐efficiency cryopreservation are proposed. This work describes the fundamental mechanisms of ice injury during cryopreservation, and introduces the state‐of‐the‐art ice‐inhibition materials and strategies, both in the cooling and thawing processes, for high‐efficiency cryopreservation. Future perspectives and challenges are also proposed to motivate the development of cell, tissue, and organ cryopreservation and offer bright new inspiration for cryobiology.

Regional Homogeneity within the Default Mode Network in Bipolar Depression: A Resting-State Functional Magnetic Resonance Imaging Study

We sought to use a regional homogeneity (ReHo) approach as an index in resting-state functional magnetic resonance imaging (fMRI) to investigate the features of spontaneous brain activity within the default mode network (DMN) in patients suffering from bipolar depression (BD).
Twenty-six patients with BD and 26 gender-, age-, and education-matched healthy subjects participated in the resting-state fMRI scans. We compared the differences in ReHo between the two groups within the DMN and investigated the relationships between sex, age, years of education, disease duration, the Hamilton Rating Scale for Depression (HAMD) total score, and ReHo in regions with significant group differences.
Our results revealed that bipolar depressed patients had increased ReHo in the left medial frontal gyrus and left inferior parietal lobe compared to healthy controls. No correlations were found between regional ReHo values and sex, age, and clinical features within the BD group.
Our findings indicate that abnormal brain activity is mainly distributed within prefrontal-limbic circuits, which are believed to be involved in the pathophysiological mechanisms underlying bipolar depression.

Association between gaming disorder and regional homogeneity in highly involved male adult gamers: A pilot resting‐state fMRI study

Background Gaming behavior can induce cerebral changes that may be related to the neurobiological features of gaming disorder (GD). Additionally, individuals with higher levels of depression or impulsivity are more likely to experience GD. Therefore, the present pilot study explored potential neurobiological correlates of GD in the context of depression and impulsivity, after accounting for video gaming behavior. Methods Using resting‐state functional magnetic resonance imaging (fMRI), a cross‐sectional study was conducted with 35 highly involved male adult gamers to examine potential associations between GD severity and regional homogeneity (ReHo) in the entire brain. A mediation model was used to test the role of ReHo in the possible links between depression/impulsivity and GD severity. Results Individuals with greater GD severity showed increased ReHo in the right Heschl's gyrus and decreased ReHo in the right hippocampus (rHip). Furthermore, depression and impulsivity were negatively correlated with ReHo in the rHip, respectively. More importantly, ReHo in the rHip was found to mediate the associations between depression/impulsivity and GD. Conclusions These preliminary findings suggest that GD severity is related to ReHo in brain regions associated with learning/memory/mood and auditory function. Higher levels of depression or impulsivity may potentiate GD through the functional activity of the hippocampus. Our findings advance our understanding of the neurobiological differences behind GD symptoms in highly involved gamers. 35 highly involved male adult gamers participated in this pilot study. Hippocampal dysfunction related to the gaming disorder severity, depression and impulsivity. Hippocampal dysfunction may play a critical role in the development of gaming disorder.

Diffusion Tensor Imaging Tractography Reveals Disrupted White Matter Structural Connectivity Network in Healthy Adults with Insomnia Symptoms

Neuroimaging studies have revealed that insomnia is characterized by aberrant neuronal connectivity in specific brain regions, but the topological disruptions in the white matter (WM) structural connectivity networks remain largely unknown in insomnia. The current study uses diffusion tensor imaging (DTI) tractography to construct the WM structural networks and graph theory analysis to detect alterations of the brain structural networks. The study participants comprised 30 healthy subjects with insomnia symptoms (IS) and 62 healthy subjects without IS. Both the two groups showed small-world properties regarding their WM structural connectivity networks. By contrast, increased local efficiency and decreased global efficiency were identified in the IS group, indicating an insomnia-related shift in topology away from regular networks. In addition, the IS group exhibited disrupted nodal topological characteristics in regions involving the fronto-limbic and the default-mode systems. To our knowledge, this is the first study to explore the topological organization of WM structural network connectivity in insomnia. More importantly, the dysfunctions of large-scale brain systems including the fronto-limbic pathways, salience network and default-mode network in insomnia were identified, which provides new insights into the insomnia connectome. Topology-based brain network analysis thus could be a potential biomarker for IS.

Association of Galanin and Major Depressive Disorder in the Chinese Han Population

This study aimed to investigate the association of galanin (GAL) gene and the development of depression in the Chinese Han population.
A total of 700 patients with depression who met the diagnostic criteria of Diagnostic and Statistical Manual of Mental Disorders, Fourth Edition (DSM-IV) and 673 healthy controls were used in this study. Ligase detection reactions were performed on 10 selected single nucleotide polymorphism (SNP) sites in the GAL gene. A series of statistical methods were carried out to investigate the correlation between the GAL gene SNP and the patient susceptibility to depression.
The SNPs of rs694066 in the GAL gene showed a positive correlation with MDD. Compared with the healthy controls, lower frequency of G/G genotype and higher frequency of A/G genotype were observed in rs694066 in MDD patients, a lower frequency of G-allele and higher frequency of A-allele were observed in rs694066. These correlations were more pronounced in the 376 female patients and 360 female control subjects than in the 324 male patients and 313 healthy male subjects.
This study investigated the relationship between the GAL gene SNP and the susceptibility to depression in the Chinese Han population. The findings clearly indicate that the GAL gene polymorphism is closely correlated to the incidence of depression in the Chinese Han female patients.

Synergistic Ice Inhibition Effect Enhances Rapid Freezing Cryopreservation with Low Concentration of Cryoprotectants

Despite recent advances in controlling ice formation and growth, it remains a challenge to design anti‐icing materials in various fields from atmospheric to biological cryopreservation. Herein, tungsten diselenide (WSe2)‐polyvinyl pyrrolidone (PVP) nanoparticles (NPs) are synthesized through one‐step solvothermal route. The WSe2‐PVP NPs show synergetic ice regulation ability both in the freezing and thawing processes. Molecularly speaking, PVP containing amides group can form hydrogen bonds with water molecules. At a macro level, the WSe2‐PVP NPs show adsorption‐inhibition and photothermal conversation effects to synergistically restrict ice growth. Meanwhile, WSe2‐PVP NPs are for the first time used for the cryopreservation of human umbilical vein endothelial cell (HUVEC)‐laden constructs based on rapid freezing with low concentrations of cryoprotectants (CPAs), the experimental results indicate that a minimal concentration (0.5 mg mL−1) of WSe2‐PVP NPs can increase the viabilities of HUVECs in the constructs post cryopreservation (from 55.8% to 83.4%) and the cryopreserved constructs can also keep good condition in vivo within 7 days. Therefore, this work provides a novel strategy to synergistically suppress the formation and growth of the ice crystalsfor the cryopreservation of cells, tissues, or organs. Disrupted Topology of Frontostriatal Circuits Is Linked to the Severity of Insomnia

Insomnia is one of the most common health complaints, with a high prevalence of 30~50% in the general population. In particular, neuroimaging research has revealed that widespread dysfunctions in brain regions involved in hyperarousal are strongly correlated with insomnia. However, whether the topology of the intrinsic connectivity is aberrant in insomnia remains largely unknown. In this study, resting-state functional magnetic resonance imaging (rsfMRI) in conjunction with graph theoretical analysis, was used to construct functional connectivity matrices and to extract the attribute features of the small-world networks in insomnia. We examined the alterations in global and local small-world network properties of the distributed brain regions that are predominantly implicated in the frontostriatal network between 30 healthy subjects with insomnia symptoms (IS) and 62 healthy subjects without insomnia symptoms (NIS). Correlations between the small-world properties and clinical measurements were also generated to identify the differences between the two groups. Both the IS group and the NIS group exhibited a small-worldness topology. Meanwhile, the global topological properties didn't show significant difference between the two groups. By contrast, participants in the IS group showed decreased regional degree and efficiency in the left inferior frontal gyrus (IFG) compared with subjects in the NIS group. More specifically, significantly decreased nodal efficiency in the IFG was found to be negatively associated with insomnia scores, whereas the abnormal changes in nodal betweenness centrality of the right putamen were positively correlated with insomnia scores. Our findings suggested that the aberrant topology of the salience network and frontostriatal connectivity is linked to insomnia, which can serve as an important biomarker for insomnia.

The Expression of Human α-like Globin Genes in Transgenic Mice Mediated by Bacterial Artificial Chromosome

After screening a bacterial artificial chromosome of human genomic DNA library with human HS-40, ζ-, α-, and γ-globin probes, a 110-kb clone bearing the whole human α-globin gene cluster was obtained and rare restriction endonuclease mapping was performed. The bacterial artificial chromosome DNA was isolated, and transgenic mice were generated. Three founders were detected from 35 newborn mice. The copy numbers were 1, 2, and 2, and the expression of human α-globin genes in various tissues at different developmental stages in the transgenic mice was assayed. The human α-globin mRNA can be detected in bone marrow, kidney, liver, brain, but not in muscle, testis, or thymus. The human ζ-globin genes were switched off, and the α-globin genes were switched at day 11.5 in mouse embryo, indicating that developmental stage-specific expression of the α-like globin genes was properly regulated. The human α-globin mRNA ranged between 17-68% of the endogenous mouse α-globin, suggesting that the expression of human α-globin genes is integration site-dependent in transgenic mice. The ratio of human α2- and α1- globin gene expression in adult transgenic mouse is about 2.5:1 similar to the expression in human.
