Silicon-based induced resistance in maize against fall armyworm Spodoptera frugiperda (Lepidoptera: Noctuidae)
The fall armyworm ( Spodoptera frugiperda ) is a major economic pest in the United States and has recently become a significant concern in African and Asian countries. Due to its increased resistance to current management strategies, including pesticides and transgenic corn, alternative management techniques have become more necessary. Currently, silicon (Si) is being used in many pest control systems due to its ability to increase plant resistance to biotic and abiotic factors and promote plant growth. The current experiments were carried out at the College of Plant Protection, Gansu Agricultural University, Lanzhou, China, to test the effect of Si on lifetable parameters and lipase activity of fall armyworm and vegetative and physiological parameters of maize plants. Two sources of Si (silicon dioxide: SiO 2 and potassium silicate: K 2 SiO 3 ) were applied on maize plants with two application methods (foliar application and soil drenching). The experiment results revealed that foliar applications of SiO 2 and K 2 SiO 3 significantly (P≤0.05) increased mortality percentage and developmental period and decreased larval and pupal biomass of fall armyworm. Similarly, both Si sources significantly (P≤0.05) reduced lipase activity of larvae, and fecundity of adults, whereas prolonged longevity of adults. Among plant parameters, a significant increase in fresh and dry weight of shoot, stem length, chlorophyll content, and antioxidant activity was observed with foliar applications of Si. Root fresh and dry weight was significantly (P ≤ 0.05) higher in plants treated with soil drenching of SiO 2 and K 2 SiO 3 . Moreover, SiO 2 performed better for all parameters as compared to K 2 SiO 3 and control treatment. The study conclusively demonstrated a significant negative effect on various biological parameters of fall armyworm when plants were treated with Si, so it can be a promising strategy to control this pest.
Effects of sex ratio on adult fecundity, longevity and egg hatchability of Bradysia difformis Frey at different temperatures
The fungus gnat Bradysia difformis Frey not only attacks edible and medicinal fungi, it also causes the damages to leek, green onion, garlic and other vegetable crops. To confirm the effects of temperature and sex ratio on adult fecundity, longevity and egg hatchability of B. difformis, we assayed the adults survival longevity and numbers of oviposition, as well as egg hatching rate under female-male ratios of 1:1, 1:2, 1:3, 2:1 and 3:1 at 10, 15, 20, 25 and 30°C. Female fecundity and egg hatchability were increased with temperature and peaked at 25°C, which, however, were adverse to adult longevity. Furthermore, female-male ratio of 1:1, 1:2 and 1:3 seemed suitable for female fecundity, of which the sex ratio of 1:1 was the most suitable ratio for its oviposition behavior. These results provide an insight for predicting the population density of B. difformis and offer a literature reference in the effective treatment of harmful insects by controlling and changing the sex ratio artificially.
Comparison of root tolerance to drought and aphid (Myzus persicae Sulzer) resistance among different potato (Solanum tuberosum L.) cultivars
This study was conducted to determine the root system architecture and biochemical responses of three potato ( Solanum tuberosum L.) cultivars to drought and aphid ( Myzus persicae Sulzer) infestation under greenhouse conditions. A factorial experiment comprising three potato cultivars (Qingshu 9, Longshu 3, and Atlantic), two levels of water (Well watered and drought) application and aphid infestation (Aphids and no aphids) was conducted. The results show that drought stress and aphid infestation significantly increased the root-projected area, root surface area, number of root tips, and number of root forks of all cultivars, relative to their corresponding control plants. The least root projected area, root surface area, number of root tips, and number of root forks occurred on DXY under both drought and aphid infestation. Nevertheless, the greatest root projected area, root surface area, number of root tips and number of root forks occurred on QS9 plants. Moreover, increased SOD, CAT, and POD activities were observed across all cultivars, under drought and aphid stress. The highest SOD, POD, and CAT activities occurred in QS9; under drought and aphid stress, while the least SOD, POD, and CAT activities was observed in DXY. The Atlantic cultivar, which possesses a root system sensitive to water deficit, demonstrated greater resistance to aphid infestation under well-watered and drought-stressed conditions. Conversely, Qingshu 9, which possesses a root system tolerant to water deficit, was highly susceptible to aphids. This study shows that the root architectural and biochemical traits that enhance potato tolerance to drought do not necessarily correlate to a plant’s tolerance to aphids.
Trehalose and glucose levels regulate feeding behavior of the phloem-feeding insect, the pea aphid Acyrthosiphon pisum Harris
Trehalose serves multifarious roles in growth and development of insects. In this study, we demonstrated that the high trehalose diet increased the glucose content, and high glucose diet increased the glucose content but decreased the trehalose content of Acyrthosiphon pisum . RNA interference (RNAi) of trehalose-6-phosphate synthase gene ( ApTPS ) decreased while RNAi of trehalase gene ( ApTRE ) increased the trehalose and glucose contents . In the electrical penetration graph experiment, RNAi of ApTPS increased the percentage of E2 waveform and decreased the percentage of F and G waveforms. The high trehalose and glucose diets increased the percentage of E2 waveform of A. pisum red biotype. The correlation between feeding behavior and sugar contents indicated that the percentage of E1 and E2 waveforms were increased but np, C, F and G waveforms were decreased in low trehalose and glucose contents. The percentage of np, E1 and E2 waveforms were reduced but C, F and G waveforms were elevated in high trehalose and glucose contents. The results suggest that the A. pisum with high trehalose and glucose contents spent less feeding time during non-probing phase and phloem feeding phase, but had an increased feeding time during probing phase, stylet work phase and xylem feeding phase.
RNA interference of trehalose-6-phosphate synthase and trehalase genes regulates chitin metabolism in two color morphs of Acyrthosiphon pisum Harris
Trehalose-6-phosphate synthase (TPS) and trehalase (TRE) directly regulate trehalose metabolism and indirectly regulate chitin metabolism in insects. Real-time quantitative PCR (RT-qPCR) and RNA interference (RNAi) were used to detect the expressions and functions of the ApTPS and ApTRE genes. Abnormal phenotypes were found after RNAi of ApTRE in the Acyrthosiphon pisum . The molting deformities were observed in two color morphs, while wing deformities were only observed in the red morphs. The RNAi of ApTPS significantly down-regulated the expression of chitin metabolism-related genes, UDP-N-acetyglucosamine pyrophosphorylase ( ApUAP ), chitin synthase 2 ( Apchs-2 ), Chitinase 2, 5 ( ApCht2 , 5 ), endo-beta-N-acetylglucosaminidase ( ApENGase ) and chitin deacetylase ( ApCDA ) genes at 24 h and 48 h; The RNAi of ApTRE significantly down-regulated the expression of ApUAP , ApCht1 , 2 , 8 and ApCDA at 24 h and 48 h, and up-regulated the expression of glucose-6-phosphate isomerase ( ApGPI ) and Knickkopf protein ( ApKNK ) genes at 48 h. The RNAi of ApTRE and ApTPS not only altered the expression of chitin metabolism-related genes but also decreased the content of chitin. These results demonstrated that ApTPS and ApTRE can regulate the chitin metabolism, deepen our understanding of the biological functions, and provide a foundation for better understanding the molecular mechanism of insect metamorphosis.
Host plant nutrient contents influence nutrient contents in Bradysia cellarum and Bradysia impatiens
The chive maggot Bradysia cellarum and the fungus gnat B. impatiens are two primary root pests of plants, which can coexist on the same host plants and are the devastating pests on liliaceous crops and edible fungi. Their growth and development are affected by the nutrient contents of their host plants. In this study, we assessed the effects of different host plant nutrients on the nutrient contents of these two Bradysia species. The nutrients of the chive (Allium tuberosum Rottl. ex Spreng.), board bean (Vicia faba L.), lettuce (Lactuca sativa L. var. ramosa Hort.), cabbage (Brassica oleracea L.), wild cabbage (Brassica oleracea var. capitata rubra) and pepper (Capsicum annuum L.) roots were determined, and their effect on nutrient content of the two Bradysia species after feeding on the host plant for three continuous generations were evaluated. The results show that chive and B-bean contained higher levels of protein, free amino acid, soluble sugar and starch than others. As a result, the soluble sugar, fat and protein contents were significantly higher in both Bradysia species reared on chive and B-bean than on cabbage, lettuce, W-cabbage and pepper, suggesting nutritional preference of these insects. Based on our results, we concluded that the two Bradysia species displayed nutrient preference toward chive and B-bean, which provides a reference for understanding their host plant range and for control of the insect species via field crop rotations.

Comparison of Synthetic Methods and Identification of Several Artificial Antigens of Deoxynivalenol
The purpose of this experiment was to study the design and modification of hapten molecules and artificial antigen molecules of deoxynivalenol (DON), and to compare the preparation and identification methods of four artificial antigens. According to the characteristics of the molecular structure of DON, four artificial antigen coupling methods were designed—namely, N,N′-carbonyldiimidazole (CDI), 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide hydrochloride (EDC), isobutyl chloroformate (IBCF), and N-hydroxysuccinimide (NHS)—to prepare artificial antigens and detection antigens. Through ultraviolet (UV), infrared (IR), and SDS–polyacrylamide gel electrophoresis (SDS–PAGE), along with other physical and chemical identification methods and animal immunisation, the best artificial antigen coupling method was screened. The results showed that the CDI method achieved the best effect among the synthesis methods. The titre of anti-DON polyclonal antibody (pAb) produced by animal immunisation reached 1: (6.4 × 103). The half inhibitory concentration (IC50) was 47.75 ng/mL, the cross-reaction rate with 3-acetyldeoxynivalenol (3-AcDON) was slightly higher at 35.3%, and there was no cross-reaction with other compounds; therefore, four artificial antigens were successfully prepared by using the molecular structure of DON. Through identification, the CDI method was screened as the best artificial antigen synthesis method, with the highest DON pAb titre, the best sensitivity, and the strongest specificity. This will lay a solid antigenic foundation for the preparation of better anti-DON monoclonal antibodies (mAbs) in the future.
Potato (Solanum tuberosum L.) Leaf Extract Concentration Affects Performance and Oxidative Stress in Green Peach Aphids (Myzus persicae (Sulzer)
This study was conducted to determine the aphicidal effect of a leaf extract of the Atlantic potato cultivar on the performance of green peach aphids. Three concentrations of the leaf extract (100, 75, and 50% potato extract), synthetic pesticide (Beta cypermethrin 4.5%), and distilled water (control) treatments were applied in a greenhouse experiment. The results showed that the synthetic pesticide, which was used as a standard check, caused the maximum aphid mortality, followed by the 100% potato leaf extract. Compared with the other botanical treatments, the 100% extract produced low mean rates of survival, aphids’ average daily reproduction, the number of nymphs per plant, and the number of nymphs per adult. This treatment also increased the accumulation of hydrogen Peroxide (H2O2) and malondialdehyde (MDA), glutathione-s-transferase, mixed-function oxidase, and carboxylesterase content in the green peach aphid. Moreover, the 100% extract also protected the host plants against green peach aphid attacks by demonstrating higher chlorophyll content, net photosynthesis, above-ground fresh weight, and above-ground dry weight of the host plant. This study demonstrates that the highest concentration of potato (Atlantic cultivar) leaf extract (100% extract) could be used as the appropriate dosage for the control of green peach aphids on potatoes, which could greatly reduce the use of synthetic insecticides and promote ecosystem sustainability.
Effects of Tartary Buckwheat Bran Flour on Dough Properties and Quality of Steamed Bread
Steamed bread is a traditional staple food of China. Replacing wheat flour (WF) with Tartary buckwheat is expected to improve the nutritional value of steamed bread. In this study, Tartary buckwheat flour (TBF), Tartary buckwheat bran flour (TBBF), and Tartary buckwheat core flour (TBCF) were prepared, their composition and physicochemical properties were compared. It was found that TBBF had higher protein and rutin contents, so its antioxidant activity and dough rheological properties were obviously superior to those of TBF and TBCF. When TBBF was mixed with WF, its weight proportion in the blend (Wbran) had a significant effect on the dough rheological properties. When Wbran was 30%, the dough had the optimal mixing tolerance, and when Wbran exceeded 30%, it caused dilution effect, weakening the gluten network. With the increase of Wbran, the color of the steamed bread developed by the TBBF–WF blend gradually darkened and yellowed, the specific volume declined, and its hardness, gumminess, and chewiness ascended gradually. The appropriate addition of TBBF (Wbran = 10% and 30%) was beneficial to cell diameter and volume of steamed bread, but the further rise of Wbran would destroy its gas retention ability. The predicted glycemic index (pGI) of steamed bread declined significantly with the increasing Wbran.
