Dissecting L-glutamine metabolism in acute myeloid leukemia: single-cell insights and therapeutic implications

Background
Acute myeloid leukemia (AML) is a rapidly progressing blood cancer. The prognosis of AML can be challenging, emphasizing the need for ongoing research and innovative approaches to improve outcomes in individuals affected by this formidable hematologic malignancy.
Methods
In this study, we used single-cell RNA sequencing (scRNA-seq) from AML patients to investigate the impact of L-glutamine metabolism-related genes on disease progression.
Results
Our analysis revealed increased glutamine-related activity in CD34 + pre-B cells, suggesting a potential regulatory role in tumorigenesis and AML progression. Furthermore, intercellular communication analysis revealed a significant signaling pathway involving macrophage migration inhibitory factor signaling through CD74 + CD44 within CD34 + pre-B cells, which transmit signals to pre-dendritic cells and monocytes. Ligands for this pathway were predominantly expressed in stromal cells, naïve T cells, and CD34 + pre-B cells. CD74, the pertinent receptor, was predominantly detected in a variety of cellular components, including stromal cells, pre-dendritic cells, plasmacytoid dendritic cells, and hematopoietic progenitors. The study’s results provide insights into the possible interplay among these cell types and their collective contribution to the pathogenesis of AML. Development and application of co-culture for ethanol production by co-fermentation of glucose and xylose: a systematic review

This article reviews current co-culture systems for fermenting mixtures of glucose and xylose to ethanol. Thirty-five co-culture systems that ferment either synthetic glucose and xylose mixture or various biomass hydrolysates are examined. Strain combinations, fermentation modes and conditions, and fermentation performance for these co-culture systems are compared and discussed. It is noted that the combination of Pichia stipitis with Saccharomyces cerevisiae or its respiratory-deficient mutant is most commonly used. One of the best results for fermentation of glucose and xylose mixture is achieved by using co-culture of immobilized Zymomonas mobilis and free cells of P. stipitis, giving volumetric ethanol production of 1.277 g/l/h and ethanol yield of 0.49-0.50 g/g. The review discloses that, as a strategy for efficient conversion of glucose and xylose, co-culture fermentation for ethanol production from lignocellulosic biomass can increase ethanol yield and production rate, shorten fermentation time, and reduce process costs, and it is a promising technology although immature.

Mixed-Dimensional Assembly Strategy to Construct Reduced Graphene Oxide/Carbon Foams Heterostructures for Microwave Absorption, Anti-Corrosion and Thermal Insulation

HighlightsReduced graphene oxide/carbon foams (RGO/CFs) vdWs heterostructures are efficiently fabricated via a simple mixed-dimensional assembly strategy.Linkage effect of optimized impedance matching and enhanced dielectric loss abilities endows the excellent microwave absorption performances of RGO/CFs vdWs heterostructures.Multiple functions such as good corrosion resistance performances and outstanding thermal insulation capabilities can be integrated into RGO/CFs vdWs heterostructures.Considering the serious electromagnetic wave (EMW) pollution problems and complex application condition, there is a pressing need to amalgamate multiple functionalities within a single substance. However, the effective integration of diverse functions into designed EMW absorption materials still faces the huge challenges. Herein, reduced graphene oxide/carbon foams (RGO/CFs) with two-dimensional/three-dimensional (2D/3D) van der Waals (vdWs) heterostructures were meticulously engineered and synthesized utilizing an efficient methodology involving freeze-drying, immersing absorption, secondary freeze-drying, followed by carbonization treatment. Thanks to their excellent linkage effect of amplified dielectric loss and optimized impedance matching, the designed 2D/3D RGO/CFs vdWs heterostructures demonstrated commendable EMW absorption performances, achieving a broad absorption bandwidth of 6.2 GHz and a reflection loss of − 50.58 dB with the low matching thicknesses. Furthermore, the obtained 2D/3D RGO/CFs vdWs heterostructures also displayed the significant radar stealth properties, good corrosion resistance performances as well as outstanding thermal insulation capabilities, displaying the great potential in complex and variable environments. Accordingly, this work not only demonstrated a straightforward method for fabricating 2D/3D vdWs heterostructures, but also outlined a powerful mixed-dimensional assembly strategy for engineering multifunctional foams for electromagnetic protection, aerospace and other complex conditions.

Intergenerational equity based optimization model joint of water allocation and water quality management
2025
Global warming and the excessive exploitation of water resources have raised significant concerns regarding water scarcity and the degradation of aquatic ecosystems. To enhance integrated water resource management, a multi-objective optimization model integrated water allocation and water quality has been developed, in which there are trades-off between average economic benefit efficiency maximization and social welfare for long-term sustainability. In the social welfare maximization objective, an adapted Bentham-Rawls criterion is incorporated to addresses intergenerational trade-offs in high-quality water resource allocation, and the overlapping generations structure is employed to analyze the intertemporal dynamics of water pollution, thereby alleviating the conflicts between economic growth and environmental quality in the basin. Besides, total economic efficiency maximization objective is also important as it ensures the effective utilization of water resources. Using the heavily polluted Tuojiang River basin in China as a case study, the feasibility and rationality of this model have been demonstrated. Scenario analysis further validates the model's effectiveness and explores the impacts of varying control parameters. Results indicate that improving sewage treatment capacity, reducing water pollution accumulation, and accounting for the reasonable needs of future generations for high-quality water can promote the sustainable development of the basin. Compared with the previous models, the proposed optimization model is capable to increase the social welfare by 13.83% and total economic efficiency by 15.97%.

Spectral Characteristic, Storage Stability and Antioxidant Properties of Anthocyanin Extracts from Flowers of Butterfly Pea (Clitoria ternatea L.)

Anthocyanins from flowers of the butterfly pea (Clitoria ternatea L.) are promising edible blue food colorants. Food processing often faces extreme pHs and temperatures, which greatly affects the color and nutritional values of anthocyanins. This study explored the color, spectra, storage stability, and antioxidant properties of C. ternatea anthocyanin extract (CTAE) at different pHs. The color and absorption spectra of CTAEs at a pH of 0.5–13 were shown, with their underlying structures analyzed. Then, the storage stability of CTAEs were explored under a combination of pHs and temperatures. The stability of CTAE declines with the increase in temperature, and it can be stored stably for months at 4 °C. CTAEs also bear much resistance to acidic and alkaline conditions but exhibit higher thermal stability at pH 7 (blue) than at pH 0.5 (magenta) or pH 10 (blue-green), which is a great advantage in food making. Antioxidant abilities for flower extracts from the butterfly pea were high at pH 4–7, as assessed by DPPH free radical scavenging assays, and decreased sharply when the pH value exceeded 7. The above results provide a theoretical basis for the application of butterfly pea flowers and imply their great prospect in the food industry.

Coupling and interaction mechanism between green urbanization and tourism competitiveness based an empirical study in the Yellow River Basin of China

Exploring the spatial coupling relationship and interaction mechanism between green urbanization (GU) and tourism competitiveness (TC) is of great significance for promoting urban sustainable development. However, the lack of research on the interaction mechanism between GU and TC limits the formulation of effective environmental management policy and urban planning. Taking 734 counties in the Yellow River Basin (YRB) as the study area, this paper analyzes the spatial coupling relationship between GU and TC on the basis of comprehensive evaluation of GU and TC. Then, the interactive mechanism between GU and TC is systematically discussed, and the synergistic development strategy of the two is proposed. The results show that the GU level presents a multicore circle structure, with provincial capitals, prefecture-level urban districts and economically developed counties in east-central regions as high-value centers. The TC at county scale presents a multi-center spatial structure. Additionally, there is a significant positive spatial coupling between GU and TC in the YRB. The analysis further reveals that green urbanization level, social progress, population development, infrastructure construction, economic development quality, and eco-environmental protection has a observably influence on TC. Tourism competitiveness, service competitiveness, location competitiveness, resource competitiveness, market competitiveness, environmental influence, and talent competitiveness has a observably influence on GU. TC can promote GU, and the improvement of green urbanization level can support the development of tourism competitiveness. According to the spatial zoning method, 734 counties are divided into 6 categories, and the coordinated development strategy of GU and TC for each type of district is proposed.

Nickel (Ni) contaminated soil is a persistent risk to plant growth and production worldwide. Therefore, to explore the Ni toxicity levels in sweetpotato production areas, we investigated the influence of different Ni treatments (0, 7.5, 15, 30, and 60 mg L -1 ) for 15 days on phenotype, Ni uptake, relative water content, gas exchange, photosynthetic pigments, oxidative stress, osmolytes, antioxidants, and enzymes of sweetpotato plants. The results presented that Ni at higher levels (30 and 60 mg L -1 ) substantially reduced growth, biomass, and root morphological traits. The Pearson correlation analysis suggested that Ni toxicity causes oxidative injuries as persistent augmentation of hydrogen peroxide (H 2 O 2 ) and malonaldehyde (MDA) and reduced RWC, gas exchange, and photosynthetic pigment. Furthermore, this study revealed that sweetpotato could tolerate moderate Ni treatment (up to 15 mg L -1 ) by reducing oxidative stress. The results also indicated that the increase in the activities of mentioned osmolytes, antioxidants, and enzymes is not sufficient to overcome the higher Ni toxicity. Based on these results, we suggest using low Ni-contaminated soil for better growth of sweetpotato and also could be used as a phytoremediator in moderate Ni-contaminated soil.

Although pediatric Burkitt lymphoma is considered curable, some patients still have a poor prognosis. Lack of early or complete response is an adverse prognostic factor. Therefore, improving patients’ sensitivity to initial treatment and achieving complete remission early are crucial for enhancing efficacy. Herein, we report two cases of children with newly diagnosed clinical stage IV Burkitt lymphoma who achieved early complete metabolic remission after only two cycles of treatment with polatuzumab vedotin plus rituximab combined with chemotherapy. Notably, this represents the first reported application of polatuzumab vedotin in pediatric patients with newly diagnosed Burkitt lymphoma.

Real-Time Detection and Short-Term Prediction of Blast Furnace Burden Level Based on Space-Time Fusion Features

Real-time, continuous and accurate blast furnace burden level information is of great significance for controlling the charging process, ensuring a smooth operation of a blast furnace, reducing energy consumption and emissions and improving blast furnace output. However, the burden level information measured by conventional mechanical stock rods and radar probes exhibit problems of weak anti-interference ability, large fluctuations in accuracy, poor stability and discontinuity. Therefore, a space-time fusion prediction and detection method of burden level based on a long-term focus memory network (LFMN) and an efficient structure self-tuning RBF neural network (ESST-RBFNN) is proposed. First, the space dimensional features are extracted by the space regression model based on radar data. Then, the LFMN is designed to predict the burden level and extract the time dimensional features. Finally, the ESST-RBFNN based on a proposed fast eigenvector space clustering algorithm (ESC) is constructed to obtain reliable and continuous burden level information with high accuracy. Both the simulation results and industrial verification indicate that the proposed method can provide real-time and continuous burden level information in real-time, which has great practical value for industrial production.
