YouTube/ Bilibili/ TikTok videos as sources of medical information on laryngeal carcinoma: cross-sectional content analysis study

Background
YouTube, a widely recognized global video platform, is inaccessible in China, whereas Bilibili and TikTok are popular platforms for long and short videos, respectively. There are many videos related to laryngeal carcinoma on these platforms. This study aims to identify upload sources, contents, and feature information of these videos on YouTube, Bilibili, and TikTok, and further evaluate the video quality.
Methods
On January 1, 2024, we searched the top 100 videos by default sort order (300 videos in total) with the terms “laryngeal carcinoma” and “throat cancer” on YouTube, “喉癌” on Bilibili and TikTok. Videos were screened for relevance and similarity. Video characteristics were documented, and quality was assessed by using the Patient Education Materials Assessment Tool (PEMAT), Video Information and Quality Index (VIQI), Global Quality Score (GQS), and modified DISCERN (mDISCERN).
Results
The analysis included 99 YouTube videos, 76 from Bilibili, and 73 from TikTok. Median video lengths were 193 s (YouTube), 136 s (Bilibili), and 42 s (TikTok). TikTok videos demonstrated higher audience interaction. Bilibili had the lowest ratio of original contents (69.7%). Treatment was the most popular topic on YouTube and Bilibili, while that was the prognosis on TikTok. Solo narration was the most common video style across all platforms. Video uploaders were predominantly non-profit organizations (YouTube), self-media (Bilibili), and doctors (TikTok), with TikTok authors having the highest certification rate (83.3%). Video quality, assessed using PEMAT, VIQI, GQS, and mDISCERN, varied across platforms, with YouTube generally showing the highest scores. Videos from professional authors performed better than videos from non-professionals based on the GQS and mDISCERN scores. Spearman correlation analysis showed no strong relationships between the video quality and the audience interaction.
Conclusions
Videos on social media platforms can help the public learn about the knowledge of laryngeal cancer to some extent. TikTok achieves the best flow, but videos on YouTube are of the best quality. However, the video quality across all platforms still needs enhancement. We need more professional uploaders to ameliorate the video quality related to laryngeal carcinoma. Content creators also should be aware of the certification, the originality, and the style of video shooting. As for the platforms, refining the algorithm will allow users to receive more high-quality videos.

A Wearable Assistive Device for Blind Pedestrians Using Real-Time Object Detection and Tactile Presentation

Nowadays, improving the traffic safety of visually impaired people is a topic of widespread concern. To help avoid the risks and hazards of road traffic in their daily life, we propose a wearable device using object detection techniques and a novel tactile display made from shape-memory alloy (SMA) actuators. After detecting obstacles in real-time, the tactile display attached to a user’s hands presents different tactile sensations to show the position of the obstacles. To implement the computation-consuming object detection algorithm in a low-memory mobile device, we introduced a slimming compression method to reduce 90% of the redundant structures of the neural network. We also designed a particular driving circuit board that can efficiently drive the SMA-based tactile displays. In addition, we also conducted several experiments to verify our wearable assistive device’s performance. The results of the experiments showed that the subject was able to recognize the left or right position of a stationary obstacle with 96% accuracy and also successfully avoided collisions with moving obstacles by using the wearable assistive device.

Association between Obesity and Puberty Timing: A Systematic Review and Meta-Analysis

This systematic review and meta-analysis examined the associations between obesity and puberty timing based on scientific evidence. Eight electronic databases were searched up to February 2017 for eligible studies, and two reviewers screened the articles and extracted the data independently. A total of 11 cohort studies with 4841 subjects met the inclusion criteria. Compared with the group of normal-weight girls, the obese group had more girls with menarche (RR: 1.87, 95% CI: 1.59–2.19, 2 studies). The number of girls with early puberty was significantly higher in the obese group than the normal weight group (RR: 2.44, 95% CI: 1.32–4.52, 5 studies). However, no differences were detected between girls who were obese or normal weight at age of menarche (WMD: −0.53 years, 95% CI: −1.24–0.19, 2 studies). There is no consistent result in the relationship between obesity and timing of pubertal onset in boys. Obesity may contribute to early onset of puberty in girls, while in boys, there is insufficient data. Given the limited number of cohort studies included in this meta-analysis, high-quality studies with strong markers of puberty onset, as well as standardized criteria for defining obesity are needed.

Organic Amendments promote saline-alkali soil desalinization and enhance maize growth

Secondary soil salinization in arid and semi-arid regions is a serious problem that severely hampers local agricultural productivity and poses a threat to the long-term sustainability of food production. the utilization of organic soil amendments presents a promising approach to mitigate yield losses and promote sustainable agricultural production in saline-alkali soil. In this study, we established four distinct treatments, chemical fertilizer (CK), humic acid with chemical fertilizer (HA), carboxymethyl cellulose with chemical fertilizer (CMC), and amino acid with chemical fertilizer (AA), to elucidate their respective impacts on the reclamation of saline soil and the growth of maize. The findings of our study reveal notable variations in desalination rates within the 0-40 cm soil layer due to the application of distinct soil amendments, ranging from 11.66% to 37.17%. Moreover, application of amendments significantly increased the percentage of soil macro-aggregates as compared to the CK treatment. Furthermore, HA and AA treatments significantly augmented soil nutrient content (HA: 48.07%; AA: 39.50%), net photosynthetic rate (HA: 12.68%; AA: 13.94%), intercellular CO 2 concentration (HA: 57.20%; AA: 35.93%) and maize yield (HA:18.32%; AA:16.81%). Correlation analysis and structural equation modeling unveiled diverse mechanisms of yield enhancement for HA, CMC, and AA treatments. HA enhanced yield by increasing organic matter and promoting soil aggregate formation, CMC improved soil water content and facilitated salt leaching due to its excellent water-holding properties, while AA increased yield by elevating soil organic matter and effective nitrogen content. Among the array of soil amendment materials scrutinized, HA treatment emerged as the most promising agent for enhancing soil conditions and is thus recommended as the preferred choice for treating local saline soils.

Integrative analyses reveal a long noncoding RNA-mediated sponge regulatory network in prostate cancer
Mounting evidence suggests that long noncoding RNAs (lncRNAs) can function as microRNA sponges and compete for microRNA binding to protein-coding transcripts. However, the prevalence, functional significance and targets of lncRNA-mediated sponge regulation of cancer are mostly unknown. Here we identify a lncRNA-mediated sponge regulatory network that affects the expression of many protein-coding prostate cancer driver genes, by integrating analysis of sequence features and gene expression profiles of both lncRNAs and protein-coding genes in tumours. We confirm the tumour-suppressive function of two lncRNAs (TUG1 and CTB-89H12.4) and their regulation of PTEN expression in prostate cancer. Surprisingly, one of the two lncRNAs, TUG1, was previously known for its function in polycomb repressive complex 2 (PRC2)-mediated transcriptional regulation, suggesting its sub-cellular localization-dependent function. Our findings not only suggest an important role of lncRNA-mediated sponge regulation in cancer, but also underscore the critical influence of cytoplasmic localization on the efficacy of a sponge lncRNA.

Prediction of HER2 expression in breast cancer by combining PET/CT radiomic analysis and machine learning

BackgroundHuman epidermal growth factor receptor 2 (HER2) expression status determination significantly contributes to HER2-targeted therapy in breast cancer (BC). The purpose of this study was to evaluate the role of radiomics and machine learning based on PET/CT images in HER2 status prediction, and to identify the most effective combination of machine learning model and radiomic features.MethodsA total of 217 BC patients who underwent PET/CT examination were involved in the study and randomly divided into a training set (n = 151) and a testing set (n = 66). For all four models, the model parameters were determined using a threefold cross-validation in the training set. Each model’s performance was evaluated on the independent testing set using the receiver operating characteristic (ROC) curve, and AUC was calculated to get a quantified performance measurement of each model.ResultsAmong the four developed machine learning models, the XGBoost model outperformed other machine learning models in HER2 status prediction. Furthermore, compared to the XGBoost model based on PET alone or CT alone radiomic features, the predictive power for HER2 status by using XGBoost model based on PET/CTmean or PET/CTconcat radiomic fusion features was dramatically improved with an AUC of 0.76 (95% confidence interval [CI] 0.69–0.83) and 0.72 (0.65–0.80), respectively.ConclusionsThe established machine learning classifier based on PET/CT radiomic features is potentially predictive of HER2 status in BC.

Genome-wide CRISPR screen identifies HNRNPL as a prostate cancer dependency regulating RNA splicing

Alternative RNA splicing plays an important role in cancer. To determine which factors involved in RNA processing are essential in prostate cancer, we performed a genome-wide CRISPR/Cas9 knockout screen to identify the genes that are required for prostate cancer growth. Functional annotation defined a set of essential spliceosome and RNA binding protein (RBP) genes, including most notably heterogeneous nuclear ribonucleoprotein L (HNRNPL). We defined the HNRNPL-bound RNA landscape by RNA immunoprecipitation coupled with next-generation sequencing and linked these RBP–RNA interactions to changes in RNA processing. HNRNPL directly regulates the alternative splicing of a set of RNAs, including those encoding the androgen receptor, the key lineage-specific prostate cancer oncogene. HNRNPL also regulates circular RNA formation via back splicing. Importantly, both HNRNPL and its RNA targets are aberrantly expressed in human prostate tumors, supporting their clinical relevance. Collectively, our data reveal HNRNPL and its RNA clients as players in prostate cancer growth and potential therapeutic targets.

The androgen receptor (AR) is a key factor that regulates the behavior and fate of prostate cancer cells. The AR-regulated network is activated when AR binds enhancer elements and modulates specific enhancer–promoter looping. Kallikrein-related peptidase 3 (KLK3), which codes for prostate-specific antigen (PSA), is a well-known AR-regulated gene and its upstream enhancers produce bidirectional enhancer RNAs (eRNAs), termed KLK3e. Here, we demonstrate that KLK3e facilitates the spatial interaction of the KLK3 enhancer and the KLK2 promoter and enhances long-distance KLK2 transcriptional activation. KLK3e carries the core enhancer element derived from the androgen response element III (ARE III), which is required for the interaction of AR and Mediator 1 (Med1). Furthermore, we show that KLK3e processes RNA-dependent enhancer activity depending on the integrity of core enhancer elements. The transcription of KLK3e was detectable and its expression is significantly correlated with KLK3 (R ² = 0.6213, P < 5 × 10 ⁻¹¹) and KLK2 (R ² = 0.5893, P < 5 × 10 ⁻¹⁰) in human prostate tissues. Interestingly, RNAi silencing of KLK3e resulted in a modest negative effect on prostate cancer cell proliferation. Accordingly, we report that an androgen-induced eRNA scaffolds the AR-associated protein complex that modulates chromosomal architecture and selectively enhances AR-dependent gene expression.

CRISPR/Cas9 screen uncovers functional translation of cryptic lncRNA-encoded open reading frames in human cancer

Emerging evidence suggests that cryptic translation within long noncoding RNAs (lncRNAs) may produce novel proteins with important developmental/physiological functions. However, the role of this cryptic translation in complex diseases (e.g., cancer) remains elusive. Here, we applied an integrative strategy combining ribosome profiling and CRISPR/Cas9 screening with large-scale analysis of molecular/clinical data for breast cancer (BC) and identified estrogen receptor α-positive (ER+) BC dependency on the cryptic ORFs encoded by lncRNA genes that were upregulated in luminal tumors. We confirmed the in vivo tumor-promoting function of an unannotated protein, GATA3-interacting cryptic protein (GT3-INCP) encoded by LINC00992, the expression of which was associated with poor prognosis in luminal tumors. GTE-INCP was upregulated by estrogen/ER and regulated estrogen-dependent cell growth. Mechanistically, GT3-INCP interacted with GATA3, a master transcription factor key to mammary gland development/BC cell proliferation, and coregulated a gene expression program that involved many BC susceptibility/risk genes and impacted estrogen response/cell proliferation. GT3-INCP/GATA3 bound to common cis regulatory elements and upregulated the expression of the tumor-promoting and estrogen-regulated BC susceptibility/risk genes MYB and PDZK1. Our study indicates that cryptic lncRNA-encoded proteins can be an important integrated component of the master transcriptional regulatory network driving aberrant transcription in cancer, and suggests that the \"hidden\" lncRNA-encoded proteome might be a new space for therapeutic target discovery.
