Histidine modulates amyloid-like assembly of peptide nanomaterials and confers enzyme-like activity
Amyloid-like assembly is not only associated with pathological events, but also leads to the development of novel nanomaterials with unique properties. Herein, using Fmoc diphenylalanine peptide (Fmoc–F–F) as a minimalistic model, we found that histidine can modulate the assembly behavior of Fmoc–F–F and induce enzyme-like catalysis. Specifically, the presence of histidine rearranges the β structure of Fmoc–F–F to assemble nanofilaments, resulting in the formation of active site to mimic peroxidase-like activity that catalyzes ROS generation. A similar catalytic property is also observed in Aβ assembled filaments, which is correlated with the spatial proximity between intermolecular histidine and F-F. Notably, the assembled Aβ filaments are able to induce cellular ROS elevation and damage neuron cells, providing an insight into the pathological relationship between Aβ aggregation and Alzheimer’s disease. These findings highlight the potential of histidine as a modulator in amyloid-like assembly of peptide nanomaterials exerting enzyme-like catalysis. In this work, the authors report that Histidine residues play a critical role in modulating amyloid-like assembly and building active sites for Fmoc–F–F and Aβ aggregates. Aβ1-42 filaments were found to perform peroxidase-like activity to enhance oxidative stress, which might also be ascribed to the interaction mode of His and F-F.
Stable peptide-assembled nanozyme mimicking dual antifungal actions
Natural antimicrobial peptides (AMPs) and enzymes (AMEs) are promising non-antibiotic candidates against antimicrobial resistance but suffer from low efficiency and poor stability. Here, we develop peptide nanozymes which mimic the mode of action of AMPs and AMEs through de novo design and peptide assembly. Through modelling a minimal building block of IHIHICI is proposed by combining critical amino acids in AMPs and AMEs and hydrophobic isoleucine to conduct assembly. Experimental validations reveal that IHIHICI assemble into helical β-sheet nanotubes with acetate modulation and perform phospholipase C-like and peroxidase-like activities with Ni coordination, demonstrating high thermostability and resistance to enzymatic degradation. The assembled nanotubes demonstrate cascade antifungal actions including outer mannan docking, wall disruption, lipid peroxidation and subsequent ferroptotic death, synergistically killing >90% Candida albicans within 10 min on disinfection pad. These findings demonstrate an effective de novo design strategy for developing materials with multi-antimicrobial mode of actions. Natural antimicrobial peptides and enzymes are good candidates for application but suffer from low stability. Here, the authors report on biomimetic self-assembling peptides which mimic both antimicrobial peptide and enzyme functionality, demonstrating application against fungal infection.
Cadmium accumulation is enhanced by ammonium compared to nitrate in two hyperaccumulators, without affecting speciation
Nitrogen fertilization could improve the efficiency of Cd phytoextraction in contaminated soil and thus shorten the remediation time. However, limited information is available on the effect of N form on Cd phytoextraction and associated mechanisms in plants. This study examined the effect of N form on Cd accumulation, translocation, and speciation in Carpobrotus rossii and Solanum nigrum. Plants were grown in nutrient solution with 5–15 μM Cd in the presence of 1000 μM NH₄⁺ or NO₃⁻. Plant growth and Cd uptake were measured, and Cd speciation was analyzed using synchrotron-based X-ray absorption spectroscopy. Shoot Cd accumulation was 30% greater with NH₄⁺ than NO₃⁻ supply. Carpobrotus rossii accumulated three times more Cd than S. nigrum. However, Cd speciation in the plants was not influenced by N form, but it did vary with species and tissues. In C. rossii, up to 91% of Cd was bound to S-containing ligands in all tissues except the xylem sap where 87–95% were Cd-OH complexes. Furthermore, the proportion of Cd-S in shoots was substantially lower in S. nigrum (44–69%) than in C. rossii (60–91%). It is concluded that the application of NH₄⁺ (instead of NO₃⁻) increased shoot Cd accumulation by increasing uptake and translocation, rather than changing Cd speciation, and is potentially an effective approach for increasing Cd phytoextraction.
Effect of Urban Built-Up Area Expansion on the Urban Heat Islands in Different Seasons in 34 Metropolitan Regions across China
The urban heat island (UHI) refers to the land surface temperature (LST) difference between urban areas and their undeveloped or underdeveloped surroundings. It is a measure of the thermal influence of the urban built-up area expansion (UBAE), a topic that has been extensively studied. However, the impact of UBAE on the LST differences between urban areas and rural areas (UHIU−R) and between urban areas and emerging urban areas (UHIU−S) in different seasons has seldom been investigated. Here, the UHIU−S and UHIU−R in 34 major metropolitan regions across China, and their spatiotemporal variations based on long-term space-borne observations during the period 2001–2020 were analyzed. The UBAE quantified by the difference in landscape metrics of built-up areas between 2020 and 2000 and their impact on UHI was further analyzed. The UBAE is impacted by the level of economic development and topography. The UBAE of cities located in more developed regions was more significant than that in less developed regions. Coastal cities experienced the most obvious UBAE, followed by plain and hilly cities. The UBAE in mountainous regions was the weakest. On an annual basis, UHIU−R was larger than UHIU−S, decreasing more slowly with UBAE than UHIU−S. In different seasons, the UHIU−S and UHIU−R were larger, more clearly varying temporally with UBAE in summer than in winter, and their temporal variations were significantly correlated with UBAE in summer but not in winter. The seasonal difference in UHIU−R was larger than that of UHIU−S. Both the UHIU−S and UHIU−R in coastal cities were the lowest in summer, decreasing the fastest with UBAE, while those in mountain cities decreased the slowest. The change in the density of built-up lands was the primary driver affecting the temporal variations in UHIU−S and UHIU−R during UBAE, followed by changes in proportion and shape, while the impact of the speed of expansion was the smallest, all of which were more obvious in summer than in winter. The decreased density of built-up lands can reduce UHI. These findings provide a new perspective for a deeper understanding of the effect of urban expansion on LST in different seasons.
Emission factors and light absorption properties of brown carbon from household coal combustion in China
Brown carbon (BrC) draws increasing attention due to its effects on climate and other environmental factors. In China, household coal burned for heating and cooking purposes releases huge amounts of carbonaceous particles every year; however, BrC emissions have rarely been estimated in a persuasive manner due to the unavailable emission characteristics. Here, seven coals jointly covering geological maturity from low to high were burned in four typical stoves as both chunk and briquette styles. The optical integrating sphere (IS) method was applied to measure the emission factors (EFs) of BrC and black carbon (BC) via an iterative process using the different spectral dependence of light absorption for BrC and BC and using humic acid sodium salt (HASS) and carbon black (CarB) as reference materials. The following results have been found: (i) the average EFs of BrC for anthracite coal chunks and briquettes are 1.08 ± 0.80 and 1.52 ± 0.16 g kg−1, respectively, and those for bituminous coal chunks and briquettes are 8.59 ± 2.70 and 4.01 ± 2.19 g kg−1, respectively, reflecting a more significant decline in BrC EFs for bituminous coals than for anthracites due to briquetting. (ii) The BrC EF peaks at the middle of coal's geological maturity, displaying a bell-shaped curve between EF and volatile matter (Vdaf). (iii) The calculated BrC emissions from China's residential coal burning amounted to 592 Gg (1 Gg  =  109 g) in 2013, which is nearly half of China's total BC emissions. (iv) The absorption Ångström exponents (AAEs) of all coal briquettes are higher than those of coal chunks, indicating that the measure of coal briquetting increases the BrC ∕ BC emission ratio and thus offsets some of the climate cooling effect of briquetting. (v) In the scenario of current household coal burning in China, solar light absorption by BrC (350–850 nm in this study) accounts for more than a quarter (0.265) of the total absorption. This implies the significance of BrC to climate modeling.
The 30 m land cover dataset for capturing land cover changes induced by ecological restoration from 1990 to 2022 on the Chinese Loess Plateau
Continuous time-series of land cover is critical for attributing runoff, sediment and carbon changes on the Chinese Loess Plateau (CLP). However, current land cover products with annal temporal resolution lack spatial identification accuracy, particularly in capturing authentic changes of cropland, forest and grassland. To address these issues, a 30 m annual land cover dataset was proposed by the Yellow River Conservancy Commission (YRCC_LPLC) for the CLP from 1990 to 2022. Different levels of land cover were classified using different combinations of spectral, monthly and annual temporal and topographic features and Random Forest classifier. Compared to other land cover products (45.64%–73.38%), the accuracy of YRCC_LPLC has a better performance with an overall accuracy of 85.16%. The YRCC_LPLC is capable of capturing not only the explicit spatial variation but also the change direction and change time of land cover, especially for the most critical conversion of cropland into forest and grassland induced by implementation of Grain to Green Program on the CLP.
Spatial–temporal patterns of inorganic nitrogen air concentrations and deposition in eastern China
Five-year (2011–2015) measurements of gaseous NH3, NO2, and HNO3 and particulate NH4+ and NO3− in air and/or precipitation were conducted at 27 sites in the Nationwide Nitrogen Deposition Monitoring Network (NNDMN) to better understand spatial and temporal (seasonal and annual) characteristics of reactive nitrogen (Nr) concentrations and deposition in eastern China. Our observations reveal annual average concentrations (16.4–32.6 µg N m−3), dry deposition fluxes (15.8–31.7 kg N ha−1 yr−1), and wet/bulk deposition fluxes (18.4–28.0 kg N ha−1 yr−1) based on land use, ranked as urban > rural > background sites. Annual concentrations and dry deposition fluxes of each Nr species in air were comparable at urban and background sites in northern and southern regions, but were significantly higher at northern rural sites. These results, together with good agreement between spatial distributions of NH3 and NO2 concentrations determined from ground measurements and satellite observations, demonstrate that atmospheric Nr pollution is heavier in the northern region than in the southern region. No significant inter-annual trends were found in the annual Nr dry and wet/bulk N deposition at almost all of the selected sites. A lack of significant changes in annual averages between the 2013–2015 and 2011–2012 periods for all land use types suggests that any effects of current emission controls are not yet apparent in Nr pollution and deposition in the region. Ambient concentrations of total Nr exhibited non-significant seasonal variation at all land use types, although significant seasonal variations were found for individual Nr species (e.g. NH3, NO2, and pNO3−) in most cases. In contrast, dry deposition of total Nr exhibited a consistent and significant seasonal variation at all land use types, with the highest fluxes in summer and the lowest in winter. Based on sensitivity tests by the GEOS-Chem model, we found that NH3 emissions from fertilizer use (including chemical and organic fertilizers) were the largest contributor (36 %) to total inorganic Nr deposition over eastern China. Our results not only improve the understanding of spatial–temporal variations of Nr concentrations and deposition in this pollution hotspot, but also provide useful information for policy-makers that mitigation of NH3 emissions should be a priority to tackle serious N deposition in eastern China.
Synthesis of Rice Husk-Based MCM-41 for Removal of Aflatoxin B1 from Peanut Oil
Edible oils, especially peanut oil, usually contain aflatoxin B1 (AFB1) at extremely high concentrations. This study focused on the synthesis of rice husk-based mesoporous silica (MCM-41) for the removal of AFB1 from peanut oil. MCM-41 was characterized by X-ray diffraction, N2 physisorption, and transmission electron microscope. MCM-41 was shown to have ordered channels with high specific surface area (1246 m2/g), pore volume (1.75 cm3/g), and pore diameter (3.11 nm). Under the optimal concentration of 1.0 mg/mL of the adsorbent dose, the adsorption behavior of MCM-41, natural montmorillonite (MONT), and commercial activated carbon (CA) for AFB1 were compared. The adsorption of AFB1 in peanut oil onto the three adsorbents was slower compared to that of AFB1 in an aqueous solution. In addition, the pseudo-second-order kinetic model better fit the adsorption kinetics of AFB1, while the adsorption mechanism followed the Langmuir adsorption isotherm on the three adsorbents. The calculated maximum adsorbed amounts of AFB1 on MONT, MCM-41, and CA were 199.41, 215.93, and 248.93 ng/mg, respectively. These results suggested that MCM-41 without modification could meet market demand and could be considered a good candidate for the removal of AFB1 from peanut oil. This study provides insights that could prove to be of economic and practical value.
Synchrotron-Based Techniques Shed Light on Mechanisms of Plant Sensitivity and Tolerance to High Manganese in the Root Environment
Plant species differ in response to high available manganese (Mn), but the mechanisms of sensitivity and tolerance are poorly understood. In solution culture, greater than or equal to 30 μM Mn decreased the growth of soybean (Glycine max), but white lupin (Lupinus albus), narrow-leafed lupin (Lupin angustifolius), and sunflower (Helianthus annuus) grew well at 100 μM Mn. Differences in species' tolerance to high Mn could not be explained simply by differences in root, stem, or leaf Mn status, being 8.6, 17.1, 6.8, and 9.5 mmol kg⁻¹ leaf fresh mass at 100 μM Mn. Furthermore, x-ray absorption near edge structure analyses identified the predominance of Mn(II), bound mostly to malate or citrate, in roots and stems of all four species. Rather, differences in tolerance were due to variations in Mn distribution and speciation within leaves. In Mn-sensitive soybean, in situ analysis of fresh leaves using x-ray fluorescence microscopy combined with x-ray absorption near edge structure showed high Mn in the veins, and manganite [Mn(III)] accumulated in necrotic lesions apparently through low Mn sequestration in vacuoles or other vesicles. In the two lupin species, most Mn accumulated in vacuoles as either soluble Mn(II) malate or citrate. In sunflower, Mn was sequestered as manganite at the base of nonglandular trichomes. Hence, tolerance to high Mn was ascribed to effective sinks for Mn in leaves, as Mn(II) within vacuoles or through oxidation of Mn(II) to Mn(III) in trichomes. These two mechanisms prevented Mn accumulation in the cytoplasm and apoplast, thereby ensuring tolerance to high Mn in the root environment.
New variants and genotype-phenotype correlation of PPP3CA-related developmental and epileptic encephalopathy
To explore the genotypic spectrum and refine the genotype-phenotype correlation of -related developmental and epileptic encephalopathy (DEE). whole-exome sequencing or whole-genome sequencing was performed to all patients. Clinical data of 15 epilepsy patients in current study and 21 epilepsy patients from published studies were collected and analyzed. In this study, 15 patients were identified with 13 variants. Among these, seven frameshift variants and one gene inversion between intron 11 and intron 13 (including exons 12 and 13) were novel. 80% of patients experiencing seizure onset before the age of one. The seizure types observed included epileptic spasms (93.3%), tonic seizures (46.7%), myoclonic seizures (46.7%), focal seizures (40.0%), atypical absence seizures (13.3%), generalized tonic-clonic seizures (6.7%) and myoclonic atonic seizures (6.7%). All patients exhibited global developmental delay. MRI abnormalities were noticed in 9 patients, including widened subarachnoid space, bilateral ventricular width, poor myelination of white matter, and dysplasia of the corpus callosum. 80% specifically diagnosed with infantile epileptic spasms syndrome (IESS). When combining data from this study and published studies, 66.7% of patients experienced seizure onset before the age of one, and 77.8% were diagnosed with IESS. In patients with variants located in the catalytic domain (CD), 45.4% patients exhibited multiple seizure types, while 45.4% patients presented only with epileptic spasms. In contrast, among patients with variants in regulatory domain (RD), 87% had multiple seizure types and only 8.7% had epileptic spasms alone. Additionally, 45.5% of patients with CD variants had comorbid autism spectrum disorders, compared to 13% patients with RD variants. Recurrent variants included p.His92Arg, p.Asp234Glu, p.Glu282Lys, and p.Ser419Asnfs*31. This study is the first to report a gene inversion in -related DEE. Patients with only epileptic spasms were more prevalent in those with CD variants, compared to those with RD variants. Conversely, patients with multiple seizure types were more common among those with RD variants. The most frequently diagnosed epileptic syndrome was IESS. Additionally, comorbid ASD were more commonly observed in patients with CD variants than in those with RD variants.
