TGF-β inhibitor Smad7 regulates dendritic cell-induced autoimmunity
TGF-β is an anti-inflammatory cytokine whose signaling is negatively controlled by Smad7. Previously, we established a role for Smad7 in the generation of autoreactive T cells; however, the function of Smad7 in dendritic cells (DCs) remains elusive. Here, we demonstrate that DC-specific Smad7 deficiency resulted in elevated expression of the transcription factors Batf3 and IRF8, leading to increased frequencies of CD8⁺CD103⁺ DCs in the spleen. Furthermore, Smad7-deficient DCs expressed higher levels of indoleamine 2,3-dioxygenase (IDO), an enzyme associated with tolerance induction. Mice devoid of Smad7 specifically in DCs are resistant to the development of experimental autoimmune encephalomyelitis (EAE) as a result of an increase of protective regulatory T cells (Tregs) and reduction of encephalitogenic effector T cells in the central nervous system. In agreement, inhibition of IDO activity or depletion of Tregs restored disease susceptibility. Intriguingly, when Smad7-deficient DCs also lacked the IFN-γ receptor, the mice regained susceptibility to EAE, demonstrating that IFN-γ signaling in DCs mediates their tolerogenic function. Our data indicate that Smad7 expression governs splenic DC subset differentiation and is critical for the promotion of their efficient function in immunity.
Increased First Pass Success with C-MAC Videolaryngoscopy in Prehospital Endotracheal Intubation—A Randomized Controlled Trial
Endotracheal intubation (ETI) with direct view laryngoscopy (DL) is the gold standard for airway management. Videolaryngoscopy (VL) can improve glottis visualization, thus facilitating ETI. The aim of this monocentric, randomized, prospective study on a physician staffed German air ambulance is to compare DL and VL for ETI in terms of number of attempts and time as well as visualization of the glottis in a prehospital setting in a physician-based rescue system in adult patients. A power analysis was performed à priori. We used consecutive on-scene randomization with a sealed envelope system for the DL and VL-group. Successful ETI with first pass success was significantly more frequent with VL than DL and three seconds faster. The percentage of glottis opening and the Cormack & Lehane classification were significantly better with VL than DL. Regarding improved first pass success in ETI with the VL, we would recommend the use of VL for prehospital airway management in physician-based rescue systems.

In March 2019, German-speaking scientists and scholars calling themselves Scientists for Future, published a statement in support of the youth protesters in Germany, Austria, and Switzerland (Fridays for Future, Klimastreik/Climate Strike), verifying the scientific evidence that the youth protestors refer to. In this article, they provide the full text of the statement, including the list of supporting facts (in both English and German) as well as an analysis of the results and impacts of the statement. Furthermore, they reflect on the challenges for scientists and scholars who feel a dual responsibility: on the one hand, to remain independent and politically neutral, and, on the other hand, to inform and warn societies of the dangers that lie ahead.
Habitat- and species-mediated short- and long-term distributional changes in waterbird abundance linked to variation in European winter weather
Aim Many species are showing distribution shifts in response to environmental change. We explored (a) the effects of inter-annual variation in winter weather conditions on non-breeding distributional abundance of waterbirds exploiting different habitats (deep-water, shallow water, farmland) and (b) the long-term shift in the population centroid of these species and investigate its link to changes in weather conditions. Location Europe. Methods We fitted generalized additive mixed Models to a large-scale, 24-year dataset (1990-2013) describing the winter distributional abundance of 25 waterbird species. We calculated the annual and long-term (3-year periods) population centroid of each species and used the winter North Atlantic Oscillation (NAO) index to explain the inter-annual and long-term shifts in their location. Results (a) Year-to-year southwestwards shifts in the population centroids of deep- and shallow-water species were linked to negative NAO values. Shallow-water species shifted northeastwards associated with positive NAO values and the distance shifted increased with increasing NAO. Deep-water species shifted northeastwards up to zero NAO indices, but showed no further increase at higher NAO values. (b) Deep-water species showed long-term northeastwards shifts in distributional abundance throughout the 1990s and the 2000s. Shallow-water species, on the other hand, shifted northeastwards during the 1990s and early 2000s, but southwestwards thereafter. There were no significant links between the NAO and year-to-year movements or long-term shifts in farmland species' population centroid. Main Conclusions We provide evidence for a link between both year-to-year and long-term changes in waterbird winter distributional abundances at large geographical scales to short- and long-term changes in winter weather conditions. We also show that species using shallow water, deep-water and farmland habitats responded differently, especially at high NAO values. As well as important ecological implications, these findings contribute to the development of future conservation measures for these species under current and future climate change.
The Influence of Former Process Steps on Changes in Hardness, Lattice and Micro Structure of AISI 4140 Due to Manufacturing Processes
The surface and subsurface conditions of components are critical for their functional properties. Every manufacturing process modifies the surface condition as a consequence of its mechanical, chemical, and thermal impact or combinations of the three. The depth of the affected zone varies for different machining operations and is related to the process parameters and characteristics. Furthermore, the initial material state has a decisive influence on the modifications that lead to the final surface conditions. With this knowledge, the collaborative research center CRC/Transregio 136 “Process Signatures” started a first joint investigation to analyze the influence of several machining operations on the surface modifications of uniformly premanufactured samples in a broad study. The present paper focusses on four defined process chains which were analyzed in detail regarding the resulting surface conditions as a function of the initial state. Two different workpiece geometries of the same initial material (AISI 4140, 42CrMo4 (1.7225) classified according to DIN EN ISO 683-2) were treated in two different heat treating lines. Samples annealed to a ferritic-perlitic microstructure were additionally deep rolled as starting condition. Quenched and tempered samples were induction hardened before further process application. These two states were then submitted to six different manufacturing processes, i.e., grinding (with mainly mechanical or thermal impact), precision turning (mainly mechanical), laser processing (mainly thermal), electrical discharge machining (EDM, mainly thermal) and electrochemical machining (ECM, (mainly chemical impact). The resulting surface conditions were investigated after each step of the manufacturing chain by specialized analysis techniques regarding residual stresses, microstructure, and hardness distribution. Based on the process knowledge and on the systematic characterizations, the characteristics and depths of the material modifications, as well as their underlying mechanisms and causes, were investigated. Mechanisms occurring within AISI 4140 steel (42CrMo4) due to thermal, mechanical or mixed impacts were identified as work hardening, stress relief, recrystallization, re-hardening and melting, grain growth, and rearrangement of dislocations.
Climate-driven changes in winter abundance of a migratory waterbird in relation to EU protected areas
Aim Species are responding to climate change by changing their distributions, creating debate about the effectiveness of existing networks of protected areas. As a contribution to this debate, we assess whether regional winter abundances and distribution of the Smew Mergellus albellus, a migratory waterbird species listed on Annex I (EU Birds Directive) that overwinters exclusively in European wetlands, changed during 1990–2011, the role of global warming in driving distributional changes and the effectiveness of the network of Special Protection Areas (SPAs, EU Birds Directive) in the context of climate change. Location Europe. Methods We used site-specific counts (6,883 sites) from 16 countries covering the entire flyway to estimate annual abundance indices and trends at country, region (north-eastern, central and south-western) and flyway scales, inside and outside SPAs. We fitted autoregressive models to assess the effect of winter temperature on the annual abundance indices whilst accounting for autocorrelation. Results The Smew wintering distribution shifted north-eastwards in Europe in accordance with the predictions of global warming, with increasing numbers in the north-eastern region and declines in the central region. Trends in wintering numbers were more positive in SPAs on the north-eastern and south-western part of the flyway. However, a large proportion of the wintering population remains unprotected in north-eastern areas outside of the existing SPA network. Main conclusions SPAs accommodated climate-driven abundance changes in the north-eastern region of the wintering distribution by supporting increasing numbers of Smew in traditional and newly colonized areas. However, we highlight gaps in the current network, suggesting that urgent policy responses are needed. Given rapid changes in species distributions, we urge regular national and international assessments of the adequacy of the EU Natura 2000 network to ensure coherence in site-safeguard networks for this and other species.
Investigating Patterns of Immune Interaction in Ovarian Cancer: Probing the O-glycoproteome by the Macrophage Galactose-Like C-Type Lectin (MGL)
Glycosylation, the posttranslational linking of sugar molecules to proteins, is notoriously altered during tumor transformation. More specifically in carcinomas, GalNAc-type O-glycosylation, is characterized by biosynthetically immature truncated glycans present on the cancer cell surface, which profoundly impact anti-tumor immune recognition. The tumor-associated glycan pattern may thus be regarded as a biomarker of immune modulation. In epithelial ovarian cancer (EOC) there is a particular lack of specific biomarkers and molecular targets to aid early diagnosis and develop novel therapeutic interventions. The aim of this study was to investigate the ovarian cancer O-glycoproteome and identify tumor-associated glycoproteins relevant in tumor–dendritic cell (DC) interactions, mediated by macrophage galactose-like C type lectin (MGL), which recognizes the tumor-associated Tn O-glycan. Lectin weak affinity chromatography (LWAC) was employed to probe the O-glycopeptidome by MGL and Vicia villosa agglutinin (VVA) lectin using glycoengineered ovarian cancer cell lines and ovarian cancer tissues as input material. Biochemical and bioinformatics analysis gave information on the glycan arrangement recognized by MGL in tumor cells. The potential MGL binders identified were located, as expected, at the cell membrane, but also within the intracellular compartment and the matrisome, suggesting that MGL in vivo may play a complex role in sensing microenvironmental cues. The tumor glycoproteins binders for MGL may become relevant to characterize the interaction between the immune system and tumor progression and contribute to the design of glycan targeting-based strategies for EOC immunotherapeutic interventions.
