Toleration
In this engaging and comprehensive introduction to the topic of toleration, Andrew Jason Cohen seeks to answer fundamental questions, such as: What is toleration? What should be tolerated? Why is toleration important? Beginning with some key insights into what we mean by toleration, Cohen goes on to investigate what should be tolerated and why. We should not be free to do everything murder, rape, and theft, for clear examples, should not be tolerated. But should we be free to take drugs, hire a prostitute, or kill ourselves? Should our governments outlaw such activities or tolerate them? Should they tolerate \"outsourcing\" of jobs or importing of goods or put embargos on other countries? Cohen examines these difficult questions, among others, and argues that we should look to principles of toleration to guide our answers. These principles tell us when limiting freedom is acceptable that is, they indicate the proper limits of toleration. Cohen deftly explains the main principles on offer and indicates why one of these stands out from the rest.
Applying systems-level spectral imaging and analysis to reveal the organelle interactome
The organization of the eukaryotic cell into discrete membrane-bound organelles allows for the separation of incompatible biochemical processes, but the activities of these organelles must be coordinated. For example, lipid metabolism is distributed between the endoplasmic reticulum for lipid synthesis, lipid droplets for storage and transport, mitochondria and peroxisomes for β-oxidation, and lysosomes for lipid hydrolysis and recycling. It is increasingly recognized that organelle contacts have a vital role in diverse cellular functions. However, the spatial and temporal organization of organelles within the cell remains poorly characterized, as fluorescence imaging approaches are limited in the number of different labels that can be distinguished in a single image. Here we present a systems-level analysis of the organelle interactome using a multispectral image acquisition method that overcomes the challenge of spectral overlap in the fluorescent protein palette. We used confocal and lattice light sheet instrumentation and an imaging informatics pipeline of five steps to achieve mapping of organelle numbers, volumes, speeds, positions and dynamic inter-organelle contacts in live cells from a monkey fibroblast cell line. We describe the frequency and locality of two-, three-, four- and five-way interactions among six different membrane-bound organelles (endoplasmic reticulum, Golgi, lysosome, peroxisome, mitochondria and lipid droplet) and show how these relationships change over time. We demonstrate that each organelle has a characteristic distribution and dispersion pattern in three-dimensional space and that there is a reproducible pattern of contacts among the six organelles, that is affected by microtubule and cell nutrient status. These live-cell confocal and lattice light sheet spectral imaging approaches are applicable to any cell system expressing multiple fluorescent probes, whether in normal conditions or when cells are exposed to disturbances such as drugs, pathogens or stress. This methodology thus offers a powerful descriptive tool and can be used to develop hypotheses about cellular organization and dynamics.

The Coevolution of Network Ties and Perceptions of Team Psychological Safety
Which comes first—team social networks or emergent team states (e.g., team climate)? We argue that team members' social network ties and team members' climate perceptions coevolve over time as a function of six reciprocal and co-occurring processes. We test our conceptual framework in a 10-month longitudinal study of perceptions of team psychological safety and social network ties in 69 work teams and find considerable support for our hypotheses. Our main results suggest that perceptions of psychological safety predict network ties. The more psychologically safe team members perceive their team to be, the more likely they are to ask their teammates for advice and to see them as friends, and the less likely they are to report difficult relationships with them. At the same time, network ties predict psychological safety. Team members adopt their friends' and advisors' perceptions of the team's psychological safety and reject the perceptions of those with whom they report a difficult relationship. Our framework and findings suggest that conceptual models and tests of unidirectional or team-level effects are likely to substantially misrepresent the mechanisms by which network ties and emergent team states coevolve.

A 'compensatory selection' effect with standardized tests: Lack of correlation between test scores and success is evidence that test scores are predictive of success
We introduce the statistical concept of 'compensatory selection', which arises when selecting a subset of applicants based on multiple predictors, such as when standardized test scores are used in combination with other predictors required in a school application (e.g., previous grades, references letters, and personal statements). Post-hoc analyses often fail to find a positive correlation between test scores and subsequent success, and this failure is sometimes taken as evidence against the predictive validity of the standardized test. The present analysis reveals that the failure to find a negative correlation indicates that the standardized test is in fact a valid predictor of success. This is due to compensation between predictors during selection: Some students are admitted despite a low test score because their application is exceptional in other respects, while other students are admitted primarily based on a high test score despite weakness in the rest of their application. This compensatory selection process introduces a negative correlation between test scores and other predictors among those admitted (a 'collider bias' or 'Berkson's paradox' effect). If test scores are valid predictors of success, this negative correlation between the predictors counteracts the positive correlation between test scores and success that would have been observed if all applicants were admitted. If test scores are not predictive of success, but were nevertheless used in a compensatory selection process, there would be a spurious negative correlation between test scores and success (i.e., an admitted student with a weak application except for a high test score would be unlikely to succeed). The selection effect that is described here is fundamentally different from the well-known 'restricted range' problem and can powerfully alter results even in situations that accept most applicants.
Ecology and Evolution of the African Great Lakes and Their Faunas
The Great Lakes of East Africa are collectively the earth's most remarkable and species-rich freshwater feature. Intrinsic biological factors and extrinsic ecological opportunities allowed much of the lakes' spectacular biological diversity to evolve through evolutionary (often adaptive) radiation and explosive speciation. Beyond evolutionary patterns and processes that led to this remarkable biodiversity and its astonishing morphological disparity, we highlight ecosystem functioning and complex biotic interactions such as coevolution. Comparative biogeographic patterns for vertebrates and invertebrates are discussed, as are patterns of diversity and disparity through the late Cenozoic. We demonstrate that the African Great Lakes, because of excellent fossil archives, are a phenomenal setting to integrate micro- and macroevolution. Unfortunately, these amazing ecosystems are also subject to various anthropogenic stressors at global and regional scales, which have already impacted their stability and threaten part of their extraordinary biodiversity with extinction.
