Playing to the Camera is the first full-length study devoted to the musical performance documentary. Its scope ranges from rock concert films to experimental video art featuring modernist music. Precision medicine integrating whole-genome sequencing, comprehensive metabolomics, and advanced imaging
Genome sequencing has established clinical utility for rare disease diagnosis. While increasing numbers of individuals have undergone elective genome sequencing, a comprehensive study surveying genome-wide disease-associated genes in adults with deep phenotyping has not been reported. Here we report the results of a 3-y precision medicine study with a goal to integrate wholegenome sequencingwith deep phenotyping. A cohort of 1,190 adult participants (402 female [33.8%]; mean age, 54 y [range 20 to 89+]; 70.6% European) had whole-genome sequencing, and were deeply phenotyped using metabolomics, advanced imaging, and clinical laboratory tests in addition to family/medical history. Of 1,190 adults, 206 (17.3%) had at least 1 genetic variant with pathogenic (P) or likely pathogenic (LP) assessment that suggests a predisposition of genetic risk. A multidisciplinary clinical team reviewed all reportable findings for the assessment of genotype and phenotype associations, and 137 (11.5%) had genotype and phenotype associations. A high percentage of genotype and phenotype associations (>75%) was observed for dyslipidemia (n = 24), cardiomyopathy, arrhythmia, and other cardiac diseases (n = 42), and diabetes and endocrine diseases (n = 17). A lack of genotype and phenotype associations, a potential burden for patient care, was observed in 69 (5.8%) individuals with P/LP variants. Genomics and metabolomics associations identified 61 (5.1%) heterozygotes with phenotype manifestations affecting serum metabolite levels in amino acid, lipid and cofactor, and vitamin pathways. Our descriptive analysis provides results on the integration of whole-genome sequencing and deep phenotyping for clinical assessments in adults.
Discovery and prioritization of variants and genes for kidney function in >1.2 million individuals
Genes underneath signals from genome-wide association studies (GWAS) for kidney function are promising targets for functional studies, but prioritizing variants and genes is challenging. By GWAS meta-analysis for creatinine-based estimated glomerular filtration rate (eGFR) from the Chronic Kidney Disease Genetics Consortium and UK Biobank (n = 1,201,909), we expand the number of eGFRcrea loci (424 loci, 201 novel; 9.8% eGFRcrea variance explained by 634 independent signal variants). Our increased sample size in fine-mapping (n = 1,004,040, European) more than doubles the number of signals with resolved fine-mapping (99% credible sets down to 1 variant for 44 signals, ≤5 variants for 138 signals). Cystatin-based eGFR and/or blood urea nitrogen association support 348 loci (n = 460,826 and 852,678, respectively). Our customizable tool for Gene PrioritiSation reveals 23 compelling genes including mechanistic insights and enables navigation through genes and variants likely relevant for kidney function in human to help select targets for experimental follow-up. Identifying causal variants and genes in genome-wide association studies remains a challenge, an issue that is ameliorated with larger sample sizes. Here the authors meta-analyze kidney function genome-wide association studies to identify new loci and fine-map loci to home in on variants and genes involved in kidney function.
Ill-Fated Merger: LTV, Dragged Down By Steel Subsidiary, Struggles to Survive --- Its Acquisition of Republic, Price War Upset Plans; Losses Approach $1 Billion --- Cost Reductions Help a Bit
LTV Corp. is fighting to survive, but you'd never know that from its posh new corporate headquarters here atop the 50-story LTV Center. A gleaming black marble staircase swirls through three of LTV's five floors, and original Western paintings grace mahogany walls. LTV Chairman Raymond Hay calls it \"one of the most prestigious landmarks west of the Mississippi River.\" It was to be a fitting companion to Mr. Hay's 1984 acquisition of Republic Steel Corp., which together with prior steel holdings made LTV the owner of the nation's second-largest steel company. The $714 million merger would become \"a landmark in the annals of America's basic industries,\" Mr. Hay predicted. Now, LTV Steel's problems are dragging down the once-freewheeling parent company, which also has aerospace and energy subsidiaries. Since the Republic acquisition, LTV has run up losses totaling nearly $1 billion. Debt has climbed to $2.6 billion from $1.6 billion. Eroding net worth has rattled suppliers and prompted LTV directors to discuss informally the possibility of a bankruptcy filing, for the parent or its steel unit. LTV officials vigorously deny that they are anywhere close to such a step, but federal bank regulators recently classified a portion of LTV bank letters of credit as \"substandard.\"
U.S. Steel Is Said To Agree to Buy Texas Oil & Gas --- Steel Company Is Expected To Announce an Accord Of About $3.78 Billion
U.S. Steel Corp. is expected to announce today an agreement to acquire Texas Oil & Gas Corp. for about $18 a share, or $3.78 billion. In composite trading on the New York Stock Exchange yesterday, Texas Oil closed at $17.875, down 12.5 cents. U.S. Steel closed at $27.50, down 62.5 cents, apparently in anticipation of a bid. A U.S. Steel bid for Texas Oil of about $18 a share would be below Wall Street estimates last week that placed the bid as high as $25 a share, or $5.3 billion. It would, however, be above the $17-a-share appraised value given Texas Oil assets by John S. Herold Inc., a Greenwich, Conn.-based appraiser of oil and gas reserves.
LTV Receives IRS Waiver for Payment Of $183 Million on Three Pension Plans
The IRS action removes a major uncertainty hanging over the steel, energy and aerospace concern. LTV had sought IRS approval to defer the payments in June. Because the plan is tax-sheltered, any change in it has to be approved by the IRS in order for it to retain its tax-qualification status. Furthermore, the IRS must approve any applications for waivers on pension-plan contributions backed by Pension Benefit Guaranty Corp., a federal agency that insures corporate pensions. LTV said that it, the IRS and Pension Benefit Guaranty agreed that LTV would \"collateralize the arrangement.\" But officials of Pension Benefit Guaranty said LTV has pledged assets of its aerospace-defense unit as collateral in case it can't meet its pension obligations. Pension Benefit Guaranty said it now isn't considering terminating LTV's pension plans. Pension Benefit Guaranty officials said last week that the agency was considering terminating the pension plans of LTV's steel unit, a move that could have forced LTV to reimburse the plans' $850 million in unfunded liabilities.

Pension Benefit Guaranty Corp. is considering terminating the pension plans of LTV Corp.'s steel unit, a move that could force the beleaguered steel, energy and aerospace concern to reimburse the plans' $850 million in liabilities. Termination of the pension plan could exacerbate the strains on LTV, which has been troubled by its 1984 acquisition of Republic Steel Corp. In terms of the money involved, it would also be the stiffest action ever by Pension Benefit Guaranty, a federal agency that guarantees corporate pension plans. The agency's threat to terminate the LTV pension plans also reflects its concern over the state of talks between it, the Internal Revenue Service and LTV. The parties have been discussing a previously reported LTV request that the IRS defer about $182 million in payments on several pension plans associated with its steel business. LTV wants to spread out the payments, which were due last month, over a 15-year period.
Clenbuterol exerts antidiabetic activity through metabolic reprogramming of skeletal muscle cells
Activation of the sympathetic nervous system causes pronounced metabolic changes that are mediated by multiple adrenergic receptor subtypes. Systemic treatment with β 2- adrenergic receptor agonists results in multiple beneficial metabolic effects, including improved glucose homeostasis. To elucidate the underlying cellular and molecular mechanisms, we chronically treated wild-type mice and several newly developed mutant mouse strains with clenbuterol, a selective β 2 -adrenergic receptor agonist. Clenbuterol administration caused pronounced improvements in glucose homeostasis and prevented the metabolic deficits in mouse models of β-cell dysfunction and insulin resistance. Studies with skeletal muscle-specific mutant mice demonstrated that these metabolic improvements required activation of skeletal muscle β 2 -adrenergic receptors and the stimulatory G protein, G s . Unbiased transcriptomic and metabolomic analyses showed that chronic β 2 -adrenergic receptor stimulation caused metabolic reprogramming of skeletal muscle characterized by enhanced glucose utilization. These findings strongly suggest that agents targeting skeletal muscle metabolism by modulating β 2 -adrenergic receptor-dependent signaling pathways may prove beneficial as antidiabetic drugs.
