Asymptotic dimension of groups
2002
Gromov introduced the asymptotic dimension (asdim) of a metric space in his study of asymptotic invariants of infinite groups. Later, Yu introduced a property of discrete metric spaces and groups called property A. Yu showed that for groups with finite asdim or with property A, the famous Novikov Higher Signature Conjecture holds. Recently Gromov and others provided means of constructing groups without either of these properties, so the problem of determining precisely which groups have these properties became interesting. In this dissertation we prove that both of these properties are preserved under the formation of certain infinite unions, amalgamated free products and HNN extensions. In the case of asdim we also give an upper bound which is tight for the HNN extension and many amalgamated free products.

Discovery of peptide-based asymmetric acyl transfer catalysts

Chapter 1. A brief overview of non-chromatographic assays used in catalyst discovery is presented. Much of the work is still in the development stages, but many of the techniques show signs of promise. Chapter 2. Small peptides based on β-turn or β-hairpin structural motifs and containing modified histidine residues were found to be effective acyl transfer catalysts in the kinetic resolution of certain amide-functionalized alcohols. Kinetic studies indicate unambiguously that enantioselectivities are due to specific acceleration of reaction for one substrate enantiomer, rather than the deceleration of the reaction for the other. The results are presented in the context of a possible enantiomer-specific hydrogen bonding interaction in the stereochemistry-determining step for these processes. Chapter 3. A fluorescence-based assay was developed to enable the high-throughput screening of catalysts in acyl transfer reactions. The assay is amenable to examining catalysts in solution as well as on the solid support. The assay has been applied to the discovery of a new class of octapeptide catalysts for the kinetic resolution of secondary alcohols. The fluorescence assay provided a lead structure on which a “directed” split and pool library was based. Screening of this “directed library” revealed several peptides with improved selectivities for the model substrate sec-phenylethanol.

Allogeneic Human Neural Stem Cells for Improved Therapeutic Delivery to Peritoneal Ovarian Cancer

Background: Immortalized, clonal HB1.F3.CD21 human neural stem/progenitor cells (NSCs), loaded with therapeutic cargo prior to intraperitoneal (IP) injection, have been shown to improve the delivery and efficacy of therapeutic agents in pre-clinical models of stage III ovarian cancer. In previous studies, the distribution and efficacy of the NSC-delivered cargo has been examined; however, the fate of the NSCs has not yet been explored. Methods: To monitor NSC tropism, we used an unconventional method of quantifying endocytosed gold nanorods to overcome the weaknesses of existing cell-tracking technologies. Results: Here, we report efficient tumor tropism of HB1.F3.CD21 NSCs, showing that they primarily distribute to the tumor stroma surrounding individual tumor foci within 3 hours after injection, reaching up to 95% of IP metastases without localizing to healthy tissue. Furthermore, we demonstrate that these NSCs are non-tumorigenic and non-immunogenic within the peritoneal setting. Conclusions: Their efficient tropism, combined with their promising clinical safety features and potential for cost-effective scale-up, positions this NSC line as a practical, off-the-shelf platform to improve the delivery of a myriad of peritoneal cancer therapeutics.

With the CND, I deliver the \"No Trident Replacement\" petition to Downing Street. Interest in the work of the CND has increased dramatically with the escalating conflicts in the Middle East and the threats presented by a lack of security of nuclear waste. On Saturday I am scheduled to attend a protest against the American position on Israel, which I am preparing for. I hear that Tony Blair has cancelled his holiday, and hope this indicates that we may get the ceasefire that is so desperately needed in Israel and Lebanon.

I have a jewellery showroom in Beirut and a workshop in Bourj Hammoud. I visit them as both are open half days. The employees at the workshop help me to connect people in need with supplies. Our spirits are not down, and we're trying to continue with a quasi- normal style of life. It gives us strength to keep things as normal as possible. We have lived through the first Lebanese war, and war is something life has taught us to handle. More aid is arriving from people in Saudi, and Jordan is giving free supplies that are helping a lot. People are grateful and the help is warming our hearts. The help from Saudi is making us think that maybe everything will be OK. Yallah!
