Lack of the Nlrp3 Inflammasome Improves Mice Recovery Following Traumatic Brain Injury

Treatment for traumatic brain injury (TBI) remains elusive despite compelling evidence from animal models for a variety of therapeutic targets. The activation of the NLRP3 (Nucleotide-binding oligomerization domain-like receptor family pyrin domain-containing 3) inflammasome has been proposed as key point in the brain damage associated with TBI. NLRP3 was tested as potential target for reducing neuronal loss and promoting functional recovery in a mouse model of TBI. Male NLRP3 -/- ( n = 20) and wild type ( n = 27) mice were used. A closed TBI model was performed and inflammatory and apoptotic markers were evaluated. A group of WT mice also received BAY 11-7082, a NLRP3 inhibitor, to further evaluate the role of this pathway. At 24 h following TBI NLRP3 -/- animals demonstrated a preserved cognitive function as compared to WT mice, additionally brain damage was less severe and the inflammatory mediators were reduced in brain lysates. The administration of BAY 11-7082 in WT animals subjected to TBI produced overlapping results. At day 7 histology revealed a more conserved brain structure with reduced damage in TBI NLRP3 -/- animals compared to WT. Our data indicate that the NLRP3 pathway might be exploited as molecular target for the short-term sequelae of TBI.

Not only climate: The importance of biotic interactions in shaping species distributions at macro scales

Abiotic factors are usually considered key drivers of species distribution at macro scales, while biotic interactions are mostly used at local scales. A few studies have explored the role of biotic interactions at macro scales, but all considered a limited number of species and obligate interactions. We examine the role of biotic interactions in large‐scale SDMs by testing two main hypotheses: (1) biotic factors in SDMs can have an important role at continental scale; (2) the inclusion of biotic factors in large‐scale SDMs is important also for generalist species. We used a maximum entropy algorithm to model the distribution of 177 bat species in Africa calibrating two SDMs for each species: one considering only abiotic variables (noBIO‐SDMs) and the other (BIO‐SDMs) including also biotic variables (trophic resource richness). We focused the interpretation of our results on variable importance and response curves. For each species, we also compared the potential distribution measuring the percentage of change between the two models in each pixel of the study area. All models gave AUC >0.7, with values on average higher in BIO‐SDMs compared to noBIO‐SDMs. Trophic resources showed an importance overall higher level than all abiotic predictors in most of the species (~68%), including generalist species. Response curves were highly interpretable in all models, confirming the ecological reliability of our models. Model comparison between the two models showed a change in potential distribution for more than 80% of the species, particularly in tropical forests and shrublands. Our results highlight the importance of considering biotic interactions in SDMs at macro scales. We demonstrated that a generic biotic proxy can be important for modeling species distribution when species‐specific data are not available, but we envision that a multi‐scale analysis combined with a better knowledge of the species might provide a better understanding of the role of biotic interactions. Traditionally abiotic factors are considered the main drivers of biodiversity at macro‐scales, whereas biotic factors are considered important almost only at local scales. Modeling the distribution of 177 African bat species, we demonstrated the importance of considering biotic interactions in SDMs at macro‐scales regardless of the species’ ecology.

A multi-disciplinary approach to identify spillover interfaces of bat coronaviruses to pig farms in Italy

Bats are recognized reservoirs of diverse coronaviruses (CoVs), but little is known about the pathways enabling their spillover into livestock. This study applied a multidisciplinary approach, combining bioacoustic surveys, landscape analysis and molecular virology, to assess the risk of CoV transmission from bats to pigs in intensive farming systems of Northern Italy. Between 2021 and 2022, we carried out bioacoustic monitoring in 14 pig farms to assess bat presence, diversity and behaviour. We also analysed landscape and farm-level variables to identify predictors of bat activity and richness. Additionally, we investigated CoV circulation in three populations of Pipistrellus kuhlii through active longitudinal surveillance, performing whole-genome sequencing on new and archival CoV strains detected in P. kuhlii and Hypsugo savii . Using these data, we explored the viral biodiversity potentially present at this interface via genetic and phylogenetic analyses. We identified eight bat species across farms, with P. kuhlii , P. pipistrellus and H. savii being the most widespread and active. Landscape and structural analysis revealed that farm features attracting insects were associated with higher bat activity, while the surrounding habitat showed little effect. Crucially, we found frequent absence of physical barriers preventing contact between bats or their droppings and pig enclosures, increasing exposure risks. Focusing on the most common bat species, we detected active CoV circulation in P. kuhlii , including colonies located near pig facilities. Two distinct CoV species were identified in P. kuhlii , suggesting potential for viral recombination. CoVs were detected throughout the active season, with amplification peaks in May and August. Phylogenetic analysis indicated that pigs could be exposed to at least eight bat CoV species in Italy. Notably, CoVs appeared to be shared between P. kuhlii and H. savii , further increasing recombination risks. Our study outlines a potential transmission route of bat CoVs to swine and highlights key risk factors, including farm structures, biosecurity gaps, bat species involved, viral diversity and seasonal patterns of virus circulation.

Trait-based approaches are becoming extremely common in ecological modeling and the availability of traits databases is increasing. However, data availability is often biased towards particular regions and taxa, with many taxa (e.g., bats) often under-represented. Here, we present the AfroBaT dataset, a compilation of trait data on 320 African bat species containing 76,914 values for 86 traits focusing on morphology, reproduction, life-history, trophic ecology, and species distributions. All data were gathered from published literature following the ecological trait-data standard procedure. Missing data for both numerical and categorical traits were imputed with a machine learning approach including species phylogeny. Trophic ecology traits showed the highest coverage in the literature (72% of the species averaged over all traits), while reproductive traits the lowest. Our data imputation improved the coverage of AfroBaT especially for reproductive traits, going from 27% to 58% of the species covered. AfroBaT has a range of potential applications in macroecology and community ecology, and the availability of open-access data on African bats will enable collaboration and data-sharing among researchers.

A comprehensive framework to assess multi‐species landscape connectivity

Due to the central role of landscape connectivity in many ecological processes, evaluating and accounting for it has gained attention in both theoretical and applied ecological sciences. To address this challenge, researchers often use generic species to simplify multi‐species connectivity assessments. Yet, this approach tends to oversimplify movement behaviour, likely reducing realism and precision of connectivity model outputs. Also, the most widely used methods and theories for assessing landscape connectivity, namely circuit and network theories, have strong limitations. Finally, uncertainty or robustness estimates are rarely integrated in connectivity assessments. Here, we propose a versatile framework, which, instead of using arbitrary defined generic species, first identifies species groups based on species' environmental niches and morphological, biological, and ecological traits. Second, it combines circuit and network theories to take the best of the two methods to assess landscape connectivity for those groups, while integrating uncertainties in modelling choices. Specifically, ecological continuities (i.e. landscape elements contributing to connectivity) are calculated for these groups and used together with group dispersal capacities to derive network‐based connectivity metrics for conservation areas. We detailed our framework through a case study where we assess the connectivity of 1619 protected areas in metropolitan France for 193 vertebrate species. Our study revealed that both the protection of ecological continuities and the connectivity of protected areas for 11 mammal and 19 bird groups, respectively, were quite low, with variations among groups. Different protection types (i.e. national parks, reserves or prefectural orders) contributed unequally to the overall connectivity of group‐specific suitable habitats. Considering uncertainty propagation was crucial, as many connectivity metrics varied among repetitions. The proposed framework combines different connectivity tools to provide a more relevant and comprehensive assessment of landscape connectivity. It can be used to inform the decision‐making process for spatial planning, particularly in the context of connectivity conservation and management, or support theoretical studies to better understand the ecological role of landscape connectivity. Its flexibility allows easy application under various environmental conditions, including future scenarios.

Bidirectional Interactions Between the Gut Microbiota and Incretin-Based Therapies

Obesity, insulin resistance, type 2 diabetes mellitus (T2DM) and metabolic syndrome have been largely correlated to a reduction in bacterial load and diversity, resulting in a condition known as intestinal dysbiosis. The recent emergence of novel antidiabetic medications has been demonstrated to exert a favourable influence on the composition of the intestinal microbiota. Incretin-based therapy exerts a multifaceted influence on the composition of the gut microbiota, leading to alterations in bacterial flora. Of particular significance is the capacity of numerous metabolites produced by the gut microbiota to modulate the activity and hormonal secretion of enteroendocrine cells. This review examines the effects of dipeptidyl peptidase 4 (DPP-4) inhibitors, glucagon-like peptide 1 (GLP-1) receptor agonists and GLP-1/gastric inhibitory polypeptide (GIP) receptor dual agonists on the composition of the gut microbiota in both mice and human subjects. The nature of this interaction is complex and bidirectional. The present study demonstrates the involvement of the incretinic axis in modulating the microbial composition, with the objective of providing novel preventative strategies and potential personalised therapeutic targets for obesity and T2DM.

Ketoacidosis is considered an emergency metabolic disorder that can be triggered by starvation and alcohol consumption in addition to diabetes. We described an unusual case of ketoacidosis in a non-diabetic young man who presented to our hospital with a complaint of rapid and shallow breathing, weakness and nausea. Fourteen days back, due to heart failure, he started sodiumglucose co-transporter 2 inhibitors (SGLT2-i) in addition to fumarate bisoprolol, sacubitril/valsartan and eplerenone. Based on clinical examination, ketoacidosis was suspected, although his glucose and glycosylated hemoglobin levels were in range. Overall, based on the clinical and laboratory findings, the diagnosis of euglycemic non-diabetic ketoacidosis due to the use of the SGLT2- i was made and SGLT2-i has been discontinued. Although SGLT2-i are effective and safe drugs, it is advisable to carefully evaluate their use also in non-diabetic patients especially when hepatic fibrosis or incorrect eating habits are present.

Evaluation of Smallpox Vaccination Coverage and Attitude towards Monkeypox Vaccination among Healthcare Workers in an Italian University Hospital

(1) Background: In 2022, monkeypox (Mpox) was declared a public health emergency. The European Medicines Agency has authorized the use of Imvanex/Jynneos, a smallpox vaccine, for coverage against pox. Healthcare workers (HCWs) are all considered by the European Centre for Disease Prevention and Control to be at risk, but in Italy, vaccination was offered only to laboratory personnel. The present study aims to investigate smallpox vaccination coverage (VC) that provides protection against Mpox among HCWs in an Italian university hospital and to assess HCWs’ attitudes towards the possibility of getting vaccinated against Mpox. (2) Methods: We conducted a cross-sectional survey. 336 HCWs from selected wards were asked to fill out a self-declaration to collect their sex, profession, ward, vaccination status, and attitude toward Mpox vaccination. (3) Results: 60.71% of HCWs involved provided the requested data; 38.7% of them were previously vaccinated against smallpox, which corresponds to 23.5% of the total HCWs in the wards considered. Considering those born before 1979 as vaccinated, VC increases from 23.5% to 41.7%; the percentage of HCWs who adhered to vaccination is 23%; laboratory technicians showed a lower willingness to be vaccinated. The ward with the highest willingness to vaccinate is proctological surgery. (4) Conclusions: Based on our experience, a variability in smallpox VC and in willingness to vaccination has emerged both among different job titles and age categories and across the wards analyzed. Additionally, our survey reveals that vaccination attitudes are higher among HCWs from wards that currently do not have free access to such vaccinations.

Semaglutide and kidney function: friends or enemies?

Semaglutide is a glucagon-like peptide-1 receptor agonist (GLP-1 RAs) approved for the treatment of type 2 diabetes mellitus (T2DM) and chronic weight management in obesity. GLP-1RAs are being investigated to slow the decline of kidney function in type 2 diabetics with chronic kidney disease. These agents prevent renal complications and have proven beneficial effects on cardiac outcomes. We describe a rare case of semaglutide-induced acute kidney injury (AKI) in a young woman with obesity, T2DM, hypertensive cardiomyopathy, and no pre-existing chronic kidney disease (CKD). This case is relevant as GLP1-RAs is used frequently in patients with and without kidney disease. Currently, only 3 cases of AKI ascribed to semaglutide of which only 1 without CKD have been described. Only in this case, kidney function improves after semaglutide discontinuation. However, because GLP1-RAs will be prescribed more and more frequently, we sought to highlight this possible, serious adverse effect of semaglutide.
