Implications of HIF-1α in the tumorigenesis and progression of pancreatic cancer
Pancreatic cancer is one of the leading causes of cancer-related deaths worldwide and is characterized by highly hypoxic tumor microenvironment. Hypoxia-inducible factor-1 alpha (HIF-1α) is a major regulator of cellular response to changes in oxygen concentration, supporting the adaptation of tumor cells to hypoxia in an oxygen-deficient tumor microenvironment. Numerous studies revealed the central role of HIF-1α in the carcinogenesis and progression of pancreatic cancer. This article reviewed the molecular mechanisms of how HIF-1α regulated tumorigenesis and progression of pancreatic cancer and suggested that targeting HIF-1α and its signaling pathways could be promising therapeutics for pancreatic cancer.

The predictive value of fibrinogen‐to‐albumin ratio in the active, severe active, and poor prognosis of systemic lupus erythematosus: A single‐center retrospective study
Objective To evaluate the prediction and effect of fibrinogen‐to‐albumin ratio (FAR) on active, severe active, and poor prognosis of systemic lupus erythematosus (SLE). Methods One hundred and sixty‐eight patients with SLE who were treated in our hospital were enrolled, the clinical data, laboratory indexes, and disease prognosis of all patients were collected and analyzed. Results Triglyceride (TG), FAR, ESR, and anti‐dsDNA (+) were the influencing factors, while complement 3 (C3) was the protective factor of active SLE, the odds ratio (OR) values were 2.968, 3.698, 2.114, 2.727, and 0.652, respectively (p < 0.05). FAR, ESR, and anti‐dsDNA (+) were the influencing factors, while C3 was the protective factor of severe active SLE, the OR values were 3.791, 1.953, 2.187, and 0.742, respectively (p < 0.05). SLE disease activity index (SLEDAI), TG, FAR, and anti‐dsDNA (+) were the influencing factors, while C3 was the protective factor of poor prognosis SLE, the OR values were 3.024, 2.293, 3.012, 2.323, and 0.801, respectively (p < 0.05). FAR and FIB were positively correlated with SLEDAI, while ALB was negatively correlated with SLEDAI, the related coefficient (r) were 0.398, 0.267, −0.270, respectively. The receiver operating curve (ROC) analysis showed that the predictive values of FAR for active, severe active and poor prognosis SLE were 0.769, 0.769, and 0.734, respectively, were significant higher than FIB and ALB (p < 0.05). Conclusion Fibrinogen‐to‐albumin ratio was an influencing factor of active, severe active, and poor prognosis SLE had higher predictive value than FIB and ALB for the activity and prognosis of SLE. As a new inflammatory marker fibrinogen‐to‐albumin ratio (FAR) has been proved to have good predictive value in the diagnosis and prognosis of diabetes nephropathy, acute renal injury, rheumatoid disease, and so on. However, there is few relevant research on whether it can predict the activity and prognosis of SLE and whether the predictive value is higher than FIB and ALB. Our study compared the predictive value of FIB, ALB, FAR, and analyzed the influencing factors of active, severe active, and poor prognosis SLE, in order to provide a new predictive biomarker for the disease activity and prognosis. Our findings demonstrated that FAR had higher predictive value than FIB and ALB, and high FAR showed great associated with an increasing probability of active, severe active, and poor prognosis SLE, in other words, FAR would be a better potential marker for predicting the severity and prognosis of SLE than the single indicators of FIB and ALB. Association of serum lipid levels, triglyceride–glucose index, systemic immune-inflammation index, and platelet-to-lymphocyte ratio with colorectal cancer: A retrospective study from China
Objective This retrospective case–control study aimed to explore the correlations of blood lipid levels, triglyceride–glucose index, systemic immune-inflammation index, and platelet-to-lymphocyte ratio with colorectal cancer and evaluate their potential value in early risk prediction. Methods A total of 120 patients admitted to Anqing First People’s Hospital between July 2022 and March 2025 were categorized into a control group (n = 60, normal colonoscopy) and an observation group (n = 60, colorectal cancer). Clinical and laboratory data, including sex, age, body mass index, lipid profile, blood glucose, and blood counts, were collected. Triglyceride–glucose index, systemic immune-inflammation index, and platelet-to-lymphocyte ratio were calculated accordingly. Results The observation group had significantly higher rates of drinking history, body mass index, triglyceride, total cholesterol, low-density lipoprotein cholesterol, triglyceride–glucose index, systemic immune-inflammation index, and platelet-to-lymphocyte ratio (P < 0.05). Triglyceride was strongly positively correlated with triglyceride–glucose index (r = 0.921) and negatively correlated with platelet-to-lymphocyte ratio (r = −0.189). Logistic regression identified drinking history (odds ratio = 5.015) and triglyceride–glucose index (odds ratio = 5.937) as independent risk factors. The receiver operating curve of triglyceride–glucose index showed an area under the curve of 0.713, with 78.3% specificity and 65% sensitivity. Conclusion Triglyceride–glucose index, along with blood lipid levels, systemic immune-inflammation index, and platelet-to-lymphocyte ratio, is closely associated with colorectal cancer. Triglyceride–glucose index may be a promising biomarker for early screening.

Prognostic and clinicopathological significance of GPRC5A in various cancers: A systematic review and meta-analysis
GPRC5A is associated with various cancer initiation and progression. Controversial findings have been reported about GPRC5A prognostic characteristics, and no meta-analysis has been conducted to assess the relationship between GPRC5A and cancer prognosis. Therefore, the objective of this meta-analysis is to evaluate the overall prognostic effectiveness of GPRC5A. We first conducted a systematic search in the PubMed, Embase, Web of Science, CNKI, Cochrane, and WangFang databases. The hazard ratio (HR) and odds ratios (OR) with 95% CI were then pooled to assess the associations between GPRC5A expression and overall survival (OS), disease-free survival (DFS), event-free survival (EFS), and clinicopathological characteristics. Chi-squared test and I2 statistics were completed to evaluate the heterogeneity in our study. A random-effects model was used when significant heterogeneity existed (I2>50% and p<0.05); otherwise, we chose the fixed-effect model. Subgroup analysis was stratified by tumor type, region, HR obtained measurements, and sample capacity to explore the source of heterogeneity. In total, 15 studies with 624 patients met inclusion criteria of this study. Our results showed that higher expression of GPRC5A is associated with worse OS (HR:1.69 95%CI: 1.20-2.38 I2 = 75.6% p = 0.000), as well as worse EFS (HR:1.45 95%CI: 1.02-1.95 I2 = 0.0% p = 0.354). Subgroup analysis indicated that tumor type might be the source of high heterogeneity. Additionally, cancer patients with enhanced GPRC5A expression were more likely to lymph node metastasis (OR:1.95, 95%CI 1.33-2.86, I2 = 43.9%, p = 0.129) and advanced tumor stage (OR: 1.83, 95%CI 1.15-2.92, I2 = 61.3%, p = 0.035), but not associated with age, sex, differentiation, and distant metastasis. GPRC5A can be a promising candidate for predicting medical outcomes and used for accurate diagnosis, prognosis prediction for patients with cancer; however, the predictive value of GPRC5A varies significantly according to cancer type. Further studies for this mechanism will be necessary to reveal novel insights into application of GPRC5A in cancers.

The higher mortality associated with low serum albumin is dependent on systemic inflammation in end-stage kidney disease
The correlation of low serum albumin with mortality in patients with chronic kidney disease (CKD) is partly linked to its association with systemic inflammation. However, it is not clear to what extent albumin's correlation with mortality depends on concomitant systemic inflammation. Here we addressed this question in patients with CKD stage 5. Serum albumin (S-Alb), systemic inflammation (high-sensitive C-reactive protein, hsCRP), cardiovascular disease (CVD) and nutritional status (subjective global assessment, SGA) were assessed at baseline in 822 patients: 523 incident dialysis patients, 212 prevalent hemodialysis (HD) and 87 prevalent peritoneal dialysis (PD) patients. Patients were divided into four groups according to hsCRP and S-Alb in each cohort: Group 1 -normal S-Alb and normal hsCRP (reference); Group 2 -low S-Alb and normal hsCRP; Group 3-normal S-Alb and high hsCRP; Group 4-low S-Alb and high hsCRP. Survival over 60 months was analyzed. In Cox analysis, Group 4 had an increased mortality risk (adjusted Hazard ratio (95% confidence interval): 1.62 (1.06-2.47); p = 0.02) whereas the augmented mortality risks for Groups 2 and 3 in univariate analyses were not significant after adjustments for age, gender, blood pressure, diabetes mellitus, smoking, SGA, renal function and renal replacement technique. Whereas mortality risk was increased in CKD stage 5 patients with low S-Alb and high CRP, it was not increased in patients with low S-Alb and normal CRP. Our observation suggests that inflammatory status should be taken into account when using S-albumin for risk assessment in CKD stage 5 patients.
