Acute myeloid leukemia (AML) is a malignancy caused by the uncontrolled and dysregulated clonal expansion of abnormal myeloid primordial cells. In general, the prognosis of AML remains poor despite new discoveries in its pathogenesis and treatment. It is crucial to find early and sensitive biomarkers and continue to explore active targeted treatments. Interferon-induced transmembrane protein (IFITM) family is an important part of the interferon signaling pathway and participate in the regulation of immune cell signaling, adhesion, cancer, and liver cell migration. However, the clinical and prognostic value of the IFITM family in AML has rarely been studied. We screened The Cancer Genome Atlas database and found 155 AML patients with IFITM family (IFITM1–5) expression data. In patients who only received chemotherapy, those with high IFITM3 expression had significantly shorter event-free survival (EFS) and overall survival (OS) than patients with low expression (all P < 0.05). Multivariate analysis demonstrated that high IFITM3 expression was an independent risk factor for EFS and OS in patients only received chemotherapy (all P < 0.05). In patients who underwent allogeneic hematopoietic stem cell transplantation (allo-HSCT), however, all IFITM members had no impact on either EFS or OS. In conclusion, our study elucidated that high IFITM3 expression could be an adverse prognostic factor for AML, whose effect might be overcome by allo-HSCT.

Overexpression of PDK2 and PDK3 reflects poor prognosis in acute myeloid leukemia

Acute myeloid leukemia (AML) is a hematological malignancy characterized by the proliferation of immature myeloid cells, with impaired differentiation and maturation. Pyruvate dehydrogenase kinase (PDK) is a pyruvate dehydrogenase complex (PDC) phosphatase inhibitor that enhances cell glycolysis and facilitates tumor cell proliferation. Inhibition of its activity can induce apoptosis of tumor cells. Currently, little is known about the role of PDKs in AML. Therefore, we screened The Cancer Genome Atlas (TCGA) database for de novo AML patients with complete clinical information and PDK family expression data, and 84 patients were included for the study. These patients did not undergo allogeneic hematopoietic stem cell transplantation (allo-HSCT). Univariate analysis showed that high expression of PDK2 was associated with shorter EFS (P = 0.047), and high expression of PDK3 was associated with shorter OS (P = 0.026). In multivariate analysis, high expression of PDK3 was an independent risk factor for EFS and OS (P < 0.05). In another TCGA cohort of AML patients who underwent allo-HSCT (n = 71), PDK expression was not associated with OS (all P > 0.05). Our results indicated that high expressions of PDK2 and PDK3, especially the latter, were poor prognostic factors of AML, and the effect could be overcome by allo-HSCT.

Pulse current-assisted flexible granular medium forming is a promising approach for manufacturing complex thin-walled components from difficult-to-deform Ti2AlNb-based alloys. In this study, the electro-thermo-mechanical deformation behavior of Ti2AlNb sheets is investigated through pulse current-assisted uniaxial tensile tests, microstructural characterization, and finite element simulations. The influences of pulse current intensity and strain rate on flow behavior, fracture characteristics, and phase evolution are clarified, and an effective forming window is identified. Numerical simulations are employed to analyze the role of granular medium friction in material flow and wall thickness distribution, providing guidance for forming box-shaped components. The results demonstrate that forming at approximately 950 °C with a strain rate of 0.001 s−1 reduces deformation resistance, while enhanced tangential interaction between the granular medium and the sheet improves wall thickness uniformity. This study provides a feasible processing route and practical guidelines for the fabrication of complex Ti2AlNb sheet components.

Distribution and Quantification of Lactic Acid Enantiomers in Baijiu

Enantiomers of lactic acid were investigated in Baijiu, including soy sauce aroma-type Baijiu (SSB), strong aroma-type Baijiu (STB), and light aroma-type Baijiu (LTB), via high-performance liquid chromatography with a chiral separation column. The natural concentration and enantiomeric distribution of lactic acid were studied, and their contribution to the flavor of Chinese Baijiu was evaluated based on recognition threshold. The results showed that there were significant differences in the content of lactic acid and the ratio of enantiomeric isomers among different aroma types and storage year. In SSB, the concentrations of D-lactic acid and L-lactic acid were higher, with the highest concentrations of 1985.58 ± 11.34 mg/L and 975.31 ± 14.03 mg/L, respectively. In STB, the highest concentrations of D-lactic acid and L-lactic acid were 1048.00 ± 11.46 mg/L and 939.83 ± 0.23 mg/L, respectively. In LTB, the highest concentrations of D-lactic acid and L-lactic acid were 760.90 ± 9.45 mg/L and 558.33 ± 3.06 mg/L, respectively. The average D/L enantiomeric ratios were 78:22 ± 16.16 and 80:20 ± 9.72 in the Commercial Baijiu products of SSB and STB, respectively. The average D/L enantiomeric ratio in LTB was 90:10 ± 6.08. D-lactic acid in JSHS vintage Baijiu showed a wave variation with aging, while L-lactic acid gradually increased during aging, and the average D/L enantiomeric ratio was 76:24 ± 4.26. The concentration of D-lactic acid in XJCT vintage Baijiu also showed a wave variation with aging, and the concentration of L-lactic acid tended to be stable during aging, with an average D/L enantiomeric ratio of 88:12 ± 2.80. The content of the two configurations of lactic acid in the LZLJ vintage Baijiu showed a decreasing trend during aging, with an average D/L enantiomeric ratio of 60:40 ± 11.99. The recognition threshold of D-lactic acid in 46% ethanol solution was 194.18 mg/L with sour taste; while the L-lactic acid was 98.19 mg/L with sour taste. The recognition threshold of L-lactic acid was about half that of D-lactic acid, indicating that L-lactic acid has a stronger sour taste. The taste activity values (TAVs) of D-lactic acid and L-lactic acid were greater than 1 in most of the Baijiu samples, and the TAV of D-lactic acid was greater than that of L-lactic acid. The study showed that the lactic acid enantiomers contributed to the taste perception of Baijiu in most of the samples, and D-lactic acid contributed more to the Baijiu taste than L-lactic acid.

Prognostic significance of PAK family kinases in acute myeloid leukemia

Acute myeloid leukemia (AML) is a clonal and heterogeneous disease characterized by a myriad of genetic defects. Genetic abnormalities are powerful prognostic factors. P21-activated kinases (PAKs) are a kind of serine/threonine protein kinases, which is regulator of plenty of oncogenic signaling pathways. The clinical and prognostic value of PAKs in AML is unclear. A total of 155 AML patients with PAK expression data from The Cancer Genome Atlas database were enrolled in this study. Eighty-four patients underwent chemotherapy only, 71 also underwent allogeneic hematopoietic stem cell transplantation (allo-HSCT). In the chemotherapy-only group, high PAK3 and PAK7 expression were both bound up with poor EFS and OS (all P < 0.05). However, high PAK2 expressers had better EFS and OS (all P < 0.05). Multivariate analysis demonstrated that high PAK7 expression was an adverse independent prognostic factor in patients who received chemotherapy only. PAKs have no influence in EFS and OS in patients who underwent allo-HSCT. In conclusion, high PAK2 expression is a favorable prognostic factor, as to the high expression of PAK3 and PAK7, they are poor prognostic factors, and PAK7 has better prognostic value, but their prognostic effects can be offset by allo-HSCT.

Characteristics of Barotropic Energy Conversion in Rapid Intensifying and Decaying Tropical Cyclones Over the Western North Pacific

The prediction of the tropical cyclone (TC) intensity remains a difficult issue. This study analyzes local instantaneous barotropic kinetic energy conversion in the lower troposphere in rapid intensifying (RI) and rapid decaying (RD) TCs over the western North Pacific (WNP) during June through November from 1979 to 2017. The kinetic energy conversion to the synoptic eddies is separated for climatological mean, interannual and intraseasonal flows. It is found that the intraseasonal cyclonic flows display a northwest‐southeast orientation in the RI TCs, but a circular feature in the RD TCs. Intraseasonal and climatological mean zonal flows contribute together to the positive kinetic energy conversion in the northwest quadrant of the RI TCs. Intraseasonal meridional flows induce positive (negative) kinetic energy conversion in the northeast (south) part of the RD TCs. The kinetic energy conversion associated with interannual flows is small for both the RI and RD TCs. Plain Language Summary The damage caused by the tropical cyclones (TCs) is closely related to their intensity. Revealing the factors of the intensification of the TCs may help to improve the skill of prediction of the TC intensity and reduce the damage of the TCs. From the energetic point of view, the intensification of the TCs requires the conversion of energy from the background flows. Here, we compare the barotropic kinetic energy conversion in the rapid intensifying (RI) and decaying (RD) TCs over the western North Pacific during June through November. Our analysis reveals that the intraseasonal flows have the largest contribution to both the RI and decaying TCs through the kinetic energy conversion to the synoptic scale eddies. The climatological mean flows have a secondary contribution to the RI TCs. The contribution of the interannual flows to the kinetic energy conversion is small for both the RI and RD TCs. Our results signify the importance of the configuration and magnitude of the background flows relative to the synoptic scale disturbances in the barotropic kinetic energy conversion for the TC intensity changes. Characterization, High-Density Fermentation, and the Production of a Directed Vat Set Starter of Lactobacilli Used in the Food Industry: A Review

Lactobacilli have been widely concerned for decades. Bacteria of the genus Lactobacillus have been commonly employed in fermented food to improve the appearance, smell, and taste of food or prolong its shelf-life. They comprise 261 species (by March 2020) that are highly diverse at the phenotypic, ecological, and genotypic levels. Some Lactobacilli strains have been documented to be essential probiotics, which are defined as a group of living microorganisms that are beneficial to the health of the host when ingested in sufficiency. However, the characterization, high-density fermentation, and the production of a directed vat set (DVS) starter of Lactobacilli strains used in the food industry have not been systematically reported. This paper mainly focuses on reviewing Lactobacilli as functional starter cultures in the food industry, including different molecular techniques for identification at the species and strain levels, methods for evaluating Lactobacilli properties, enhancing their performance and improving the cell density of Lactobacilli, and the production techniques of DVS starter of Lactobacilli strains. Moreover, this review further discussed the existing problems and future development prospects of Lactobacilli in the food industry. The viability and stability of Lactobacilli in the food industry and gastrointestinal environment are critical challenges at the industrial scale. The new production equipment and technology of DVS starter of Lactobacilli strains will have the potential for large-scale application, for example, developing low-temperature spray drying, freezing granulation drying, and spray freeze-drying.

Contributions of Various Temporal Components of Large-Scale Parameters in Tropical Cyclone Genesis over the North Indian Ocean

This work compares the contributions of synoptic, intraseasonal, and interannual components of large-scale parameters to tropical cyclone (TC) genesis over the North Indian Ocean (NIO) from April to December from 1979 to 2020. A composite analysis is employed with respect to TC genesis time and location. It is shown that most TCs occur when the total sea surface temperature (SST) is between 28 and 30 °C and SST anomalies in three time ranges are small (with the magnitude less than 0.2 °C). The TCs form mostly when the anomalies of vertical zonal wind shear are between −6 and 6 m s−1 and total vertical zonal wind shear falls within −12 and −3 m s−1, with the synoptic component being a positive contributor. The intraseasonal component of vorticity and convergence in the low level, vertical motion and specific humidity in the middle level, and convection contributes dominantly to the TC genesis. Synoptic-scale tropical disturbances obtain barotropic kinetic energy from the climatological mean and intraseasonal flows, with the former dominant in the southeastern sector, and the latter dominant in the northwestern sector. The contributions of the three temporal components of environmental factors are compared for TC genesis between the Arabian Sea (AS) and Bay of Bengal (BOB) and between the early season (April through June) and late season (September through December). The relative contributions of the three temporal components of factors are also compared for the TC formation among the NIO, northern tropical Atlantic Ocean (NTA), Northwestern Pacific (WNP), and Northeastern Pacific (ENP).

Westward shift of tropical cyclogenesis over the southern Indian Ocean

Tropical cyclones (TCs), commonly called cyclones in the southern Indian Ocean (SIO), represent one of the most devastating disasters in the oceanfront regions of Africa. The present study explores the long-term tendency of annual mean TC genesis location in the SIO. A notable westward shift is detected in the SIO TC genesis longitude since 1979, which is linked to an increase in the TC genesis frequency in the southwestern SIO and a decrease in the TC genesis frequency in the northeastern SIO. The dipole trend pattern of the TC genesis frequency in the SIO is intimately linked to the weakening of the westerly vertical wind shear over the western SIO and the strengthening of the easterly vertical wind shear over the eastern SIO, resulting from a reduced meridional temperature gradient. The weakened meridional temperature gradient is attributed to the enhanced warming of the subtropical troposphere that is a response of atmospheric temperature to global warming. Our study implies a potential increase in the risks faced by coastal and island countries in eastern Africa.
