Divertor Tokamak Test facility project: status of design and implementation
An overview is presented of the progress since 2021 in the construction and scientific programme preparation of the Divertor Tokamak Test (DTT) facility. Licensing for building construction has been granted at the end of 2021. Licensing for Cat. A radiologic source has been also granted in 2022. The construction of the toroidal field magnet system is progressing. The prototype of the 170 GHz gyrotron has been produced and it is now under test on the FALCON facility. The design of the vacuum vessel, the poloidal field coils and the civil infrastructures has been completed. The shape of the first DTT divertor has been agreed with EUROfusion to test different plasma and exhaust scenarios: single null, double null, X-divertor and negative triangularity plasmas. A detailed research plan is being elaborated with the involvement of the EUROfusion laboratories.
The Dynamical Asymmetry in SARS-CoV2 Protease Reveals the Exchange Between Catalytic Activity and Stability in Homodimers
The molecular approach to understanding the mechanisms of emerging diseases, like COVID-19, has largely accelerated the search for successful therapeutical strategies. In this work, we present an extensive molecular dynamics (MD) analysis of two forms of the SARS-CoV-2 main protease MPro. We analyzed the free form (apo) and compared the results with those coming from the (holo) form bound to the inhibitor Boceprevir, an FDA-approved drug repurposed for COVID-19 therapy. We applied Dynamic Cross Correlation (DCC) analysis to the MD simulations to trace the concerted motion patterns within the protein structure. Although symmetric, the homodimer in the bound form showed clearly asymmetric dynamical behavior. In particular, the presence of concerted motions was detected in the protomer where the expulsion of the substrate from the active site happened. Such behavior was not observed in the same time lapses in the apo form. These results highlight a sort of ‘symmetry breaking’, making a symmetric structure to display functional induced asymmetric behavior in response to a perturbation. This highly coordinated dynamics in response to an external cue confirms the character of ‘complex molecular machines’ of biopolymers.
Family ownership and the export performance of SMEs: the moderating role of financial constraints and flexibility
PurposeThis article investigates the relationship between family ownership and export performance in the context of SMEs while also considering the moderating role of the financial dimension and, in particular, financial constraints and financial flexibility.Design/methodology/approachWe select a sample of 1,132 Italian SMEs to examine through an econometric analysis the role and impact of family ownership and the financial moderating variables being used on their export performance.FindingsThe results indicate that there is a U-shaped relationship between family ownership and export performance: the highest levels of export performance correspond to the lowest and highest family ownership levels, whereas when a mixture of family and nonfamily ownership exists, the performance suffers because of “conflicting voices” dominating strategic visions and approaches, harming the firm's export commitment. Moreover, the findings show that lower financial constraints and/or stronger financial flexibility improve the relationship between family ownership and export performance.Research limitations/implicationsOur findings show that the ownership structure is important for export performance; in particular, firms should avoid a mixture between family and nonfamily ownership because it is detrimental to export performance. Moreover, Italian SMEs need to develop sources of financing other than the banking channel, and policy makers should favour this process to overcome financial constraint problems and improve financial flexibility. Limitations concern the use of other econometric approaches and measurement variables to further investigate the connection between family ownership and export performance.Originality/valueThe present study enhances the comprehension of the complex relationship between family ownership and export performance by documenting the relevance of the level of family ownership and considering the moderating role of financial constraints and flexibility.
Collective exploitation of large prey by group foraging shapes aggregation and fitness of cnidarian polyps
Group living is widespread and beneficial to metazoans. It improves protection and survival opportunities, reinforcing interspecific competitiveness. Benthic cnidarians often colonize large surfaces. Evidence of collective capture and exploitation of large prey by small, clumped polyps suggests that aggregation is functional to access food resources hardly achievable by isolated individuals. In turn, the chance to catch large prey may represents a driver of aggregation in polyps, whether beneficial to their fitness. Here, the effects of group foraging on aggregation, asexual reproduction, and growth rates of Aurelia coerulea von Lendenfeld 1884 polyps were experimentally tested by providing them with either small or large prey, or a mix of both to simulate the co-occurrence of preys at sea. As expected, some polyps were not able to reach the large prey. Hence, the population was a posteriori divided into group-foragers and solitary-feeders. In general, the large prey diet resulted in higher population fitness and when simultaneously supplied with the small prey represented an energetic booster resulting beneficial for all group-members. The decrease of interindividual distances was reported among group-foragers, that converged towards each other. Cnidarians are basal in metazoan evolution, and the comprehension of their collective foraging behavior, as well as the processes leading to the selective feature driving them to forage in group or not, may be essential to better understand the evolution and spread of social foraging in animals. Moreover, the access to large prey by sessile polyps of Aurelia coerulea could be pivotal in determining the increase in abundance of adult bloom-forming medusae.
Fetal-Type Rhabdomyoma of the Cheek: A Conservative Management
Extracardiac rhabdomyomas are rare benign mesenchymal tumors diagnosed upon radiological and hystologic investigations and the treatment of choice is surgical exertion. There aren’t any similar cases managed conservatively reported in literature as in our case, to the best of our knowledge. We present a rare case of fetal cheek rhabdomyoma diagnosed in a healthy 2 months-old boy, with asymptomatic mass over the left masseter. The lesion could not be removed, due to the size and dimensions and the young age of the patient. However, the lesion did not show signs of spreading or progression over a 36 months follow-up. Fetal rhabdomyoma is a benign tumor, often located in the head and neck district, where surgery, especially in very young children, is associated with a high risk of complications and long-term sequelae. Our case report demonstrates the possibility to manage these lesions conservatively in the first years of life with close sonographic and clinical follow-up.
The detection of specific prey cues triggers distinct predatory behaviour in Aurelia coerulea polyps (Cnidaria: scyphozoa)
HIGHLIGHTS Preys of different size are captured by cnidarian polyps with distinct predatory behaviours; Collective capture of large prey consists of active grabbing and rapid movements; Contact among polyps has been reported during collective large prey capture; The detection of prey-produced cues could trigger specific predatory behaviour in polyps; In addition to tactile stimulation, olfactory signalling could play a role in eliciting predation in benthic animals.
Mechanically‐Driven Vase–Kite Conformational Switch in Cavitand Cross‐Linked Polyurethanes
The eligibility of tetraquinoxaline cavitands (QxCav) as molecular grippers relies on their unique conformational mobility between a closed (vase) and an open (kite) form, triggered in solution by conventional stimuli like pH, temperature and ion concentration. In the present paper, the mechanochemical conformational switching of ad hoc functionalized QxCav covalently embedded in an elastomeric polydimethylsiloxane and in a more rigid polyurethane matrix is investigated. The rigid polymer matrix is more effective in converting mechanical force into a conformational switch at the molecular level, provided that all four quinoxaline wings are covalently connected to the polymer. Controlled switch: The Vase‐kite conformational switching of polymer embedded tetraquinoxaline cavitands is triggered by mechanical force.
Single and combined effects of two trace elements (Cd and Cu) on the asexual reproduction of Aurelia sp. polyps
Jellyfish blooms are an increasingly common event in our seas. Occurring via polyps’ asexual reproduction induced by human stresses, they represent a hazard for ecosystems equilibrium. The aim of this study is to highlight polyps underrated role during these events by investigating cadmium (Cd) and copper (Cu) exposure on Aurelia sp. polyps under laboratory conditions. Cd treatments seemed to not cause toxic effects on polyps up to 1000 μg/L, while 150 μg/L of Cu resulted above polyps’ tolerance for the metal, leading to a 62% of mortality and inducing the regression-regeneration cycle of polyps. Surprisingly, combined treatments of Cd and Cu had lesser effects, working antagonistically. Our results show how, in natural conditions, the chosen concentrations and combinations could not represent a hazard for polyps, instead, they stimulate the asexual reproduction, supporting the hormesis and, through jellyfish blooms, altering the Good Environmental Status aimed by the Marine Strategy Framework Directive.
Pleural drainage vs video-assisted thoracoscopic debridement in children affected by pleural empyema
Background Both thoracic drainage and video-assisted thoracic surgery (VATS) are available treatment for pleural empyema in pediatric patients. Materials and methods This retrospective multicenter study includes pediatric patients affected by pleural empyema treated from 2004 to 2021 at two Italian centers. Patients were divided in G1 (traditional approach) and G2 (VATS). Demographic and recovery data, laboratory tests, imaging, surgical findings, post-operative management and follow-up were analyzed. Results 70 patients with a mean age of 4.8 years were included; 12 (17.1%) in G1 and 58 (82.9%) in G2. Median surgical time was 45 min in G1, 90 in G2 ( p  < 0.05). Mean duration of thoracic drainage was 7.3 days in G1, 6.2 in G2 ( p  > 0.05). Patients became afebrile after a mean of 6.4 days G1, 3.9 in G2 ( p  < 0.05). Mean duration of antibiotic therapy was 27.8 days in G1, 25 in G2 ( p  < 0.05). Mean duration of postoperative hospital stay was 16 days in G1, 12.1 in G2 ( p  < 0.05). There were 4 cases (33.3%) of postoperative complications in G1, 17 (29.3%) in G2 ( p  > 0.05). 2 (16.7%) patients of G1 needed a redosurgery with VATS, 1 (1.7%) in G2. Conclusions VATS is an effective and safe procedure in treatment of Pleural Empyema in children: it is associated to reduction of chest tube drainage, duration of fever, hospital stay, time of antibiotic therapy and recurrence rate.
