Variability of Calodium hepaticum eggs from sigmodontine host species through geometric morphometric analysis

Calodium hepaticum is a zoonotic nematode with a worldwide distribution. Although the host range of C. hepaticum includes a wide spectrum of mammals (including humans), this parasite is predominantly associated with the families Muridae and Cricetidae. Several Sigmodontinae species from Argentina were found to be infected by C. hepaticum, with a high prevalence in Akodon azarae. The present study focuses on C. hepaticum eggs from natural infection of three species of sigmodontine rodents from Argentina. Eggs were genetically characterized (intergenic 18S rRNA region). The objectives of this work are: (i) to propose a new analytical methodology; and (ii) to morphologically characterize C. hepaticum eggs, from three Sigmodontinae species (A. azarae, Calomys callidus and Oligoryzomys flavescens). Analyses were made by the Computer Image Analysis System based on the new standardized measurements and geometric morphometric tools. The resulting factor maps clearly illustrate global size differences in the parasite eggs from the three Sigmodontinae species analysed. The degree of similarity between egg populations was assessed through pairwise Mahalanobis distances, showing that the largest distances were detected between parasite eggs from C. callidus and O. flavescens. Herein, the phenotypical plasticity of C. hepaticum eggs is shown. Significant positive correlations were obtained between each egg parasite principal component 1 and rodent corporal characteristics: weight; liver weight; rodent length; and rodent body condition. The usefulness of the geometric morphometric analysis in studies of the relationship between C. hepaticum and its host must be highlighted. The high prevalence observed in A. azarae, associated with the wide size range of the parasite eggs evidenced by principal component analysis, suggests A. azarae to be the Sigmodontinae host species that plays the most important role as reservoir host for C. hepaticum in the New World.

Calodium hepaticum (Trichuroidea: Capillariidae) Infection in Akodon azarae: First Description of Histopathological Lesions in Sigmodontinae

Calodium hepaticum (Trichinellida: Capillaridae) is a parasitic nematode of mammals distributed worldwide. Although this parasite can infect the liver of a wide diversity of mammals (including humans), it is mostly associated with Muroidea hosts. Sigmodontinae rodents were recently recognized as important hosts of this parasite in Argentina, but the impact of this parasitism on these hosts has not been established. Here we report results of histopathological analyses of 40 livers of Akodon azarae infected with C. hepaticum. Lesions were classified into 4 categories: level 0, absence of lesions; level 1, with focal granulomatous hepatitis; level 2, presence of multifocal granulomatous hepatitis, fibrosis and focal necrosis with neutrophils, and level 3, absence of intact adult parasites, diffuse distribution, necrosis, and fibrosis. Most samples presented lesions of level 2 (55%), but all categories of lesions were found. This is the first study to describe the lesions caused by C. hepaticum in the liver of Sigmodontinae rodents, and the results suggest that infection by this parasite is costly to A. azarae populations.

The effects of postural training on postural stability and vestibulospinal reflexes (VSRs) were investigated in normal subjects. A period (23 minutes) of repeated episodes (n = 10, 50 seconds) of unipedal stance elicited a progressive reduction of the area covered by centre of pressure (CoP) displacement, of average CoP displacement along the X and Y axes and of CoP velocity observed in this challenging postural task. All these changes were correlated to each other with the only exception of those in X and Y CoP displacement. Moreover, they were larger in the subjects showing higher initial instability in unipedal stance, suggesting that they were triggered by the modulation of sensory afferents signalling body sway. No changes in bipedal stance occurred soon and 1 hour after this period of postural training, while a reduction of CoP displacement was apparent after 24 hours, possibly due to a beneficial effect of overnight sleep on postural learning. The same period of postural training also reduced the CoP displacement elicited by electrical vestibular stimulation (EVS) along the X axis up to 24 hours following the training end. No significant changes in postural parameters of bipedal stance and VSRs could be observed in control experiments where subjects were tested at identical time points without performing the postural training. Therefore, postural training led to a stricter control of CoP displacement, possibly acting through the cerebellum by enhancing feedforward mechanisms of postural stability and by depressing the VSR, the most important reflex mechanism involved in balance maintenance under challenging conditions.

Predicting neuropsychological late effects in pediatric brain tumor survivors using the Neurological Predictor Scale and the Pediatric Neuro-Oncology Rating of Treatment Intensity

The Neurological Predictor Scale (NPS) quantifies cumulative exposure to tumor- and treatment-related neurological risks. The Pediatric Neuro-Oncology Rating of Treatment Intensity (PNORTI) measures the intensity of different treatment modalities, but research is needed to establish whether it is associated with late effects. This study evaluated the predictive validity of the NPS and PNORTI for neuropsychological outcomes in pediatric brain tumor survivors.
A retrospective chart review was completed of pediatric brain tumor survivors (PBTS) (
= 161,
= 13.47,
= 2.80) who were at least 2 years from the end of tumor-directed treatment. Attention, intellectual functioning, perceptual reasoning, processing speed, verbal reasoning, and working memory were analyzed in relation to the NPS and PNORTI.
NPS scores ranged from 1 to 11 (
= 5.57,
= 2.27) and PNORTI scores ranged from 1 (
= 101; 62.7%) to 3 (
= 18; 11.2%). When controlling for age, sex, SES factors, and time since treatment, NPS scores significantly predicted intellectual functioning [
(7,149) = 12.86,
< .001,
= .38] and processing speed [
(7,84) = 5.28,
< .001,
= .31]. PNORTI scores did not significantly predict neuropsychological outcomes.
The findings suggest that the NPS has value in predicting IF and processing speed above-and-beyond demographic variables. The PNORTI was not associated with neuropsychological outcomes. Future research should consider establishing clinical cutoff scores for the NPS to help determine which survivors are most at risk for neuropsychological late effects and warrant additional assessment.

To date, the literature has failed to individuate a clear motivation for the performance decrement after a mental fatigue-inducing task. This study aimed to evaluate biomechanical and perceptual variables during a swimming training session in different mental fatigue states. Seven amateur triathletes watched a documentary, utilized a smartphone, or performed an AX-CPT for 45 min randomly on three different days. After, they performed a 15-min warm-up followed by 6 × 200 m at constant pre-set speed plus one 200 m at maximal effort. The mental fatigue status was assessed by the visual analog scale (VAS) and short-Stroop task results before, post-mental task, and post-swimming session. The biomechanical and motor coordination variables during swimming were assessed using five IMU sensors and video analysis. The heart rate and rate of perceived exertion were monitored during the task. No differences in biomechanical and perceptual variables were found between and within conditions. Higher mental fatigue was found only in the AX-CPT condition at post task by VAS. In this preliminary study, no changes in swimming biomechanics were highlighted by mental fatigue, but the warm-up performed may have counteracted its negative effects. Further studies are recommended.

Apparent diffusion coefficient obtained by magnetic resonance imaging as a prognostic marker in glioblastomas: correlation with MGMT promoter methylation status

Objective
To evaluate whether apparent diffusion coefficient (ADC) values can predict the status of MGMT of glioblastoma multiforme (GBM) and correlate with overall survival (OS) and progression-free survival (PFS).
Methods
This retrospective study included 47 patients with pathologically proven glioblastoma. All of them underwent MR DWI study before surgery (mean time 1 week) and the status of methylguanine-DNA-methyltransferase (MGMT) promoter methylation was searched for. Minimum apparent diffusion coefficient (ADC) values were evaluated. OS and PSF parameters were calculated, and Student’s t-test, Kaplan-Meier curves, linear and Cox regression were performed.
Results
Twenty-five patients showed positive methylation of the MGMT promoter. Patients showing MGMT promoter methylation had higher minimum ADC values, and they survived longer than those without MGMT promoter methylation. The median ADCmin value of 0.80 represents the cutoff value able to distinguish between methylated and un-methylated patients. Patients showing minimum ADC values higher than 0.80 survived longer than patients with minimum ADC values lower than 0.80. A linear correlation between minimum ADC values vs. the OS and PFS was observed.
Conclusions
Minimum ADC values in glioblastoma multiforme could be used as a preoperative parameter to estimate the status of MGMT promoter methylation and the survival of patients.

Impact of species hybridization on the clinical management of schistosomiasis: A prospective study

Species hybridization represents a real concern in terms of parasite transmission, epidemiology and morbidity of schistosomiasis. It is greatly important to better understand the impact of species hybridization for the clinical management.
A prospective observational study was carried out in sub-Saharan migrants who were diagnosed with confirmed genitourinary schistosomiasis. A tailored protocol was applied, including Schistosoma serology, a specific urine LAMP tests for schistosomiasis and an ultrasound examination before treatment with praziquantel. A scheduled follow-up was performed at 3, 6 and 12 months to monitor treatment response, comparing patients carriers of Schistosoma hybrids with carriers of only genetically pure forms.
A total of 31 male patients from West Africa were included in the study with a mean age of 26.5 years. Twelve (38.7 %) of the patients were carriers of Schistosoma hybrids. As compared with patients infected with S. haematobium alone, hybrid carriers had lower haemoglobin levels (13.8 g/dL [SD 1.8] vs 14.8 g/dL [SD 1.4], p = 0.04), a greater frequency of hematuria (100 % vs 52.6 %, p = 0.005), a higher ultrasound score (2.64, SD 2.20 vs 0.89, SD 0.99; p = 0.02). However, the presence of hybrids did not result in differences in clinical and analytical responses after treatment.
The presence of Schistosoma hybrids seems to cause increased morbidity in infected individuals. However, it does not appear to result in differences in diagnostic tests or in clinical and analytical responses after treatment.
•Hybridization of schistosomes is prevalent in patients with imported schistosomiasis.•Migrants with the presence of hybrid schistosomes present a higher morbidity.•Hybridization does not appear to affect the sensitivity of diagnostic methods.•Hybrid schistosomes does not cause differences in the clinical response to treatment.

Calodium hepaticum (Nematoda; Capillariidae) is a parasitic nematode of mammals with a cosmopolitan distribution. Adults of this nematode can infect the liver of many mammalian species, including humans, but the main hosts are members of the superfamily Muroidea. Among these, Rattus spp. have the highest apparent prevalences reported worldwide. There are only two reports of C. hepaticum infecting Sigmodontinae species (Muroidea: Cricetidae). In this survey, we examined the occurrence of C. hepaticum in two assemblages of Sigmodontinae rodents from Argentina (Santa Fe and Entre Ríos provinces). The diagnosis was made by morphological features, histopathological exam, and molecular characterization of 18S ribosomal RNA gene region. Here, we show that C. hepaticum is a common parasite of Sigmodontinae. We report the infection in six species (all new hosts): Akodon azarae, Calomys callidus, Calomys venustus, Oligoryzomys flavescens, Oligoryzomys nigripes, and Oxymycterus rufus. This is the first report of C. hepaticum in Sigmodontinae rodents from Argentina and the second record in this subfamily for South America. It is also the first confirmation of C. hepaticum infection in Sigmodontinae by molecular diagnosis. The genetic findings and the prevalences observed, together with the existing information on C. hepaticum, lead us to propose that Rattus spp. brought C. hepaticum to the New World exposing Sigmodontinae rodents, which are frequently infected by this parasite. The high prevalence of infection in A. azarae (41.2%) suggests that this host may be playing an important role in C. hepaticum dynamics in the New World.

Geological and geotechnical models definition for 3rd level seismic microzonation studies in Central Italy

The 2016–2017 seismic events that struck central Italy led the Government to carry out a project to produce the third level Seismic Microzonation studies in 138 municipalities. These activities have involved many experts in different disciplines such as geology, geomorphology, geophysics, seismology and geotechnical engineering. This project represented the first opportunity to perform nationally coordinated third level Seismic Microzonation studies over a wide area in a quite short time (6 months). It provided the chance to improve methodological procedures, to test the reliability of methods and models for site response analyses and to produce a huge amount of validated data. This paper focuses on the contribution of geological disciplines and concerns: (a) the definition of the main “morphostructural domains” of the Central-Northern Apennines; (b) the creation of an archive of all the lithostratigraphic units occurring in the study area with their conversion into engineering-geological units and their distribution in the different morphostructural domains; (c) the construction of the reference geological and geotechnical models, which are essential to classify the territory into seismically homogeneous microzones and to perform the successive 1D and 2D numerical analyses of the local site response. The geophysical dataset acquired for the study allowed a first statistical characterization of the Vs values typical of the engineering-geological units identified in this study. Some examples of the recurrent geological and geotechnical models are shown to explain the complexity and variety of the geological and geomorphological features of the investigated area and to highlight the different seismostratigraphic behavior of rocks and cover terrains. The analysis of third level Seismic Microzonation data made it possible to identify recurrent subsoil models and to note the main stratigraphic and morphological control-factors of the ground motion modification in the different morphostructural domains.
