The most detailed anatomical reconstruction of a Mesozoic coelacanth

Although the split of coelacanths from other sarcopterygians is ancient, around 420 million years ago, the taxic diversity and the morphological disparity of the clade have remained relatively low, with a few exceptions. This supposedly slow evolutionary pace has earned the extant coelacanth Latimeria the nickname “living fossil”. This status generated much interest in both extinct and extant coelacanths leading to the production of numerous anatomical studies. However, detailed descriptions of extinct taxa are made difficult due to the quality of the fossil material which generally prevents fine comparisons with the extant Latimeria . Here we describe a new genus and species of coelacanth, Graulia branchiodonta gen. et sp. nov. from the Middle Triassic of Eastern France, based on microtomographical imaging using synchrotron radiation. Through exquisite 3D preservation of the specimens, we reconstructed the skeletal anatomy of this new species at an unprecedented level of detail for an extinct coelacanth, and barely achieved for the extant Latimeria . In particular, we identified a well-developed trilobed ossified lung whose function is still uncertain. The skeletal anatomy of G . branchiodonta displays the general Bauplan of Mesozoic coelacanths and a phylogenetic analysis resolved it as a basal Mawsoniidae, shedding light on the early diversification of one of the two major lineages of Mesozoic coelacanths. However, despite its exquisite preservation, G . branchiodonta carries a weak phylogenetic signal, highlighting that the sudden radiation of coelacanths in the Early and Middle Triassic makes it currently difficult to detect synapomorphies and resolve phylogenetic interrelationships among coelacanths in the aftermath of the great Permo-Triassic biodiversity crisis.

Evaluating Recent Advances in Affective Intelligent Tutoring Systems: A Scoping Review of Educational Impacts and Future Prospects
Affective intelligent tutoring systems (ATSs) are gaining recognition for their role in personalized learning through adaptive automated education based on students’ affective states. This scoping review evaluates recent advancements and the educational impact of ATSs, following PRISMA guidelines for article selection and analysis. A structured search of the Web of Science (WoS) and Scopus databases resulted in 30 studies covering 27 distinct ATSs. These studies assess the effectiveness of ATSs in meeting learners’ emotional and cognitive needs. This review examines the technical and pedagogical aspects of ATSs, focusing on how emotional recognition technologies are used to customize educational content and feedback, enhancing learning experiences. The primary characteristics of the selected studies are described, emphasizing key technical features and their implications for educational outcomes. The discussion highlights the importance of emotional intelligence in educational environments and the potential of ATSs to improve learning processes. This review identifies gaps in the current research and suggests future directions, including broader implementation across diverse educational settings and deeper integration of affective data to refine system responsiveness. Future research should investigate the integration of advanced natural dialogue modules and generative AI to create more sophisticated interfaces, underscoring the role of affective adaptation in educational technology.

In the present article, we describe a study of antitumor activity in breast cell lines using silver nanoparticles (Ag NPs) synthesized by a microbiological method. These Ag NPs were tested for their antitumor activity against MCF7 and T47D cancer cells and MCF10-A normal breast cell line. We analyzed cell viability, apoptosis induction, and endocytosis activity of those cell lines and we observed that the effects of the biosynthesized Ag NPs were directly related with the endocytosis activity. Moreover, Ag NPs had higher inhibition efficacy in tumor lines than in normal lines of breast cells, which is due to the higher endocytic activity of tumor cells compared to normal cells. In this way, we demonstrate that biosynthesized Ag NPs can be an alternative for the treatment of tumors.

Dependence and resource commitment as antecedents of supply chain integration
PurposeThe purpose of this paper is to explore and further the existing knowledge on supply chain integration (SCI). This study proposes a model and several hypotheses to better understand some SCI antecedents, dependence and resource commitment and their relationships with performance.Design/methodology/approachBased on diverse theoretical approaches, the author develops and tests an integrated model in which dependence and resource commitment are proposed to enhance external integration, leading to an increase in economic performance. This study's empirical validity is reinforced by collecting data from 142 manufacturing firms in Spain and Germany and testing the model using structural equation model (SEM).FindingsThe results support dependence and resource commitment as antecedents of SCI, both with a positive effect. Also, discrepancies in the effect of external integration on performance are found where supplier integration seems not to have any effect on performance.Originality/valueThis study helps to better understand SCI antecedents. It makes both theoretical and managerial contributions by empirically analyzing both antecedents. This furthers extant knowledge regarding the joined impact of resource commitment and dependence on SCI. In particular, it incorporates resource commitment by considering it as the sacrifice firms need to implement to get involved in a long-term relationship.

Payments for environmental services supported social capital while increasing land management

Payments for environmental services (PES) programs incentivize landowners to protect or improve natural resources. Many conservationists fear that introducing compensation for actions previously offered voluntarily will reduce social capital (the institutions, relationships, attitudes, and values that govern human interactions), yet little rigorous research has investigated this concern. We examined the land cover management and communal social capital impacts of Mexico’s federal conservation payments program, which is a key example for other countries committed to reducing deforestation, protecting watersheds, and conserving biodiversity. We used a regression discontinuity (RD) methodology to identify causal program effects, comparing outcomes for PES participants and similar rejected applicants close to scoring cutoffs. We found that payments increased land cover management activities, such as patrolling for illegal activity, building fire breaks, controlling pests, or promoting soil conservation, by ∼50%. Importantly, increases in paid activities as a result of PES did not crowd out unpaid contributions to land management or other prosocial work. Community social capital increased by ∼8–9%, and household-level measures of trust were not affected by the program. These findings demonstrate that major environmental conditional cash transfer programs can support both land management and the attitudes and institutions underpinning prosocial behavior. Rigorous empirical research on this question can proceed only country by country because of methodological limitations, but will be an important line of inquiry as PES continues to expand worldwide.

Discovery and Functional Validation of EP3 Receptor Ligands with Therapeutic Potential in Cardiovascular Disease

The prostaglandin E2 receptor EP3 is emerging as a promising therapeutic target in cardiovascular diseases because of its involvement in vascular inflammation, platelet aggregation, and vasoconstriction. However, selective EP3 ligands with validated biological activities are scarce. In this study, we combined computational and experimental strategies to identify and validate novel EP3 receptor ligands with therapeutic potential. We implemented a high-throughput, structure- and ligand-based virtual screening pipeline, enabling efficient exploration of approved drugs and natural compounds from DrugBank and FooDB libraries. Top-scoring candidates were prioritised based on binding energy and pharmacophoric similarity. Selected hits were subjected to in silico ADME/Tox profiling using QikProp to identify molecules with favourable pharmacokinetic and safety parameters. TUCA, masoprocol, and pravastatin sodium have emerged as lead candidates and were validated in vitro using endothelial migration and platelet aggregation assays. TUCA exhibited the most consistent inhibitory effect on endothelial migration, whereas masoprocol and hydrocortisone significantly reduced platelet aggregation. These findings establish a multidimensional workflow for the rational identification of EP3 ligands and support their potential use in cardiovascular therapeutics.

Interdisciplinary Reservoir Management—A Tool for Sustainable Water Resources Management

Reservoirs are a common way to store and retain water serving for a multitude of purposes like storage of drinking and irrigation water, recreation, flood protection, navigation, and hydropower production, and have been built since centuries. Today, few reservoirs serve only one purpose, which requires management of present demands and interests. Since each reservoir project will cause negative impacts alongside desired advantages both on a local, regional and global scale, it is even more urgent to develop a common management framework in an attempt to mitigate negative impacts, incorporate different demands and make them visible within the discourse in order to avoid conflicts from early on. The scientific publications on reservoirs are manifold, yet a comprehensive and integrative holistic tool about management of this infrastructure is not available. Therefore, a comprehensive and integrated conceptual tool was developed and proposed by the authors of this paper that can contribute to the sustainable management of existing reservoirs. The tool presented herein is based on the results from the interdisciplinary CHARM (CHAllenges of Reservoir Management) project as well as the condensed outcome of relevant literature to aid and enhance knowledge of reservoir management. The incorporated results are based on field, laboratory and empirical social research. The project CHARM focused on five different aspects related to existing reservoirs in southern Germany (Schwarzenbachtalsperre, Franconian Lake District), namely: sedimentation of reservoirs, biostabilisation of fine sediments, toxic cyanobacteria(l) (blooms), greenhouse gas emissions from reservoirs and social contestation, respectively consent. These five research foci contributed to the topics and setup of a conceptual tool, put together by the research consortium via delphi questioning, which can be found alongside this publication to provide insights for experts and laymen. Conceptualising and analysing the management in combination with quantitative and qualitative data in one descriptive tool presents a novelty for the case studies and area of research. The distribution within the scientific community and interested public will possibly make a positive contribution to the goal of sustainable water resources management in the future.

Highly pathogenic H5N1 avian influenza viruses (HPAIV) belonging to lineage 2.3.4.4b emerged in Chile in December 2022, leading to mass mortality events in wild birds, poultry, and marine mammals and one human case. We detected HPAIV in 7,33% (714/9745) of cases between December 2022–April 2023 and sequenced 177 H5N1 virus genomes from poultry, marine mammals, a human, and wild birds spanning >3800 km of Chilean coastline. Chilean viruses were closely related to Peru’s H5N1 outbreak, consistent with north-to-south spread down the Pacific coastline. One human virus and nine marine mammal viruses in Chile had the rare PB2 D701N mammalian-adaptation mutation and clustered phylogenetically despite being sampled 5 weeks and hundreds of kilometers apart. These viruses shared additional genetic signatures, including another mammalian PB2 adaptation (Q591K,
n
= 6), synonymous mutations, and minor variants. Several mutations were detected months later in sealions in the Atlantic coast, indicating that the pinniped outbreaks on the west and east coasts of South America are genetically linked. These data support sustained mammal-to-mammal transmission of HPAIV in marine mammals over thousands of kilometers of Chile’s Pacific coastline, which subsequently continued through the Atlantic coastline.
Highly pathogenic H5N1 avian influenza lineage 2.3.4.4b has spread through the Americas in birds since 2021 with frequent spillover into mammals. Here, the authors characterise the dissemination of the virus in Chile and find evidence of sustained transmission between mammals.
