Strong graphene oxide nanocomposites from aqueous hybrid liquid crystals

Combining polymers with small amounts of stiff carbon-based nanofillers such as graphene or graphene oxide is expected to yield low-density nanocomposites with exceptional mechanical properties. However, such nanocomposites have remained elusive because of incompatibilities between fillers and polymers that are further compounded by processing difficulties. Here we report a water-based process to obtain highly reinforced nanocomposite films by simple mixing of two liquid crystalline solutions: a colloidal nematic phase comprised of graphene oxide platelets and a nematic phase formed by a rod-like high-performance aramid. Upon drying the resulting hybrid biaxial nematic phase, we obtain robust, structural nanocomposites reinforced with graphene oxide.
Reinforcing polymers with carbon-based nanofillers is non-trivial due to incompatibilities between matrix and filler. Here, the authors report highly reinforced graphene oxide–aramid nanocomposites utilizing a water-based hybrid biaxial nematic mixture.

We report on the rheology of isotropic and nematic aqueous solutions of a sulfonated all-aromatic polyamide, poly(2,2′-disulfonyl-4,4′-benzidine terephthalamide) (PBDT), that forms high-aspect-ratio rod-like assemblies. For quiescently isotropic solutions, the concentration dependence of the zero-shear viscosity, longest relaxation time, and terminal modulus shows deviations in comparison to the Doi-Edwards theory for hard rods. For quiescently nematic solutions, we characterize the flow behavior through steady-state and transient nonlinear rheological measurements in conjunction with small-angle neutron scattering under shear. The steady-state flow curve is characterized by two anomalous shear thickening responses, one at moderate shear rates and the other immediately prior to flow alignment at high shear rates. We assign the origin of these shear thickening response to director “kayaking” and “out-of-plane steady” states, using predictions from prior high-resolution numerical simulations of sheared nematic rods. Utilizing transient shear flow reversals and step-down experiments, we characterize the oscillatory response of the nematic director through these flow regimes. When the first normal stress difference is plotted versus the shear stress during a transient step-down, the so-called dynamic stress path, the counterclockwise versus clockwise rotation has previously been shown to reveal the relative dominance of viscous versus elastic contributions to the stress tensor, respectively. Our measurements strongly suggest that the anomalous shear thickening behavior in nematic PBDT solutions arises from viscous stresses developed as the ensemble of rods undergoes periodic oscillatory motion under shear, rather than elastic stresses due to broadening of the orientational distribution function.

Detection of X-ray emission at a location coincident with the kilonova transient of the gravitational-wave event GW170817 provides the missing observational link between short gamma-ray bursts and gravitational waves from neutron-star mergers.
When neutron stars collide
Merging neutron stars are potential sources of gravitational waves and have long been predicted to produce jets of material as part of a low-luminosity transient known as a 'kilonova'. There is growing evidence that neutron-star mergers also give rise to short, hard gamma-ray bursts. A group of papers in this issue report observations of a transient associated with the gravitational-wave event GW170817—a signature of two neutron stars merging and a gamma-ray flash—that was detected in August 2017. The observed gamma-ray, X-ray, optical and infrared radiation signatures support the predictions of an outflow of matter from double neutron-star mergers and present a clear origin for gamma-ray bursts. Previous predictions differ over whether the jet material would combine to form light or heavy elements. These papers now show that the early part of the outflow was associated with lighter elements whereas the later observations can be explained by heavier elements, the origins of which have been uncertain. However, one paper (by Stephen Smartt and colleagues) argues that only light elements are needed for the entire event. Additionally, Eleonora Troja and colleagues report X-ray observations and radio emissions that suggest that the 'kilonova' jet was observed off-axis, which could explain why gamma-ray-burst detections are seen as dim.

Ocean community warming responses explained by thermal affinities and temperature gradients

As ocean temperatures rise, species distributions are tracking towards historically cooler regions in line with their thermal affinity1,2. However, different responses of species to warming and changed species interactions make predicting biodiversity redistribution and relative abundance a challenge3,4. Here, we use three decades of fish and plankton survey data to assess how warming changes the relative dominance of warm-affinity and cold-affinity species5,6. Regions with stable temperatures (for example, the Northeast Pacific and Gulf of Mexico) show little change in dominance structure, while areas with warming (for example, the North Atlantic) see strong shifts towards warm-water species dominance. Importantly, communities whose species pools had diverse thermal affinities and a narrower range of thermal tolerance showed greater sensitivity, as anticipated from simulations. The composition of fish communities changed less than expected in regions with strong temperature depth gradients. There, species track temperatures by moving deeper2,7, rather than horizontally, analogous to elevation shifts in land plants8. Temperature thus emerges as a fundamental driver for change in marine systems, with predictable restructuring of communities in the most rapidly warming areas using metrics based on species thermal affinities. The ready and predictable dominance shifts suggest a strong prognosis of resilience to climate change for these communities.

Plasma phospho-tau181 in presymptomatic and symptomatic familial Alzheimer’s disease: a longitudinal cohort study

Blood biomarkers have great potential to advance clinical care and accelerate trials in Alzheimer’s disease (AD). Plasma phospho-tau181 (p-tau181) is a promising blood biomarker however, it is unknown if levels increase in presymptomatic AD. Therefore, we investigated the timing of p-tau181 changes using 153 blood samples from 70 individuals in a longitudinal study of familial AD (FAD). Plasma p-tau181 was measured, using an in-house single molecule array assay. We compared p-tau181 between symptomatic carriers, presymptomatic carriers, and non-carriers, adjusting for age and sex. We examined the relationship between p-tau181 and neurofilament light and estimated years to/from symptom onset (EYO), as well as years to/from actual onset in a symptomatic subgroup. In addition, we studied associations between p-tau181 and clinical severity, as well testing for differences between genetic subgroups. Twenty-four were presymptomatic carriers (mean baseline EYO −9.6 years) while 27 were non-carriers. Compared with non-carriers, plasma p-tau181 concentration was higher in both symptomatic (p < 0.001) and presymptomatic mutation carriers (p < 0.001). Plasma p-tau181 showed considerable intra-individual variability but individual values discriminated symptomatic (AUC 0.93 [95% CI 0.85–0.98]) and presymptomatic (EYO ≥ −7 years) (AUC 0.86 [95% CI 0.72–0.94]) carriers from non-carriers of the same age and sex. From a fitted model there was evidence (p = 0.050) that p-tau181 concentrations were higher in mutation carriers than non-carriers from 16 years prior to estimated symptom onset. Our finding that plasma p-tau181 concentration is increased in symptomatic and presymptomatic FAD suggests potential utility as an easily accessible biomarker of AD pathology.

Lesion network localization of free will

Our perception of free will is composed of a desire to act (volition) and a sense of responsibility for our actions (agency). Brain damage can disrupt these processes, but which regions are most important for free will perception remains unclear. Here, we study focal brain lesions that disrupt volition, causing akinetic mutism (n = 28), or disrupt agency, causing alien limb syndrome (n = 50), to better localize these processes in the human brain. Lesion locations causing either syndrome were highly heterogeneous, occurring in a variety of different brain locations. We next used a recently validated technique termed lesion network mapping to determine whether these heterogeneous lesion locations localized to specific brain networks. Lesion locations causing akinetic mutism all fell within one network, defined by connectivity to the anterior cingulate cortex. Lesion locations causing alien limb fell within a separate network, defined by connectivity to the precuneus. Both findings were specific for these syndromes compared with brain lesions causing similar physical impairments but without disordered free will. Finally, our lesion-based localization matched network localization for brain stimulation locations that disrupt free will and neuroimaging abnormalities in patients with psychiatric disorders of free will without overt brain lesions. Collectively, our results demonstrate that lesions in different locations causing disordered volition and agency localize to unique brain networks, lending insight into the neuroanatomical substrate of free will perception.

Effect of IBD medications on COVID-19 outcomes: results from an international registry

ObjectiveWe sought to evaluate COVID-19 clinical course in patients with IBD treated with different medication classes and combinations.DesignSurveillance Epidemiology of Coronavirus Under Research Exclusion for Inflammatory Bowel Disease (SECURE-IBD) is a large, international registry created to monitor outcomes of IBD patients with confirmed COVID-19. We used multivariable regression with a generalised estimating equation accounting for country as a random effect to analyse the association of different medication classes with severe COVID-19, defined as intensive care unit admission, ventilator use and/or death.Results1439 cases from 47 countries were included (mean age 44.1 years, 51.4% men) of whom 112 patients (7.8%) had severe COVID-19. Compared with tumour necrosis factor (TNF) antagonist monotherapy, thiopurine monotherapy (adjusted OR (aOR) 4.08, 95% CI 1.73 to 9.61) and combination therapy with TNF antagonist and thiopurine (aOR 4.01, 95% CI 1.65 to 9.78) were associated with an increased risk of severe COVID-19. Any mesalamine/sulfasalazine compared with no mesalamine/sulfasalazine use was associated with an increased risk (aOR 1.70, 95% CI 1.26 to 2.29). This risk estimate increased when using TNF antagonist monotherapy as a reference group (aOR 3.52, 95% CI 1.93 to 6.45). Interleukin-12/23 and integrin antagonists were not associated with significantly different risk than TNF antagonist monotherapy (aOR 0.98, 95% CI 0.12 to 8.06 and aOR 2.42, 95% CI 0.59 to 9.96, respectively).ConclusionCombination therapy and thiopurines may be associated with an increased risk of severe COVID-19. No significant differences were observed when comparing classes of biologicals. The causes of phenotypic heterogeneity in familial Alzheimer’s disease with autosomal dominant inheritance are not well understood. We aimed to characterise clinical phenotypes and genetic associations with APP and PSEN1 mutations in symptomatic autosomal dominant familial Alzheimer’s disease (ADAD).
We retrospectively analysed genotypic and phenotypic data (age at symptom onset, initial cognitive or behavioural symptoms, and presence of myoclonus, seizures, pyramidal signs, extrapyramidal signs, and cerebellar signs) from all individuals with ADAD due to APP or PSEN1 mutations seen at the Dementia Research Centre in London, UK. We examined the frequency of presenting symptoms and additional neurological features, investigated associations with age at symptom onset, APOE genotype, and mutation position, and explored phenotypic differences between APP and PSEN1 mutation carriers. The proportion of individuals presenting with various symptoms was analysed with descriptive statistics, stratified by mutation type.
Between July 1, 1987, and Oct 31, 2015, age at onset was recorded for 213 patients (168 with PSEN1 mutations and 45 with APP mutations), with detailed history and neurological examination findings available for 121 (85 with PSEN1 mutations and 36 with APP mutations). We identified 38 different PSEN1 mutations (four novel) and six APP mutations (one novel). Age at onset differed by mutation, with a younger onset for individuals with PSEN1 mutations than for those with APP mutations (mean age 43·6 years [SD 7·2] vs 50·4 years [SD 5·2], respectively, p<0·0001); within the PSEN1 group, 72% of age at onset variance was explained by the specific mutation. A cluster of five mutations with particularly early onset (mean age at onset <40 years) involving PSEN1’s first hydrophilic loop suggests critical functional importance of this region. 71 (84%) individuals with PSEN1 mutations and 35 (97%) with APP mutations presented with amnestic symptoms, making atypical cognitive presentations significantly more common in PSEN1 mutation carriers (n=14; p=0·037). Myoclonus and seizures were the most common additional neurological features; individuals with myoclonus (40 [47%] with PSEN1 mutations and 12 [33%] with APP mutations) were significantly more likely to develop seizures (p=0·001 for PSEN1; p=0·036 for APP), which affected around a quarter of the patients in each group (20 [24%] and nine [25%], respectively). A number of patients with PSEN1 mutations had pyramidal (21 [25%]), extrapyramidal (12 [14%]), or cerebellar (three [4%]) signs.
ADAD phenotypes are heterogeneous, with both age at onset and clinical features being influenced by mutation position as well as causative gene. This highlights the importance of considering genetic testing in young patients with dementia and additional neurological features in order to appropriately diagnose and treat their symptoms, and of examining different mutation types separately in future research.
