Evaluation of Isolates of Entomopathogenic Fungi in the Genera Metarhizium,Beauveria, and Isaria, and Their Virulence to Thaumastocoris peregrinus (Hemiptera: Thaumastocoridae)

Thaumastocoris peregrinus Carpintero and Dellapé, or bronze bug (Hemiptera: Thaumastocoridae), is a pest found in eucalyptus plantations worldwide. Use of entomopathogenic fungi has emerged recently as an alternative means of control for T. peregrinus. Here we report laboratory experiments conducted to select isolates of entomopathogenic fungi for use to control this pest. Seven isolates of Beauveria bassiana (Bals.-Criv.) Vuill. (Hypocreales: Cordycipitaceae), 3 of Metarhizium anisopliae (Metschn.) Sorokīn (Hypocreales: Clavicipitaceae), 2 of Metarhizium robertsii J.F. Bisch., S.A. Rehner & Humber (Hypocreales: Clavicipitaceae), and 2 of Isaria fumosorosea Wize (Hypocreales: Cordycipitaceae) at a concentration of 1 × 108 conidia per mL were assessed for their virulence to adults of T. peregrinus. In vitro growth and sporulation of the isolates also were evaluated through the analysis of colony development and conidia production in PDA culture medium, at 26 ± 1 °C, 70% RH, and a 12:12 h (L:D) photoperiod. All tested isolates were found to be pathogenic to T. peregrinus. The isolates Bb3, Bb4, Bb7, Ma1, Ma2, Mr2, and If1 exhibited significant virulence to the insect. The greatest growth (longest mean diam of colony) was for the isolate Ma2, whereas the highest sporulation rates were observed in the isolates If2 and Ma2. This study demonstrated that isolates of Beauveria, Metarhizium, and Isaria can cause infection in T. peregrinus. The Ma2 isolate of Metarhizium stood out for its pathogenic effect on the adult insect, as well as for its capacity to grow rapidly and produce large numbers of conidia on an artificial medium, indicating its potential for use as a microbial control agent for T. peregrinus, and its suitability for mass production.

Mercury is a severe environmental pollutant with neurotoxic effects, especially when exposed for long periods. Although there are several evidences regarding mercury toxicity, little is known about inorganic mercury (IHg) species and cerebellum, one of the main targets of mercury associated with the neurological symptomatology of mercurial poisoning. Besides that, the global proteomic profile assessment is a valuable tool to screen possible biomarkers and elucidate molecular targets of mercury neurotoxicity; however, the literature is still scarce. Thus, this study aimed to investigate the effects of long-term exposure to IHg in adult rats’ cerebellum and explore the modulation of the cerebellar proteome associated with biochemical and functional outcomes, providing evidence, in a translational perspective, of new mercury toxicity targets and possible biomarkers. Fifty-four adult rats were exposed to 0.375 mg/kg of HgCl2 or distilled water for 45 days using intragastric gavage. Then, the motor functions were evaluated by rotarod and inclined plane. The cerebellum was collected to quantify mercury levels, to assess the antioxidant activity against peroxyl radicals (ACAPs), the lipid peroxidation (LPO), the proteomic profile, the cell death nature by cytotoxicity and apoptosis, and the Purkinje cells density. The IHg exposure increased mercury levels in the cerebellum, reducing ACAP and increasing LPO. The proteomic approach revealed a total 419 proteins with different statuses of regulation, associated with different biological processes, such as synaptic signaling, energy metabolism and nervous system development, e.g., all these molecular changes are associated with increased cytotoxicity and apoptosis, with a neurodegenerative pattern on Purkinje cells layer and poor motor coordination and balance. In conclusion, all these findings feature a neurodegenerative process triggered by IHg in the cerebellum that culminated into motor functions deficits, which are associated with several molecular features and may be related to the clinical outcomes of people exposed to the toxicant.

Association of frailty with short-term outcomes, organ support and resource use in critically ill patients

PurposeFrail patients are known to experience poor outcomes. Nevertheless, we know less about how frailty manifests itself in patients’ physiology during critical illness and how it affects resource use in intensive care units (ICU). We aimed to assess the association of frailty with short-term outcomes and organ support used by critically ill patients.MethodsRetrospective analysis of prospective collected data from 93 ICUs in Brazil from 2014 to 2015. We assessed frailty using the modified frailty index (MFI). The primary outcome was in-hospital mortality. Secondary outcomes were discharge home without need for nursing care, ICU and hospital length of stay (LOS), and utilization of ICU organ support and transfusion. We used mixed logistic regression and competing risk models accounting for relevant confounders in outcome analyses.ResultsThe analysis consisted of 129,680 eligible patients. There were 40,779 (31.4%) non-frail (MFI = 0), 64,407 (49.7%) pre-frail (MFI = 1–2) and 24,494 (18.9%) frail (MFI ≥ 3) patients. After adjusted analysis, frailty was associated with higher in-hospital mortality (OR 2.42, 95% CI 1.89–3.08), particularly in patients admitted with lower SOFA scores. Frail patients were less likely to be discharged home (OR 0.36, 95% CI 0.54–0.79) and had higher hospital and ICU LOS than non-frail patients. Use of all forms of organ support (mechanical ventilation, non-invasive ventilation, vasopressors, dialysis and transfusions) were more common in frail patients and increased as MFI increased.ConclusionsFrailty, as assessed by MFI, was associated with several patient-centered endpoints including not only survival, but also ICU LOS and organ support.

Microglial immunosurveillance of the brain parenchyma to detect local perturbations in homeostasis, in all species, results in the adoption of a spectrum of morphological changes that reflect functional adaptations. Here, we review the contribution of these changes in microglia morphology in distantly related species, in homeostatic and non-homeostatic conditions, with three principal goals (1): to review the phylogenetic influences on the morphological diversity of microglia during homeostasis (2); to explore the impact of homeostatic perturbations (Dengue virus challenge) in distantly related species ( Mus musculus and Callithrix penicillata ) as a proxy for the differential immune response in small and large brains; and (3) to examine the influences of environmental enrichment and aging on the plasticity of the microglial morphological response following an immunological challenge (neurotropic arbovirus infection). Our findings reveal that the differences in microglia morphology across distantly related species under homeostatic condition cannot be attributed to the phylogenetic origin of the species. However, large and small brains, under similar non-homeostatic conditions, display differential microglial morphological responses, and we argue that age and environment interact to affect the microglia morphology after an immunological challenge; in particular, mice living in an enriched environment exhibit a more efficient immune response to the virus resulting in earlier removal of the virus and earlier return to the homeostatic morphological phenotype of microglia than it is observed in sedentary mice.

ObjectiveTo develop recombinant factor IX (FIX) variants with augmented clotting activity.ResultsWe generated three new variants, FIX-YKALW, FIX-ALL and FIX-LLW, expressed in SK-Hep-1 cells and characterized in vitro and in vivo. FIX-YKALW showed the highest antigen expression level among the variants (2.17 µg−mL), followed by FIX-LLW (1.5 µg−mL) and FIX-ALL (0.9 µg−mL). The expression level of FIX variants was two–five fold lower than FIX-wild-type (FIX-WT) (4.37 µg−mL). However, the biological activities of FIX variants were 15–31 times greater than FIX-WT in the chromogenic assay. Moreover, the new variants FIX-YKALW, FIX-LLW and FIX-ALL also presented higher specific activity than FIX-WT (17, 20 and 29-fold higher, respectively). FIX variants demonstrated a better clotting time than FIX-WT. In hemophilia B mice, we observed that FIX-YKALW promoted hemostatic protection.ConclusionWe have developed three improved FIX proteins with potential for use in protein replacement therapy for hemophilia B.

Objective
To describe the clinicopathological features of 3 new cases of soft-tissue keratocyst (STK) of the buccal mucosa from Brazil.
Materials and methods
Clinical data from 3 patients diagnosed with STK were obtained from the archives of the Laboratory of Oral Pathology at the Federal University of Rio de Janeiro– Brazil from 2020 to 2023. Two oral pathologists reviewed conventional hematoxylin and eosin-stained slides of each case. Immunohistochemical stainings for CK19, Bcl-2, CD138, D2-40, EMA, and Ki-67 were performed in all cases.
Results
Case 1 was a 58-year-old man with a 2-year history of painless swelling of the right buccal mucosa, measuring approximately 5 cm. Case 2 was of a 44-year-old man with a painful swelling in the left buccal mucosa lasting 3 years with drainage to the oral cavity. Case 3 was of a 74-year-old woman with a painful swelling in the left buccal mucosa of unknown duration. Microscopic evaluation of all 3 cases revealed a cyst lined with a few cell layers composed of columnar palisading basal cells and a corrugated parakeratin surface. The capsule comprised fibrous connective tissue with variable amounts of blood vessels with hemorrhage, nerve bundles, fat tissue, striated muscle fibers, and the absence of pilosebaceous units. No recurrence was observed after complete surgical removal. All epithelial layers of the 3 cases expressed positivity for CK19 and CD138, the basal cells were positive for Bcl-2 and D2-40, and the superficial epithelial layer was positive for EMA. All cases demonstrated a low proliferation index by Ki-67.
Conclusion
This study represents a series of 3 cases of STK that affected the posterior buccal mucosa of middle-aged patients from Brazil, sharing histopathological and immunohistochemical features with odontogenic keratocyst.

An extensive literature search among six eletronic databases and Grey Literature was used to identify systematic reviews (S) that could respond if: in individuals diagnosed with OKC (P), is there any influence of the treatment method (I/C) on the recurrence rate of the lesion (O)? After evaluating all titles and abstracts and then applying the eligibility criteria, the included studies were read in full, and data were extracted based on a standardized sheet ordered in the PICO sequence. The assessment of the quality of the systematic reviews included, was determined by AMSTAR2, and final synthesis were descriptively made based on the results and quality of the systematic reviews.
Results
From a total of 19 included systematic reviews, it was observed that the most used treatment for OKC was enucleation, followed by enucleation with adjuvant techniques and marsupialization. The mean percentage of recurrence was 16,2%, and the highest OKC recurrence rate was 43.2% after simple enucleation. The use of adjuvant techniques promoted reductions in OKC recurrence rates. The overall methodological quality of systematic reviews was critically low, and this parameter demonstrate the need for more studies to facilitate the choose of the treatment.
Conclusion
Despite being the most used treatment, simple enucleation is related to the highest rate of recurrence, except when performed after marsupialization/decompression. In addition, the use of adjuvant techniques has a strong impact on reducing the likelihood of recurrence. However, these findings are not conclusive because of the critically low quality of the systematic reviews.

Three-Dimensional Organoid-like Co-Culture of Human Endometrial Endothelial and Stromal Cells to Study Endometriosis-Associated Responses

Three-dimensional (3D) endothelium-stromal co-cultures were established using human endometrial cells from biopsy of healthy women (
= 13) and serum samples from both healthy and endometriotic women (
= 5). For 3D construction, stromal cells were mixed with extracellular matrix components, followed by endothelial cell seeding. Morphological analysis confirmed the organization of tissue-like structures. Immunofluorescence and flow cytometry verified the expression of specific stromal and endothelial markers (Cytokeratin, Vimentin, Insulin-like growth factor-binding protein 1, and von Willebrand factor). Cell viability and proliferation increased over time, with minimal cell death. To test functional responsiveness, these co-cultures were exposed to inflammatory serum from endometriotic patients. After 48 h, cytometric bead array showed elevated levels of IL-1β, IL-6, and IL-8 in cultures treated with inflammatory serum, indicating preserved functional activity and responsiveness. By allowing detailed investigation of functional endometrial states within a physiologically relevant cellular network, this approach provides a valuable organoid-like tool to explore conditions such as implantation failure and infertility and to study the cellular interactions underlying reproductive pathologies.

In Brazil, the coronavirus disease 2019 (COVID-19) epidemic spread rapidly in a heterogeneous way, mainly due to the different socioeconomic and behavioral characteristics of different regional populations and different evaluation periods. We performed a cross-sectional study including 1,337 individuals (first wave = 736/second wave = 601) after the first two waves of COVID-19 in the city of Belém, the capital of the state of Pará. The detection of IgG anti-SARS-CoV-2 antibodies was performed using an enzyme-linked immunosorbent assay test followed by statistical analysis using the RStudio program. Our results showed an increase in the seroprevalence (first wave= 39.1%/second wave= 50.1%) of anti-severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) IgG antibodies in the population of Belém from the first to the second pandemic wave. Advanced age, primary or secondary education level, lack of social isolation, and a low frequency of protective mask use were considered risk factors for SARS-CoV-2 infection during the first wave compared to the second wave. This study is one of the firsts to provide important information about the dynamics of virus circulation and the groups vulnerable to exposure in the two major periods. Our data emphasize the socioeconomic characteristics of the affected population and that nonpharmacological prevention measures are crucial for combating the pandemic.

Genetic parameters for egg production in meat quails through partial periods

We estimated genetic and phenotypic parameters for egg production in meat-type quails aiming to propose an optimal age for selection through partial record egg production. Data of 3,503 female quails from two strains (namely, UFV1, with 1,811 and UFV2 with 1,692 females) were used. Egg production was assessed by the number of eggs recorded after 42 days of age and each partial period consisted on 35 days of egg production forward. Covariance components were estimated by using single and bivariate animal model, comprising each partial period of egg production and full egg production period (one year of egg laying). Regarding strain UFV1, heritability estimates ranges from 0.03 to 0.16, and for strain UFV2 0.20 to 0.25. The highest genetic correlation with full egg production was with second period (0.64) for strain UFV1 and with third period (0.47) for UFV2. Therefore, animal selection based on egg production until 112 days for the strain UFV1 and 147 days for the strain UFV2 provided increased genetic gain by reducing generation interval. RESUMO: Este estudo teve como objetivo estimar os parâmetros genéticos e fenotípicos para a produção de ovos de codornas de corte e propor uma idade ideal para a seleção através da característica de produção de ovos. Os dados utilizados neste estudo vieram de 3.503 codornas de duas linhagens (1.811 fêmeas UFV1 e 1.692 fêmeas UFV2). A produção de ovos foi avaliada pelo número de ovos coletados a partir do 42o dia de vida em diante, usando modelo animal uni ou bicaracterístico, em períodos parciais de 35 dias cada e período total (um ano de postura). Para a linhagem UFV1, as estimativas de herdabilidade variaram de 0,03 a 0,16, e para a linhagem UFV2, de 0,20-0,25. A correlação genética maior foi entre o segundo período e total (0,64) para a linhagem UFV1 e 0,47 (terceiro período e total) para UFV2. Portanto, recomenda-se usar o período de produção de ovos até 112 dias para a linhagem UFV1 e o período de produção de ovos até 147 dias para a linhagem UFV2, permitindo, assim, uma redução no intervalo de geração.
