Photodynamic therapy compared with loop electrosurgical excision procedure in patients with cervical high-grade squamous intraepithelial lesion

The study aimed to compare the effectiveness between Hiporfin-photodynamic therapy (PDT) and loop electrosurgical excision procedure (LEEP) in treating cervical high-grade squamous intraepithelial lesion (HSIL). We conducted a retrospective analysis of 104 women diagnosed with cervical HSIL at our hospital from April 2019 to July 2023. With 1:1 matched case-control, 52 cases received Hiporfin-PDT and 52 cases underwent LEEP. All PDT-treated cases received Hiporfin (2 mg/kg) I.V. and a diode laser at a wavelength of 630-nm. T-test, Chi-square test, Fisher’s exact test, and Yates’s correction for continuity were conducted for statistical. A P value < 0.05 was considered significant. The complete response (CR) post-PDT at 3rd-6th months,10th-12th months and 24th months was in 98.1% (51/52),100.0% (45/45) and 100.0% (22/22) patients respectively, compared to 98.1% (51/52), 100.0% (46/46) and 100.0% (24/24) patients respectively after LEEP. HPV clearance rates after PDT at 3rd-6th months,10th-12th months and 24th months was 76.9% (40/52), 88.9% (40/45) and 95.5% (21/22) respectively, compared to 69.2% (36/52), 93.5% (43/46) and 95.8% (23/24) respectively after LEEP, with no significant difference between the two groups (

= 1.000, respectively). Postoperative complications in the PDT group were tolerable. The cure rates and HPV eradication of Hiporfin-PDT were comparable with that of LEEP in the management of cervical HSIL. Hiporfin-PDT may serve an alternative method for organ-saving in cervical HSIL.

Lung adenocarcinoma (LUAD) is the most common and aggressive subtype of lung cancer, and coronavirus disease 2019 (COVID-19) has become a serious public health threat worldwide. Patients with LUAD and COVID-19 have a poor prognosis. Therefore, finding medications that can be used to treat COVID-19/LUAD patients is essential. Bioinformatics analysis was used to identify 20 possible metformin target genes for the treatment of COVID-19/LUAD. PTEN and mTOR may serve as hub target genes of metformin. Metformin may be able to cure COVID-19/LUAD comorbidity through energy metabolism, oxidoreductase NADH activity, FoxO signalling pathway, AMPK signalling system, and mTOR signalling pathway, among other pathways, according to the results of bioinformatic research. Metformin has ability to inhibit the proliferation of A549 cells, according to the results of colony formation and proliferation assays. In A549 cells, metformin increased glucose uptake and lactate generation, while decreasing ATP synthesis and the NAD
+
/NADH ratio. In summary, PTEN and mTOR may be potential targets of metformin for the treatment of COVID-19/LUAD. The mechanism by which metformin inhibits lung adenocarcinoma cell proliferation may be related to glucose metabolism regulated by PI3K/AKT signalling and mTOR signalling pathways. Our study provides a new theoretical basis for the treatment of COVID-19/LUAD.

Boron and nitrogen co-doped carbon dots as the dual functional fluorescent probe for Fe3+ and pH detection

Carbon dots doped with boron and nitrogen atoms (B, N-CDs) were designed and prepared by hydrothermal strategy using citric acid, urea and borax as raw materials. The structure and optical properties of B, N-CDs were characterized by various characterization technologies. The results showed that the B, N-CDs exhibited good mono-dispersion, excellent fluorescence stability, and high fluorescence intensity (quantum yield up to 32%). A dual-functional Fe3+ and pH detection platform based on B, N-CDs was constructed. The Fe3+ sensing method based on B, N-CDs fluorescence quenching showed an excellent linear relationship between 0.044–70μM (R2=0.994), with a detection limit as low as 0.044μM. Then, this detection method was successfully applied to the high-accuracy detection of Fe3+ in water samples and iron supplement tablets. The study of the quenching mechanism showed that the fluorescence of B, N-CDs was quenched by Fe3+ in two ways: static quenching and dynamic quenching. Moreover, B, N-CDs can also be used as a pH fluorescence sensor in the ranges of pH 2.0–4.8 and 5.2–7.6. The developed sensing system based on B, N-CDs has high selectivity and sensitivity for the detection of Fe3+, and a wide detection range for pH.

Longitudinal Relations Between Students’ Engagement and Their Perceived Relationships with Teachers and Peers in a Chinese Secondary School

The purpose of this study was to examine the reciprocal relationships between students’ engagement and their relationships with teachers and peers in the context of Chinese educational reform. The participants were 628 7th-grade students from a public secondary school in Beijing. All of the students completed self-reported student engagement, teacher–student relationships and peer relationships measures at three time points within 3 years (at the end of the 7th grade, 8th grade and 9th grade). Through cross-lagged models, this study found that prior-year teacher–student relationships could predict the cognitive component of student engagement only in the 9th grade. However, higher levels of behavioural, emotional and cognitive engagement predicted subsequent higher-quality teacher–student relationships from the 7th to the 9th grades. Moreover, previous peer relationships predicted student behavioural and cognitive engagement in the 8th grade and emotional engagement in the 9th grade. Prior-year behavioural, emotional and cognitive engagement predicted peer relationships in the 8th grade, but the prediction was significant only for the influence of behavioural engagement on peer relationships for older students between the 8th grade and the 9th grade. Implications for supportive interpersonal relationships with teachers and peers that facilitate student engagement are discussed.

Chronic intermittent hypoxia (CIH) could cause neuronal damage, accelerating the progression of dementia. However, safe and effective therapeutic drugs and delivery are needed for successful CIH therapy.
To investigate the neuroprotective effect of Huperzine A (HuA) packaged with nanoliposomes (HuA-LIP) on neuronal damage induced by CIH.
The stability and release of HuA-LIP in vitro were identified. Mice were randomly divided into the Control, CIH, HuA-LIP, and HuA groups. The mice in the HuA and HuA-LIP groups received HuA (0.1 mg/kg, i.p.), and HuA-LIP was administered during CIH exposure for 21 days. HuA-LIP contains the equivalent content of HuA.
We prepared a novel formulation of HuA-LIP that had good stability and controlled release. First, HuA-LIP significantly ameliorated cognitive dysfunction and neuronal damage in CIH mice. Second, HuA-LIP elevated T-SOD and GSH-Px abilities and decreased MDA content to resist oxidative stress damage induced by CIH. Furthermore, HuA-LIP reduced brain iron levels by downregulating TfR1, hepcidin, and FTL expression. In addition, HuA-LIP activated the PKAα/Erk/CREB/BDNF signaling pathway and elevated MAP2, PSD95, and synaptophysin to improve synaptic plasticity. Most importantly, compared with HuA, HuA-LIP showed a superior performance against neuronal damage induced by CIH.
HuA-LIP has a good sustained-release effect and targeting ability and efficiently protects against neural injury caused by CIH.

Down-regulation of interleukin-2 predicts poor prognosis and associated with immune escape in lung adenocarcinoma

Background and Objective: Lung adenocarcinoma (LUAD) is the most common subtype of lung cancer and has a poor prognosis. Interleukin-2 (IL2) is a cytokine that stimulates lymphocyte proliferation. However, its role in LUAD remains unclear. Methods: The UALCAN, human protein atlas (HPA), and tumor immune estimation resource (TIMER) databases were used to investigate IL2 expression in samples from patients with LUAD. The HPA, PrognoScan, and Kaplan–Meier plotter databases were used to examine the prognostic value of IL2 in LUAD. Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathway analyses were performed to analyze IL2-interacting genes identified through the GeneMANIA database. TIMER was used to analyze the correlation of IL2 expression with immune cell infiltration and immune checkpoint expression levels in LUAD. Results: Bioinformatic analysis using the TIMER, The University of Alabama at Birmingham Cancer data analysis Portal (UALCAN), and HPA public databases showed that IL2 expression was lower in patients with LUAD than in the normal control group. Moreover, patients with low IL2 expression exhibited poor overall survival. Furthermore, IL2 expression was significantly positively correlated with various immune cells, including B cells, CD8+ T cells, CD4+ T cells, macrophages, neutrophils, and dendritic cells, in patients with LUAD. Additionally, IL2 expression was markedly positively associated with the above-mentioned immune cells. Furthermore, IL2 expression was positively correlated with PD-1, PD-L1, and CTLA-4 expression. Conclusion: Our results indicate that down-regulation of IL2 predicts poor prognosis and is associated with immune escape in LUAD, and IL2 could serve as a potential novel prognostic biomarker of LUAD.

Factors associated with negative conversion of viral RNA in hospitalized children infected with SARS-CoV-2 Omicron variant in Shanghai, China: a retrospective analysis

Objectives
This study aimed to identify the related risk factors and potential predictors of SARS-CoV-2 RNA negative conversion by describing the dynamics of viral shedding in infected children admitted to two hospitals from Shanghai during the Omicron variant outbreak.
Methods
This retrospective cohort included laboratory-confirmed cases of SARS-CoV-2 infection from Shanghai between March 28 and May 31, 2022. Clinical characteristics, personal vaccination, and household vaccination rates were collected through electronic health records and telephone interviews.
Results
A total of 603 paediatric patients confirmed to have COVID-19 were included in this study. Both univariate and multivariate analyses were performed to filter independent factors for the duration to viral RNA negative conversion. Data on the redetection of SARS-CoV-2 in the patients after they showed negative results on the RT‒PCR test (intermittent negative status) were also analysed. The median duration of virus shedding was 12 (interquartile range, IQR: 10–14) days. The severity of clinical outcome, personal vaccination-2doses, household vaccination rates, and abnormal defecation were factors indecently affecting negative conversion of SARS-CoV-2 RNA, suggesting that patients who had abnormal defecation or with more severe conditions would have delayed virological clearance, while patients who previously had 2 doses of vaccination or had higher household vaccination rates would have accelerated virological clearance. Loss of appetite (odds ratio (OR): 5.343; 95% CI: 3.307–8.632) and abnormal defecation (OR: 2.840; 95% CI: 1.736–4.645) were significantly associated with intermittent negative status.
Conclusion
These findings could provide clues for the early identification of paediatric patients with prolonged viral shedding and could enrich the evidence for the development of prevention and control strategies, especially vaccination policies for children and adolescents.

Assessment of Conventional and Low Gossypol Cottonseed Meal as Alternative Protein Sources in Low-Fishmeal Diets of Hybrid Grouper (Epinephelus fuscoguttatus♀× Epinephelus lanceolatus♂): Growth, Feed Utilization, Gut Histology, and Immunity

A 9-week growth trial was carried out to assess the influence of replacing poultry by-product meal protein with conventional cottonseed meal protein (CCMP) or low gossypol cottonseed meal protein (LGCMP) on growth, feed utilization, gut micromorphology, and immunity of hybrid grouper (Epinephelus fuscoguttatus♀× Epinephelus lanceolatus♂) juveniles fed low-fish meal (18.53%, dry matter) diets. Eleven experimental diets were prepared. The control diet (PBMP) contained 46.15% poultry by-product meal protein. Both conventional cottonseed meal protein (CCMP) and low-gossypol cottonseed meal protein (LGCMP) were used in replacement ratios of 20, 40, 60, 80, and 100% of poultry by-product meal protein (PBMP) from the control diet, forming ten experimental diets (CCMP20, CCMP40, CCMP60, CCMP80, CCMP100, LGCMP20, LGCMP40, LGCMP60, LGCMP80, and LGCMP100). Results demonstrated that weight-gain percentage (WG%) was not different between different sources of cottonseed meal (CCMP and LGCMP). However, values of WG% significantly differed among different replacement levels, with CCMP80 and LGCMP40 having significantly higher values compared to other treatments. Fish fed CCMP80 and LGCMP40 exhibited higher protein efficiency ratios (PERs) than fish fed other experimental diets. The regression analysis from a second-order or third-order polynomial model based on WG% showed that the optimal PBMP replacement levels by CCMP and LGCMP are 74% and 33%, respectively. The whole-body lipid contents remarkably decreased as dietary CCMP or LGCMP inclusion levels increased. The relative mRNA expression of insulin-like growth factor-1(IGF-1) in liver was higher in fish fed CCMP80 and LGCMP40 diets compared to fish fed other diets. Generally, in low-FM diets of hybrid grouper, CCMP and LGCMP could replace 74% and 33% of PBMP, respectively.

Nasal delivery of nanoliposome-encapsulated ferric ammonium citrate can increase the iron content of rat brain

Background
Iron deficiency in children can have significant neurological consequences, and iron supplementation is an effective treatment of choice. However, traditional routes of iron supplementation do not allow efficient iron delivery to the brain due to the presence of the blood–brain barrier. So an easily delivered iron formulation with high absorption efficiency potentially could find widespread application in iron deficient infants.
Results
In this study, we have developed and characterized a nanovesicular formulation of ferric ammonium citrate (ferric ammonium citrate nanoliposomes, FAC-LIP) and have shown that it can increase brain iron levels in rats following nasal administration. FAC was incorporated into liposomes with high efficiency (97%) and the liposomes were small (40 nm) and stable. Following intranasal delivery in rats, FAC-LIP significantly increased the iron content in the olfactory bulb, cerebral cortex, striatum, cerebellum and hippocampus, and was more efficient at doing so than FAC alone. No signs of apoptosis or abnormal cell morphology were observed in the brain following FAC-LIP administration, and there were no significant changes in the levels of SOD and MDA, except in the cerebellum and hippocampus. No obvious morphological changes were observed in lung epithelial cells or tracheal mucosa after nasal delivery, suggesting that the formulation was not overtly toxic.
Conclusions
In this study, nanoscale FAC-LIP proved an effective system delivering iron to the brain, with high encapsulation efficiency and low toxicity in rats. Our studies provide the foundation for more detailed investigations into the applications of niosomal nasal delivery of liposomal formulations of iron as a simple and safe therapy for iron deficiency anemia.

High Sensitive Visual Protein Detection by Microfluidic Lateral Flow Assay with On-Stripe Multiple Concentration

Lateral flow assays (LAFs) especially integrated with a microfluidic chip, provides a simple, rapid, user-friendly, potable robust, and cost-effective technology for broad assays. However, this technology suffers from low sensitivity. In this paper, one kind of automatic roller of tap, which can be precisely controlled to replace sample tap was integrated into the microfluidic LAFs platform. And then, on-stripe repeated injection and concentration were realized with this simple mechanic unit. The minimum detection concentration for human chorionic gonadotropin (HCG) was 1.26 ng/mL, comparable with literature using complex enzyme/chemical reaction-based signal amplification. The linear relationship between the signal intensity and enrichment times reflected the good reproducibility of the novel device. At the same time, the good linear relationship between the predicted accumulation quantity of HCG and the gray value of bands is very meaningful for quantitative detection. Consequently, this novel universal approach shows great potential in the rapid trace analysis and broaden the application of LAFs with its attractive characteristics.
