Risk factors for Toxoplasma gondii infection in dogs: a serological survey

Background
Dogs, as well as a wide variety of other warm-blooded animals, act as intermediate host of
Toxoplasma gondii
. In dogs, most cases of toxoplasmosis are subclinical, although clinical disease has been sporadically reported. Beyond its role in diagnostic pathways, seropositivity also functions as a reflection of the parasite’s spread within the dog’s living environment. The aim of the present study was to evaluate the possible risk factor associated with seropositivity to
T. gondii
in dogs in Central-Northern Italy, analysing 120 dogs sera for the presence of IgG antibodies by indirect fluorescence antibody test (IFAT).
Results
The population examined was composed of 54.2% hunting dogs, 24.2% companion dogs, 14.2% truffle dogs and 7.5% watchdogs. Thirty-four (29.2%) dogs tested positive for
T. gondii
IgG, with titres ranging from 1:40 to 1:1280. Seroprevalence and antibodies titres were not related to dog gender, age or function. The logistic regression and ordered logistic regression results indicated that seroprevalence, and antibody titres were significantly higher in dogs cohabiting with cats, exhibiting coprophagy habits, and living constantly outdoors. Notably, the lifestyle factor showed the highest odds-ratios in the study: dogs living constantly outdoors were found to be at approximately 5 times greater risk of testing positive and having higher antibody titres compared to dogs living both indoors and outdoors.
Conclusion
Both logistic and ordered logistic regression results support the key role of living with cats, engaging in coprophagy behaviours, and maintaining an outdoor lifestyle in increasing the risk of
T. gondii
infection in dogs. These identified risk factors collectively suggest that both ingesting oocysts, as observed through cat cohabitation and coprophagy, and engaging in predatory behaviours, as possible for outdoor living dogs, are indicating likely sources of
T. gondii
infection in this host species.

Systemic Aspergillosis in Dogs: A Historical and Current State-of-the-Art Review

Canine systemic aspergillosis is a rare but highly serious condition, often associated with a fatal outcome. This review encompasses all reported cases of canine systemic aspergillosis from 1978 to the present, focusing exclusively on studies in which the diagnosis was confirmed through fungal culture. A total of 155 clinical cases reported in the literature were included. Among these, the German Shepherd was the most frequently affected breed (65.16%), followed by mixed-breed dogs (7.74%). The predominant Aspergillus species isolated was A. terreus (57.69%), although other species were also reported, including A. deflectus, A. fumigatus, A. niger, A. caninus, A. versicolor, A. alabamensis, A. citrinoterreus, and A. floccosus. Recognizing clinical signs and accurately interpreting laboratory findings are crucial for early diagnosis and timely intervention, both of which can potentially improve outcomes. This review provides a detailed discussion of these aspects.

Hepatozoon martis in Italy: first evidence based on histopathological characterization and molecular confirmation in Martes foina

Background
Hepatozoon martis
is an apicomplexan parasite infecting mustelids, but its geographical distribution and pathological relevance remain incompletely understood. To date, its presence in Italy has not been documented. This study aimed to investigate the occurrence of
H. martis
in martens from northern and central Italy using an integrated pathological and molecular approach.
Methods
Between 2023 and 2025, carcasses of 17 martens (
Martes
spp.) collected within wildlife health surveillance programs were examined. Tissue samples were screened for apicomplexan DNA by qPCR, and conventional PCR targeting the 18S rDNA, followed by sequencing and phylogenetic analyses, were applied for species characterization. Histopathological examinations were performed on tissues from freshly deceased animals.
Results
Eight of 17 martens (47%), 16 identified as
Martes foina
and one
Martes martes
, tested positive for
Hepatozoon
infection. Sequencing confirmed
H. martis
in multiple tissues, including spleen, heart, lymph nodes, and muscle. Histopathological analysis revealed multifocal granulomatous myocarditis and myositis, with intracellular zoites and type II meronts observed within inflammatory lesions. Phylogenetic analyses of short and long 18S rDNA fragments showed that Italian isolates clustered with
H. martis
sequences from other European countries, displaying low genetic variability.
Hepatozoon
DNA was also detected in one
Ixodes
sp. collected from an infected animal.
Conclusions
This study provides the first evidence to our knowledge of
H. martis
infection in Italy and highlights its pathogenic potential in stone martens. The low genetic diversity observed among European isolates suggests widespread circulation and gene flow across regions. Further investigations are required to elucidate transmission pathways, identify competent vectors, and assess the pathological significance of
H. martis
in wildlife.

The genus Trypanosoma includes flagellated protozoa belonging to the family Trypanosomatidae (Euglenozoa, Kinetoplastida) that can infect humans and several animal species. The most studied species are those causing severe human pathology, such as Chagas disease in South and Central America, and the human African trypanosomiasis (HAT), or infections highly affecting animal health, such as nagana in Africa and surra with a wider geographical distribution. The presence of these Trypanosoma species in Europe has been thus far linked only to travel/immigration history of the human patients or introduction of infected animals. On the contrary, little is known about the epidemiological status of trypanosomes endemically infecting mammals in Europe, such as Trypanosomatheileri in ruminants and Trypanosomalewisi in rodents and other sporadically reported species. This brief review provides an updated collection of scientific data on the presence of autochthonous Trypanosoma spp. in mammals on the European territory, in order to support epidemiological and diagnostic studies on Trypanosomatid parasites.

Humans and dogs commonly share the same domestic environment. Europe, and Italy specifically, have a substantial and growing dog population. Potentially zoonotic parasites may be harbored even by dogs receiving regular veterinary care. Thus, transmission of zoonotic or potentially zoonotic parasites to owners and their families should not be underestimated. Frequently, endoparasite infections occur as a subclinical infection and clinicopathological alterations have been documented including anemia, hypoalbuminemia, and eosinophilia. The aim of this large retrospective secondary data study was to analyze coprological endoparasite results and putative risk factors obtained from owned dogs, through a 9-year-period (2011–2019). Possible associations between diagnosed endoparasites and sex, age, seasonality, and year of examination were evaluated. Additionally, parasitological diagnoses were combined to complete blood count parameters and biochemical profiles, when available, to check for any possible hematological alteration from parasitism. A total of 1,972 dogs were evaluated for endoparasites using common fecal diagnostic tests over a 9-year period. The overall proportion of endoparasite-positive animals was 10%. The most common endoparasites detected were Cystoisospora spp. (3%), Toxocara canis (2.8%), Giardia duodenalis (1.6%), and Trichuris vulpis (1.2%). Of these parasites detected, Toxocara poses the greatest zoonotic risk, while Giardia species are considered to have a low potential to be zoonotic. There was no significant diagnostic trend across the years through the study period. Dogs were more frequently diagnosed endoparasite-positive when young and during cold seasons compared to the baselines of mature dogs and warm seasons. The clinicopathological profiles indicated that parasitized dogs had mild hematological alterations. The frequency of detected potentially zoonotic endoparasites in this study highlights that the risk should not be underestimated. Parasitic infection was found to be mostly dependent on age and season. Having this information may help clinicians to develop anthelmintic protocols to reduce the risk of transmission.

Field Application of an Innovative Approach to Assess Honeybee Health and Nutritional Status

Environment, forage quality, management practices, pathogens, and pesticides influence honeybee responses to stressors. This study proposes an innovative approach to assess colony health and performance using molecular diagnostic tools by correlating hemolymph proteins with common measures of colony strength, prevalent honeybee pathogens (Varroa destructor and Nosema spp.), and essential trace elements (iron, zinc and copper). Colonies were selected from four apiaries located in different environmental and foraging conditions in the province of Bologna (Italy). Hemolymph samples were taken from June to October 2019. The Varroa infestation of the colonies was estimated by assessing the natural mortality of the mites, while the bees were tested for Nosema spp. spores using a microscopic method. Hemolymph proteins were quantified and separated using SDS-PAGE, and colony performance was assessed by determining adult bees, total brood, honey, and pollen reserves. The biomarkers measured proved to be useful for monitoring changes in performance and trophic conditions during summer and early autumn. Significant correlations were found between hemolymph proteins and colony performance measures. A positive correlation between pollen reserves, vitellogenin, and hexamerin 70a highlights the importance of these proteins for successful overwintering. In October, Varroa infestation was negatively correlated with total proteins, vitellogenin, apolipophorin II, transferrin, and hexamerin 70a, with negative implications for overwintering; furthermore, Varroa infestation was also negatively correlated with iron content, potentially affecting iron homeostasis.

Synanthropic rodents are species well adapted to coexisting in anthropogenically influenced environments. This coexistence raises concerns about the potential risks of pathogen’s transmission due to their close proximity to human habitats. This study presents an epidemiological survey of the gastrointestinal helminth fauna in synanthropic rodents (Mus musculus, Rattus rattus, and Rattus norvegicus) from the Emilia-Romagna Region (Italy), aiming to provide updated data on the endoparasitic populations in these species. A total of 111 rodents, sampled from 2019 to 2021 during pest control programs, were examined for parasitic infections. Helminths were extracted through necropsy and microscopic analysis of gastrointestinal tracts and sediment, with species identification based on morphological characteristics. Overall, 72.1% of the rodents were found to be parasitized, with nematodes being the most prevalent. Syphacia muris, Aspiculuris tetraptera, Nippostrongylus brasiliensis, and Heterakis spumosa were the most frequently identified nematodes. Tapeworms, including Rodentolepis nana and Hymenolepis diminuta, were also detected, albeit in lower frequencies. The trematode Brachylaima recurva was recovered only in one R. rattus. Co-infection was common, particularly among rats, with 51.8% of black rats and 22% of brown rats harboring multiple parasitic species. Mice exhibited lower levels of polyparasitism, with only two individuals showing mixed infections. Interestingly, disparities between the detection of adult helminths and parasitic eggs were noted, especially in cases where no adults were observed, but eggs were found through sediment analysis. These findings suggest that traditional necropsy, especially with poorly preserved carcasses, may underestimate parasite prevalence. This highlights the importance of combining necropsy with microscopic techniques, such as flotation and sedimentation, for a more thorough assessment. Using these methods, nematodes with direct life cycles, such as Syphacia spp., Nippostrongylus brasiliensis, and Heterakis spumosa, have been confirmed as widespread and cosmopolitan among rodent populations. The detection of zoonotic parasites raises concerns about potential transmission to humans, particularly in areas with poor sanitation and high rodent densities. These findings underscore the need for integrated rodent control and environmental sanitation to reduce zoonotic risks.

Prevalence and risk factors associated with cat parasites in Italy: a multicenter study

Background
Parasites that infect cats include protozoa, helminths and arthropods, many of which are transmissible to humans. Effective control relies on a good knowledge of parasite distribution and the risk factors for infection. The present study was aimed at evaluating the prevalence of major feline parasites in Italy and the risk factors associated with their occurrence.
Methods
Over a 12-month study period, feces, hair and ectoparasites from naturally infected cats from feral colonies, shelters and private households were analyzed at 13 study centers across Italy. Samples from these cats (
n
= 987) were analyzed at all centers using the same diagnostic methods. Prevalence values and risk factors were evaluated statistically for the identification of predictors of risk.
Results
The overall prevalence of gastro-intestinal and broncho-pulmonary (BP) nematodes was 35.9% (354/987).
Toxocara cati
was the most prevalent species (253/987; 25.6%), followed by Ancylostomatidae (98/987; 9.9%). Among BP nematodes,
Aelurostrongylus abstrusus
was the most common (76/987; 7.7%). Approximately 35.7% (352/987) of the study population was infested by ectoparasites, of which the most common were fleas (29.4%, 290/987), followed by ear mites
Otodectes cynotis
(9.8%, 97/987). Predictors of risk for parasite infection included age, a predominantly or exclusively outdoor lifestyle, geographic area and lack of antiparasitic treatment.
Conclusions
Both ecto- and endoparasites are still common in cats throughout Italy, many of them being of zoonotic concern and vectors of pathogens to humans. Given the presence of parasites throughout the entire study period, year-round treatment should be considered. Furthermore, data confirm the need to protect the human–animal bond using proper endo- and ectoparasiticides to reduce the risk of human infection, in application of the One-Health concept.

Migratory Wild Birds as Potential Long-Distance Transmitters of Toxoplasma gondii Infection

Toxoplasma gondii is a worldwide distributed zoonotic protozoan capable of infecting a wide range of mammals (including humans) and birds as intermediate hosts. Migratory wild birds, through interconnecting countries along their flyways, can play a role in the spatial spread of T. gondii and could contribute to its sylvatic cycle. Additionally, hunted wild birds used for meat consumption could represent a further source of human infection. To determine the presence of T. gondii in wild birds, a total of 50 individuals belonging to the Anseriformes and Charadriiformes orders were sampled during the 2021–2022 hunting season in Northern Italy. Cardiac muscle samples of three Northern shovelers (Anas clypeata), two wild mallards (A. platyrhynchos), one Eurasian teal (A. crecca), and one Northern lapwing (Vanellus vanellus) were positive for the molecular detection of T. gondii based on a targeted amplification of the B1 gene. A 14% (7/50) overall positivity was observed in the sampled population. Results from this study suggest a moderate exposure of wild aquatic birds to T. gondii, highlighting the importance of a further characterization of T. gondii in its wildlife hosts.

First Report of Bronchial Oestriasis in a Domestic Dog

This study reports the first case of bronchial myiasis in a dog caused by Oestrus ovis, a parasite known to infest sheep, and occasionally other mammals. A 9‐year‐old dog, from a rural area near Ancona (Italy), exhibited persistent coughing despite antibiotic and anti‐inflammatory treatments. Bronchoscopy revealed a live first‐instar larva of O. ovis, confirmed through morphological and molecular analyses. Larval removal and treatment with isoxazolines resulted in a rapid resolution of symptoms. This case highlights the critical importance of including atypical parasitic infestations in the differential diagnosis of respiratory symptoms in dogs. It underscores the importance of heightened vigilance, particularly in areas with sheep flocks, to prevent and manage such infestation effectively. Oestrus ovis bronchial myasis is described in a dog for the first time. Bronchoscopy proved essential for larval visualization and removal. Isoxazoline compounds should be considered both as a treatment and as a preventive option.
