A New Coupling Method for PM2.5 Concentration Estimation by the Satellite-Based Semiempirical Model and Numerical Model
Aerosol optical and chemical properties play a major role in the retrieval of PM2.5 concentrations based on aerosol optical depth (AOD) data from satellites in the conventional semiempirical model (SEM). However, limited observation information hinders the high-resolution estimation of PM2.5. Therefore, a new method for evaluating near-surface PM2.5 at high spatial resolution is developed by coupling the SEM and the chemical transport model (CTM)-based numerical (CSEN) model. The numerical model can provide large-scale information for aerosol properties with high spatial resolution at a large scale based on emissions and meteorology, though it can still be biased in simulating absolute PM2.5 concentrations. Therefore, the two crucial aerosol characteristic parameters, including the coefficient integrated humidity effect (γ′) and the comprehensive reference value of aerosol properties (K) in SEM, have been redefined using the WRF-Chem numerical model. Improved model performance was observed for these results compared with the original SEM results. The monthly averaged correlation coefficients (R) by CSEN were 0.92, 0.82, 0.84, and 0.83 in January, April, July, and October, respectively, whereas those of the SEM were 0.80, 0.77, 0.72, and 0.72, respectively. All the statistical metrics of the model validation showed significant improvements in all seasons. The reduced biases of estimated PM2.5 by CSEN indicated the effect of hygroscopic growth and aerosol properties affected by the meteorology on the relationship between AOD and estimated PM2.5 concentrations, especially in winter and summer. The better performance of the CSEN model provides insight for air quality monitoring at different scales, which supplies important information for air pollution control policies and health impact analysis.
Iris lactea var. chinensis plant drought tolerance depends on the response of proline metabolism, transcription factors, transporters and the ROS-scavenging system
Iris lactea var. chinensis, a perennial herbaceous species, is widely distributed and has good drought tolerance traits. However, there is little information in public databases concerning this herb, so it is difficult to understand the mechanism underlying its drought tolerance. In this study, we used Illumina sequencing technology to conduct an RNA sequencing (RNA-seq) analysis of I. lactea var. chinensis plants under water-stressed conditions and rehydration to explore the potential mechanisms involved in plant drought tolerance. The resulting de novo assembled transcriptome revealed 126,979 unigenes, of which 44,247 were successfully annotated. Among these, 1187 differentially expressed genes (DEGs) were identified from a comparison of the water-stressed treatment and the control (CK) treatment (T/CK); there were 481 upregulated genes and 706 downregulated genes. Additionally, 275 DEGs were identified in the comparison of the rehydration treatment and the water-stressed treatment (R/T). Based on Quantitative Real-time Polymerase Chain Reaction (qRT-PCR) analysis, the expression levels of eight randomly selected unigenes were consistent with the transcriptomic data under water-stressed and rehydration treatment, as well as in the CK. According to Gene Ontology (GO) annotation and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis, proline metabolism-related DEGs, including those involved in the 'proline catabolic process', the 'proline metabolic process', and 'arginine and proline metabolism', may play important roles in plant drought tolerance. Additionally, these DEGs encoded 43 transcription factors (TFs), 46 transporters, and 22 reactive oxygen species (ROS)-scavenging system-related proteins. Biochemical analysis and histochemical detection showed that proline and ROS were accumulated under water-stressed conditions, which is consistent with the result of the transcriptomic analysis. In summary, our transcriptomic data revealed that the drought tolerance of I. lactea var. chinensis depends on proline metabolism, the action of TFs and transporters, and a strong ROS-scavenging system. The related genes found in this study could help us understand the mechanisms underlying the drought tolerance of I. lactea var. chinensis.
A Novel Variant of Avian Reovirus Is Pathogenic to Vaccinated Chickens
Avian reovirus (ARV) infections, characterized by severe arthritis, tenosynovitis, pericarditis, and poor weight gain, have become increasingly serious in recent years. The economic impact is significant as it causes growth inhibition and immunosuppression. Some commercial poultry in China have been widely vaccinated with available ARV vaccines; however, infections continue to occur even after vaccination. This study aimed to isolate a novel variant, ARV-SD19/11103, from the joint tissues of infected broiler chickens vaccinated with ARV vaccines in Shandong Province. Genetic evolution analysis of the major protective antigen σC gene in ARVs showed that ARV-SD19/11103 was located in the genotype cluster I but not in the same sub-cluster as the S1133 vaccine strain. The amino acid sequence similarity between SD19/11103 and vaccine strains S1133, 1733, and 2408 was <80%. After analyzing the amino acid sequences of the σC protein, 33 amino acid differences were found between the new variant isolate and the vaccine strains. This novel variant showed obvious pathogenicity in specific pathogen-free chicken embryos and chicks and could cause serious disease in chickens vaccinated with commercially available ARV vaccines. Cross-neutralization experiments further demonstrated a significant antigenic difference between the novel variant and genotype cluster I ARV strains. The novel variant strain isolated in this study provides an important theoretical basis for understanding the prevalence and genetic evolutionary characteristics of ARV variant strains in our country. This study identified the causes of ARVs circulating and emphasizes the needs for developing new vaccines against novel ARV variants.
Impact of Taihu Lake on City Ozone in the Yangtze River Delta
The lake-breeze at Taihu Lake generates a different specific heat capacity between the water body and the surrounding land. Taihu Lake has a significant impact on the atmospheric conditions and the air quality in the Yangtze River Delta. This phenomenon is referred to as the Taihu Lake effect. In this study, two simulations were conducted to determine the impact of the Taihu Lake effect in the reference experiment (R-E) and sensitivity experiments (NO_TH). The control simulations demonstrated that the meteorological field and the spatial distribution of ozone (03) concentrations over Taihu lake obviously changed once the land-use type of water body was substituted by cropland. The surface temperature of Taihu Lake was reduced under the impact of Taihu Lake, and a huge temperature difference caused a strong lake-breeze effect. The results also showed that the difference in the average concentrations of 03 between the R-E and NO_TH experiments reached 12 ppbv in most areas of Taihu Lake, all day, on 20 May 2014. During daytime (0800-1600 LST, LST=UTC+8), the influence of the Taihu Lake effect on 03 in the Suzhou region was not significant. However, the influence of the Taihu Lake effect on 03 in the Suzhou region was obvious during nighttime (1800-2400 LST). The larger changes in the physical and chemical processes were horizontal and vertical advections under the influence of the Taihu Lake effect in Taihu Lake.
Extracellular Enzyme Stoichiometry Reveals Soil Microbial Carbon and Phosphorus Limitations in the Yimeng Mountain Area, China
Soil extracellular enzymes are considered key components in ecosystem carbon and nutrient cycling, and analysing their stoichiometry is an effective way to reveal the resource limitations on soil microbial metabolism. In this study, the soil and litter of Quercus acutissima plots, Pinus thunbergii plots, Quercus acutissima–Pinus thunbergii mixed-plantation plots, herb plots, and shrub plots in the state-owned Dawa Forest Farm in the Yimeng Mountain area were studied. The total carbon (C), nitrogen (N), and phosphorus (P) contents of litter and the physical and chemical properties of soil were analyzed, along with the activities of four extracellular enzymes related to the soil C, N, and P cycle: β-1,4-glucosidase (BG), β-1,4-N-acetylglucosaminidase (NAG), L-leucine aminopeptidase (LAP), and acid phosphatase (AP). The extracellular enzyme stoichiometric model was used to study and compare the metabolic limitations of soil microorganisms in different plots, and the driving factors of microbial metabolic limitations were explored by redundancy and linear regression analyses. The results showed that the values of BG/(NAG + LAP) were all higher than 1, the values of (NAG + LAP)/AP all lower than 1, and the vector angles of the five plots all greater than 45°, which indicated that the soil microorganisms were relatively limited by C and P. Redundancy and linear regression analysis revealed that soil physical properties (e.g., soil moisture) and litter total C make greater contributions to soil extracellular enzymes and stoichiometry than the other investigated soil parameters, whereas soil chemical properties (e.g., soil organic C and available P) predominantly controlled vector properties. Therefore, microbial metabolism limitations are greatly regulated by soil physical and chemical properties and litter total C and N. Compared with the forest plots, the soil microbial C (1.67) and P (61.07°) limitations of herb plots were relatively higher, which means that the soil microbial communities of forest plots are more stable than those of herb plots in the Yimeng Mountain area. Forest plots were more conducive than other plots to the improvement of soil microbial ecology in this area. This study could be important for illuminating soil microbial metabolism and revealing soil nutrient cycling in the Yimeng Mountain area ecosystem of China.
Detrital Zircon LA-ICP-MS U-Pb Ages of the North Liaohe Group from the Lianshanguan Area, NE China: Implications for the Tectonic Evolution of the Paleoproterozoic Jiao-Liao-Ji Belt
The Liaohe Group, which is a significant lithostratigraphic unit within the Paleoproterozoic Jiao-Liao-Ji Belt situated between the Longgang and Liaonan-Nangrim blocks, comprises the Langzishan, Li’eryu, Gaojiayu, Dashiqiao, and Gaixian formations, which are characterized mainly by a clastic-rich sequence with an interlayered bimodal-volcanic sequence, carbonate-rich sequence, and (meta-)pelite-rich sequence. Currently, the tectonic background and evolution of the Liaohe Group remain contentious. Based on the study of detrital zircon geochronology and the zircon trace element characteristics in the Langzishan and Li’eryu formations in the North Liaohe Group in the Lianshanguan area, NE China, this paper reveals the formations’ provenances, depositional ages, and relationships with Paleoproterozoic granitoids (the Liao-Ji granites). The present results, in conjunction with previous studies, indicate that the depositional age of the Langzishan Formation is 2136 Ma and that of the Li’eryu Formation is 1974 Ma. The provenances of the Langzishan Formation and the Li’eryu Formation are mainly characterized by Neoarchean-to-early-Paleoproterozoic basement rocks (~2.6–2.4 Ga) and the Liao-Ji granites (~2.2–2.0 Ga), respectively. Moreover, the coeval mafic and metasedimentary rocks of the Liaohe Group exhibit characteristics of an extensional environment, which is represented by the tectonic setting of a back-arc basin. Notably, the Upper Langzishan Formation records a prominent shift in sedimentary environment from a passive continental margin to an active continental margin. In terms of the tectonic evolution of the North Liaohe Group and the Jiao-Liao-Ji Belt, our proposed model suggests that the Archean basement rocks in the northern part of the continental block, along with a limited contribution from the Paleoproterozoic Liao-Ji granites, served as the primary sources for the Langzishan Formation. Subsequently, the rapid deposition of the Li’eryu Formation was influenced by intense magmatism and subsequent erosion of the subduction-related magmatic arc (the Liao-Ji granites) within a back-arc basin environment. Lastly, the deposition of clastic materials from the Longgang blocks and the Liao-Ji granites resulted in the formation of the Gaojiayu, Dashiqiao, and Gaixian formations.
Chromosome-level genome assembly and annotation of the prickly nightshade Solanum rostratum Dunal
The prickly nightshade Solanum rostratum, an annual malignant weed, is native to North America and has globally invaded 34 countries, causing serious threats to ecosystems, agriculture, animal husbandry, and human health. In this study, we constructed a chromosome-level genome assembly and annotation of S. rostratum. The contig-level genome was initially assembled in 898.42 Mb with a contig N50 of 62.00 Mb from PacBio high-fidelity reads. With Hi-C sequencing data scaffolding, 96.80% of the initially assembled sequences were anchored and orientated onto 12 pseudo-chromosomes, generating a genome of 869.69 Mb with a contig N50 of 72.15 Mb. We identified 649.92 Mb (72.26%) of repetitive sequences and 3,588 non-coding RNAs in the genome. A total of 29,694 protein-coding genes were predicted, with 28,154 (94.81%) functionally annotated genes. We found 99.5% and 91.3% complete embryophyta_odb10 genes in the pseudo-chromosomes genome and predicted gene datasets by BUSCO assessment. The present genomic resource provides essential information for subsequent research on the mechanisms of environmental adaptation of S. rostratum and host shift in Colorado potato beetles.
Source apportionment of atmospheric water over East Asia - a source tracer study in CAM5.1
The atmospheric water tracer (AWT) method is implemented in the Community Atmosphere Model version 5.1 (CAM5.1) to quantitatively identify the contributions of various source regions to precipitation and water vapour over East Asia. Compared to other source apportionment methods, the AWT method was developed based on detailed physical parameterisations, and can therefore trace the behaviour of atmospheric water substances directly and exactly. According to the simulation, the northern Indian Ocean (NIO) is the dominant oceanic moisture source region for precipitation over the Yangtze River valley (YRV) and southern China (SCN) in summer, while the north-western Pacific (NWP) dominates during other seasons. Evaporation over the South China Sea (SCS) is responsible for only 2.7-3.7% of summer precipitation over the YRV and SCN. In addition, the Indo-China Peninsula is an important terrestrial moisture source region (annual contribution of ∼ 10%). The overall relative contribution of each source region to the water vapour amount is similar to the corresponding contribution to precipitation over the YRV and SCN. A case study for the SCS shows that only a small part ( ≤ 5.5%) of water vapour originates from local evaporation, whereas much more water vapour is supplied by the NWP and NIO. In addition, because evaporation from the SCS represents only a small contribution to the water vapour over the YRV and SCN in summer, the SCS mainly acts as a water vapour transport pathway where moisture from the NIO and NWP meet.
Quantifying Arctic lower stratospheric ozone sources in winter and spring
The dynamical and chemical characteristics of unusually low Arctic ozone events in 2005 and 2011 have been well-studied. However, the quantitative identification of Arctic ozone sources is lacking. Here, we use tagged ozone tracers in a numerical simulation to quantify the contributions to Arctic lower stratospheric ozone (ARCLS_O ) at diverse latitudes in winter and spring from 2005-2011. We demonstrate that the northern mid-latitudinal stratosphere steadily contributes approximately half of ARCLS_O . The absolute contributions during February have evident variations, which are smaller in cold years (151.3 ± 7.0 Dobson units (DU) in 2005 and 139.0 ± 7.4 DU in 2011) and greater in warm years (182.6 ± 7.3 DU in 2006 and 164.6 ± 7.4 DU in 2009). The tropical stratosphere is also an important source. During February, its absolute contributions are 66.5 ± 11.5 DU (2005), 73.1 ± 4.7 DU (2011), 146.0 ± 9.0 DU (2006), and 153.7 ± 7.0 DU (2009). Before and after stratospheric warming, variations in the tropical components of ARCLS_O (51.8 DU in 2006 and 77.0 DU in 2009) are significantly larger than those in the mid-latitudinal components (17.6 DU in 2006 and 18.1 DU in 2009). These results imply that although the mid-latitudinal components of ARCLS_O are larger, the tropical components control stratospheric temperature-induced ARCLS_O anomalies in winter and spring.
Pharmacological EZH2 inhibition combined with retinoic acid treatment promotes differentiation and apoptosis in rhabdomyosarcoma cells
Abstract Background Rhabdomyosarcomas (RMS) are predominantly paediatric sarcomas thought to originate from muscle precursor cells due to impaired myogenic differentiation. Despite intensive treatment, 5-year survival for patients with advanced disease remains low (< 30%), highlighting a need for novel therapies to improve outcomes. Differentiation therapeutics are agents that induce differentiation of cancer cells from malignant to benign. The histone methyltransferase, Enhancer of Zeste Homolog 2 (EZH2) suppresses normal skeletal muscle differentiation and is highly expressed in RMS tumours. Results We demonstrate combining inhibition of the epigenetic modulator EZH2 with the differentiating agent retinoic acid (RA) is more effective at reducing cell proliferation in RMS cell lines than single agents alone. In PAX3-FOXO1 positive RMS cells this is due to an RA-driven induction of the interferon pathway resulting in apoptosis. In fusion negative RMS, combination therapy led to an EZH2i-driven upregulation of myogenic signalling resulting in differentiation. In both subtypes, EZH2 is significantly associated with enrichment of trimethylated lysine 27 on histone 3 (H3K27me3) in genes that are downregulated in untreated RMS cells and upregulated with EZH2 inhibitor treatment. These results provide insight into the mechanism that drives the anti-cancer effect of the EZH2/RA single agent and combination treatment and indicate that the reduction of EZH2 activity combined with the induction of RA signalling represents a potential novel therapeutic strategy to treat both subtypes of RMS. Conclusions The results of this study demonstrate the potential utility of combining EZH2 inhibitors with differentiation agents for the treatment of paediatric rhabdomyosarcomas. As EZH2 inhibitors are currently undergoing clinical trials for adult and paediatric solid tumours and retinoic acid differentiation agents are already in clinical use this presents a readily translatable potential therapeutic strategy. Moreover, as inhibition of EZH2 in the poor prognosis FPRMS subtype results in an inflammatory response, it is conceivable that this strategy may also synergise with immunotherapies for a more effective treatment in these patients.
