Discourse, grammar and ideology : functional and cognitive perspectives
Researchers in critical discourse analysis (CDA) have often pointed to grammar as a locus of ideology in discourse. This book illustrates the role that grammars as models of language (and image) can play in revealing ideological properties of texts and discourse in social and political contexts. The book takes the reader through three distinct grammatical frameworks - functional grammar, multimodal grammar and cognitive grammar. Using examples taken from a range of discourses relating to globalisation, including discourses of immigration, war, corporate practice and political protests, the book demonstrates the individual utility and the interconnectedness of these models inside CDA. A key argument advanced is that the cognitive processes necessarily involved in making sense of language are based in visual experience. This position offers new ways of understanding the ideological effects of grammatical choices in texts and suggests a reassessment of the relationship between linguistic and multimodal grammars in CDA. The book will appeal to students and researchers interested in CDA and the relationship between discourse, cognition and social action.

Point-of-care antimicrobial coating protects orthopaedic implants from bacterial challenge
Implant related infections are the most common cause of joint arthroplasty failure, requiring revision surgeries and a new implant, resulting in a cost of $8.6 billion annually. To address this problem, we created a class of coating technology that is applied in the operating room, in a procedure that takes less than 10 min, and can incorporate any desired antibiotic. Our coating technology uses an in situ coupling reaction of branched poly(ethylene glycol) and poly(allyl mercaptan) (PEG-PAM) polymers to generate an amphiphilic polymeric coating. We show in vivo efficacy in preventing implant infection in both post-arthroplasty infection and post-spinal surgery infection mouse models. Our technology displays efficacy with or without systemic antibiotics, the standard of care. Our coating technology is applied in a clinically relevant time frame, does not require modification of implant manufacturing process, and does not change the implant shelf life.
Implant infection is the most common mode of joint replacement failure with serious complications. Here, the authors report on the in vivo application of a prophylactic coating technology that can incorporate a range of antibiotics and be applied in the operating room prior to implantation.

Chemical modification of PS-ASO therapeutics reduces cellular protein-binding and improves the therapeutic index
The molecular mechanisms of toxicity of chemically modified phosphorothioate antisense oligonucleotides (PS-ASOs) are not fully understood. Here, we report that toxic gapmer PS-ASOs containing modifications such as constrained ethyl (cEt), locked nucleic acid (LNA) and 2′-O-methoxyethyl (2′-MOE) bind many cellular proteins with high avidity, altering their function, localization and stability. We show that RNase H1–dependent delocalization of paraspeckle proteins to nucleoli is an early event in PS-ASO toxicity, followed by nucleolar stress, p53 activation and apoptotic cell death. Introduction of a single 2′-O-methyl (2′-OMe) modification at gap position 2 reduced protein-binding, substantially decreasing hepatotoxicity and improving the therapeutic index with minimal impairment of antisense activity. We validated the ability of this modification to generally mitigate PS-ASO toxicity with more than 300 sequences. Our findings will guide the design of PS-ASOs with optimal therapeutic profiles.Chemical modification of PS-ASO therapeutics reduces binding to cellular proteins and decreases toxic side-effects.

Contemporary Critical Discourse Studies
CDS is a multifarious field constantly developing different methodological frameworks for analysing dynamically evolving aspects of language in a broad range of socio-political and institutional contexts. This volume is a cutting edge, interdisciplinary account of these theoretical and empirical developments. It presents an up-to-date survey of Critical Discourse Studies (CDS), covering both the theoretical landscape and the analytical territories that it extends over. It is intended for critical scholars and students who wish to keep abreast of the current state of the art. The book is divided into two parts. In the first part, the chapters are organised around different methodological perspectives for CDS (history, cognition, multimodality and corpora, among others). In the second part, the chapters are organised around particular discourse types and topics investigated in CDS, both traditionally (e.g. issues of racism and gender inequality) and only more recently (e.g. issues of health, public policy, and the environment).
