The inhibition of advanced glycation end-products by five fractions and three main flavonoids from Camellia nitidissima Chi flowers

Camellia nitidissima Chi (CNC), belonging to Camellia genus (Theaceae family), is a medicinal and edible plant in China. Among the whole plant, the CNC flowers are especially precious, but the biological activities and the compositions of the CNC flowers are unknown. In this study, inhibiting effects on the formation of advanced glycation end-products (AGEs) of five CNC flowers fractions and three isolated compounds were investigated, these three compounds are two flavonoid glycosides and one flavanol, namely kaempferol 3-O-[2,3,4-Tri-O-acetyl-α-L-rhamnopyranosyl-(1→3)-2,4-di-O-acetyl-α-L-rhamnopyranosyl-(1→6)]-β-D-glucopyranoside, kaempferol 3-O-[2,3,4-Tri-O-acetyl-α-L-rhamnopyranosyl-(1→3)-4-O-acetyl-α-L-rhamnopyranosyl-(1→6)]-β-D-glucopyranoside and catechin. Among these five fractions, the ethyl acetate fraction showed the highest total phenolic contents and inhibiting effects on AGE formation. Bovine serum albumin (BSA)-glucose and BSA-methylglyoxal assay showed that the ethyl acetate fraction inhibited AGE formation by 74.49% and 34.3% at 1 mg/mL, respectively. As the main components, these three compounds also showed remarkable inhibiting effects on AGE formation by scavenging methylglyoxal, next two catechin-carbonyl adducts were identified using HPLC-ESI-MS/MS. The results showed that the CNC flowers had remarkable inhibiting effects on the formation of AGEs. The primary structure-activity relationship showed (1) the glycosides could reduce the inhibiting effects compared to kaempferol and (2) the acetyl at position 2‴ in compound 1 had no remarkable influence of the inhibiting effects on AGE formation compared to compound 2.

Efficacy of Qingpeng ointment (a Tibetan medicine) for acute gouty arthritis: a multi-center, randomized, double-blind, placebo-controlled trial

Background
This study aims to assess the efficacy and safety of Qingpeng ointment (QPO), a Tibetan medicine for alleviating symptoms in individuals with acute gouty arthritis (AGA).
Methods
This study was a randomized, double-blind, placebo-controlled trial that involved individuals with AGA whose joint pain, as measured on a visual analog scale (VAS) from 0 to 10, was equal to or greater than 3. The participants were randomly assigned to either the QPO or the placebo group and received their respective treatments twice daily for seven consecutive days. In case of intolerable pain, the participants were allowed to use diclofenac sodium sustained-release tablets as a rescue medicine. The primary outcomes measured were joint pain and swelling, while the secondary outcomes included joint mobility, redness, serum uric acid levels, C-reactive protein levels, and the amount of remaining rescue medicine. Any adverse events that occurred during the trial were also recorded.
Results
A total of 203 cases were divided into two groups, with balanced baselines: 102 in the QPO group and 101 in the placebo group. For joint pain, differences between the groups were notable in the VAS scores [1.75 (0, 3.00) versus 2.00 (1.00, 3.50);

= 0.038], changes in VAS [5.00 (3.00, 6.00) versus 4.00 (2.00, 6.00);
P
= 0.036], and disappearance rate [26.47% compared to 15.84%;

= 0.046] after treatment. Concerning joint swelling, significant between-group differences were observed in the VAS scores [1.00 (0, 2.30) versus 2.00 (0.70, 3.00);

= 0.032] and disappearance rate [33.33% compared to 21.78%;

= 0.046] at treatment completion. The QPO group exhibited a statistically significant mobility improvement compared to the placebo group (

= 0.004). No significant differences were found in other secondary outcomes. Five patients, four from the QPO group and one from the other, encountered mild adverse events, primarily skin irritation. All of these cases were resolved after dosage reduction or discontinuation of the medication.
Conclusions
Compared to the placebo, QPO exhibits positive effects on AGA by alleviating pain, reducing swelling, and enhancing joint mobility, without causing significant adverse effects.

Background:Sarpogrelate is a selective 5-hydroxytryptamine (5-HT) receptor subtype 2A antagonist which blocks 5-HT induced platelet aggregation and proliferation of vascular smooth muscle cells.We compared the efficacy of sarpogrelate-based dual antiplatelet therapies for the prevention of restenosis and target lesion revascularization (TLR) rates comparing with that of clopidogrel after percutaneous endovascular interventions (EVIs) of femoropopliteal (FP) arterial lesions.Methods:This prospective,multicenter,randomized clinical trial recruited a total of 120 patients with successful EVI of FP lesions at seven centers across China between January 2011 and June 2012.Patients were randomized to receive either sarpogrelate (100 mg trice daily for 6 months,n =63) or clopidogrel (75 mg once daily for 6 months,n =57).All patients also received oral aspirin (100 mg once daily for 12 months).Clinical follow-up was conducted up to 12 months postprocedure.Results:There was no significant difference between the two groups in basic demographic data.The restenosis rate was higher in the clopidogrel group (22.80%) than in sarpogrelate group (17.50%),but there was no significant difference between these two groups (P =0.465).The TLR rate,ipsilateral amputation rate,mortality in all-cause and bleeding rate were also similar in the two groups (P 〉 0.05).Conclusions:Aspirin plus sarpogrelate is a comparable antithrombotic regimen to aspirin plus clopidogrel after EVI of FP arterial lesions.Dual antiplatelet therapies might play an important role in preventing restenosis after successful EVI of FP lesions.

Study on Mix Proportion of C80 High-Strength Tri-Admixture Concrete

This paper studies the engineering application of C80 concrete with the feasibility of the raw material and production process. According to the design principle of high strength concrete and the method of the orthogonal design, The C80 high-strength tri-admixture concrete which includes fly ash, slag and volatilized silica was developed by using the local admixture. Further, the results show that the designed of the high-strength concrete based on the local material is available by adjusting and optimizing the amount of admixture reasonably. At last, the green production goal is achieved by reducing the dosage of cement, consuming industrial waste materials which playing an important role in its activity.

Failure experiments were carried out through a stretch-bending test system for advanced high strength steels, i.e. dual-phase (DP) steels and martensitic steels (MS). The die radius in this system was designed from 1 to 15 mm to investigate the failure mode under different geometries. Two failure modes were observed during the ex- periments. As a result, critical relative radii (the ratio of inner bending radius R to sheet thickness t) for DP590 and DP780 steels were obtained. The stretch-bending tests of DP980 display some trends unlike DP590 and DP780 steels, and curve of DP980 in different thicknesses does not coincide well. High blank holder force exhibits more possibility of shear fracture tendency than low blank holder force. The unique character of high strength martensitic steel (1500MS) is that no shear fracture is found especially over small bending radius (R =2 mm) under the same experi- mental conditions. Microstructure analysis indicates that there are obviously elongated grains on shear fracture sur- face. It shows smaller diameter and shallower depth of the dimples than the necking failure.

Study on the Impact of Polycarboxylic Acid Additive Experimental Process Parameters on Cement Adaptability

By analyzing two Polycarboxylate cement admixture of different polymerization process adaptability, the cement paste experiment is carried out to show the influence of Polycarboxylate superplasticizer, slump agent and the cement synthesis process on cement adaptability. Through orthogonal design method, result is drawn from the comparison of the influence of initial liquidity and an hour liquidity under different conditions,which is base on treating Polymeric acid process parameters as effecting factor within a certain range. It is shown that the preferably optimized process parameters of Polycarboxylic acid exist in the same kind of cement.The system error caused by the impact of cement composition is indicated in different kinds of cement.The adaptability of Polycarboxylic acid cement admixture of the mother liquor is available in the process optimization of the parameters.

Research on the Optimization Method of Compatibility between Polycarboxylic Admixture and Cement

In this paper, the method to adjust the production process of polycarboxylic admixtures mother liquid is researched according to the production of polycarboxylic admixtures. One polycarboxylic admixture cannot be adapted to all cements because of the influence of the mother liquid, although it could be adjusted by composite stabilizer. This paper studies the method of parameter adjustment on the level of factor analysis according to the process of polycarboxylic admixtures. The fluidity of cement slurry test is carried out when the type of cement is changed. On the basis of the test results the parameters are analyzed to design the molecular structure of the polycarboxylic admixtures, which could solve the problem of compatibility between polycarboxylic admixture and cement fundamentally.

In this paper,study the influence from concrete mixture proportions to poly carboxylate super plasticizer utility, discuss the net of gelled material pulp of liquidity and concrete slump collapse performance issues,analysis fresh concrete and gradation impact on the performance of concrete ,according to the concrete engineering practice.The conclusion is that the above situation can give an explanation in the super plasticizer mechanism level,and can be used in the selective use of admixture ,as well as reference for the choice of materials of concrete .

Aim: Fatty acid-binding protein 4 (FABP4) plays an important role in maintaining glucose and lipid homeostasis. The aim of this study was to find new inhibitors of FABP4 for the treatment of type 2 diabetes. Methods: Human FABP4 protein was expressed, and its inhibitors were detected in 1,8-ANS displacement assay. The effect of the inhibitor on lipolysis activity was examined in mouse 3T3-L1 preadipocytes. The db/db mice were used to evaluate the anti-diabetic activity of the inhibitor. Molecular docking and site-directed mutagenesis studies were carried out to explore the binding mode between the inhibitor and FABP4. Results: From 232 compounds tested, benzbromarone (BBR), an old uricosuric drug, was discovered to be the best inhibitor of FABP4 with an IC~o value of 14.8 pmol/L. Furthermore, BBR (25 pmol/L) significantly inhibited forskolin-stimulated lipolysis in 3T3-L1 cells. Oral administration of BBR (25 or 50 mg/kg, for 4 weeks) dose-dependently reduced the blood glucose level and improved glucose tolerance and insulin resistance in db/db mice. Molecular docking revealed that the residues Ser55, Asp76, and Arg126 of FABP4 formed important interactions with BBR, which was confirmed by site-directed mutagenesis studies. Conclusion: BBR is an inhibitor of FABP4 and a potential drug candidate for the treatment of type 2 diabetes and atherosclerosis.
