A New Adsorbent Composite Material Based on Metal Fiber Technology and Its Application in Adsorption Heat Exchangers

In order to achieve process intensification for adsorption chillers and heat pumps, a new composite material was developed based on sintered aluminum fibers from a melt-extraction process and a dense layer of silico-aluminophosphate (SAPO-34) on the fiber surfaces. The SAPO-34 layer was obtained through a partial support transformation (PST) process. Preparation of a composite sample is described and its characteristic pore size distribution and heat conductivity are presented. Water adsorption data obtained under conditions of a large pressure jump are given. In the next step, preparation of the composite was scaled up to larger samples which were fixed on a small adsorption heat exchanger. Adsorption measurements on this heat exchanger element that confirm the achieved process intensification are presented. The specific cooling power for the adsorption step per volume of composite is found to exceed 500 kW/m3 under specified conditions.

Hybrid Cascade Heat Pump and Thermal-Electric Energy Storage System for Residential Buildings: Experimental Testing and Performance Analysis

The need for innovative heating and cooling systems to decarbonize the building sector is widely recognized. It is especially important to increase the share of renewables at building level by maximizing self-consumption and reducing the primary energy demand. Accordingly, in the present paper, the results on a wide experimental campaign on a hybrid system are discussed. The system included a sorption module working as the topping cycle in a cascade configuration with a DC-driven vapor compression heat pump. A three-fluids heat exchanger with a phase change material (PCM), i.e., RT4 with nominal melting temperature of 4 °C, was installed on the evaporator side of the heat pump, for simultaneous operation as thermal storage and heat pumping purposes. The heat pump was connected to a DC-bus that included PV connection and electricity storage (batteries). Results showed that the energy efficiency of the heat pump in cascade operation was double compared to compression-only configuration and that, when simultaneously charging and discharging the latent storage in cascade configuration, no penalization in terms of efficiency compared to the compression-only configuration was measured. The self-sufficiency of the system was evaluated for three reference weeks in summer conditions of Athens climate and it was found that up to 100% of the electricity needed to drive the system could be self-produced for a modest cooling demand and up to 67% for the warmer conditions with high cooling demand.

Radiotheranostics, injectable radiopharmaceuticals with antitumour effects, have seen rapid development over the past decade. Although some formulations are already approved for human use, more radiopharmaceuticals will enter clinical practice in the next 5 years, potentially introducing new therapeutic choices for patients. Despite these advances, several challenges remain, including logistics, supply chain, regulatory issues, and education and training. By highlighting active developments in the field, this Review aims to alert practitioners to the value of radiotheranostics and to outline a roadmap for future development. Multidisciplinary approaches in clinical trial design and therapeutic administration will become essential to the continued progress of this evolving therapeutic approach.

The Centers for Medicare and Medicaid Services has required minimum procedural volumes as a condition of reimbursement for TAVR. CMS is reviewing this policy. In this analysis of data from 2015 through 2017, mortality at 30 days was higher among patients treated at hospitals or by operators with a lower volume of TAVR procedures.

Artemisinin kills malaria parasites by damaging proteins and inhibiting the proteasome

Artemisinin and its derivatives (collectively referred to as ARTs) rapidly reduce the parasite burden in
Plasmodium falciparum
infections, and antimalarial control is highly dependent on ART combination therapies (ACTs). Decreased sensitivity to ARTs is emerging, making it critically important to understand the mechanism of action of ARTs. Here we demonstrate that dihydroartemisinin (DHA), the clinically relevant ART, kills parasites via a two-pronged mechanism, causing protein damage, and compromising parasite proteasome function. The consequent accumulation of proteasome substrates, i.e., unfolded/damaged and polyubiquitinated proteins, activates the ER stress response and underpins DHA-mediated killing. Specific inhibitors of the proteasome cause a similar build-up of polyubiquitinated proteins, leading to parasite killing. Blocking protein synthesis with a translation inhibitor or inhibiting the ubiquitin-activating enzyme, E1, reduces the level of damaged, polyubiquitinated proteins, alleviates the stress response, and dramatically antagonizes DHA activity.
Artemisinin (ART) is a widely used antimalarial drug, but its mechanism of action is poorly understood. Here, Bridgford et al. show that ART kills parasites by a two-pronged mechanism, causing protein damage and compromising proteasome function, and that accumulation of proteasome substrates activates the ER stress response.

Transcatheter aortic valve replacement (TAVR) with the SAPIEN 3 valve demonstrates good 30 day clinical outcomes in patients with severe aortic stenosis who are at intermediate risk of surgical mortality. Here we report longer-term data in intermediate-risk patients given SAPIEN 3 TAVR and compare outcomes to those of intermediate-risk patients given surgical aortic valve replacement.
In the SAPIEN 3 observational study, 1077 intermediate-risk patients at 51 sites in the USA and Canada were assigned to receive TAVR with the SAPIEN 3 valve [952 [88%] via transfemoral access) between Feb 17, 2014, and Sept 3, 2014. In this population we assessed all-cause mortality and incidence of strokes, re-intervention, and aortic valve regurgitation at 1 year after implantation. Then we compared 1 year outcomes in this population with those for intermediate-risk patients treated with surgical valve replacement in the PARTNER 2A trial between Dec 23, 2011, and Nov 6, 2013, using a prespecified propensity score analysis to account for between-trial differences in baseline characteristics. The clinical events committee and echocardiographic core laboratory methods were the same for both studies. The primary endpoint was the composite of death from any cause, all strokes, and incidence of moderate or severe aortic regurgitation. We did non-inferiority (margin 7·5%) and superiority analyses in propensity score quintiles to calculate pooled weighted proportion differences for outcomes.
At 1 year follow-up of the SAPIEN 3 observational study, 79 of 1077 patients who initiated the TAVR procedure had died (all-cause mortality 7·4%; 6·5% in the transfemoral access subgroup), and disabling strokes had occurred in 24 (2%), aortic valve re-intervention in six (1%), and moderate or severe paravalvular regurgitation in 13 (2%). In the propensity-score analysis we included 963 patients treated with SAPIEN 3 TAVR and 747 with surgical valve replacement. For the primary composite endpoint of mortality, strokes, and moderate or severe aortic regurgitation, TAVR was both non-inferior (pooled weighted proportion difference of −9·2%; 90% CI −12·4 to −6; p<0·0001) and superior (−9·2%, 95% CI −13·0 to −5·4; p<0·0001) to surgical valve replacement.
TAVR with SAPIEN 3 in intermediate-risk patients with severe aortic stenosis is associated with low mortality, strokes, and regurgitation at 1 year. The propensity score analysis indicates a significant superiority for our composite outcome with TAVR compared with surgery, suggesting that TAVR might be the preferred treatment alternative in intermediate-risk patients.

A highly immunogenic and effective measles virus-based Th1-biased COVID-19 vaccine

The COVID-19 pandemic is caused by severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) and has spread worldwide, with millions of cases and more than 1 million deaths to date. The gravity of the situation mandates accelerated efforts to identify safe and effective vaccines. Here, we generated measles virus (MeV)-based vaccine candidates expressing the SARS-CoV-2 spike glycoprotein (S). Insertion of the full-length S protein gene in two different MeV genomic positions resulted in modulated S protein expression. The variant with lower S protein expression levels was genetically stable and induced high levels of effective Th1-biased antibody and T cell responses in mice after two immunizations. In addition to neutralizing IgG antibody responses in a protective range, multifunctional CD8⁺ and CD4⁺ T cell responses with S protein-specific killing activity were detected. Upon challenge using a mouse-adapted SARS-CoV-2, virus loads in vaccinated mice were significantly lower, while vaccinated Syrian hamsters revealed protection in a harsh challenge setup using an early-passage human patient isolate. These results are highly encouraging and support further development of MeV-based COVID-19 vaccines.

Chimpanzees adapt their exploration to key properties of the environment

Exploration is an important strategy for reducing the uncertainty that pervades daily life. Yet the evolutionary roots of adaptive exploration are poorly understood. We harness and adapt the human decisions-from-experience paradigm to investigate exploration under uncertainty in chimpanzees. In our study, chimpanzees (
N
= 15; eight females) are simultaneously confronted with an uncertain option (with outcome variance) and a safe option (without outcome variance) and tested in both stable and changing environments. Results reveal that, as in human exploration, how and how much chimpanzees explore depends on the environment. One key environmental property is change: Chimpanzees explore more across trials in changing than in stable conditions. Consistent with the assumption of classic economic models that variance indicates risk, chimpanzees also explore more when they experience variance in the options’ outcomes. Individual risk and uncertainty preferences did not have a statistically significant effect on exploratory efforts. These findings suggest that chimpanzees and humans share key similarities in the way they respond to risk and uncertainty.
Exploration helps reduce uncertainty in daily life, but the evolutionary roots of adaptive exploration are unclear. Here, the authors show that chimpanzees, like humans, tailor their exploration depending on their environment.

Worldwide, more than one million people die on the roads each year. A third of these fatal accidents are attributed to speeding, with properties of the individual driver and the environment regarded as key contributing factors. We examine real-world speeding behavior and its interaction with illuminance, an environmental property defined as the luminous flux incident on a surface. Drawing on an analysis of 1.2 million vehicle movements, we show that reduced illuminance levels are associated with increased speeding. This relationship persists when we control for factors known to influence speeding (e.g., fluctuations in traffic volume) and consider proxies of illuminance (e.g., sight distance). Our findings add to a long-standing debate about how the quality of visual conditions affects drivers' speed perception and driving speed. Policy makers can intervene by educating drivers about the inverse illuminance‒speeding relationship and by testing how improved vehicle headlights and smart road lighting can attenuate speeding.
