Primate mosaic brain evolution reflects selection on sensory and cognitive specialization

The mammalian brain is composed of numerous functionally distinct structures that vary in size within and between clades, reflecting selection for sensory and cognitive specialization. Primates represent a particularly interesting case in which to examine mosaic brain evolution since they exhibit marked behavioural variation, spanning most social structures, diets and activity periods observed across mammals. Although studies have consistently demonstrated a trade-off between visual and olfactory specialization in primates, studies of some regions (for example, the neocortex) have produced conflicting results. Here, we analyse the socioecological factors influencing the relative size of 33 brain regions, using updated statistical techniques and data from more species and individuals than previous studies. Our results confirm that group-living species and those with high-quality diets have expanded olfactory or visual systems, depending on whether they are nocturnal or diurnal. Conversely, regions associated with spatial memory are expanded in solitary species and those with low-quality diets, suggesting a trade-off between visual processing and spatial memory. Contrary to previous work, we show that diet quality predicts relative neocortex size at least as well as, if not better than, social complexity. Overall, our results demonstrate that primate brain structure is largely driven by selection on sensory and cognitive specializations that develop in response to divergent socioecological niches.

Weather-related disasters are increasing in frequency and severity, leaving survivors to cope with ensuing mental, financial, and physical hardships. This adversity can exacerbate existing morbidities, trigger new ones, and increase the risk of mortality—features that are also characteristic of advanced age—inviting the hypothesis that extreme weather events may accelerate aging. To test this idea, we examined the impact of Hurricane Maria and its aftermath on immune cell gene expression in large, age-matched, cross-sectional samples from free-ranging rhesus macaques (Macaca mulatta) living on an isolated island. A cross section of macaques was sampled 1 to 4 y before (n = 435) and 1 y after (n = 108) the hurricane. Hurricane Maria was significantly associated with differential expression of 4% of immune-cell-expressed genes, and these effects were correlated with age-associated alterations in gene expression. We further found that individuals exposed to the hurricane had a gene expression profile that was, on average, 1.96 y older than individuals that were not—roughly equivalent to an increase in 7 to 8 y of a human life. Living through an intense hurricane and its aftermath was associated with expression of key immune genes, dysregulated proteostasis networks, and greater expression of inflammatory immune cell-specific marker genes. Together, our findings illuminate potential mechanisms through which the adversity unleashed by extreme weather and potentially other natural disasters might become biologically embedded, accelerate age-related molecular immune phenotypes, and ultimately contribute to earlier onset of disease and death.

The rhesus macaque as a success story of the Anthropocene

Of all the non-human primate species studied by researchers, the rhesus macaque ( Macaca mulatta ) is likely the most widely used across biological disciplines. Rhesus macaques have thrived during the Anthropocene and now have the largest natural range of any non-human primate. They are highly social, exhibit marked genetic diversity, and display remarkable niche flexibility (which allows them to live in a range of habitats and survive on a variety of diets). These characteristics mean that rhesus macaques are well-suited for understanding the links between sociality, health and fitness, and also for investigating intra-specific variation, adaptation and other topics in evolutionary ecology.

A fundamental mistake in receptor theory has led to an enduring misunderstanding of how to estimate the affinity and efficacy of an agonist. These properties are inextricably linked and cannot be easily separated in any case where the binding of a ligand induces a conformation change in its receptor. Consequently, binding curves and concentration–response relationships for receptor agonists have no straightforward interpretation. This problem—the affinity–efficacy problem—remains overlooked and misunderstood despite it being recognized in 1987. To avoid the further propagation of this misunderstanding, we propose in this review that the affinity–efficacy problem should be included in the core curricula for pharmacology undergraduates proposed by the British Pharmacological Society and the International Union of Basic and Clinical Pharmacology (IUPHAR).

Strong social bonds in gregarious adult animals have been associated with lower levels of glucocorticoids. However, similar research is lacking for juvenile primates. We examined relationships between social bonds and mean concentrations of fecal glucocorticoid metabolites (fGCMs) in 44 free-ranging juvenile rhesus macaques (
Macaca mulatta
) on Cayo Santiago, Puerto Rico. We measured frequencies of affiliative behavior (grooming, play, approaches and proximity) with other same-sex, same-aged juveniles (peers) and the total number of affiliative peer relationships. We found a positive relationship between fGCMs and grooming frequencies. Females that spent more time in proximity to peers also had higher fGCMs. In contrast, among juveniles with more closely related peers, those with more affiliative peers or more frequent play bouts had lower fGCMs. However, strong peer bonds in most juveniles did not appear to be associated with reduced glucocorticoid levels. fGCMs were higher for females than males, but were unassociated with physical activity, aggression, or peer seeking tendencies. We propose that the establishment and navigation of some peer bonds at this life stage may involve increased metabolic demand.

Vocalizations differ substantially between the sexes in many primates, and low-frequency male vocalizations may be favored by sexual selection because they intimidate rivals and/or attract mates. Sexual dimorphism in fundamental frequency may be more pronounced in species with more intense male mating competition and in those with large group size, where social knowledge is limited and efficient judgment of potential mates and competitors is crucial. These non-mutually exclusive explanations have not been tested simultaneously across primate species. In a sample of vocalizations (
n
= 1914 recordings) across 37 anthropoid species, we investigated whether fundamental frequency dimorphism evolved in association with increased intensity of mating competition (H1), large group size (H2), multilevel social organization (H3), a trade-off against the intensity of sperm competition (H4), and/or poor acoustic habitats (H5), controlling for phylogeny and body size dimorphism. We show that fundamental frequency dimorphism increased in evolutionary transitions towards larger group size and polygyny. Findings suggest that low-frequency male vocalizations in primates may have been driven by selection to win mating opportunities by avoiding costly fights and may be more important in larger groups, where limited social knowledge affords advantages to rapid assessment of status and threat potential via conspicuous secondary sexual characteristics.
Sexual dimorphism in the fundamental frequency of primate vocalizations is variable. Here, the authors examine 1914 vocalizations from 37 anthropoid species to find that fundamental frequency dimorphism increased with larger group size and polygyny, due to sexual selection.

Females influence offspring paternity through diverse pre- and post-copulatory mechanisms. Sperm discrimination—the differential physiological response to ejaculates based on male or sperm characteristics—can bias fertilization outcomes, but in vivo evidence of this process in large-bodied mammals is lacking. Here, in a study of nine females and four males, we tested whether two aspects of female physiology that affect sperm survival—vaginal immune response and pH—are modulated by male genetic makeup in a non-human primate, the olive baboon ( Papio anubis ). Our findings suggest post-copulatory differences in vaginal gene expression and pH, with the strongest immune responses and largest pH decreases, harmful to sperm, exhibited by females mating with genetically similar males. These findings are consistent with genetically-based post-copulatory mate discrimination, offering new insights into how interactions between male gametes and the female reproductive tract may shape conception probability in primates.

Primates appear unusual among mammals in the expression of female colorful ornaments in the absence of sex role reversal. Most studies of female ornamentation in primates have focused on the sexual signaling function of female exaggerated anogenital swellings in female-female competition and male attraction, but other female colorful ornaments, such as red skin coloration, may also contain information about reproductive status and individual characteristics. We analyzed variation in facial and hindquarter coloration (redness and luminance) according to the timing of the fertile phase (intra-cycle variation), the cycle number, whether the cycle was conceptive or non-conceptive (inter-cycle variation), and in relation to individual characteristics (social rank, parity, and body mass) in 12 captive female Japanese macaques (Macaca fuscata). While facial and hindquarter coloration did not accurately indicate the timing of the fertile phase, variation in hindquarter luminance signaled perceptible differences between pre- and post-ovulation stages. Hindquarters became less red, and faces were lighter as the number of consecutive cycles increased. Hindquarters were redder during non-conceptive cycles compared with conceptive ones. Individual variation in skin redness and luminance appeared perceptible under good light conditions. Higher-ranking females had darker hindquarters. We also found that variation in female skin coloration may contain information about differences in body mass but not in parity. Female skin coloration in Japanese macaques may thus be more indicative of inter-cycle variation and various specific female characteristics than the timing of the fertile phase. Our study provides insight into the potential information content of this signal and demonstrates the characteristics that males might be selecting for should males prefer redder females.

Character displacement of Cercopithecini primate visual signals

Animal visual signals have the potential to act as an isolating barrier to prevent interbreeding of populations through a role in species recognition. Within communities of competing species, species recognition signals are predicted to undergo character displacement, becoming more visually distinctive from each other; however, this pattern has rarely been identified. Using computational face recognition algorithms to model primate face processing, we demonstrate that the face patterns of guenons (tribe: Cercopithecini) have evolved under selection to become more visually distinctive from those of other guenon species with whom they are sympatric. The relationship between the appearances of sympatric species suggests that distinguishing conspecifics from other guenon species has been a major driver of diversification in guenon face appearance. Visual signals that have undergone character displacement may have had an important role in the tribe’s radiation, keeping populations that became geographically separated reproductively isolated on secondary contact.
