Integrating Methods and Empirical Findings from Social and Behavioural Sciences into Energy System Models—Motivation and Possible Approaches
The transformation of the energy system is a highly complex process involving many dimensions. Energy system models help to understand the process and to define either target systems or policy measures. Insights derived from the social sciences are not sufficiently represented in energy system models, but address crucial aspects of the transformation process. It is, therefore, necessary to develop approaches to integrate results from social science studies into energy system models. Hence, as a result of an interdisciplinary discourse among energy system modellers, social scientists, psychologists, economists and political scientists, this article explains which aspects should be considered in the models, how the respective results can be collected and which aspects of integration into energy system models are conceivable to provide an overview for other modellers. As a result of the discourse, five facets are examined: Investment behaviour (market acceptance), user behaviour, local acceptance, technology innovation and socio-political acceptance. Finally, an approach is presented that introduces a compound of energy system models (with a focus on the macro and micro-perspective) as well as submodels on technology genesis and socio-political acceptance, which serves to gain a more fundamental knowledge of the transformation process.
El significado del desarrollo anidado de las literacidades y numeracidades académicas en el currículum universitario
Ante el gran desafío de los deficientes niveles de literacidad y numeracidad en Chile, la Facultad de Educación de la Universidad Santo Tomás impulsó desde el año 2011 un modelo anidado o transversal de desarrollo de estas habilidades. El presente artículo detalla los resultados en torno al significado de esta estrategia para sus estudiantes y académicos, que se sustanciaron a través de grupos focales, analizados a través de categorización y triangulación. Los hallazgos sustentan la relevancia de estas habilidades en la profesión, su presencia en las actividades prácticas y su anidamiento en contextos concretos, lo que reduce la visibilidad de las actividades de apoyo. Futuras investigaciones debieran abordar el desarrollo de la numeracidad académica, y las literacidades profesionales.
Progression of lipase activity and pancreatic lipase immunoreactivity in dogs hospitalized for acute pancreatitis and correlation with clinical features
Abstract Background Lipase activity and pancreatic lipase immunoreactivity (PLI) have not been compared in dogs hospitalized for acute pancreatitis (AP). Objectives To describe the progression of lipase activity and PLI, and correlations with clinicopathologic features in dogs with AP. Animals Thirty-nine dogs with AP based on clinical signs and lipase activity >350 U/L (reference interval [RI], 24-108 U/L). Methods Retrospective study. Lipase activity (LIPC Roche), PLI (SpecPL), and clinical signs were recorded daily. Admission (d1) data (clinical, laboratory, and ultrasound [US] findings), and clinical signs during hospitalization (d2-d3) were assessed for correlation with lipases. Results Median (range) duration of clinical signs before presentation was 2 days (1-7 days). Median (range) lipase activity and PLI at d1 were 1070 U/L (range, 357-1500 U/L) and 1111 μg/L (range, 292-1500 μg/L). Strong correlation between assays at d1 (rs 0.96; P < .0001; n = 39), remained equally strong on d2 (rs 0.964; P < .0001; n = 39), and d3 (rs 0.966; P < .0001; n = 22). On d2, lipase activity and PLI were within RI in 13/39 (33%) and 18/39 (46%) of cases. Lipase activities were minimally increased (median, 124 U/L) in 5 dogs with d2 PLI <200 μg/L. On d3, 4 more dogs had normal lipase activity and PLI, and the nature and magnitude of change were always the same for both assays. Clinical signs were not associated with lipases. Only a hyperechoic mesentery, but not an US diagnosis of AP, correlated significantly with lipase activity and PLI. Conclusions and Clinical Importance Lipase decreases rapidly to near or within RI within 2 days of treatment in the majority of dogs with AP. Both lipase assays yielded virtually identical results. Mesenteric echogenicity may be an early marker of AP in dogs.
Longitudinal assessment of thyroid function in dogs with hypoadrenocorticism: Clinical outcomes and prevalence of autoantibodies
Abstract Background Knowledge about primary hypoadrenocorticism coexisting with immune-mediated thyroiditis (Schmidt's syndrome) in dogs is limited. Objective To evaluate thyroid function in dogs with naturally occurring hypoadrenocorticism before and during treatment. Animals Sixty-six client-owned dogs. Methods Measurement of canine thyroid stimulating hormone (cTSH), total thyroxine (T4), free thyroxine, and autoantibodies against thyroglobulin, T4, and total triiodothyronine. Results Thirty-eight dogs were assessed before and 28 during treatment. Follow-up data were available for 24/38 and 17/28 dogs, with median follow-up duration of 3.8 years (range, <1.0-8.8 years) and 4 years (range, 1.1 weeks to 10.5 years), respectively. Canine thyroid stimulating hormone was above the reference range at the time of diagnosis of hypoadrenocorticism in 10 of 38 dogs but decreased into the reference range in 7 for which follow-up data was available. Hypothyroidism was confirmed in 5 dogs at a median age of 11 years (range, 7-15 years). In 4 dogs, the condition was diagnosed after a median treatment duration of 5.75 years (range, 2.6-10 years), while in 1 dog, the diagnosis was made concurrently. One dog had detectable thyroid autoantibodies. Conclusions and Clinical Relevance Hypothyroidism occurs as a rare concurrent condition in dogs with hypoadrenocorticism, potentially at any phase of treatment. Close monitoring of cTSH levels in these dogs could be beneficial, as early changes might indicate the onset of hypothyroidism. The low prevalence of detectable thyroid autoantibodies suggests that nonimmune mechanisms might contribute to thyroid dysfunction.
The emergence of functional microcircuits in visual cortex
A study of mouse visual cortex relating patterns of excitatory synaptic connectivity to visual response properties of neighbouring neurons shows that, after eye opening, local connectivity reorganizes extensively: more connections form selectively between neurons with similar visual responses and connections are eliminated between visually unresponsive neurons, but the overall connectivity rate does not change. Development in the neonatal visual cortex Intrinsic and experiential factors guide the patterning of neural pathways and the establishment of sensory response properties during postnatal development. Although sensory processing is known to depend on the precise wiring of cortical microcircuits, how this functional connectivity develops remains unclear. Based on electrical recordings of neighbouring neurons and changing network dynamics measures using calcium imaging, Thomas Mrsic-Flogel and colleagues offer a proposal that neuronal feature preference is initially acquired before sensory experience in a feed-forward manner, and with patterned input later driving the formation of precision within the network following the appropriate re-arrangement of synaptic connections. Sensory processing occurs in neocortical microcircuits in which synaptic connectivity is highly structured 1 , 2 , 3 , 4 and excitatory neurons form subnetworks that process related sensory information 5 , 6 . However, the developmental mechanisms underlying the formation of functionally organized connectivity in cortical microcircuits remain unknown. Here we directly relate patterns of excitatory synaptic connectivity to visual response properties of neighbouring layer 2/3 pyramidal neurons in mouse visual cortex at different postnatal ages, using two-photon calcium imaging in vivo and multiple whole-cell recordings in vitro . Although neural responses were already highly selective for visual stimuli at eye opening, neurons responding to similar visual features were not yet preferentially connected, indicating that the emergence of feature selectivity does not depend on the precise arrangement of local synaptic connections. After eye opening, local connectivity reorganized extensively: more connections formed selectively between neurons with similar visual responses and connections were eliminated between visually unresponsive neurons, but the overall connectivity rate did not change. We propose a sequential model of cortical microcircuit development based on activity-dependent mechanisms of plasticity whereby neurons first acquire feature preference by selecting feedforward inputs before the onset of sensory experience—a process that may be facilitated by early electrical coupling between neuronal subsets 7 , 8 , 9 —and then patterned input drives the formation of functional subnetworks through a redistribution of recurrent synaptic connections.
Evaluation of a low-dose desoxycorticosterone pivalate treatment protocol for long-term management of dogs with primary hypoadrenocorticism
Abstract Background Lowering the dose of desoxycorticosterone pivalate (DOCP) for the treatment of dogs with primary hypoadrenocorticism (PH) decreases costs and could lead to increased owner motivation to treat their affected dogs. Objective To evaluate the efficacy of a low-dose DOCP treatment protocol in dogs with PH. Animals Prospective study, 17 client-owned dogs with naturally occurring PH (12 newly diagnosed, 5 previously treated with fludrocortisone acetate [FC]). Methods Dogs with newly diagnosed PH were started on 1.5 mg/kg DOCP SC; dogs previously treated with FC were started on 1.0-1.8 mg/kg DOCP SC. Reevaluations took place at regular intervals for a minimum of 3 months and included clinical examination and determination of serum sodium and potassium concentrations. The DOCP dosage was adjusted to obtain an injection interval of 28-30 days and to keep serum electrolyte concentrations within the reference interval. Results Median (range) follow-up was 16.2 months (4.5-32.3 months). The starting dosage was sufficient in all but 2 dogs and had to be significantly decreased after 2-3 months to a median dosage (range) of 1.1 mg/kg (0.7-1.8). Dogs 3 years of age or younger needed significantly higher dosages compared to older dogs. None of them, however, needed the 2.2 mg/kg DOCP dosage, recommended by the manufacturer. Conclusions and Clinical Importance A starting dosage of 1.5 mg/kg DOCP is effective in controlling clinical signs and serum electrolyte concentrations in the majority of dogs with PH. An additional dose reduction often is needed to maintain an injection interval of 28-30 days. Young and growing animals seem to need higher dosages.
Clinical features and long-term management of cats with primary hypoadrenocorticism using desoxycorticosterone pivalate and prednisolone
Abstract Background Primary hypoadrenocorticism (PH) is rare in cats and knowledge about treatment is sparse. Objective To describe cats with PH with a focus on long-term treatment. Animals Eleven cats with naturally occurring PH. Methods Descriptive case series with data on signalment, clinicopathological findings, adrenal width, and doses of desoxycorticosterone pivalate (DOCP) and prednisolone during a follow-up period of >12 months. Results Cats ranged from 2 to 10 years (median 6.5); 6 cats were British Shorthair. Most common signs were reduced general condition and lethargy, anorexia, dehydration, obstipation, weakness, weight loss, and hypothermia. Adrenal glands on ultrasonography were judged small in 6. Eight cats could be followed for 14 to 70 months (median: 28). Two were started on DOCP doses ≥2.2 mg/kg (2.2; 2.5) and 6 < 2.2 mg/kg (1.5-2.0 mg/kg, median 1.8) q28 days. Both high-dose cats and 4 low-dose cats needed a dose increase. Desoxycorticosterone pivalate and prednisolone doses at the end of the follow-up period were 1.3 to 3.0 mg/kg (median: 2.3) and 0.08 to 0.5 mg/kg/day (median: 0.3), respectively. Conclusions and Clinical Importance Desoxycorticosterone pivalate and prednisolone requirements in cats were higher than what is currently used in dogs; thus, a DOCP starting dose of 2.2 mg/kg q28 days and a prednisolone maintenance dose of 0.3 mg/kg/day titrated to the individual need seems warranted. Small adrenal glands (width < 2.7 mm) on ultrasonography in a cat suspected of hypoadrenocorticism can be suggestive of the disease. The apparent predilection of British Shorthaired cats for PH should be further evaluated.
Post-mortem magnetic resonance imaging with computed tomography-guided biopsy for foetuses and infants: a prospective, multicentre, cross-sectional study
Background Post-mortem imaging has been suggested as an alternative to conventional autopsy in the prenatal and postnatal periods. Noninvasive autopsies do not provide tissue for histological examination, which may limit their clinical value, especially when infection-related morbidity and mortality are suspected. Methods We performed a prospective, multicentre, cross-sectional study to compare the diagnostic performance of post-mortem magnetic resonance imaging with computed tomography-guided biopsy (Virtopsy®) with that of conventional autopsy in foetuses and infants. Cases referred for conventional autopsy were eligible for enrolment. After post-mortem imaging using a computed tomography scanner and a magnetic resonance imaging unit, computed tomography-guided tissue sampling was performed. Virtopsy results were compared with conventional autopsy in determining the likely final cause of death and major pathologies. The primary outcome was the proportion of cases for which the same cause of death was determined by both methods. Secondary outcomes included the proportion of false positive and false negative major pathological lesions detected by virtopsy and the proportion of computed tomography-guided biopsies that were adequate for histological examination. Results Overall, 101 cases (84 fetuses, 17 infants) were included. Virtopsy and autopsy identified the same cause of death in 91 cases (90.1%, 95% CI 82.7 to 94.5). The sensitivity and specificity of virtopsy for determining the cause of death were 96.6% (95% CI 90.6 to 98.8) and 41.7% (95% CI 19.3 to 68.0), respectively. In 32 cases (31.7%, 95% CI 23.4 to 41.3), major pathological findings remained undetected by virtopsy, and in 45 cases (44.6%, 95% CI 35.2 to 54.3), abnormalities were diagnosed by virtopsy but not confirmed by autopsy. Computed tomography-guided tissue sampling was adequate for pathological comments in 506 of 956 biopsies (52.7%) and added important diagnostic value in five of 30 cases (16.1%) with an unclear cause of death before autopsy compared with postmortem imaging alone. In 19 of 20 infective deaths (95%), biopsies revealed infection-related tissue changes. Infection was confirmed by placental examination in all fetal cases. Conclusions Virtopsy demonstrated a high concordance with conventional autopsy for the detection of cause of death but was less accurate for the evaluation of major pathologies. Computed tomography-guided biopsy had limited additional diagnostic value. Trial registration ClinicalTrials.gov (NCT01888380).
Ice nucleation efficiency of natural dust samples in the immersion mode
A total of 12 natural surface dust samples, which were surface-collected on four continents, most of them in dust source regions, were investigated with respect to their ice nucleation activity. Dust collection sites were distributed across Africa, South America, the Middle East, and Antarctica. Mineralogical composition has been determined by means of X-ray diffraction. All samples proved to be mixtures of minerals, with major contributions from quartz, calcite, clay minerals, K-feldspars, and (Na, Ca)-feldspars. Reference samples of these minerals were investigated with the same methods as the natural dust samples. Furthermore, Arizona test dust (ATD) was re-evaluated as a benchmark. Immersion freezing of emulsion and bulk samples was investigated by differential scanning calorimetry. For emulsion measurements, water droplets with a size distribution peaking at about 2 µm, containing different amounts of dust between 0.5 and 50 wt % were cooled until all droplets were frozen. These measurements characterize the average freezing behaviour of particles, as they are sensitive to the average active sites present in a dust sample. In addition, bulk measurements were conducted with one single 2 mg droplet consisting of a 5 wt % aqueous suspension of the dusts/minerals. These measurements allow the investigation of the best ice-nucleating particles/sites available in a dust sample. All natural dusts, except for the Antarctica and ATD samples, froze in a remarkably narrow temperature range with the heterogeneously frozen fraction reaching 10 % between 244 and 250 K, 25 % between 242 and 246 K, and 50 % between 239 and 244 K. Bulk freezing occurred between 255 and 265 K. In contrast to the natural dusts, the reference minerals revealed ice nucleation temperatures with 2–3 times larger scatter. Calcite, dolomite, dolostone, and muscovite can be considered ice nucleation inactive. For microcline samples, a 50 % heterogeneously frozen fraction occurred above 245 K for all tested suspension concentrations, and a microcline mineral showed bulk freezing temperatures even above 270 K. This makes microcline (KAlSi3O8) an exceptionally good ice-nucleating mineral, superior to all other analysed K-feldspars, (Na, Ca)-feldspars, and the clay minerals. In summary, the mineralogical composition can explain the observed freezing behaviour of 5 of the investigated 12 natural dust samples, and partly for 6 samples, leaving the freezing efficiency of only 1 sample not easily explained in terms of its mineral reference components. While this suggests that mineralogical composition is a major determinant of ice-nucleating ability, in practice, most natural samples consist of a mixture of minerals, and this mixture seems to lead to remarkably similar ice nucleation abilities, regardless of their exact composition, so that global models, in a first approximation, may represent mineral dust as a single species with respect to ice nucleation activity. However, more sophisticated representations of ice nucleation by mineral dusts should rely on the mineralogical composition based on a source scheme of dust emissions.
