Development of a capsid protein-based ELISA for the detection of PCV2 antibodies in swine serum

Porcine circovirus type 2 (PCV2) is the major causative agent of postweaning multisystemic wasting syndrome which leads to significant economic losses in the global swine industry. In China, there is a widespread dissemination of PCV2 infection in the pig population. Serological diagnosis of the disease is considered as an effective control measure. Here, we developed a capsid protein (Cap)-based enzyme-linked immunosorbent assay (Cap-ELISA) for the detection of PCV2 antibodies in swine serum using a nuclear localization signal-truncated capsid protein produced in Escherichia coli. The Cap protein was expressed as water-soluble and purified using nickel-nitrilotriacetic acid (Ni-NTA) chromatography. After the optimization of the working conditions of the Cap-ELISA using chessboard titrations, a total of 649 serum samples were tested using the Cap-ELISA and a commercial ELISA kit. The diagnostic sensitivity (DSN), diagnostic specificity (DSP) and accuracy of the Cap-ELISA were determined to be 96.7%, 94.1% and 99.5%, respectively. Cross-reactivity analysis indicated that the Cap-ELISA was PCV2-specific and possessed no cross-reactions with antibodies against other common swine pathogens including porcine circovirus type 1 (PCV1), porcine reproductive and respiratory syndrome virus (PRRSV), classical swine fever virus (CSFV), porcine parvovirus (PPV), foot and mouth disease virus (FMDV), porcine epidemic diarrhea virus (PEDV) and pseudorabies virus (PRV). Repeatability of the experiment showed that Cap-ELISA was highly repeatable with the intra- and inter-plate coefficients of variation less than 10%. Hence, the Cap-ELISA has the potential for the swine industry to monitor PCV2 epidemiology and to evaluate PCV2 vaccine efficacy.

The full data set of the Daya Bay reactor neutrino experiment is used to probe the effect of the charged current non-standard interactions (CC-NSI) on neutrino oscillation experiments. Two different approaches are applied and constraints on the corresponding CC-NSI parameters are obtained with the neutrino flux taken from the Huber-Mueller model with a 5% uncertainty. Study on the modeling and application of arc structure's profile tolerance
Profile tolerance can play a comprehensive control role, and then it can replace some other kinds of geometric tolerances, so it is widely used in practical engineering. However, there are few studies on profile tolerance modeling. There are only several papers in public, and the modeling methods in these papers all have certain deficiencies. In addition, the arc structure is much more used in practical engineering to deal with approximation of free-form surfaces, and the arc structure is often controlled by profile tolerance. Therefore, a modeling method specifically for the arc profile tolerance is given, and a tolerance analysis method including arc structure is given by combining the profile tolerance modeling method and Vector Loop and Monte Carlo method. Statistical distribution of each tolerance is included in the tolerance analysis method, and an example is given.

Effectiveness of Exercise, Cognitive Behavioral Therapy, and Pharmacotherapy on Improving Sleep in Adults with Chronic Insomnia: A Systematic Review and Network Meta-Analysis of Randomized Controlled Trials

Despite the well-established treatment effectiveness of exercise, cognitive behavioral therapy for insomnia (CBT-I), and pharmacotherapy on improving sleep, there have been no studies to compare their long-term effectiveness, which is of clinical importance for sustainable management of chronic insomnia. This study compared the long-term effectiveness of these three interventions on improving sleep in adults with chronic insomnia. MEDLINE, PsycINFO, Embase, and SPORTDiscus were searched for eligible reports. Trials that investigated the long-term effectiveness of these three interventions on improving sleep were included. The post-intervention follow-up of the trial had to be ≥6 months to be eligible. The primary outcome was the long-term effectiveness of the three interventions on improving sleep. Treatment effectiveness was the secondary outcome. A random-effects network meta-analysis was carried out using a frequentist approach. Thirteen trials were included in the study. After an average post-intervention follow-up period of 10.3 months, both exercise (SMD, −0.29; 95% CI, −0.57 to −0.01) and CBT-I (−0.48; −0.68 to −0.28) showed superior long-term effectiveness on improving sleep compared with control. Temazepam was the only included pharmacotherapy, which demonstrated superior treatment effectiveness (−0.80; −1.25 to −0.36) but not long-term effectiveness (0.19; −0.32 to 0.69) compared with control. The findings support the use of both exercise and CBT-I for long-term management of chronic insomnia, while temazepam may be used for short-term treatment.

Comparison between Sevoflurane/Remifentanil and Propofol/Remifentanil Anaesthesia in Providing Conditions for Somatosensory Evoked Potential Monitoring during Scoliosis Corrective Surgery

Somatosensory evoked potential (SSEP) monitoring is an important tool in spinal corrective surgery. Anaesthesia has a significant influence on SSEP monitoring and a technique which has the least and shortest suppressant effect on SSEP while facilitating a fast recovery from anaesthesia is ideal. We compared the effect of sevoflurane/remifentanil and propofol/remifentanil anaesthesia on SSEPs during scoliosis corrective surgery and assessed patients’ clinical recovery profiles.
Twenty patients with idiopathic scoliosis receiving surgical correction with intraoperative SSEP monitoring were prospectively randomised to receive sevoflurane/remifentanil anaesthesia or propofol/remifentanil anaesthesia. During surgery, changes in anaesthesia dose and physiological variables were recorded, while SSEP was continuously monitored. A simulated ‘wake-up’ test was performed postoperatively to assess speed and quality of recovery from anaesthesia.
The effects of propofol and sevoflurane resulted in SSEP amplitude variability between 18.0% ± 3.5% to 28.7% ± 5.9% and SSEP latency variability within 1.3% ± 0.4% to 2.6% ± 1.2%. Patients receiving sevoflurane had faster suppression and faster recovery of SSEP amplitude compared to propofol (P <0.05), although propofol anaesthesia showed less within-patient variability in Cz amplitude and latency (P <0.05). On cessation of anaesthesia, time to eye-opening (5.2 vs. 16.5 minutes) and toe movement (5.4 vs. 17.4 minutes) was shorter following sevoflurane (all P <0.05).
These findings indicate that propofol produces a better SSEP signal than sevoflurane. However, adjustments in sevoflurane concentration result in faster changes in the SSEP signal than propofol. Assessment of neurological function was facilitated more rapidly after sevoflurane anaesthesia.

Mayor Michael R. Bloomberg, who has refused in his two years in office to disclose his personal views on gay and lesbian marriage, told 80 journalists at a lesbian and gay fund-raising dinner in Manhattan Thursday night that he favored changing state law to legalize same-sex unions, four people who were there said yesterday. On a day when the struggle for gay and lesbian marriage rights in New York moved into the courts with the first of many anticipated lawsuits and into the streets with auto caravans and protests on Long Island, the mayor of an upstate community said he would temporarily suspend his performing of same-sex marriages and Gov. George E. Pataki vowed again to uphold the existing marriage law. After receiving a flurry of calls yesterday, Edward Skyler, the mayor's press secretary, summoned the City Hall press corps to his office to view a taped interview with the mayor to be aired Sunday on Channel 11. Asked to illuminate his views on gay marriage, Mr. Bloomberg said he was opposed to discrimination and said he had always thought civil unions should have the same rights as marriages.
