Altered protein secretion in Batten disease
The neuronal ceroid lipofuscinoses (NCLs), collectively known as Batten disease, are a group of neurological diseases that affect all ages and ethnicities worldwide. There are 13 different subtypes of NCL, each caused by a mutation in a distinct gene. The NCLs are characterized by the accumulation of undigestible lipids and proteins in various cell types. This leads to progressive neurodegeneration and clinical symptoms including vision loss, progressive motor and cognitive decline, seizures, and premature death. These diseases have commonly been characterized by lysosomal defects leading to the accumulation of undigestible material but further research on the NCLs suggests that altered protein secretion may also play an important role. This has been strengthened by recent work in biomedical model organisms, including Dictyostelium discoideum, mice, and sheep. Research in D. discoideum has reported the extracellular localization of some NCL-related proteins and the effects of NCL-related gene loss on protein secretion during unicellular growth and multicellular development. Aberrant protein secretion has also been observed in mammalian models of NCL, which has allowed examination of patient-derived cerebrospinal fluid and urine for potential diagnostic and prognostic biomarkers. Accumulated evidence links seven of the 13 known NCL-related genes to protein secretion, suggesting that altered secretion is a common hallmark of multiple NCL subtypes. This Review highlights the impact of altered protein secretion in the NCLs, identifies potential biomarkers of interest and suggests that future work in this area can provide new therapeutic insight.

Molecular networking in the neuronal ceroid lipofuscinoses: insights from mammalian models and the social amoeba Dictyostelium discoideum
The neuronal ceroid lipofuscinoses (NCLs), commonly known as Batten disease, belong to a family of neurological disorders that cause blindness, seizures, loss of motor function and cognitive ability, and premature death. There are 13 different subtypes of NCL that are associated with mutations in 13 genetically distinct genes ( CLN1-CLN8 , CLN10-CLN14 ). Similar clinical and pathological profiles of the different NCL subtypes suggest that common disease mechanisms may be involved. As a result, there have been many efforts to determine how NCL proteins are connected at the cellular level. A main driving force for NCL research has been the utilization of mammalian and non-mammalian cellular models to study the mechanisms underlying the disease. One non-mammalian model that has provided significant insight into NCL protein function is the social amoeba Dictyostelium discoideum . Accumulated data from Dictyostelium and mammalian cells show that NCL proteins display similar localizations, have common binding partners, and regulate the expression and activities of one another. In addition, genetic models of NCL display similar phenotypes. This review integrates findings from Dictyostelium and mammalian models of NCL to highlight our understanding of the molecular networking of NCL proteins. The goal here is to help set the stage for future work to reveal the cellular mechanisms underlying the NCLs.
Assessing the relative importance of psychological and demographic factors for predicting climate and environmental attitudes
In this paper, we seek to identify robust predictors of individuals’ attitudes towards climate change and environmental degradation. While much of the extant literature has been devoted to the individual explanatory potential of individuals’ characteristics, we focus on the extent to which these characteristics provide robust predictions of climate and environmental attitudes. Thereby, we adjudicate the relative predictive power of psychological and sociodemographic characteristics, as well as the predictive power of combinations of these attributes. To do so, we use a popular machine learning technique, Random Forests, on three surveys fielded in China, Switzerland, and the USA, using a variety of outcome variables. We find that a psychological construct, the consideration of future consequences (CFC) scale, performs well in predicting attitudes, across all contexts and better than traditional explanations of climate attitudes, such as income and education. Given recent advances suggesting potential psychological barriers of behavioural change Public (Weaver, Adm Rev 75:806–816, 2015) and the use of psychological constructs to target persuasive messages (Abrahamse et al., J Environ Psychol 265–276, 2007; Hirsh et al., Psychol Sci 23:578–581, 2012), identifying important predictors, such as the CFC may allow to better understand public’s appetite for climate and environmental policies and increase demand for these policies, in an area where existing efforts have shown to be lacking (Bernauer and McGrath, Nat Clim Chang 6:680–683, 2016; Chapman et al., Nat Clim Chang 7:850–852, 2017).
Economic value of three grassland ecosystem services when managed at the regional and farm scale
Grasslands cover a major share of the world’s agricultural land and their management influences ecosystem services. Spatially targeted policy instruments can increase the provision of ecosystem services by exploiting how they respond to spatial differences in environmental characteristics such as altitude, slope, or soil quality. However, most policy instruments focus on individual farms, where spatial differences are small. Here we assess the economic value of three grassland ecosystem services (i.e., forage provision, carbon sequestration, and habitat maintenance) and its variability in a Swiss region of 791 km 2 that consists of 19,000 farmland parcels when managed at the regional and farm scale, respectively. Our spatially explicit bio-economic simulation approach combines biophysical information on grassland ecosystem services and their economic values. We find that in our case study region, spatial targeting on a regional scale management increases the economic value of ecosystem services by 45% compared to targeting at farm scale. We also find that the heterogeneity of economic values coming from prices and willingness to pay estimates is higher than the economic gains from spatial targeting that make use of the spatial difference in environmental characteristics. This implies that heterogeneity in prices and/or societal demand of these three ecosystem services is more important for grassland management than spatial heterogeneity in our case study region. The here applied framework allows for an ex-ante assessment of economic gains from spatial targeting and thus provides basic information for the implementation of incentive mechanisms addressing the nexus of food production and ecosystem service provision in grasslands.
Calmodulin binding proteins and neuroinflammation in multiple neurodegenerative diseases
Calcium dysregulation (“Calcium Hypothesis”) is an early and critical event in Alzheimer’s and other neurodegenerative diseases. Calcium binds to and regulates the small regulatory protein calmodulin that in turn binds to and regulates several hundred calmodulin binding proteins. Initial and continued research has shown that many calmodulin binding proteins mediate multiple events during the onset and progression of Alzheimer’s disease, thus establishing the “Calmodulin Hypothesis”. To gain insight into the general applicability of this hypothesis, the involvement of calmodulin in neuroinflammation in Alzheimer’s, amyotrophic lateral sclerosis, Huntington’s disease, Parkinson’s disease, frontotemporal dementia, and other dementias was explored. After a literature search for calmodulin binding, 11 different neuroinflammatory proteins (TREM2, CD33, PILRA, CR1, MS4A, CLU, ABCA7, EPHA1, ABCA1, CH3L1/YKL-40 and NLRP3) were scanned for calmodulin binding domains using the Calmodulin Target Database. This analysis revealed the presence of at least one binding domain within which visual scanning demonstrated the presence of valid binding motifs. Coupled with previous research that identified 13 other neuroinflammation linked proteins (BACE1, BIN1, CaMKII, PP2B, PMCA, NOS, NMDAR, AchR, Ado A2AR, Aβ, APOE, SNCA, TMEM175), this work shows that at least 24 critical proteins involved in neuroinflammation are putative or proven calmodulin binding proteins. Many of these proteins are linked to multiple neurodegenerative diseases indicating that calmodulin binding proteins lie at the heart of neuroinflammatory events associated with multiple neurodegenerative diseases. Since many calmodulin-based pharmaceuticals have been successfully used to treat Huntington’s and other neurodegenerative diseases, these findings argue for their immediate therapeutic implementation.
On the Distinct Effects of Left-Wing and Right-Wing Populism on Democratic Quality
This study examines the differences and commonalities of how populist parties of the left and right relate to democracy. The focus is narrowed to the relationship between these parties and two aspects of democratic quality, minority rights and mutual constraints. Our argument is twofold: first, we contend that populist parties can exert distinct influences on minority rights, depending on whether they are left-wing or right-wing populist parties. Second, by contrast, we propose that the association between populist parties and mutual constraints is a consequence of the populist element and thus, we expect no differences between the left-wing and right-wing parties. We test our expectations against data from 30 European countries between 1990 and 2012. Our empirical findings support the argument for the proposed differences regarding minority rights and, to a lesser extent, the proposed similarities regarding mutual constraints. Therefore we conclude that, when examining the relationship between populism and democracy, populism should not be considered in isolation from its host ideology.
