Antigen-specific cytokine profiles for pulmonary Mycobacterium avium complex disease stage diagnosis
Controlling pulmonary complex (MAC) disease is difficult because there is no way to know the clinical stage accurately. There have been few attempts to use cell-mediated immunity for diagnosing the stage. The objective of this study was to characterize cytokine profiles of CD4+T and CD19+B cells that recognize various -associated antigens in different clinical stages of MAC. A total of 47 MAC patients at different stages based on clinical information (14 before-treatment, 16 on-treatment, and 17 after-treatment) and 17 healthy controls were recruited. Peripheral blood mononuclear cells were cultured with specific antigens (MAV0968, 1160, 1276, and 4925), and the cytokine profiles (IFN-γ, TNF-α, IL-2, IL-10, IL-13, and IL-17) of CD4+/CD3+ and CD19+ cells were analyzed by flow cytometry. The response of Th1 cytokines such as IFN-γ and TNF-α against various antigens was significantly higher in both the on-treatment and after-treatment groups than in the before-treatment group and control (P < 0.01-0.0001 and P < 0.05-0.0001). An analysis of polyfunctional T cells suggested that the presence of IL-2 is closely related to the stage after the start of treatment (P = 0.0309-P < 0.0001) and is involved in memory function. Non-Th1 cytokines, such as IL-10 and IL-17, showed significantly higher responses in the before-treatment group (P < 0.0001 and P < 0.01-0.0001). These responses were not observed with purified protein derivative (PPD). CD19+B cells showed a response similar to that of CD4+T cells. There is a characteristic cytokine profile at each clinical stage of MAC.
Association between bone mineral density, muscle volume, walking ability, and geriatric nutritional risk index in hemodialysis patients
Background and Objectives: Hemodialysis patients are at risk for bone loss and sarcopenia, characterized by reduced muscle mass and limited mobility/function. Osteoporosis and sarcopenia both increase the risk of hospitalization and death in affected individuals. Malnutrition also occurs as a complication of hemodialysis and has been identified as a risk factor for osteoporosis and sarcopenia. In this study, we examined the relationship between osteoporosis, muscle volume, walking ability, and malnutrition in hemodialysis patients. Methods and Study Design: Forty-five hemodialysis patients were evaluated. Bone mineral density (BMD) and muscle volume were measured by dual-energy X-ray absorptiometry. Muscle volume and strength were evaluated using lean mass index (LMI), handgrip strength, and walking ability. The time required for a patient to walk 10 meters was measured to evaluate walking ability. The geriatric nutritional risk index (GNRI) was used to assess malnutrition. Results: Multiple linear regression analysis showed that older age, female sex, lower LMI, and higher total type I procollagen N-terminal propeptide were correlated with lower BMD of lumbar spine. Higher age and lower LMI were correlated with lower BMD of the femoral neck. Female sex and lower GNRI were correlated with lower LMI. Longer duration of hemodialysis was correlated with lower walking ability. Conclusions: Our findings suggest that muscle preservation is required to maintain both lumbar spine and femoral neck BMD. Similarly, nutritional management is necessary to maintain BMD via preservation of muscle volume. Complementary nutritional therapies are needed to improve osteoporosis and sarcopenia in high-risk hemodialysis patients.
Symmetries of hyperbolic spatial graphs and realization of graph symmetries
In a closed connected orientable 3-manifold associated with an orientation-preserving smooth finite group action, we construct setwise invariant hyperbolic spatial graphs with given singularity. As an application, we provide a condition under which symmetries of abstract graphs are realizable by symmetries of the 3-sphere through hyperbolic spatial embeddings.
Finite group actions on homologically peripheral 3-manifolds
We generalize the notion of totally peripheral 3-manifolds to define homologically peripheral 3-manifolds. The homologically peripheral property survives canonical decompositions of 3-manifolds as well as it defines a sufficiently large class of 3-manifolds containing link exteriors. The aim of this paper is to study finite group actions on a homologically peripheral 3-manifold, which agree on the boundary, up to equivalence relative to the boundary. As an application, we generalize Sakuma's theorems on the uniqueness of symmetries of knots to the case of symmetries of links.
INVOLUTIONS OF GRAPH LINK EXTERIORS WHOSE FIXED POINT SETS ARE CLOSED SURFACES
A link L in S3 possibly admits an involution of the exterior E(L) with fixed point set a closed surface, which is not extendable to an involution of S3. In this paper, we focus on the case of graph links and show that the genus of the surface provides a lower estimate of the number of link components.
Stress intensity factors for interface cracks between orthotropic and monoclinic material
Different expressions are used in the literature for the stress intensity factors of interface cracks between anisotropic material. In particular, two of these approaches will be discussed and compared for orthotropic and monoclinic materials. Relations between the stress intensity factors will be found. Expressions for the interface energy release rate are presented. Although the expressions appear different, they are shown to be the same by using the relations between the stress intensity factors. Phase angles are defined which may be used in a fracture criterion.
Stress intensity factor analysis of an interface crack between dissimilar anisotropic materials under thermal stress using the finite element analysis
New numerical methods were presented for stress intensity factor analyses of two-dimensional interfacial crack between dissimilar anisotropic materials subjected to thermal stress. The virtual crack extension method and the thermal M-integral method for a crack along the interface between two different materials were applied to the thermoelastic interfacial crack in anisotropic bimaterials. The moving least-squares approximation was used to calculate the value of the thermal M-integral. The thermal M-integral in conjunction with the moving least-squares approximation can calculate the stress intensity factors from only nodal displacements obtained by the finite element analysis. The stress intensity factors analyses of double edge cracks in jointed dissimilar isotropic semi-infinite plates subjected to thermal load were demonstrated. Excellent agreement was achieved between the numerical results obtained by the present methods and the exact solution. In addition, the stress intensity factors of double edge cracks in jointed dissimilar anisotropic semi-infinite plates subjected to thermal loads were analyzed. Their results appear reasonable.
Symmetries of Spatial Graphs and Rational Twists along Spheres and Tori
A symmetry group of a spatial graph Γ in S3 is a finite group consisting of orientation-preserving self-diffeomorphisms of S3 which leave Γ setwise invariant. In this paper, we show that in many cases symmetry groups of Γ which agree on a regular neighborhood of Γ are equivalent up to conjugate by rational twists along incompressible spheres and tori in the exterior of Γ.
Stress intensity factor analyses of three-dimensional interface cracks using tetrahedral finite elements
A numerical method is proposed for evaluating the stress intensity factors of a three-dimensional bimaterial interfacial crack using tetrahedral finite elements. This technique is based on the M 1 -integral method and employs the moving least-squares approximation. Stress or strain in the M 1 -integral equation is automatically approximated from the nodal displacements obtained by the finite element analysis using the moving least-squares method. Therefore, the presented method needs no elemental information from the finite element analysis. In this study, stress intensity factor analyses of some typical three-dimensional interface crack problems using the tetrahedral finite elements are demonstrated.
