Today's world poses a triple threat to the American population: infectious diseases, contamination of food and water, and bioattacks (biowarfare or bioterrorism). At least 17 countries are producing weapons of mass destruction using viruses, bacteria, or their toxins. AIDS, E. coli contamination, drug-resistant tuberculosis, and virulent flu strains are perhaps the best known of a host of disease threats. What these dangers have in common is the amount of data required to achieve solutions; in some cases, as much as a petabit (1 followed by 15 zeros) of data is required to study large numbers of samples from widespread locations.
Firepower in the Lab examines how the nation can combat this triple threat by improving our ability to detect, measure, and monitor harmful biological agents. Taming Wildcat Stablecoins

Cryptocurrencies, including stablecoins, are all the rage. Investors are exploring ways to profit off of them. Governments are considering ways to regulate them. While the technology underlying cryptocurrencies is new, the economics is centuries old. Oftentimes, lawmakers are so focused on understanding a new technological innovation that they fail to ask what exactly is being created.
In this case, the new technology has recreated circulating private money in the form of stablecoins, which are similar to the banknotes that circulated in many countries during the nineteenth century. The implication is that stablecoin issuers are unregulated banks. Based on lessons learned from economic theory and financial history, we argue that circulating private money is not an effective medium of exchange because it is not always accepted at par and its issuers are vulnerable to destabilizing bank runs.
We also explore the treatment of stablecoins under the existing legal framework and examine the upsides and downsides of interpretive, regulatory, and legislative options that attempt to mitigate the financial-stability risks associated with stablecoins. These options include requiring the issuance of stablecoins through banks, backing stablecoins one-for-one with safe assets, and establishing sovereign digital money to compete against private digital money.

Ancestral origin of ApoE ε4 Alzheimer disease risk in Puerto Rican and African American populations

The ApoE ε4 allele is the most significant genetic risk factor for late-onset Alzheimer disease. The risk conferred by ε4, however, differs across populations, with populations of African ancestry showing lower ε4 risk compared to those of European or Asian ancestry. The cause of this heterogeneity in risk effect is currently unknown; it may be due to environmental or cultural factors correlated with ancestry, or it may be due to genetic variation local to the ApoE region that differs among populations. Exploring these hypotheses may lead to novel, population-specific therapeutics and risk predictions. To test these hypotheses, we analyzed ApoE genotypes and genome-wide array data in individuals from African American and Puerto Rican populations. A total of 1,766 African American and 220 Puerto Rican individuals with late-onset Alzheimer disease, and 3,730 African American and 169 Puerto Rican cognitively healthy individuals (> 65 years) participated in the study. We first assessed average ancestry across the genome (\"global\" ancestry) and then tested it for interaction with ApoE genotypes. Next, we assessed the ancestral background of ApoE alleles (\"local\" ancestry) and tested if ancestry local to ApoE influenced Alzheimer disease risk while controlling for global ancestry. Measures of global ancestry showed no interaction with ApoE risk (Puerto Rican: p-value = 0.49; African American: p-value = 0.65). Conversely, ancestry local to the ApoE region showed an interaction with the ApoE ε4 allele in both populations (Puerto Rican: p-value = 0.019; African American: p-value = 0.005). ApoE ε4 alleles on an African background conferred a lower risk than those with a European ancestral background, regardless of population (Puerto Rican: OR = 1.26 on African background, OR = 4.49 on European; African American: OR = 2.34 on African background, OR = 3.05 on European background). Factors contributing to the lower risk effect in the ApoE gene ε4 allele are likely due to ancestry-specific genetic factors near ApoE rather than non-genetic ethnic, cultural, and environmental factors.

Among the different types of natural disasters, floods are the most devastating, widespread, and frequent. Floods account for approximately 30% of the total loss caused by natural disasters. Accurate flood-risk mapping is critical in reducing such damages by correctly predicting the extent of a flood when coupled with rain and stage gage data, supporting emergency-response planning, developing land use plans and regulations with regard to the construction of structures and infrastructures, and providing damage assessment in both spatial and temporal measurements. The reliability and accuracy of such flood assessment maps is dependent on the quality of the digital elevation model (DEM) in flood conditions. This study investigates the quality of an Unmanned Aerial Vehicle (UAV)-based DEM for spatial flood assessment mapping and evaluating the extent of a flood event in Princeville, North Carolina during Hurricane Matthew. The challenges and problems of on-demand DEM production during a flooding event were discussed. An accuracy analysis was performed by comparing the water surface extracted from the UAV-derived DEM with the water surface/stage obtained using the nearby US Geologic Survey (USGS) stream gauge station and LiDAR data.

Viral genome sequencing has guided our understanding of the spread and extent of genetic diversity of SARS-CoV-2 during the COVID-19 pandemic. SARS-CoV-2 viral genomes are usually sequenced from nasopharyngeal swabs of individual patients to track viral spread. Viral genome sequencing has guided our understanding of the spread and extent of genetic diversity of SARS-CoV-2 during the COVID-19 pandemic. SARS-CoV-2 viral genomes are usually sequenced from nasopharyngeal swabs of individual patients to track viral spread. Recently, RT-qPCR of municipal wastewater has been used to quantify the abundance of SARS-CoV-2 in several regions globally. However, metatranscriptomic sequencing of wastewater can be used to profile the viral genetic diversity across infected communities. Here, we sequenced RNA directly from sewage collected by municipal utility districts in the San Francisco Bay Area to generate complete and nearly complete SARS-CoV-2 genomes. The major consensus SARS-CoV-2 genotypes detected in the sewage were identical to clinical genomes from the region. Using a pipeline for single nucleotide variant calling in a metagenomic context, we characterized minor SARS-CoV-2 alleles in the wastewater and detected viral genotypes which were also found within clinical genomes throughout California. Observed wastewater variants were more similar to local California patient-derived genotypes than they were to those from other regions within the United States or globally. Additional variants detected in wastewater have only been identified in genomes from patients sampled outside California, indicating that wastewater sequencing can provide evidence for recent introductions of viral lineages before they are detected by local clinical sequencing. These results demonstrate that epidemiological surveillance through wastewater sequencing can aid in tracking exact viral strains in an epidemic context.
