Structure–Activity Relationship of PAD4 Inhibitors and Their Role in Tumor Immunotherapy

Protein arginine deiminase 4 (PAD4) plays an important role in cancer progression by participating in gene regulation, protein modification, and neutrophil extracellular trap (NET) formation. Many reversible and irreversible PAD4 inhibitors have been reported recently. In this review, we summarize the structure–activity relationships of newly investigated PAD4 inhibitors to bring researchers up to speed by guiding and describing new scaffolds as optimization and development leads for new effective, safe, and selective cancer treatments. In addition, some recent reports have shown evidence that PAD4 inhibitors are expected to trigger antitumor immune responses, regulate immune cells and related immune factors, enhance the effects of immune checkpoint inhibitors, and enhance their antitumor efficacy. Therefore, PAD4 inhibitors may potentially change tumor immunotherapy and provide an excellent direction for the development and clinical application of immunotherapy strategies for related diseases.

Cerebral ischemia–reperfusion injury significantly contributes to global morbidity and mortality. Loganin is a natural product with various neuroprotective effects; however, it lacks targeted specificity for particular cells or receptors, which may result in reduced therapeutic efficacy and an increased risk of side effects. To address the limitations of loganin, we developed LA-1, a novel compound incorporating an Arg-Gly-Asp (RGD) peptide to target integrin receptor αvβ3, enhancing brain-targeting efficacy. LA-1 exhibited optimal nanoscale properties, significantly improved cell viability, reduced ROS production, and enhanced survival rates in vitro. In vivo, LA-1 decreased infarct sizes, improved neurological function, and reduced oxidative stress and neuroinflammation. Proteomic analysis showed LA-1 modulates PI3K/Akt and Nrf2/HO-1 pathways, providing targeted neuroprotection. These findings suggest LA-1’s potential for clinical applications in treating cerebral ischemia–reperfusion injury.

Importance of glutamine in synaptic vesicles revealed by functional studies of SLC6A17 and its mutations pathogenic for intellectual disability

Human mutations in the gene encoding the solute carrier (SLC) 6A17 caused intellectual disability (ID). The physiological role of SLC6A17 and pathogenesis of SLC6A17 -based-ID were both unclear. Here, we report learning deficits in Slc6a17 knockout and point mutant mice. Biochemistry, proteomic, and electron microscopy (EM) support SLC6A17 protein localization in synaptic vesicles (SVs). Chemical analysis of SVs by liquid chromatography coupled to mass spectrometry (LC-MS) revealed glutamine (Gln) in SVs containing SLC6A17. Virally mediated overexpression of SLC6A17 increased Gln in SVs. Either genetic or virally mediated targeting of Slc6a17 reduced Gln in SVs. One ID mutation caused SLC6A17 mislocalization while the other caused defective Gln transport. Multidisciplinary approaches with seven types of genetically modified mice have shown Gln as an endogenous substrate of SLC6A17, uncovered Gln as a new molecule in SVs, established the necessary and sufficient roles of SLC6A17 in Gln transport into SVs, and suggested SV Gln decrease as the key pathogenetic mechanism in human ID.

A radiohybrid theranostics ligand labeled with fluorine-18 and lutetium-177 for fibroblast activation protein-targeted imaging and radionuclide therapy

PurposeA series of radiotracers targeting fibroblast activation protein (FAP) with great pharmacokinetics have been developed for cancer diagnosis and therapy. Nevertheless, the use of dominant PET tracers, gallium-68–labeled FAPI derivatives, was limited by the short nuclide half-life and production scale, and the therapeutic tracers exhibited rapid clearance and insufficient tumor retention. In this study, we developed a FAP targeting ligand, LuFL, containing organosilicon-based fluoride acceptor (SiFA) and DOTAGA chelator, capable of labeling fluorine-18 and lutetium-177 in one molecular with simple and highly efficient labeling procedure, to achieve cancer theranostics.MethodsThe precursor LuFL (20) and [natLu]Lu-LuFL (21) were successfully synthesized and labeled with fluorine-18 and lutetium-177 using a simple procedure. A series of cellular assays were performed to characterize the binding affinity and FAP specificity. PET imaging, SPECT imaging, and biodistribution studies were conducted to evaluate pharmacokinetics in HT-1080-FAP tumor-bearing nude mice. A comparison study of [177Lu]Lu-LuFL ([177Lu]21) and [177Lu]Lu-FAPI-04 was carried out in HT-1080-FAP xenografts to determine the cancer therapeutic efficacy.ResultsLuFL (20) and [natLu]Lu-LuFL (21) demonstrated excellent binding affinity towards FAP (IC50: 2.29 ± 1.12 nM and 2.53 ± 1.87 nM), compared to that of FAPI-04 (IC50: 6.69 ± 0.88 nM). In vitro cellular studies showed that 18F-/177Lu-labeled 21 displayed high specific uptake and internalization in HT-1080-FAP cells. Micro-PET, SPECT imaging and biodistribution studies with [18F]/[177Lu]21 revealed higher tumor uptake and longer tumor retention than those of [68 Ga]/[177Lu]Ga/Lu-FAPI-04. The radionuclide therapy studies showed significantly greater inhibition of tumor growth for the [177Lu]21 group, than for the control group and the [177Lu]Lu-FAPI-04 group.ConclusionThe novel FAPI-based radiotracer containing SiFA and DOTAGA was developed as a theranostics radiopharmaceutical with simple and short labeling process, and showed promising properties including higher cellular uptake, better FAP binding affinity, higher tumor uptake and prolong retention compared to FAPI-04. Preliminary experiments with 18F- and 177Lu-labeled 21 showed promising tumor imaging properties and favorable anti-tumor efficacy.

Shikonin as a therapeutic agent in renal cell carcinoma: insights from TEK-related causal association with glaucoma

Renal cell carcinoma (RCC) is a lethal malignancy with rising incidence, while glaucoma, a chronic eye disease, shares systemic mechanisms such as oxidative stress and inflammation with cancers. This study aimed to investigate the causal link between glaucoma and RCC and explore molecular intersections to identify novel therapeutic targets.
A two-step Mendelian randomization (MR) analysis using genetic data from the NHGRI-EBI GWAS Catalog and FinnGen database was performed, supplemented by NHANES data. Gene expression analysis (GSE53757, E-MTAB-1980) identified glaucoma-related genes in RCC. Molecular docking and functional assays evaluated shikonin's effects on TEK and AKT/mTOR signaling.
MR revealed a significant causal relationship between glaucoma and RCC. TEK, a glaucoma-related gene, was downregulated in RCC tissues and correlated with advanced tumor stage and metastasis. Shikonin and acetylshikonin upregulated TEK expression, inhibited RCC cell proliferation/migration, and suppressed AKT/mTOR phosphorylation.
These findings support a role for glaucoma-associated genes in RCC development and progression, highlighting shikonin as a promising therapeutic agent targeting this molecular axis.

CircIQGAP1-CARM1 axis promotes renal cell carcinoma progression through glycolytic reprogramming

Renal cell carcinoma (RCC) relies heavily on aerobic glycolysis for rapid proliferation and metastasis; however, the role of circular RNAs (circRNAs) in this process remains unclear. This study identified circIQGAP1 as a key regulator; its expression was upregulated under glucose deficiency, with U2AF2 promoting its biosynthesis. Functional assays showed that circIQGAP1 enhances RCC cell proliferation, motility, invasion, and glycolytic flux. Mechanistically, circIQGAP1 binds to CARM1, inhibiting its K48-linked ubiquitination and prolonging its half-life. Elevated CARM1 then catalyses COL5A1 promoter dimethylation, driving COL5A1 transcription. Rescue experiments confirmed that both CARM1 and COL5A1 were essential for circIQGAP1-mediated metabolic reprogramming and malignant phenotypes. Clinically, circIQGAP1 is overexpressed in RCC tissues and is correlated with poor outcomes. These findings revealed that circIQGAP1 promotes glycolysis-dependent RCC progression by stabilizing CARM1 to activate COL5A1, highlighting that this regulatory axis may provide an innovative strategy for RCC treatment.

Crosslinked polyarylene ether nitrile film as flexible dielectric materials with ultrahigh thermal stability

Dielectric film with ultrahigh thermal stability based on crosslinked polyarylene ether nitrile is prepared and characterized. The film is obtained by solution-casting of polyarylene ether nitrile terminated phthalonitrile (PEN-Ph) combined with post self-crosslinking at high temperature. The film shows a 5% decomposition temperature over 520 °C and a glass transition temperature (
T
g
) around 386 °C. Stable dielectric constant and low dielectric loss are observed for this film in the frequency range of 100–200 kHz and in the temperature range of 25–300 °C. The temperature coefficient of dielectric constant is less than 0.001 °C
−1
even at 400 °C. By cycling heating and cooling up to ten times or heating at 300 °C for 12 h, the film shows good reversibility and robustness of the dielectric properties. This crosslinked PEN film will be a potential candidate as high performance film capacitor electronic devices materials used at high temperature.

High-dose vitamin C (VC) exhibits anti-tumor effects, and the cytotoxicity of VC is correlated with oxidative stress. However, iron, as a redox metal, plays an important effect in redox cycling and free radical formation in cells. This study addresses the role of iron ion in the cytotoxicity of VC. We found that iron supplementation increases the anti-tumor effect of VC, which was influenced by the cellular iron uptake pathway–transferrin (TF)/transferrin receptor (TFR) system. The TFR expression of tumors can be assessed by 68Ga-citrate PET imaging, and it would be helpful to screen out the tumor type which is more sensitive to VC combined with an iron supplementation treatment.

Research on Volume Measurement Technology for Rail Tanker Based on Computer Vision

In this paper, a computer vision based volume measurement system for rail tanker is proposed. Considering the complex environment, we propose an accurate identification algorithm of coded point and a precise localization algorithm. By investigating the epipolar geometry among three views, this paper develops a novel method for metric reconstruction of a scene based on the trilinear relations. Through the method of ordering incomplete scattered data presented, we construct a series of cross-section contours for a tanker and achieve a precise reconstruction of the surface of the tanker. Experiments show that, the volume measurement method for rail tanker based on computer vision can be operated with simplicity and high accuracy.
