Phosphaturic mesenchymal tumors (PMTs) are extremely rare mesenchymal tumors of soft tissue and bone that cause tumor-induced osteomalacia (TIO). Some of these tumors are completely asymptomatic and may grow undetected unless they become large enough to cause pain or discomfort. This type of tumor is crucial to diagnose in patients being treated for phosphate metabolism disorders and are a rare reason why patients seek medical help owing to pain. Here, we report the details of a patient with progressive bone pain caused by a PMT originating in the left femur.

Case Report and Literature Review on Skin Toxicity Induced by PD-1 Inhibitor in a Penile Cancer with Massive Ulceration of Chemoradiotherapy-Resistant and Successful Treatment by Immunotherapy

Penile cancer is a rare malignant tumor with a poor prognosis in advanced stages. Immune checkpoint inhibitors (ICIs) have demonstrated promising efficacy in patients with advanced penile cancer, but it can also induce immune-related adverse events (irAEs). This article reports a patient who achieved almost a complete response to the PD-1 inhibitor sintilimab as third-line treatment for advanced penile squamous cell cancer with massive ulceration of chemoradiotherapy-resistant, and successful treatment by immunotherapy. One year into maintenance therapy with sintilimab, skin toxicity in the form or grade-2 skin rashes and grade-3 pruritus occurred. Sintilimab was permanently discontinued. The skin toxicity was effectively controlled by oral prednisone at a daily dosage of 15 mg. At the last follow-up of 16 months after sintilimab discontinuation, the patient remained in partial response, with total progression-free survival exceeding 30 months. We also conducted a comprehensive literature search, and summarized skin toxicity of ICIs administration. These articles suggested that immune-related skin toxicity may be indicative of good treatment response.

Pathological features of gastric‑type endocervical adenocarcinoma: A report of two cases

Gastric-type endocervical adenocarcinoma (GEA) is an uncommon form of uterine cervical adenocarcinoma with an unfavorable prognosis. The tumor consists of glands exhibiting a morphological resemblance to gastric cells and occasionally manifests features akin to pancreaticobiliary mucinous adenocarcinoma. GEA differs from the typical cervical cancer, particularly in its lack of association with the human papillomavirus. Immunophenotypic analysis suggests intestinal differentiation. The present study reports two cases of GEA occurring in postmenopausal individuals who were diagnosed in Lishui Central Hospital (Lishui, China) between January 2015 and January 2023. Microscopic examination revealed cysts lined with mucinous cells within the tumors. Immunohistochemical assays confirmed the positivity of the tumors for cytokeratin 7, mucin (MUC)5AC, and mutant tumor protein p53, while the results were negative for tumor suppressor p16, and in one case for paired box protein 8, consistent with characteristics of mucinous adenocarcinoma originating from the gastrointestinal tract. Programmed death-ligand 1 expression was also negative. The proto-oncogene K-ras was identified using amplification refractory mutation system polymerase chain reaction. Both cases were negative for mutations in codons 12 and 13 of exon 2, codon 61 of exon 3 and codon 146 of exon 4, but were positive for wild-type K-ras. Clinical follow-up revealed a potential association between histopathological features and resistance to chemotherapeutic drugs. The infrequency of this tumor type may contribute to diagnostic challenges.

Investigation on the Edge Doping Process of Nitrogen-Doped Carbon Materials by In Situ Pyrolysis Mass Spectrometry and Laser-Induced Acoustic Desorption Mass Spectrometry

Nitrogen-doped carbon materials demonstrate high performance as electrodes in fuel cells and higher oxygen reduction reactivity than traditional Pt-based electrodes. However, the formation process of nitrogen-doped carbon materials has long been a mystery. In this study, the formation mechanism of nitrogen-doped carbon materials from polyaniline (PANI) pyrolysis was studied by the combination of in situ pyrolysis vacuum ultraviolet photoionization time-of-flight mass spectrometry (Py-VUVPI-TOF MS) and substrate-enhanced, laser-induced acoustic desorption source time-of-flight mass spectrometry (SE-LIAD-TOF MS). The initial pyrolysis species, including free radicals and intermediates, were investigated via in situ Py-VUVPI-TOF MS during the temperature-programmed desorption process (within tens of microseconds). The pyrolysis residues were collected and further investigated via SE-LIAD-TOF MS, revealing the product information of the initial pyrolysis products. The results show that the edge doping of carbon materials depends on free radical reactions rather than the direct substitution of carbon atoms by nitrogen atoms. Meanwhile, pyrrole nitrogen and pyridine nitrogen are formed by the free radical cyclization reaction and the amino aromatization reaction at the initial stage of pyrolysis, while the formation of graphitic nitrogen depends on the further polymerization reaction of pyrrole nitrogen and pyridine nitrogen.

人长链非编码RNA基因SPRY4-IT1对肺癌细胞A549侵袭和迁移能力的影响

背景与目的人长链非编码RNA基因的异常表达与多种肿瘤有关。本研究旨在探讨人长链非编码RNA基因SPRY4-IT1对肺癌细胞A549侵袭和迁移能力的影响及机制。方法将SPRY4-IT1转染入肺癌A549细胞系,采用RT-PCR方法检测重组质粒在细胞中表达水平比较;采用MTT和Transwell检测A549细胞侵袭和迁移能力的变化,采用Western blot方法检测SPRY4-IT1对MMP-2和MMP-9蛋白的影响。结果转染pc DNA3.1-SPRY4-IT1细胞划痕两侧细胞间距较转染pc DNA3.1细胞明显变窄,其穿膜细胞数为(207±34)个/视野,相比对照组明显增多(P<0.05),且转染pc DNA3.1-SPRY4-IT1细胞中的基质金属蛋白酶(matrix metalloproteinase,MMP)-2及MMP-9蛋白表达较转染空质粒组均有升高。结论在肺癌A549细胞中过表达SPRY4-IT1可增强细胞的侵袭、迁移能力,且细胞内MMP-2及MMP-9表达升高,提示SPRY4-IT1可能通过调控MMP影响肺癌的侵袭和转移能力。
