Lungworms of the genus Dictyocaulus are causative agents of parasitic bronchitis in domestic and wild ungulates. This study investigates the distribution, morphology and genetic diversity of D. cervi and a new lungworm species, Dictyocaulus skrjabini n. sp. infecting red deer Cervus elaphus, fallow deer Dama dama and moose Alces alces in Poland and Sweden. The study was conducted on 167 red deer from Poland and on the DNA of lungworms derived from 7 fallow deer, 4 red deer and 2 moose collected in Sweden. The prevalence of D. cervi and D. skrjabini n. sp. in dissected red deer in Poland was 31.1% and 7.2%, respectively. Moreover, D. skrjabini n. sp. was confirmed molecularly in 7 isolates of fallow deer lungworms and 1 isolate of red deer lungworms from Sweden. Dictyocaulus skrjabini n. sp. was established based on combination of their distinct molecular and morphological features; these included the length of cephalic vesicle, buccal capsule (BC), buccal capsule wall (BCW), distance from anterior extremity to the nerve ring, the width of head, oesophagus, cephalic vesicle, BC and BCW, as well as the dimensions of reproductive organs of male and female. Additionally, molecular analyses revealed 0.9% nucleotide sequence divergence for 1,605 bp SSU rDNA, and 16.5–17.3% nucleotide sequence divergence for 642 bp mitochondrial cytB between D. skrjabini n. sp. and D. cervi, respectively, and 18.7–19% between D. skrjabini n. sp. and D. eckerti, which translates into 18.2–18.7% amino acid sequence divergence between D. skrjabini n. sp. and both lungworms.

Background
European bison,

, is a strictly protected species of large mammal, with 25% of the world’s population living in Poland. The most numerous populations of European bison live in the Białowieża Primeval Forest, northeastern Poland, and in the Bieszczady Mountains, southeastern Poland. The purpose of this study was to investigate the occurrence of
Anaplasma
spp. in
B. bonasus
from Poland.
Methods
Tissue samples were collected from 45 European bisons between 2021 and 2024 in the Białowieża and Bieszczady areas. Two genetic markers,
16S
ribosomal DNA (rDNA) and
msp4
, were used for the detection, genotyping, and phylogenetic analysis of bacteria from the genus
Anaplasma
.
Results
The prevalence of
Anaplasma
spp. was 40% (18/45) in the examined samples.
Anaplasma phagocytophilum
was detected in 10 samples, and eight samples were found to be positive for the presence of
Anaplasma marginale
DNA.
Conclusions
This study is the first report of
A. marginale
occurrence in Poland and the first report of
A. marginale
occurrence in
B. bonasus
in Europe. Infection by the pathogenic
A. marginale
in strictly protected species such as the European bison may have an impact on the health, ecology, and conservation of this endangered species.

Comorbidities and concentration of trace elements in livers of European bison from Bieszczady Mountains (Poland)

European bison is a species for which health monitoring is essential in conservation activities. So far, little research has been carried out on the concentration of elements in this species. Most previous studies did not associate the concentration of elements with susceptibility to diseases. In this study we investigate the relationship between comorbidities in European bison and concentrations of a wide spectrum of elements in the liver. Samples were collected during the monitoring of the European bison population in Bieszczady (southeast Poland) over the 2020–2022 period. Each individual was also visually inspected by a veterinarian in the field for the presence of lesions as a part of a post-mortem examination. The animals were divided into 3 groups: group A—one type of clinical sign; group B—two types of clinical signs; group C—three or more types of clinical signs. The ICP-OES method was applied to assess the concentration of 40 elements in livers. Discriminant analysis showed clear differences between the mineral status of individuals in the groups with one, two, and at least three types of clinical signs. Detailed analysis of selected elements showed that, in the case of eight elements, there was a relationship with age, sex, or comorbidities. Cu, Se, and Zn showed significant differences in relation to comorbidities, but only Cu concentration was lower when the frequency of lesions was higher. We concluded that in research on the mineral status of the population, apart from the availability of trace elements in the environment, the health condition of the studied individuals should also be considered. However, inferring the mineral status of the population on the basis of randomly obtained samples from dead individuals may give an incomplete view of the population, especially in the case of species susceptible to diseases, such as European bison.

Microbiological and molecular monitoring for bovine tuberculosis in the Polish population of European bison (Bison bonasus)

In recent years, bovine tuberculosis (BTB) has become one of the major health hazards facing the European bison (EB, Bison bonasus), a vulnerable species that requires active protection, including regular and effective health monitoring. Monitoring of zoonotic disease in wildlife is also an important part of public health protection. The aim of the study was to determine whether BTB still influences the EB population in Poland.
During 2017-2019, mandibular, retropharyngeal and mediastinal lymph nodes were collected from 90 EB during post-mortem examination, and then cultivated on Lowenstein-Jensen and Stonebrink media. Isolated strains were subjected to molecular analysis to determine the species, spoligotype and MIRU-VNTR pattern.
Lesions were found in lymph nodes originating from eight EB (8.89%). Positive microbiological cultures for mycobacteria were obtained in samples from six (6.67%) EB. The isolated strains were identified as Mycobacterium caprae (material from four EB) and atypical mycobacteria (material from two EB). For M. caprae strains spoligotype M. bovis 4_CA 1600 was identified and the MIRU-VNTR pattern was identified as 345751355413232.
It is recommended that this potentially dangerous disease should be monitored in EB via a comprehensive strategy based on a combination of microbiological and molecular methods. Such monitoring will protect the health of both animals and humans.

First report of Fasciola hepatica seroprevalence and risk factors in European bison (Bison bonasus)

); however, complex population health monitoring is particularly important in protected species such as this. Addressing the need for this surveillance, the aim of this study was to assess for the first time the seroprevalence of
in Polish free-living European bison populations and identify risk factors for infection.
Between 2020 and 2024, serum samples were collected from 119 free-ranging European bison from mountains and lowland areas. Serum samples were tested with a commercial ELISA to detect antibodies to
, and the data yielded were statistically analysed.
The study revealed
seropositivity in 20/119 animals (16.8%), with higher herd seroprevalence in lowland groups, and higher individual seroprevalence in females and animals from lowland areas.
Our study shows that a serological examination may be a useful and convenient diagnostic tool in assessing the
exposure of the European bison population, especially when performing epizootic and retrospective studies.

Background
The majority of animal tuberculosis (TB) cases reported in wildlife in Poland over the past 20 years have concerned the European bison inhabiting the Bieszczady Mountains in Southeast Poland: an area running along the border of Southeast Poland. As no TB cases have been reported in domestic animals in this region since 2005, any occurrence of TB in the free-living animals inhabiting this area might pose a real threat to local livestock and result in the loss of disease-free status. The aim of the study was to describe the occurrence of tuberculosis in the wildlife of the Bieszczady Mountains and determine the microbiological and molecular characteristics of any cultured strains. Lymph node samples were collected for analysis from 274 free-living animals, including European bison, red foxes, badgers, red deer, wild boar and roe deer between 2011 and 2017. Löwenstein–Jensen and Stonebrink media were used for culture. Molecular identification of strains was performed based on

sequence analysis, the GenoType®MTBC (Hain Lifescience, Germany
)
test, spoligotyping and MIRU-VNTR analysis.
Results
Mycobacterium caprae
was isolated from the lymph nodes of 21 out of 55 wild boar (38.2%; CI 95%: 26.5%, 51.4%) and one roe deer. Since 2014, no new TB cases have been reported in the Bieszczady European bison population.
Conclusions
The identification of TB in wild boar in the Bieszczady is an alarming phenomenon, which requires further investigation. The Bieszczady mountains are a precious, unique area, home to many protected species. However, it is also the only area in Poland where TB cases have been reported in free-living animals. The occurrence of TB in wild boar inhabiting this area might pose a real threat to local livestock and many of the protected species (for example European bison that can share feeding places with wild boar). Given this situation, ongoing monitoring of the prevalence of TB should be conducted, and protective measures should be considered.

Wild large ungulates, like European bison (Bison bonasus) and Eurasian moose (Alces alces), form an important part of the circulation of Anaplasma phagocytophilum, a Gram-negative, intracellular, tick-transmitted bacterium, in the natural environment. Bison and moose tissue samples were subjected to 16S rDNA, groEL and ankA partial gene marker amplification with specific primers using various variants of PCR. Out of 42 examined individuals, Anaplasma sp. were detected in 4/13 Eurasian moose (31%) and 7/29 European bison (24%). In addition, 12 groEL and 5 ankA partial gene positive samples were obtained from the examined animals. The phylogenetic analysis of the groEL partial gene classified samples from European bison to ecotype I, and samples from Eurasian moose to ecotype I and II; the analysis of the ankA partial gene assigned the samples to clusters I and IV. This study extends knowledge about A. phagocytophilum in wild large ungulates in Poland. This is the first report about the occurrence of Anaplasma sp. in one of the largest populations of free living European bison in the world. Our findings confirm that strains of A. phagocytophilum from Bison bonasus and Alces alces may constitute a natural reservoir of pathogenic HGA Anaplasma strains.

Ten Years of Animal Tuberculosis Monitoring in Free-Living European Bison (Bison bonasus) in Poland

In the period 1996–2012, two outbreaks of animal tuberculosis were noted in the population of free-living European bison (Bison bonasus caucasicus) in the Bieszczady Mountains, Southern Poland. As the European bison is an endangered species and particularly susceptible to tuberculosis, not to mention a national icon, the decision was made to test all deceased bison for TB in Poland. The screened bison were obtained by elimination due to poor health or natural death. A total of 159 European bison have been examined over the last 10 years. The individuals came from four regions of Poland (Białowieża Forest, Bieszczady Mountains, Borecka Forest, Knyszyńska Forest), not only from the area where tuberculosis is still endemic. Mycobacterium bovis and Mycobacterium avium spp. hominisuis were identified in two different herds. The isolation of M. bovis from European bison was the first case described in Poland. So far, the only causative agent of tuberculosis identified in European bison in Poland, both in the wild and in captive herds, was Mycobacterium caprae. The isolated M. bovis spoligotype has not previously been registered in international spoligotype databases so far. The obtained results highlight the need to monitor TB in European bison in Poland.

Mycobacterium caprae transmission to free-living grey wolves (Canis lupus) in the Bieszczady Mountains in Southern Poland

The aim of this study was to assess whether animal tuberculosis (TB) is transmitted between free-living European bison (
Bison bonasus caucasicus
), wild boars (
Sus scrofa
), and protected carnivores such as grey wolves (
Canis lupus
), brown bears (
Ursus arctos
), and Eurasian lynx (
Lynx lynx
) in the Bieszczady Mountains in Southern Poland. Results of animal studies suggest that TB transmission from bison or wild boars to grey wolves is possible. These are the first described cases where
Mycobacterium caprae
was detected in samples collected from grey wolves.

Mechanical, Thermal and Microstructural Characteristic of 3D Printed Polylactide Composites with Natural Fibers: Wood, Bamboo and Cork

In the study, polylactide-based (PLA) composites modified with natural particles (wood, bamboo, and cork) and with different levels of infilling (100%, 80%, and 60%) obtained by fused deposition modeling were tested. The effect of fiber type, infill level and crystallization rate on the mechanical properties were investigated by using tensile, flexural, and impact tests. The materials were subjected to mechanical tests carried out at 23 and 80 °C. Differential scanning calorimetry were employed to analyze crystallization behavior of composite. Furthermore, hydrothermal degradation was performed, and its effect on the properties was analyzed. The addition of natural fillers and different levels of infilling result in a similar level of reduction in the properties. However, the addition of natural fillers resulted in a slightly lower drop than the lowered infilling rate − 40% and 50% for tensile strength, respectively. Moreover, it was found that, composites made of PLA are more sensitive to high temperatures than to water. The decrease in Young's modulus of PLA at 80 °C was 90%, while after 28 days of hydrodegradation ~ 9%. The addition of fibers reduced this decrease at elevated temperatures. Importantly, in the case of a brittle material such as PLA, the impact strength has been improved by 50% for composites with cork particles and other lignocellulosic composites remained at the same level as for resin. Generally, the thermal treatment of composites increased the degree of crystallinity of the materials, as reflected in the higher results of mechanical tests.
