Response of upstream migrating juvenile European eel (Anguilla anguilla) to electric fields: Application of the marginal gains concept to fish screening

The decline in European eel (
Anguilla anguilla
) recruitment over the past half-century is partly due to river infrastructure that delays or blocks upstream migration to rearing habitat. Stimuli, such as electricity, can be used to modify the behaviour of downstream moving fish and guide them to preferred routes of passage at river infrastructure; but research on upstream migrating juvenile eel remains limited. The response of upstream migrating juvenile eel exposed to pulsed direct current (PDC) electric fields was investigated using a recirculatory flume. Eel were presented a choice of two routes upstream under either: (1) a treatment condition, in which the selection of one route resulted in exposure to High Electric Field (HEF) strength that was between 1.5–2 times stronger than the Low Electric Field (LEF) strength encountered in the alternative route; or (2) a control in which the electric field was absent in both routes. Under the treatment, five different mean HEF strengths (0.53, 0.77, 1.22, 2.17 and 3.74 Vcm
-1
) were tested at one of two frequencies (2 and 10 Hz). Route choice, distance downstream of the first set of electrodes at which an initial response was observed and avoidance behaviours (
acceleration
,
retraction
,
switching
and
rejection
) were compared among treatments. For the 1.22, 2.17 and 3.74 Vcm
-1
and under 2 Hz, eel preferred to pass the LEF route. Avoidance was greater in the HEF route and positively related to field strength. The distance of the initial response did not differ between routes, field strengths or frequency. Upstream migrating eel avoided electric fields indicating potential to develop this approach for fish guidance. Further work is needed to test prototypes in field settings, particularly in combination with traditional physical screens to water intakes as part of a process of applying the concept of marginal gains to advance environmental impact mitigation technology.

The influence of flow velocity on the response of rheophilic fish to visual cues

The strong association with visual cues exhibited by fish that prefer to inhabit flowing water (rheophilic species) may help reduce the energetic costs of maintaining position due to the provision of spatial points of reference. If this “Station Holding Hypothesis” is true, a positive relationship between the association with visual cues and flow velocity is expected. This hypothesis was tested experimentally by quantifying the response of common minnow (
Phoxinus phoxinus
) and brown trout (
Salmo trutta
) to visual cues under three flow velocities. In contradiction to the prediction, there was no evidence that the association with strong visual cues was positively related to flow velocity when fish were presented with vertical black stripes in an open channel flume, although interspecific variation in response was observed. The association with visual cues was relatively weak in trout, compared to minnow that spent 660% more time associated with the zone in which visual cues were present during the treatment, than the control when visual cues were absent. Trout tended to be more exploratory and made short visits to the area where visual cues were present, whereas minnow associated with the cues for longer. The strong association with visual cues independent of flow velocity exhibited by minnow and the weak association across all velocities by trout suggest that this behaviour is unlikely to reflect a strategy to minimise the energetic cost of maintaining position in flowing water. Minnow may have used the visual cues as a proxy indicator of physical structure that provides alternative benefits, such as refuge from predators. Trout may have employed alternative cues (e.g. mechanosensory) to seek more energetically favourable regions of the experimental area, reducing the importance of stationary visual stimuli.

Magmatic and Crustal Differentiation History of Granitic Rocks from Hf-O Isotopes in Zircon

Granitic plutonism is the principal agent of crustal differentiation, but linking granite emplacement to crust formation requires knowledge of the magmatic evolution, which is notoriously difficult to reconstruct from bulk rock compositions. We unlocked the plutonic archive through hafnium (Hf) and oxygen (O) isotope analysis of zoned zircon crystals from the classic hornblende-bearing (I-type) granites of eastern Australia. This granite type forms by the reworking of sedimentary materials by mantle-like magmas instead of by remelting ancient metamorphosed igneous rocks as widely believed. I-type magmatism thus drives the coupled growth and differentiation of continental crust.

Retinoblastoma (Rb) protein is a tumor suppressor that is dysregulated in a majority of human cancers. Rb functions to inhibit cell cycle progression in part by directly disabling the E2F family of cell cycle-promoting transcription factors. Because the
de novo
synthesis of multiple glutamine-derived anabolic precursors is required for cell cycle progression, we hypothesized that Rb also may directly regulate proteins involved in glutamine metabolism. We examined glutamine metabolism in mouse embryonic fibroblasts (MEFs) isolated from mice that have triple knock-outs (TKO) of all three Rb family members (Rb-1, Rbl1 and Rbl2) and found that loss of global Rb function caused a marked increase in

C-glutamine uptake and incorporation into glutamate and tricarboxylic acid cycle (TCA) intermediates in part via upregulated expression of the glutamine transporter ASCT2 and the activity of glutaminase 1 (GLS1). The Rb-controlled transcription factor E2F-3 altered glutamine uptake by direct regulation of ASCT2 mRNA and protein expression, and E2F-3 was observed to associate with the ASCT2 promoter. We next examined the functional consequences of the observed increase in glutamine uptake and utilization and found that glutamine exposure potently increased oxygen consumption, whereas glutamine deprivation selectively decreased ATP concentration in the Rb TKO MEFs but not the wild-type (WT) MEFs. In addition, TKO MEFs exhibited elevated production of glutathione from exogenous glutamine and had increased expression of gamma-glutamylcysteine ligase relative to WT MEFs. Importantly, this metabolic shift towards glutamine utilization was required for the proliferation of Rb TKO MEFs but not for the proliferation of the WT MEFs. Last, addition of the TCA cycle intermediate α-ketoglutarate to the Rb TKO MEFs reversed the inhibitory effects of glutamine deprivation on ATP, GSH levels and viability. Taken together, these studies demonstrate that the Rb/E2F cascade directly regulates a major energetic and anabolic pathway that is required for neoplastic growth.

The impact of reintroduced Eurasian beaver (Castor fiber) dams on the upstream movement of brown trout (Salmo trutta) in upland areas of Great Britain

The return of Eurasian beaver (Castor fiber) to large areas of Europe represents a conservation success with the current population estimated to be around 1.2 million individuals. Their reintroduction to many areas, including Great Britain, has in some cases been controversial. Despite numerous documented benefits to biodiversity, concerns relate to localised flooding, adverse impacts on land use and engineered structures (e.g. culvert blockage), disease transfer, and the influence of beaver habitat modifications on fisheries, particularly in relation to salmonids. This study investigated the impacts of a series of four beaver dams on the upstream movement of brown trout during the spawning period (October-December) at a field site in Scotland. The study site comprised two streams entering a common loch, one modified by a series of four beaver dams, the other remaining unaltered during the Study Period. Trout were captured using electric fishing, fyke nets and rod and line and were tagged with Passive Integrated Transponders (PIT) before release. PIT telemetry antennas were installed below and above each dam to establish successful passage of trout during the monitoring period that included trout spawning movements in 2015 (high flows) and 2016 (low flow). There was a distinct difference in passage success between years, with high flows (using prior rainfall as a proxy measure) and larger fish size being important positive predictors of upstream passage success. A combination of environmental (prior rainfall and water temperature) and biotic (fish size) factors influenced passage success with high flows being a significant covariate at all four dams in two models used to define trout passage dynamics (Weibull and exponential base models), providing the best explanatory variable for fish passage at two of the four dams. Survival analysis and associated modelling indicated that migratory delay was inversely related to previous passage success, whilst motivation was also a determinant of success, with greatest passage in highly motivated trout. Our findings indicate that given the right environmental and biotic factors, brown trout are adept at passing beaver dams, although under certain conditions, beaver dams can impede the movement of brown trout and the magnitude of impact is influenced by these factors. In particular, the barrier effects of beaver dams are exacerbated under low flow conditions, and this may become a greater challenge in the future due to shifting climatic conditions if periods of warmer and drier weather persist and coincide with peak migratory movements of fish.

Chalk stream restoration: Physical and ecological responses to gravel augmentation

To mitigate the morphological and ecological impacts of direct (e.g. dredging) and indirect (e.g. damaged river function) sediment loss, gravel augmentation is commonly practiced in river systems globally. Despite this, the effectiveness of this practice remains poorly understood, especially in less often considered systems such as chalk streams which present uncommon conditions (e.g. low stream power, stable flow) and may respond to interventions in ways that differ from systems more commonly studied. This study quantified immediate (0–1 years) and short-term (1–2 years) physical and ecological responses to gravel augmentation at two English chalk stream restoration sites: Home Stream (HS; River Test) and East Lodge (EL; River Itchen). We quantified habitat (depth, velocity, substrate composition), cover of different macrophytes, and macroinvertebrate (before-after-control-impact) abundance and community structure. Restoration reduced depth and increased gravel cover in both sites and decreased the cover of filamentous green algae in HS. Macroinvertebrate communities became more dominated by silt-intolerant taxa, while abundance [HS only] and taxon richness increased 1–2 years post-restoration. Whilst the responses found were generally positive in light of the restoration goals, the effects varied across sites, post-restoration time periods and ecological groups, emphasising the need for the more holistic monitoring of restoration projects considering community-level responses at different sites and systems over ecologically relevant timescales. This will help inform on the generality and longevity of responses and provide the evidence needed to develop sound restoration practice.

The Maillard reaction (MR) between sugars and amino acids, peptides, or proteins is understood to occur gradually during the production and aging of sparkling wines, where it contributes to caramel, roasted, and toasted aromas. Divalent metal ions can accelerate the MR, although this has not been previously reported in wine or wine-like conditions. In this work, the effect of calcium (Ca) and magnesium (Mg) ions on the concentration of 10 Maillard reaction-associated products (MRPs) was measured in modified sparkling base wine during accelerated aging at 50 °C for four weeks. Chardonnay base wine was modified by the addition of fructose (0.02 M) and a single amino acid (lysine, glycine, cysteine; 0.01 M) in combination with Ca2+ or Mg2+ at zero, low (10 mg/L), or high (50 mg/L) dose levels. MRPs were quantified by headspace solid-phase microextraction coupled with gas chromatography–mass spectrometry (HS-SPME-GC/MS), sugar concentration was measured by enzymatic assay, and amino acids and free metal ions were monitored by capillary electrophoresis. Fructose levels did not substantially decrease during aging despite increases in all MRPs, suggesting that trace sugars or α-dicarbonyl species present in the wine matrix likely play a greater role in MRP formation than fructose. Aging duration and amino acid content had a greater effect than metal addition on the composition of the MRPs. Treatments containing cysteine and 50 mg/L Ca2+ had elevated concentrations of benzaldehyde and furfural ethyl ether following 4 weeks of accelerated aging. This work identified key MRPs that increase during base wine accelerated aging and informs future research on the relationship between wine composition and aging markers.

The accuracy and reproducibility of video assessment in the pitch-side management of concussion in elite rugby

To investigate the accuracy and reliability of side-line video review of head impact events to aid identification of concussion in elite sport.
Diagnostic accuracy and inter-rater agreement study.
Immediate care, match day and team doctors involved in the 2015 Rugby Union World Cup viewed 20 video clips showing broadcaster’s footage of head impact events occurring during elite Rugby matches. Subjects subsequently recorded whether any criteria warranting permanent removal from play or medical room head injury assessment were present. The accuracy of these ratings were compared to consensus expert opinion by calculating mean sensitivity and specificity across raters. The reproducibility of doctor’s decisions was additionally assessed using raw agreement and Gwets AC1 chance corrected agreement coefficient.
Forty rugby medicine doctors were included in the study. Compared to the expert reference standard overall sensitivity and specificity of doctors decisions were 77.5% (95% CI 73.1–81.5%) and 53.3% (95% CI 48.2–58.2%) respectively. Overall there was raw agreement of 67.8% (95% CI 57.9–77.7%) between doctors across all video clips. Chance corrected Gwets AC1 agreement coefficient was 0.39 (95% CI 0.17–0.62), indicating fair agreement.
Rugby World Cup doctors’ demonstrated moderate accuracy and fair reproducibility in head injury event decision making when assessing video clips of head impact events. The use of real-time video may improve the identification, decision making and management of concussion in elite sports.
